Abstract We report the fabrication of heterostructure white light–emitting diode (LED) comprised of n-ZnO nanotubes (NTs) aqueous chemically synthesized on p-GaN substrate. Room temperature electroluminescence (EL) of the LED demonstrates strong broadband white emission spectrum consisting of predominating peak centred at 560 nm and relatively weak violet–blue emission peak at 450 nm under forward bias. The broadband EL emission covering the whole visible spectrum has been attributed to the large surface area and high surface states of ZnO NTs produced during the etching process. In addition, comparison of the EL emission colour quality shows that ZnO nanotubes have much better quality than that of the ZnO nanorods. The colour-rendering index of the white light obtained from the nanotubes was 87, while the nanorods-based LED emit yellowish colour.

Abstract ZnO nanorods (NRs) with high surface area to volume ratio and biocompatibility is used as an efficient photosensitizer carrier system and at the same time providing intrinsic white light needed to achieve cancer cell necrosis. In this letter, ZnO nanorods used for the treatment of breast cancer cell (T47D) are presented. To adjust the sample for intracellular experiments, we have grown the ZnO nanorods on the tip of borosilicate glass capillaries (0.5 μm diameter) by aqueous chemical growth technique. The grown ZnO nanorods were conjugated using protoporphyrin dimethyl ester (PPDME), which absorbs the light emitted by the ZnO nanorods. Mechanism of cytotoxicity appears to involve the generation of singlet oxygen inside the cell. The novel findings of cell-localized toxicity indicate a potential application of PPDME-conjugated ZnO NRs in the necrosis of breast cancer cell within few minutes.

Abstract Long-read sequencing technology has enabled variant detection in difficult-to-map regions of the genome and enabled rapid genetic diagnosis in clinical settings. Rapidly evolving third-generation sequencing platforms like Pacific Biosciences (PacBio) and Oxford Nanopore Technologies (ONT) are introducing newer platforms and data types. It has been demonstrated that variant calling methods based on deep neural networks can use local haplotyping information with long-reads to improve the genotyping accuracy. However, using local haplotype information creates an overhead as variant calling needs to be performed multiple times which ultimately makes it difficult to extend to new data types and platforms as they get introduced. In this work, we have developed a local haplotype approximate method that enables state-of-the-art variant calling performance with multiple sequencing platforms including PacBio Revio system, ONT R10.4 simplex and duplex data. This addition of local haplotype approximation simplifies long-read variant calling with DeepVariant.

This current study investigates the biological efficiency of essential oils extracted from Bistorta affinis and Malcolmia cabulica on human and foodborne pathogens as well as against insects and termites. The oils were obtained by steam distillation using a Clevenger-type system and analyzed for their constituents using GC-MS. Twenty compounds were identified, wherein carvacrol and thymol were the main constituents in both plants. Essential oils exhibited significant activity against all tested foodborne bacteria, fungi, and other pathogens. In addition, essential oils from both plants have shown promising activity against all tested insects, which is a positive sign of being used as an insect repellent. In contrast, no activity was observed against termites. The bioactivities are mainly due to carvacrol and thymol’s levels in the essential oils that known for their effectiveness against foodborne pathogens and pests. The present study constitutes a basis for further investigation and development of bioactive compounds in Bistorta affinis and Malcolmia cabulica.

