This study unveils histological features of the intestinal tract of juvenile striped catfish Pseudoplatystoma fasciatum (Linnaeus, 1776) in three size classes (weight, standard length): I – 36.84 ± 10.19 g, 14.52 ± 1.54 cm; II – 59.03 ± 11.47 g, 17.17 ± 1.06 cm; III – 89.72 ± 18.70 g, 20.79 ± 1.55 cm, respectively. Histological organization of the juvenile speckled catfish intestine bears features common to the carnivorous fish, but the organ presents some convolutions that indicate a certain degree of dietary flexibility, a surprising trend, common only to omnivorous Siluriforms. The architecture of the mucosa of the speckled catfish intestine indicates that the species concentrates digestion and absorption of nutrients in the medium intestine, a common feature among carnivorous Teleosts. New technologies for more effective microbiological assays are being adopted by the food industry to intervene more rapidly in its production chain. The aim of this study was to evaluate the alternative methods of TEMPO ® CAM and real-time PCR (rtPCR) Biotecon ® in comparison with the ISO 10272-2:2017 reference method for Campylobacter spp. quantification in turkey meat, aiming to validate a quick and easily replicable method in these meat matrices. A total of 416 samples were analyzed over a one-year period. The TEMPO ® methodology showed inadequate performance with a significant difference ( p < 0.05) compared with the reference methodology; therefore, its use was not recommended for turkey meat matrices. However, the performance of the rtPCR Biotecon ® methodology showed adequate performance with no significant difference ( p > 0.05), and its use was recommended in turkey meat matrices. The study was limited to exclusive research in turkey meat matrices, and expansion of the research into other matrices is recommended to verify whether the behavior of alternative methodologies is similar. The findings of this study illustrate the necessity for a thorough and comprehensive evaluation during the implementation of alternative methodologies that may potentially supplant conventional approaches.

This study aimed to assess the impact of a commercial blend of functional oils, specifically cashew nutshell liquid and castor oil (FO), in two physical forms (solid: P; liquid: S), in comparison to a combination of virginiamycin and anticoccidials on the gut health of broilers challenged with coccidiosis. A total of 1760 1-day-old male chicks were randomly distributed in a study design with eight treatments. The treatments included: a control group (without additive), OFS&#95;0.75&#95;kg/t (FO spray), OFP&#95;1.0&#95;kg/t (FO powder), OFP&#95;1.5&#95;kg/t (FO liquid spray), Sal (anticoccidials), Sal&#95;Vir (virginiamycin and anticoccidials), Sal&#95;OFS&#95;0.5&#95; kg/t (anticoccidials plus FO spray), and Sal&#95;OFP&#95;1.0&#95;kg/t (anticoccidials plus FO powder). All birds were challenged with Eimeria spp. at 14 days. The physical form of FO did not affect performance and intestinal health parameters. At 42 days, broilers from the control and OFS&#95;0.75 treatments were the lightest, while those from the Sal&#95;Vir and Sal&#95;OFP&#95;1.0 treatments were the heaviest (P < 0.05). FO reduced the presence of Clostridium perfringens. The individual phytogenic additives did not prevent weight loss in birds challenged with Eimeria, but they mitigated the effects of the infection by modulating the intestinal microbiota. We aimed to evaluate the effect of different grazing management strategies on carcass characteristics traits, meat quality and CH 4 intensity and yield of lambs grazing Italian ryegrass pastures in Southern Brazil. A grazing trial was performed (2014 and 2015) in a randomized complete block design with two grazing management targets and four replicates. Treatments were traditional rotational stocking (RT), with pre- and post-grazing sward heights of 25 and 5 cm, respectively, and 'Rotatinuous' stocking (RN), with pre- and post-grazing sward heights of 18 and 11 cm, respectively. Castrated crossbred Texel and Polwarth lambs were used. Results indicated that diet cost per kg of dry matter (p = 0.001) and per hectare (p < 0.001) were lower for RN than for RT treatment. Final live weight (p = 0.022) and hot and cold carcass weight (p = 0.006) were greater for the RN treatment. All commercial cuts were greater for RN than for RT treatment. The RN treatment presented greater (p < 0.001) production of carcass, edible food and crude protein. Feed efficiency and feed cost conversion were better for RN than for RT treatment. CH 4 intensity per kg of carcass, edible food and crude protein gain were 2.6, 2.7 and 2.1 times lower (p < 0.001) for RN. Moreover, CH 4 yield was lower (p = 0.014) for RN than for RT treatment, with an average of 7.6 and 8.3% of the gross energy intake, respectively. We conclude that the 'Rotatinuous' stocking results in a greater carcass production, carcass quality and lower diet cost, and CH 4 intensity and yield of grazing lambs. Muscle satellite cells (MSCs) are myogenic stem cells that play a critical role in post-hatch skeletal muscle growth and regeneration. Activation of regeneration pathways to repair muscle fiber damage requires both the proliferation and differentiation of different MSC populations as well as the function of resident phagocytic cells such as anti-inflammatory and pro-inflammatory macrophages. The Wooden Breast (WB) phenotype in broiler chickens is characterized by myofiber degeneration and extensive fibrosis. Previous work indicates that the resident MSC populations expressing the myogenic regulatory factors, Myf-5 and Pax7 are larger and more proliferative in broilers severely affected with WB vs. unaffected broilers. To further characterize the cellular and molecular changes occurring in WB-affected muscles, samples from pectoralis major (PM) muscles with varying severity of WB (WB score 0 = normal; 1 = mildly affected; 2 = severely affected) were collected at 25 and 43 days post-hatch ( n = 8 per score per age) and processed for cryohistological and protein expression analyses. Collagen per field and densities of macrophages and MyoD+, Myf-5+, and Pax7+ MSC populations were quantified on immunofluorescence-stained cryosections. Relative collagen protein expression was quantified by fluorescent Western Blotting. In both 25 and 43-days-old broilers, the proportion of collagen per field ( P ≤ 0.021) and macrophage density ( P ≤ 0.074) were greater in PM exhibiting severe WB compared with normal. At day 43, populations of MyoD+, Myf-5+:MyoD+ MSC were larger and relative collagen protein expression was greater in WB-affected vs. unaffected broilers ( P ≤ 0.05). Pax7+ MSC relative to total cells was also increased as WB severity increased in 43-days-old broilers ( P ≤ 0.05). Densities of Myf-5+ ( P = 0.092), MyoD+ ( P = 0.030), Myf5+:MyoD+ ( P = 0.046), and Myf-5+:MyoD+:Pax7+ ( P = 0.048) MSC were greater in WB score 1 birds compared with WB score 0 and 2 birds. Overall, alterations in the resident MSC and macrophage populations and collagen protein content were observed in WB-affected muscle. The aim of study was evaluate the effect of using rice protein coating with different plasticizers types on the quality of eggs storaged at 20 °C for 6 weeks. Three hundred eggs were coated with rice protein at 8% solution combined with glycerol, propylene glycol or sorbitol. Uncoated eggs showed the highest weight loss (5.31%), while rice protein with glycerol (4.29%) propylene glycol (4.13%) and sorbitol (4.07%) solutions were effective in preventing weight lost. Uncoated eggs had the worst HU (58.40), albumen (9.52) and yolk (7.06) pH, and YI (0.33) after 6 weeks of storage. The eggs coated of rice protein with glycerol, propylene glycol and sorbitol presented results with similar intern quality between them during all the storage period. However, the use of sorbitol