Abstract

The overexpression of murine double minute 2 (MDM2) is found in several human tumors, and increased expression of MDM2 inactivates the apoptotic and cell cycle arrest function of p53. Interleukin-16 (IL-16) is a pleiotrophic cytokine and the properties of IL-16 suggest that it involve in the pathophysiological process of chronic inflammatory diseases. In this study, we investigated the expression of MDM2 in intestinal metaplasia and gastric cancer as well as the effect of H. pylori infection and IL-16 on epithelial cell proliferation and MDM2 expression in gastric cells in vitro. The expression of MDM2 on gastric biopsies was studied immunohistochemistry. AGS cells were incubated with a combination of IL-16 and Helicobacter pylori (H. pylori). Gastric epithelial cell proliferation was studied by BrdU uptake and the expressions of MDM2 were studied by ELISA. There was no significant difference on the expression of MDM2 between with and without H. pylori infected chronic gastritis. In H. pylori infected gastric mucosa; the MDM2 expression was higher on intestinal metaplasia and gastric cancer than chronic gastritis. IL-16 administration was increased MDM2 expression and cell proliferation on AGS cells, which was decreased by H. pylori infection. In conclusion, the expression of MDM2 in long-term H. pylori infected gastric mucosa may indicate a risk for carcinogenesis. IL-16 secretion in H. pylori infected mucosa is one of the factors for gastric cancer. The expression of MDM2 on mucosa can be a mediator for gastric cancer.

Abstract

Inflammatory bowel disease (IBD) is a chronic disease of the digestive tract, with complicated and multifactoral etiology. An exaggerated intestinal immune response to otherwise innocuous stimuli plays a key role in the pathophysiology of this intestinal disorder. Nutritional intervention is an important therapy for patients with IBD. But the ratio of n-3 and n-6 fatty acids on the modulation of intestinal inflammation is not clear. Interleukin-16 (IL-16) is a pleiotrophic cytokine secreted mainly by CD8+ T cells. The properties of IL-16 suggest that it may be involved in pathophysiological process of chronic inflammatory diseases. The aim was to determine the effect of an n-3 fatty acid-rich diet (RACOL®) on the expressions of cytokines and mucosal integrity in trinitrobenzene surufonic acid (TNBS) induced rat colitis. 7 week-male Wistar rats were divided into 4 groups. 1. Normal laboratory diet (n = 10), 2. RACOL® diet only (n = 10), 3. TNBS induced colitis with normal laboratory diet (n = 10), 4. TNBS induced colitis with RACOL® diet (n = 10). TNBS induced colitis rats were given an intracolonic injection of 50 mg of TNBS dissolved in 50% ethanol. From day 2, rats were fed 80 kcal/day of RACOL® and/or normal laboratory diet, and sacrified on the 14th and the 28th days. One hour before sacrifice, 0.2 mg/g/rat of BrdU was injected. The expressions of BrdU labeled cells, tumor necrosis factor (TNF)-α, interferon (IFN)-γ and IL-16 were studied by the ABC staining method and DAB staining lesions in colonic mucosa were estimated by an image analyzer system. Apoptosis was assessed by the TUNEL method. Macroscopic and histological findings were classified by the Morris and Vilaseca method. TNBS application increased colonic epithelial cell proliferation, but nutrition by RACOL® reduced this stimulation. Number of apoptosis cells was also increased by TNBS application, but nutrition of RACOL® reduced the number of apoptosis cells, which had been increased by TNBS treatment. The expressions of cytokines had been increased in TNBS-colitis rats, but were reduced by the RACOL®. In conclusion, the effect of nutrition by n-3 fatty acid-rich diet, RACOL®, is an important treatment through the inhibition of inflammatory mediators, and colonic hyper-proliferation.

Abstract

This paper discusses the preventive controls of the generator outputs to improve security in the decentralized power system. Preventive controls of the power system can improve security level. However, the preventive control requires too much computation time because of too many considerable faults in the power system. Multiarea decentralization can be considered to reduce computation time of the power flow calculation. It is assumed that the effects of disturbances in the external systems are relatively smaller than the effects of disturbances in the study area. Linearized sensitivity analysis is used to evaluate the effects of the external systems to reduce computation time. Linear programming is used to determine the control signals of generators considering the incremental cost of generators. The New England 38 buses system is used as an example system to check the validity of the algorithm in this paper. The calculations of the example system show satisfactory results.

Abstract

A 38-year-old woman was referred to our institution due to epigastralgia. She presented with obstructive jaundice and eosinophilia. Endoscopic retrograde cholangiopancreatography showed diffuse narrowing from the distal common bile duct to the bifurcation of the hepatic ducts. An endoscopic plastic biliary stent was inserted; the specimen obtained from the common bile duct wall revealed dense infiltration by eosinophils. Treatment was started with prednisolone 60 mg daily. The patient's biliary stenosis and eosinophilia gradually improved. Eosinophilic infiltration in the lungs or stomach is relatively common, but it is rare in the common bile duct. Most of the reported cases of eosinophilic cholangitis presented with eosinophilia; our patient's eosinophil count was over 1000/mm(3). Since our patient had allergies to pollen and house dust, a relationship between the allergies and the eosinophilic cholangitis was suspected, but no cause was identified.