The low water solubility of an active pharmaceutical ingredient (aripiprazole) is one of the most critical challenges in pharmaceutical research and development. This antipsychotic drug has an inadequate therapeutic impact because of its minimal and idiosyncratic oral bioavailability to treat schizophrenia. The main objective of this study was to improve the solubility and stability of the antipsychotic drug aripiprazole (ARP) via forming binary as well as ternary inclusion complexes with hydroxypropyl-β-cyclodextrin (HPβCD) and L-Arginine (LA) as solubility enhancers. Physical mixing and lyophilization were used in different molar ratios. The developed formulations were analyzed by saturation solubility analysis, and dissolution studies were performed using the pedal method. The formulations were characterized by FTIR, XRD, DSC, SEM, and TGA. The results showcased that the addition of HPβCD and LA inclusion complexes enhanced the stability, in contrast to the binary formulations and ternary formulations prepared by physical mixing and solvent evaporation. Ternary formulation HLY47 improved dissolution rates by six times in simulated gastric fluid (SGF). However, the effect of LA on the solubility enhancement was concentration-dependent and showed optimal enhancement at the ratio of 1:1:0.27. FTIR spectra showed the bond shifting, which confirmed the formation of new complexes. The surface morphology of complexes in SEM studies showed the rough surface of lyophilization and solvent evaporation products, while physical mixing revealed a comparatively crystalline surface. The exothermic peaks in DSC diffractograms showed diminished peaks previously observed in the diffractogram of pure drug and LA. Lyophilized ternary complexes displayed significantly enhanced thermal stability, as observed from the thermograms of TGA. In conclusion, it was observed that the preparation method and a specific drug-to-polymer and amino acid ratio are critical for achieving high drug solubility and stability. These complexes seem to be promising candidates for novel drug delivery systems development.

A general investigation of sub-tropical forests, from Pir Chinasi National Park, Tolipir National Park, Dhirkot Nature Reserve and Banjosa Game Reserve was carried out during different months from February 2008 to May 2010. The relative abundance of species was calculated using line transects of 50m. A total of five different species (Abies pindrow. Cedrus deodara, Pinus wallichiana, Pinus roxburgii and Picea smithiana) from the Pinaceae family were recorded. The main reported use of Cedrus deodara and Pinus wallichiana by the local people was for furniture and construction purposes. Pinus wallichiana was observed as the dominant species from all the selected sites.

Fog Computing has emerged as an extension to cloud computing by providing an efficient infrastructure to support IoT. Fog computing acting as a mediator provides local processing of the end-users' requests and reduced delays in communication between the end-users and the cloud via fog devices. Therefore, the authenticity of incoming network traffic on the fog devices is of immense importance. These devices are vulnerable to malicious attacks. All kinds of information, especially financial and health information travel through these devices. Attackers target these devices by sending malicious data packets. It is imperative to detect these intrusions to provide secure and reliable service to the user. So, an effective Intrusion Detection System (IDS) is essential for the secure functioning of fog without compromising efficiency. In this paper, we propose a method (Auto-IF) for intrusion detection based on deep learning approach using Autoencoder (AE) and Isolation Forest (IF) for the fog environment. This approach targets only binary classification of the incoming packets as fog devices are more concerned about differentiating attack from normal packets in real-time. We validate the proposed method on the benchmark NSL-KDD dataset. Our technique of intrusion detection achieves a high accuracy rate of 95.4% as compared to many other state-of-art intrusion detection methods.

Vaccination protects people from serious illness and associated complications. Conspiracy theories and misinformation on vaccines have been rampant during the COVID-19 pandemic and are considered significant drivers of vaccine hesitancy. Since vaccine hesitancy can undermine efforts to immunize the population against COVID-19 and interferes with the vaccination rate, this study aimed to ascertain the COVID-19-vaccine-related conspiracy beliefs, vaccine hesitancy, views regarding vaccine mandates, and willingness to pay for vaccines among the general population. A web-based, cross-sectional survey was conducted (April–August 2021) among the adult population in six countries (Pakistan, Saudi Arabia, India, Malaysia, Sudan, and Egypt). Participants were recruited using an exponential, non-discriminate snowball sampling method. A validated self-completed electronic questionnaire was used for the data collection. All the participants responded to questions on various domains of the study instrument, including conspiracy beliefs, vaccine hesitancy, and willingness to pay. The responses were scored according to predefined criteria and stratified into various groups. All data were entered and analyzed using SPSS version 22. A total of 2481 responses were included in the study (Pakistan 24.1%, Saudi Arabia 19.5%, India 11.6%, Malaysia 8.1%, Sudan 19.3%, and Egypt 17.3%). There was a preponderance of participants ≤40 years old (18–25 years: 55.8%, 26–40 years: 28.5%) and females (57.1%). The average score of the COVID-19 vaccine conspiracy belief scale (C19V-CBS) was 2.30 ± 2.12 (median 2; range 0–7). Our analysis showed that 30% of the respondents were found to achieve the ideal score of zero, indicating no conspiracy belief. The mean score of the COVID-19 vaccine hesitancy scale (C19V-HS) was 25.93 ± 8.11 (range: 10–50). The majority (45.7%) had C19V-HA scores of 21–30 and nearly 28% achieved a score greater than 30, indicating a higher degree of hesitancy. There was a significant positive correlation between conspiracy beliefs and vaccine hesitancy (Spearman’s rho = 0.547, p < 0.001). Half of the study population were against the vaccine mandate. Respondents in favor of governmental enforcement of COVID-19 vaccines had significantly (p < 0.001) lower scores on the C19V-CBS and C19V-HS scale. Nearly 52% reported that they would only take vaccine if it were free, and only 24% were willing to pay for COVID-19 vaccines. A high prevalence of conspiracy beliefs and vaccine hesitancy was observed in the targeted countries. Our findings highlight the dire need for aggressive measures to counter the conspiracy beliefs and factors underlying this vaccine hesitancy.