as a plasticizer in the coating is more efficient in maintaining control of the increase in albumen pH. Scanning electron microscopy demonstrated a lower surface porosity in coated eggshell, indicating that the use of the coating may provide a protective barrier against the transfer of gases and moisture. A study was conducted to investigate Fe requirements of broiler breeders. One-hundred-fifty-six Cobb 500 broiler breeder hens were individually placed in electrostatically painted cages at 22 weeks. The study was composed of an adaptation phase, in which hens were fed corn-soy-wheat bran diets until 35 wks. An Fe deficient mash diet (24.6 ppm Fe) was provided from 35 to 46 wk in order to induce a partial body Fe depletion. A production phase followed from 47 to 70 wk when hens were fed 6 diets with increasing Fe sulfate supplementation, which, upon analyses had 24.6, 48.6, 74.3, 99.6, 125.6, and 148.2 ppm Fe. Thirty hatching eggs from each treatment were randomly collected in the last wk of each production period and incubated. Hemoglobin and hematocrit were analyzed from 6 hens as well as all hatched chicks per treatment. Analyses of production and hatching data were conducted using quadratic polynomial (QP), broken-line (BL), and exponential asymptotic (EA) models. Effects of dietary Fe were observed for total eggs and total hatching eggs, egg yolk Fe content, and hen and chick hematocrit and hemoglobin (P < 0.05). These responses to added Fe were optimized when dietary Fe were 96.8, 97.1, 130.6, 122.6, 120.0, and 125.0 ppm (QP) and 76.4, 89.3, 135.0, 128.4, 133.8, and 95.0 ppm (BL) for total hatching eggs, egg yolk Fe content, and hen and chick hematocrit and hemoglobin, respectively. Optimization with the EA model was obtained for total hatching eggs, egg yolk Fe, and hen and chick hemoglobin at 97.9, 111.0, 77.9, and 96.3 ppm Fe for total hatching eggs, egg yolk Fe, and hen and chick hemoglobin, respectively. Adequate Fe levels are needed to maintain egg production as well as hatching chicks' indexes. Fe concentration in the yolk and diet are positively influenced. Three experiments were conducted separately to estimate the digestible Lys (dig. Lys) requirements of Cobb × Cobb 500 male broilers using different statistical models. For each experiment, 1,200 chicks were housed in 48 floor pens in a completely randomized design with 6 treatments and 8 replicates. Broilers were fed diets with increasing dig. Lys levels from 1 to 12 d (Exp. 1), from 12 to 28 d (Exp. 2), and 28 to 42 d (Exp. 3). Increasing dig. Lys levels were equally spaced from 0.97 to 1.37% in Exp. 1, 0.77 to 1.17% in Exp. 2, and 0.68 to 1.07% in Exp. 3. The lowest dig. Lys diets were not supplemented with L-Lysine and all other essential AA met or exceeded recommendations. In Exp. 3, six birds per pen were randomly selected from each replication to evaluate carcass and breast yields. Digestible Lys requirements were estimated by quadratic polynomial (QP), linear broken-line (LBL), quadratic broken-line (QBL), and exponential asymptotic (EA) models. Overall, dig. Lys requirements varied among response variables and statistical models. Increasing dietary dig. Lys had a positive effect on BW, carcass and breast yields. Levels of dig. Lys that optimized performance using QP, LBL, QBL, and EA models were 1.207, 1.036, 1.113, and 1.204% for BWG and 1.190, 1.027, 1.100, and 1.172% for FCR in Exp. 1; 1.019, 0.853, 0.944; 1.025% for BWG and 1.050, 0.879, 1.032, and 1.167% for FCR in Exp. 2; and 0.960, 0.835, 0.933, and 1.077% for BWG, 0.981, 0.857, 0.963, and 1.146% for FCR in Exp. 3. The QP, LBL, QBL, and EA also estimated dig. Lys requirements as 0.941, 0.846, 0.925, and 1.070% for breast meat yield in Exp. 3. In conclusion, Lys requirements vary greatly according to the statistical analysis utilized; therefore, the origin of requirement estimation must be taken into account in order to allow adequate comparisons between references.
