Abstract

Since 2007, the Oncofertility Consortium Annual Conference has brought together a diverse network of individuals from a wide range of backgrounds and professional levels to disseminate emerging basic and clinical research findings in fertility preservation. This network also developed enduring educational materials to accelerate the pace and quality of field-wide scientific communication. Between 2007 and 2019, the Oncofertility Consortium Annual Conference was held as an in-person event in Chicago, IL. The conference attracted approximately 250 attendees each year representing 20 countries around the world. In 2020, however, the COVID-19 pandemic disrupted this paradigm and precluded an in-person meeting. Nevertheless, there remained an undeniable demand for the oncofertility community to convene. To maintain the momentum of the field, the Oncofertility Consortium hosted a day-long virtual meeting on March 5, 2021, with the theme of "Oncofertility Around the Globe" to highlight the diversity of clinical care and translational research that is ongoing around the world in this discipline. This virtual meeting was hosted using the vFairs ® conference platform and allowed over 700 people to participate, many of whom were first-time conference attendees. The agenda featured concurrent sessions from presenters in six continents which provided attendees a complete overview of the field and furthered our mission to create a global community of oncofertility practice. This paper provides a synopsis of talks delivered at this event and highlights the new advances and frontiers in the fields of oncofertility and fertility preservation around the globe from clinical practice and patient-centered efforts to translational research.

Abstract

PURPOSE: Today, male and female adult and pediatric cancer patients, individuals transitioning between gender identities, and other individuals facing health extending but fertility limiting treatments can look forward to a fertile future. This is, in part, due to the work of members associated with the Oncofertility Consortium. METHODS: The Oncofertility Consortium is an international, interdisciplinary initiative originally designed to explore the urgent unmet need associated with the reproductive future of cancer survivors. As the strategies for fertility management were invented, developed or applied, the individuals for who the program offered hope, similarly expanded. As a community of practice, Consortium participants share information in an open and rapid manner to addresses the complex health care and quality-of-life issues of cancer, transgender and other patients. To ensure that the organization remains contemporary to the needs of the community, the field designed a fully inclusive mechanism for strategic planning and here present the findings of this process. RESULTS: This interprofessional network of medical specialists, scientists, and scholars in the law, medical ethics, religious studies and other disciplines associated with human interventions, explore the relationships between health, disease, survivorship, treatment, gender and reproductive longevity. CONCLUSION: The goals are to continually integrate the best science in the service of the needs of patients and build a community of care that is ready for the challenges of the field in the future.

9. A Conserved Role for H15-elated T-Box Transcription Factors in Zebrafish and Drosophila Heart Formation

Abstract

T-box transcription factors are critical regulators of early embryonic development. We have characterized a novel zebrafish T-box transcription factor, hrT (H15-related T box) that is a close relative of Drosophila H15 and a recently identified human gene. We show that Drosophila H15 and zebrafish hrT are both expressed early during heart formation, in strong support of previous work postulating that vertebrate and arthropod hearts are homologous structures with conserved regulatory mechanisms. The timing and regulation of zebrafish hrT expression in anterior lateral plate mesoderm suggest a very early role for hrT in the differentiation of the cardiac precursors, hrT is coexpressed with gata4 and nkx2.5 not only in anterior lateral plate mesoderm but also in noncardiac mesoderm adjacent to the tail bud, suggesting that a conserved regulatory pathway links expression of these three genes in cardiac and noncardiac tissues. Finally, we analyzed hrT expression in pandora mutant embryos, since these have defects in many of the tissues that express hrT, including the heart, hrT expression is much reduced in the early heart fields of pandora mutants, whereas it is ectopically expressed subsequently. Using hrT expression as a marker, we describe a midline patterning defect in pandora affecting the anterior hindbrain and associated midline mesendodermal derivatives. We discuss the possibility that the cardiac ventricular defect previously described in pandora and the midline defects described here are related.