Iron-oxide nanoparticles are synthesized by the Green Chemistry method and using Gooseberry leaves extract as reducing agent. The characterization of the nanoparticles was carried by the common tools which include UV–Vis, SEM, EDX, XRD, FTIR and CV. UV–Vis confirms the formation of iron oxide nanoparticles. SEM verifies the spherical morphology of the nanoparticles. Elemental composition was determined by the EDX. XRD peaks details the spinal state crystal structure of the as-synthesized iron-oxide nanoparticles. CV results confirm the electrochemical nature of the iron-oxide nanoparticles useful for biosensing. Finally Bioassay tests reveal that these nanoparticles can be potent candidate to be used as biosensors. The Rosaceae family is also considered as the rose family. There are 27 genera and nearly 160 species from this family in Pakistan. This study was carried out to determine the diversity and population size of species belonging to the Rosaceae family in selected national parks from Azad Jammu and Kashmir, Pakistan. Unbiased stage-sampling was carried out. Four species belonging to the Rosaceae family were observed in Banjosa Game Reserve. Rosa brunonii and Duchesnea indica present, were not enough to calculate relative abundance. Only two species from this family were recorded from Dhirkot National Park, Rubus fruticosus and Fragaria nubicola. Pir Chanasi National Park and Tolipir National Park had the most diversity out of all the National Parks surveyed.

A quick visit of the protected forests of Banjosa, Dhirkot, Pir Chanasi, Pir Lassura and Tolipir was carried out. Berberidaceae belong to a family also known as the Barberry family (Elpel, n.d.). In Pakistan, there are 3 genera and 22 species belonging to this group (Jafri, n.d.). Species of this family are extensively cultivated due to their aesthetic and medicinal value. In the past, researchers would consider this family to get an idea about the habitat degradation of temperate areas. This study was aimed to produce a comprehensive list of species richness of this family in five National Parks of Pakistan. Overall three species from this family were observed in the study sites. The greatest diversity was seen in Banjosa Game Reserve. DNR, PLNP and TNP each reported only the presence of Berberis lycium. However, the constancy percentage of this species represented a good density. Using this data, further research may be conducted to ascertain the level of protection the National Parks have been able to afford this family and their surrounding biodiversity as an extension

Five protected areas from temperate highlands of Pakistan were gauged for biodiversity of the Moraceae family. The National Park in Pir Lasura was studied from June to July 2009, in Banjosa from May to June 2009, in Pir Chanasi from April to May 2010, in Dhirkot (February 2008) and in Tolipir from April to May 2008. From the five protected areas surveyed, only three had species belonging to the family Moraceae. Three species were observed from Banjosa Game Reserve, two from Tolipir Nature Reserve and six from Pir Lasura National Park. The species commonly found were Ficus carica, Ficus Palmate, Ficus religiosa, Ficus bengalensis, Ficus spp., Ficus virens and Morus spp.

