Community recreation centers (CRCs) are gaining popularity across the country. While many support the argument that CRCs improve quality of life by building social capital and improving the physical health of residents, that support does not necessarily translate into community support for the funding and construction of CRCs. Some communities undertake surveys to try to gauge the support for CRCs but have had mixed results on the insights provided by those surveys. Furthermore, public perceptions about the accuracy and usefulness of survey data could lead to stakeholder disregard for the data produced by them. This case study focuses on the benefits of a robust partnership between a local community and a regional university in developing a survey research project to analyze support for building a CRC. In this paper, we emphasize the value of producing trusted data and provide lessons on how community-university partnerships can achieve similar objectives. The first lesson is that community-university partnerships, by emphasizing local connections, can help foster trust in survey research projects. Second, strategic community-university partnerships can derive additional benefits such as division of labor, expertise, and cost savings. Third, by embracing the above two benefits, collaboration is not only possible, but can produce tailored survey research projects that provide stakeholders insights into critical questions. Finally, community-university partnerships that are structured around shared values can help communities solve critical policy issues effectively and efficiently. This case study details these lessons through an examination of a robust communityuniversity partnership that led to the passage of a ballot referendum to build a CRC. The idea that medical treatment costs and outcomes might be connected is not new. Likewise, as long as researchers have been designing clinical trials and public opinion polls, there has been interest in the sample size necessary to obtain a desired level of precision and certainty before collecting the data. However, researchers continue to adapt cost-effectiveness models for scenarios of ever-increasing complexity, and equally adaptable sample size determination schemes are required. Of interest for the current study are those instances wherein a non-trivial proportion of patients incur zero costs associated with their treatment. We propose a sample size determination scheme for a cost-effectiveness model fit to such a scenario. Abstract Conjugated linoleic acid (CLA) supplementation has shown convincing effects at reducing body fat in animals; yet human study results have been somewhat inconclusive. The purpose of this study is to determine whether four weeks of CLA supplementation, the approximate length of a commercial package, can result in a positive change in visceral adipose tissue in resistance-trained middle-aged men. Thirty overweight and moderately obese, but otherwise healthy male subjects (aged 35 to 55 years) currently involved in resistance training, were randomly assigned into CLA and placebo groups in a double-blind, placebo controlled approach. The study lasted for 12 weeks and consisted of three four-week periods. During the first four weeks (run-in period) each subject received placebo (4 g safflower oil). Throughout the next four weeks (supplementation period), the placebo group continued receiving placebo, while the CLA group received 3.2 g/d of CLA. During the final four weeks (run-out period) all subjects received the placebo. Computed tomography (CT) scans were used to measure visceral adipose tissue (VAT) at weeks 4, 8 and 12. No significant reduction in VAT cross-sectional area was determined in the CLA group during the study. On the contrary, a significant reduction in cross-sectional area of VAT of 23.12 cm2 during the supplementation period was measured in the placebo group, which was abated during the run-out period. Our results suggest that CLA supplementation of 3.2 g/d for four weeks does not promote decreases in VAT in middle-aged men currently participating in a resistance-training program.

Abstract Background Lung cancer remains the leading cause of cancer-related mortality despite continuous efforts to find effective treatments. Data from the American Cancer Society indicate that while the overall incidence of lung cancer is declining, it continues to rise in women. Stem cell-based therapy has been an emerging strategy to treat various diseases. The purpose of this paper is to determine the efficacy of an intrinsic anti-cancer effect of rat umbilical cord matrix stem cells (UCMSCs) on lung cancer. Methods A mouse syngeneic lung carcinoma model was used to test the basic ability of UCMSCs to control the growth of lung cancer. Lung tumors were experimentally induced by tail vein administration of Lewis lung carcinoma (LLC) cells derived from the lung of C57BL/6 mouse. Rat UCMSCs were then administered intratracheally five days later or intravenously on days 5 and 7. The tumor burdens were determined by measuring lung weight three weeks after the treatment. Results Co-culture of rat UCMSCs with LLC significantly attenuated the proliferation of LLC cells as monitored by MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide), a tetrazole cell proliferation assay, thymidine uptake, and direct cell counts. In vitro colony assays with rat UCMSCs as feeder layers markedly reduced LLC colony size and number. Co-culture of rat UCMSCs with LLCs causes G0/G1 arrest of cancer cells. This is evident in the decrease of cyclin A and CDK2 expression. The in vivo studies showed that rat UCMSC treatment significantly decreased tumor weight and the total tumor mass. Histological study revealed that intratracheally or systemically administered rat UCMSCs homed to tumor areas and survived for at least 3 weeks without any evidence of differentiation or adverse effects. Conclusions These results indicate that rat UCMSCs alone remarkably attenuate the growth of lung carcinoma cells in vitro and in a mouse syngeneic lung carcinoma graft model and could be used for targeted cytotherapy for lung cancer.

Quantile regression (QR) allows one to model the effect of covariates across the entire response distribution, rather than only at the mean, but QR methods have been almost exclusively applied to continuous response variables produced by a single data-generating process. Of the few studies that have performed QR on count data, none have accounted for excess zeros from a Bayesian perspective, as does the hurdle model that we propose. In this article, we propose a Bayesian two-part QR model for count data with excess zeros. The proposed model is compared to a frequentist approach via simulation, and its usefulness is displayed on two real datasets. In each application, multiple covariates are found to have differing effects across the response distribution, with special attention given to the nature of those effects in the outermost response distribution quantiles. Quantile regression (QR) allows one to model the effect of covariates across the entire response distribution, rather than only at the mean, but QR methods have been almost exclusively applied to continuous response variables and without considering spatial effects. Of the few studies that have performed QR on count data, none have included random spatial effects, which is an integral facet of the Bayesian spatial QR model for areal counts that we propose. Additionally, we introduce a simplifying alternative to the response variable transformation currently employed in the QR for counts literature. The efficacy of the proposed model is demonstrated via simulation study and on a real data application from the Texas Department of Family and Protective Services (TDFPS). Our model outperforms a comparable non-spatial model in both instances, as evidenced by the deviance information criterion (DIC) and coverage probabilities. With the TDFPS data, we identify one of four covariates, along with the intercept, as having a nonconstant effect across the response distribution. [ABSTRACT FROM AUTHOR]

Dental caries induced by Streptococcus mutans is one of the most prevalent chronic infectious diseases worldwide. The pathoge-nicity of S. mutans relies on the bacterium's ability to colonize tooth surfaces and survive a strongly acidic environment. We performed an I SS1 transposon mutagenesis to screen for acid-sensitive mutants of S. mutans and identified an SMU.746-SMU.747 gene cluster that is needed for aciduricity. SMU.746 and SMU.747 appear to be organized in an operon and encode a putative membrane-associated permease. SMU.746- and SMU.747-deficient mutants showed a reduced ability to grow in acidified medium. However, the short-term or long-term acid survival capacity and F1F0 ATPase activity remained unaffected in the mutants. Furthermore, deletion of both genes did not change cell membrane permeability and the oxidative and heat stress responses. Growth was severely affected even with slight acidification of the defined medium (pH 6.5). The ability of the mutant strain to acidify the defined medium during growth in the presence of glucose and sucrose was significantly reduced, although the glycolysis rate was only slightly affected. Surprisingly, deletion of the SMU.746-SMU.747 genes triggered increased biofilm formation in low-pH medium. The observed effects were more striking in a chemically defined medium. We speculate that the SMU.746-SMU.747 complex is responsible for amino acid transport, and we discuss its possible role in colonization and survival in the oral environment. [ABSTRACT FROM AUTHOR]
