Abstract

Neoangiogenesis is a pivotal therapeutic target in glioblastoma. Tumor monitoring requires imaging methods to assess treatment effects and disease progression. Until now mapping of the tumor vasculature has been difficult. We have developed a combined magnetic resonance and optical toolkit to study neoangiogenesis in glioma models. We use in vivo magnetic resonance imaging (MRI) and correlative ultramicroscopy (UM) of ex vivo cleared whole brains to track neovascularization. T2* imaging allows the identification of single vessels in glioma development and the quantification of neovessels over time. Pharmacological VEGF inhibition leads to partial vascular normalization with decreased vessel caliber, density, and permeability. To further resolve the tumor microvasculature, we performed correlated UM of fluorescently labeled microvessels in cleared brains. UM resolved typical features of neoangiogenesis and tumor cell invasion with a spatial resolution of ~5 µm. MR-UM can be used as a platform for three-dimensional mapping and high-resolution quantification of tumor angiogenesis.

Abstract

Background: The San Francisco Project is a prospective study aiming to determine the most important cardiovascular risk factors in the Chilean population. Aim: To report the results of the San Francisco Project after a mean follow up of five years. Material and methods: Between 1997 and 1999, 920 subjects (aged 39.5 ± 16.3 years, 382 males), free of cardiovascular events, were examined to determine their cardiovascular risk factors. The prevalence of risk factors was compared with the Chilean National Health Survey. All non fatal cardiovascular events were registered until 2003. A logistic regression model including gender, obesity, family history, educational level and classical cardiovascular risk factors, was performed to identify independent predictors of cardiovascular events. Results: The prevalence of cardiovascular risk factors in the studied population was similar to that of the Chilean National Health Survey, except for serum lipid levels, that were lower in the latter. The mean follow up was 5.3 ± 0.3 years (range 4-6 years), and 26 cardiovascular events were registered. The independent predictors of these events were age, with a relative risk (RR) of 4.3 and 95% confidence intervals (CI) of 1.9 to 9.5 (p

40. Evaluation of two immunoassay procedures for drug testing in hair samples.

Abstract

Abstract: A preliminary initial enzyme-linked immunosorbent assay (LUCIO®-Direct ELISA kit) and a preliminary DRI® enzyme immunoassay were evaluated for drug detection in head hair with respect to lowered cutoff values recommended in Germany for the control of abstinence in cases of re-granting of drivers’ licences. Following drug classes were included: cannabinoids, opiates, cocaine like substances, amphetamine, methamphetamine (and methylenedioxyamphetamines), methadone, and benzodiazepines. 759 analyses were performed using LUCIO®-Direct ELISA kits and 936 analyses using DRI® enzyme immunoassay tests. Sample size for each drug group and immunoassay test reached from 74 to 178. The LUCIO®-Direct ELISA kit revealed a sensitivity of 91% for amphetamine up to 98% for methadone (methamphetamine 92%, cocaine 94%, opiates 94%, benzodiazepines 96%) and values of specificity of 72% for methadone up to 89% for amphetamine and benzodiazepines. The test was not useful for a preliminary screening for tetrahydrocannabinol (sensitivity of 65%) in consideration of a suggested cutoff of 0.02ng/mg. The DRI® enzyme immunoassay test was only useful for morphine and cocaine testing at low recommended new cutoff values (0.1ng/mg) revealing sensitivities of 94% and 99%, respectively. Abstract

The preparation and reactions of several new (η3-allyl)molybdenum complexes are described. Functionalization of a carbon−carbon double bond adjacent to the π-allyl unit proceeds stereoselectively, and the stereochemical outcome of this reaction is discussed in light of the Curtin−Hammett principle. It is shown that carbonyl groups can also be functionalized with moderate stereoselectivity, and this behavior is rationalized on the basis of conformational analysis using molecular mechanics calculations.