A detailed survey was conducted in five national parks and game reserves of Azad Jammu and Kashmir, Pakistan to assess the biodiversity of family Fagaceae. Identification of the species was done using literary sources and comparison of samples with herbaria from Quaid-i-Azam University, Islamabad and Pakistan Museum of Natural History, Islamabad, Pakistan. A total of eight different species belonging to two genera were identified, including Quercus spp., Castanea sativa, Quercus glauca, Quercus baloot, Quercus incana, Quercus velutina, Quercus alba and Quercus dilatata. The greatest biodiversity was found in Banjosa Game Reserve (BGR) and Tolipir National Park (TNP), while Pir Chinasi National Park (PCNP), Pir Lasura National Park (PLNP) and Dhirkot National Reserve (DNR) had the least number of species.

Survey carried out in June-July 2009 recorded a minimum of 159 plant species with 48 species of trees. Out of these only two species are of gymnosperms; Pinus roxburghii and Pinus wallichiana. One species of trees, Pinus roxburghii is widely distributed. It is present in all communities, and can be found in different combinations with different other species. Both of these species have ethnobotanical value and are used by local population for different purposes.

A total of 65 maize grain samples (Zea Mays L.) collected from cold and hot climatic zones of Pakistan lead to isolation of 10 Fusarium supposing deep freezing method. Occurrence of fusarium spp. was high (75%) in cold climatic zones as compared to hot zones (25%). Fumonisins were recorded in both climatic zones, though their incidence was higher in hot climatic zone (range 4,379-9,990 μg/kg). Incidence of A and B trichothecenes was higher in cool climatic zone. Zearalenone (ZON) with mean level 500 μg/kg was positive in only 10% samples of hot climatic zone. Furthermore, co-contamination of fusariotoxins was also observed for fumonisins with ZON, A and B trichothecenes. Among 33 recovered isolates, F. proliferatum, F. tricinctum, F. subglutinans, F. poae, F. nivale and F. acuminatum were found toxigenic.

The saprophytic hyphomycetous fungi: Beniowskia sphaeroidea, Graphium putredinis, Phaeoisaria clematidis, and Tilachlidium ramosum were recorded in Rawalpindi from 2008 to 2010. These were studied in the laboratories of PMAS-Arid Agriculture University campus Rawalpindi. The first records from Pakistan were marked with an asterisk.

Objective: The present study was used to design simple, accurate and sensitive reversed phase-high-performance liquid chromatography RP-HPLC and high-performance thin-layer chromatography (HPTLC) methods for the development of quantification of khellin present in the seeds of Ammi visnaga. Materials and Methods: RP-HPLC analysis was performed on a C18 column with methanol: Water (75: 25, v/v) as a mobile phase. The HPTLC method involved densitometric evaluation of khellin after resolving it on silica gel plate using ethyl acetate: Toluene: Formic acid (5.5:4.0:0.5, v/v/v) as a mobile phase. Results: The developed HPLC and HPTLC methods were validated for precision (interday, intraday and intersystem), robustness and accuracy, limit of detection and limit of quantification. The relationship between the concentration of standard solutions and the peak response was linear in both HPLC and HPTLC methods with the concentration range of 10–80 μg/mL in HPLC and 25–1,000 ng/spot in HPTLC for khellin. The % relative standard deviation values for method precision was found to be 0.63–1.97%, 0.62–2.05% in HPLC and HPTLC for khellin respectively. Accuracy of the method was checked by recovery studies conducted at three different concentration levels and the average percentage recovery was found to be 100.53% in HPLC and 100.08% in HPTLC for khellin. Conclusions: The developed HPLC and HPTLC methods for the quantification of khellin were found simple, precise, specific, sensitive and accurate which can be used for routine analysis and quality control of A. visnaga and several formulations containing it as an ingredient.

