6. Sumoylation of Pdx1 is associated with its nuclear localization and insulin gene activation

Abstract

Pancreatic duodenal homeobox-1 (Pdx1) is a transcription factor, and its phosphorylation is thought to be essential for activation of insulin gene expression. This phosphorylation is related to a concomitant shift in molecular mass from 31 to 46 kDa. However, we found that Pdx1 was modified by SUMO-1 (small ubiquitin-related modifier 1) in [beta]-TC-6 and COS-7 cells, which were transfected with Pdx1 cDNA. This modification contributed to the increase in molecular mass of Pdx1 from 31 to 46 kDa. Additionally, sumoylated Pdx1 localized in the nucleus. The reduction of SUMO-1 protein by use of RNA interference (SUMO-iRNAs) resulted in a significant decrease in Pdx1 protein in the nucleus. A 34-kDa form of Pdx1 was detected by the cells exposed to SUMO-iRNAs in the presence of lactacystin, a proteasome inhibitor. Furthermore, the reduced nuclear sumoylated Pdx1 content was associated with significant lower transcriptional activity of the insulin gene. Abstract

Bhutanese Buddhism and the nature of the country are supposed to support the happiness of the Bhutanese. This report introduces the former studies and my own experiences in Bhutan, which supports the idea. There are at least two ways in Bhutanese Buddhism supporting the spirits of the Bhutanese. One is that they think not only about this life but also the afterlife, so they don’t only think about the happiness in this life. The other is that the Buddhism contains the idea that one should be content to be as it is. I think the natural environment also supports the spirits of the people. Abstract

ブータンと言えば「幸せの国」という言葉を思い浮かべる人が多いだろう。これは、ブータンは国民総所得が低いにも関わらず、2005年の国勢調査で国民の約97％が「幸せ」と回答したという事実に影響されているためだろう。では物質的な豊かさはなくとも、精神的には幸せと感じるブータン人の暮らしとは一体どのようなものだろうか。この疑問に対する答えを探るべく、今回、ブータン滞在中に私が感じたことも交えながら、ブータンの文化や暮らしを考察していこうと思う。, It is often said tha"t Bhutan is a happy country". Then, what the life in Bhutan is like? In this essay, I would like to write about Bhutan's lifestyle. People in Bhutan wear folk costumes in daily lives. Male costumes are called" Go", and female costumes are called" Kira". They are similar to kimono, Japanese costume. I think that by wearing the same costumes, people in Bhutan feel strong affinities with each other and strengthen their bonds. In Bhutan, people mainly eat rice and meat, vegetables, and mushrooms which are boiled with cheese. The most popular vegetable is red pepper, and they eat red pepper boiled with cheese every meal. At first, I did not like red pepper very much because it was hot. However, I gradually came to like it, and, now, I even miss that hot red pepper. During our trip, we stayed at a farmer's house in Ura Area for two days. During the stay, I was impressed that farmers were really considerate to each other, whether they were families or not. Then, I wondered why they were so kind to each other and asked the driver the reason. He immediately answered, "It is because people in Bhutan believe that others’ happiness is their own happiness". " When you are in trouble, someone is sure to help you." People in Bhutan believe so and help each other in daily lives. I think that such sense of security leads them to feel that they are happy. 15. Limitations of using generalized transport costs to estimate changes in trip demand: a bias caused by the endogenous value of time

Abstract

ABSTRACTWe show that constancy of value of time (VoT), which is assumed in generalized transport costs, can generate a significant bias in trip demand forecasts when travel time and travel fee change, because VoT endogenously changes in reality. Modeling shopping behaviors, we demonstrate that the change in trip demand through changes in travel time and travel fees differs even if the change in the travel fee is equivalent to the changes in the travel time in terms of generalized transport cost. This is because a decrease in travel time decreases VoT, whereas a decrease in travel fee increases VoT. Our simulation shows that the difference is large when there is a high substitutability between sites. This implies that demand estimation assuming a fixed VoT could be inadequate, and that it should treat VoT endogenously.

Abstract

ABSTRACT Aim Materials and Methods Results Conclusions Nocturia impairs the quality of life in patients with type 2 diabetes mellitus. Although sodium glucose co‐transporter 2 inhibitors (SGLT2i) such as tofogliflozin increase urine volume, their impact on nocturia, in conjunction with dietary salt restriction, is less clear.This multicenter, open‐label, randomized, parallel‐group trial included 80 subjects with type 2 diabetes and nocturia. The patients were divided into two groups: one receiving tofogliflozin, the shortest half‐life, without salt restriction, and the other receiving both tofogliflozin and dietary salt restriction. The primary endpoint was nocturia frequency at 12 weeks. The secondary outcomes included changes in daytime urination frequency, urine volume, and home blood pressure.At 12 weeks, there were no significant differences in nocturia changes between both groups. Nocturia frequency did not change in the tofogliflozin without salt restriction group from 1.5 ± 0.8 to 1.3 ± 1.1 times per night (P = 0.297), and significantly decreased from 1.6 ± 1.0 to 1.3 ± 0.7 times per night in the tofogliflozin and dietary salt restriction group (P = 0.049). There was a trend toward increased urine volume and frequency during the daytime in the group with salt restriction, indicating a time‐shift effect of the short half‐life tofogliflozin and salt restriction on urinary time.The frequency of nocturia after tofogliflozin did not increase. Tofogliflozin reduced nocturia when combined with salt restriction. Furthermore, daytime urine volume and frequency showed an increasing trend, suggesting a shift in urine production to daytime hours due to the short half‐life of tofogliflozin. Dietary modifications can enhance the therapeutic benefits of tofogliflozin in managing nocturia in people with type 2 diabetes. [ABSTRACT FROM AUTHOR]

31. Ultrastructure of the Gelatinous Layer in Tension Wood Fibers from Populus euramericana

Abstract

木材の細胞膜構造についてより深い知見を得るための一つの課題として, Populus euramericanaの引張あて材のゼラチン層 (以下Gと略記) の微細構造を電子顕微鏡, X線回折法および赤外分光分析法を用いて研究した。試料のゼラチン層は, NorbergとMeierの方法を用いて引張あて材から分離して得た。引張あて材のゼラチン繊維の膜層構成はP+S_1+S_2+Gである。Gの解体試料を負染色して電子顕微鏡で観察した結果, Gのミクロフィブリルは長さの方向に非晶あるいは準結晶領域の存在を推定させるような周期構造は認められない。ミクロフィブリルの幅は50 - 70Åである。反射法によるGのX線回折強度曲線から, Gの結晶幅は37Å, 結晶化度は60％の値が測定された。またGの赤外分光分析の結果, Gにはリグニンがほとんど存在しないこと, また結晶性の高いセルロースからなることがわかる。