A simple, sensitive, specific, rapid and accurate high performance thin layer chromatographic (HPTLC) method has been developed for the quantification of podophyllotoxin and etoposide. Podophyllotoxin was quantified in the roots of Podophyllum hexandrum whereas etoposide in a marketed formulation. The method involved densitometric evaluation of both podophyllotoxin and etoposide after resolving it on silica gel plate using dichloromethane–methanol–formic acid (9.5:0.5:0.5 v/v/v) as the mobile phase. The method was validated for precision (inter-day, intra-day and inter-system), robustness, accuracy, limit of detection and limit of quantification. The relationship between the concentration of standard solutions and the peak response (area) was linear within the concentration range of 150–2400 ng spot−1 for podophyllotoxin and 200–2000 ng spot−1 for etoposide. Instrumental precision was found to be 1.03–1.80 (% RSD) and 0.79–1.99 (% RSD) for podophyllotoxin and etoposide, respectively. Accuracy of the method was checked by recovery studies conducted at three different concentration levels and the average percentage recovery was found to be 100.71% for podophyllotoxin and 100.43% for etoposide, respectively. The HPTLC method for the quantification of podophyllotoxin and etoposide was found to be simple, precise, specific, sensitive and accurate which can be used for routine analysis and quality control of P. hexandrum and several formulations containing these markers.

Seven hymenomycetous fungi (mushrooms) were collected from the twin cities area during July 2008. These were identified as the members of the order Agaricales, class Basidiomycetes. They belong to five genera: *Clitocybe fragrans, *Collybia cookie, *Coprinus radians, *Coprinus sterculinus, *Volvariella bingensis, Volvariella parvula, and Termitomyces microcarpus. Five of them, marked with asterisk were reported for the first time from Pakistan.

Among the edible mushrooms, Morchella esculenta (Pers.) is a highly prized mushroom which consists of a short stalk and a conical, pitted, hollow pileus. It is commonly known as a morel or sponge mushroom, or gucchi, etc. It belongs to the class Ascomycetes, order Pezizales and family Morchellaceae. It is found throughout the world. In Pakistan, it is found in northern areas from Murree, Patriata, and Gilgit under pine vegetation. It is therapeutic in nature; therefore its medicinal and biochemical properties are under investigation throughout the world, especially in regard to treating the hazardous diseases such as cancer, tumors, etc. The present in vitro study was conducted for the first time in Pakistan. The three media soil and soil extracts Agar and Luria Agar were used. Mycellial growth was obtained on all these media, but the LA medium showed better growth. Microscopic studies had shown pigmentation is most likely due to the secondary mycelium. Among these mycelium ascogenous hyphae are produced that may lead to the formation of ascogonium and antheridium with the dilated tips. The compact mass of prosenchymatous and pseudoparenchyma tissues was observed.

Objective: A simple, sensitive, and specific thin layer chromatography (TLC) densitometry method has been developed for the simultaneous quantification of strychnine and brucine in the seeds of Strychnos nux-vomica. Materials and Methods: The method involved simultaneous estimation of strychnine and brucine after resolving it by high performance TLC (HPTLC) on silica gel plate with chloroform-methanol-formic acid (8.5:1.5:0.4 v/v/v) as the mobile phase. Results: The method was validated as per the ICH guidelines for precision (interday, intraday, intersystem), robustness, accuracy, limit of detection, and limit of quantitation. The relationship between the concentration of standard solutions and the peak response was linear within the concentration range of 50-1000 ng/spot for strychnine and 100-1000 ng/spot for brucine. The method precision was found to be 0.58-2.47 (% relative standard deviation [RSD]) and 0.36-2.22 (% RSD) for strychnine and brucine, respectively. Accuracy of the method was checked by recovery studies conducted at three different concentration levels and the average percentage recovery was found to be 100.75% for strychnine and 100.52% for brucine, respectively. Conclusions: The HPTLC method for the simultaneous quantification of strychnine and brucine was found to be simple, precise, specific, sensitive, and accurate and can be used for routine analysis and quality control of raw material of S. nux-vomica and several unani and ayurvedic formulations containing this as an ingredient.

Introduction Home parenteral nutrition (HPN) is essential for patients with intestinal failure requiring long-term nutritional support. The Amerita Quality Improvement Project for HPN Patients (QIP-PN) explored the effect of a physician nutrition expert (PNE)-led multidisciplinary nutritional support team (MNST) on HPN care for patients under its service.Objective To determine an MNST effect on adherence to protocols, outcomes and quality of life (QOL) in HPN.Methods The study was divided into three phases: data review (phases 1a and 1b), observation (phase 2) and intervention (phase 3). Seven Amerita locations were selected as ‘study branches’ (population), from which all study patients and controls were drawn. The quality improvement project employed a quasi-experimental case-matched control group (control) design. Data were collected on demographics, treating physicians PNE status, HPN care variables, recommended interventions, quality-of-life assessment, adverse outcomes and hospitalisations. Paired t-test compared continuous data between phases 2 and 3. Comparisons between study and control groups used a negative binomial regression model.Results Thirty-four patients were reviewed in phase 1a and 197 in phase 1b. Forty study patients completed phase 2 and progressed into phase 3, of whom 30 completed ≥60 therapy days. Patients were lost to follow-up if they discontinued HPN for any reason. Improvements in weight, body mass index and QOL were seen in the study patients during intervention. Recommendations made and accepted by treating physicians differed based on PNE status. Study patients had fewer adverse outcomes and related hospitalisations than controls.Conclusion MNST recommendations improved clinical, biochemical parameters and patients’ self-reported overall health. MNST input reduced adverse outcomes, hospitalisation and the length of stay at the hospital. This study highlights the potential for MNST to have a significant impact on the quality and overall cost of HPN management.

Background: The lack of reliable tobacco healthcare and economic cost estimates leaves the tobacco industry undertaxed and thriving in Pakistan and makes the country as one of the top tobacco-consuming nations. To facilitate effective tobacco tax policymaking, this study estimates the economic cost of smoking-attributable diseases and deaths in Pakistan., Methods: A nationally representative sample survey of 13 000 households was administered to gather the data required to estimate different cost components of smoking-attributable diseases through the prevalence-based approach., Findings: The total smoking-attributable economic cost of all diseases and deaths in Pakistan in the year 2018-2019 for persons aged 35 years or older is 615.07 billion ($3.85 billion). Similarly, three major diseases, namely cancer, cardiovascular disease and respiratory disease, along with associated deaths, cost the nation PKR437.8 billion ($2.7 billion) of which 77% is the indirect cost. The three major diseases make 71% of the total estimated cost, nearly two-thirds of which is borne by rural residents, nine-tenth by males and more than four-fifths by the citizens in the 35-64 years age group., Conclusion: The total annual economic costs of all smoking-attributable diseases and deaths and those of the three major diseases equal 1.6% and 1.15% of Pakistan's gross domestic product, respectively. The tax contribution of tobacco sector is merely 20% of the total estimated cost. The finding of huge economic and health costs of smoking makes a convincing case for policymakers to realise the true value of the industry's contribution and raise tobacco taxes to the level of full cost recovery., Competing Interests: Competing interests: None declared., (© Author(s) (or their employer(s)) 2024. Workers in informal employment suffered significant out-of-pocket healthcare expenditures (OOPHEs) due to their low earnings and a lack of a social safety net or health insurance. There is little or no evidence of impoverishment caused by OOPHEs in the context of labor market categorization. Therefore, this study examines the economic burden of OOPHEs and its associated consequences on households, whose members are in informal employment. This study estimates the incidence of catastrophic health expenditures (CHEs) and impoverishment across the households in formal and informal employment and their key determinants in Pakistan by employing the data from the two rounds of the Household Integrated Economic Survey (2015-16, 2018-19). For measuring CHEs and impoverishment, the budget share and capacity-to-pay approaches are applied. Various thresholds are used to demonstrate the sensitivity of catastrophic measures. We found a higher incidence of catastrophic healthcare payments among the informal workers, that is, 4.03% and 7.11% for 2015-16 and 2018-19, respectively, at a 10% threshold, while at a 40% threshold, the incidence of CHEs is found to be 0.40% and 2.34% for 2015-16 and 2018-19, respectively. These OOPHEs caused 1.53% and 3.66% of households who are in informal employment to become impoverished, compared with their formal counterparts. The study demonstrates that the probability of incurring CHEs and becoming impoverished is high among informal workers, compared with their formal counterparts. This result has clear policy implications, in which to protect the informal workers, it is necessary to expand the insurance coverage, particularly during the COVID-19 response and recovery efforts., (© 2023 The Authors.