Abstract

Background The evolution of animal segmentation is a major research focus within the field of evolutionary–developmental biology. Most studied segmented animals generate their segments in a repetitive, anterior-to-posterior fashion coordinated with the extension of the body axis from a posterior growth zone. In the current study we ask which selection pressures and ordering of evolutionary events may have contributed to the evolution of this specific segmentation mode. Results To answer this question we extend a previous in silico simulation model of the evolution of segmentation by allowing the tissue growth pattern to freely evolve. We then determine the likelihood of evolving oscillatory sequential segmentation combined with posterior growth under various conditions, such as the presence or absence of a posterior morphogen gradient or selection for determinate growth. We find that posterior growth with sequential segmentation is the predominant outcome of our simulations only if a posterior morphogen gradient is assumed to have already evolved and selection for determinate growth occurs secondarily. Otherwise, an alternative segmentation mechanism dominates, in which divisions occur in large bursts through the entire tissue and all segments are created simultaneously. Conclusions Our study suggests that the ancestry of a posterior signalling centre has played an important role in the evolution of sequential segmentation. In addition, it suggests that determinate growth evolved secondarily, after the evolution of posterior growth. More generally, we demonstrate the potential of evo-devo simulation models that allow us to vary conditions as well as the onset of selection pressures to infer a likely order of evolutionary innovations. 13. Regulation of Mesoderm Induction and Involution in Zebrafish

Abstract

A fundamental aspect of early embryogenesis is the formation of the hypoblast, the cell layer in which mes-endodermal differentiation subsequently occurs. Despite the importance of hypoblast formation, comparatively little is known about the molecular-genetic control of this extensive morphogenetic rearrangement. Mutant analysis in mouse and zebrafish suggests that the Nodal and FGF signaling pathways function together with members of the T-box transcription factor family in this process. We have begun to examine the genetic interplay between these factors to dissect the control of mesoderm induction and hypoblast formation. In zebra fish, nonnotochord mesoderm progenitors continuously express three T-box transcription factors: no tail (which is predominantly expressed in epiblast cells but is required only for notochord and tail development), spadetail (which is predominantly expressed in hypoblast cells and is required for trunk somite formation), and tbx6 (which is coexpressed with spadetail and thought to be important for tail somite formation). A combination of double-mutant analysis and gene expression patterns supports a two-step model in which spadetail and no tail are initially required for the formation of mesodermal progenitors, but subsequently spadetail and tbx6 promote the formation of hypoblast. We find that Nodal signaling has an important role in hypoblast formation, by maintaining expression of spadetail and tbx6, which is clearly separate from the role of Nodal signaling in mesoderm induction. Our studies demonstrate that hypoblast formation is a multistep process involving independently acting pathways.

14. Characterization of the Roles of a Novel T-Box Transcription Factor, hrT, during Cardiovascular Development in Zebrafish

Abstract

Our laboratory has recently identified a novel T-box transcription factor, hrT, which is likely an ortholog of the Drosophila H15 gene. The initial characterization of the hrT gene has shown that it is expressed throughout all of the key steps of cardiogenesis. The expression patterns of hrT are very similar to that of the nkx 2.5 gene, a cardiac-specific marker. In addition, the expression of hrT has been detected in a number of other tissues, including the roof of the dorsal aorta. The potential roles of hrT in vasculogenesis and cardiogenesis were investigated by using the morpholino antisense oligonucleotide injection approach. The phenotypic consequences of zebrafish embryos injected with the hrT-specific morpholino oligonucleotide include anomalous tail development, absence of blood circulation, and dismorphic heart formation. The anomalous tail development is marked by a dramatic decrease in tbx6 expression which is potentially required for the tail mesoderm formation. The absence of blood circulation is marked by altered gene expression of flk-1 and scl-1, which have been shown to be essential for dorsal aorta development. The dismorphic heart defect is marked by morphological alterations in looping and chamber formation. These observations provide strong evidences that hrT plays an important role in cardiovascular development and tail mesoderm formation.

Abstract

Posterior mesoderm formation in zebrafish embryos requires FGF signaling, and the effects of FGF signaling are mediated by the T-box transcription factors Spadetail (Spt) and the Brachyury orthologue No Tail (Ntl). Spt and ntl are expressed throughout trunk and tail mesoderm progenitors but Spt is required only for trunk paraxial mesoderm, whereas ntl is required only for tail mesoderm and the notochord. The discrepancy between the broad expression patterns of these genes and their mutant phenotypes suggests that interactions with other pathways masks their full developmental roles. Consistent with this, ntl interacts synergistically with one-eyed pinhead (oep), an extracellular factor required for Nodal-related TGF[Beta] signaling, causing oep;ntl mutants to lack most trunk mesoderm. We show that oep similarly interacts with spt, resulting in embryos with little remaining mesoderm. Analysis of gene expression in oep;spt embryos indicates that mesoderm induction occurs normally, but that mesoderm involution is blocked. We used genetic and pharmacological methods to understand how oep interacts with spt and ntl. Using the FGFR inhibitor SU5402, and a null mutant allele of fgf8, we establish an important role for FGF8 in posterior mesoderm, and show that the FGF and Nodal signaling pathways act synergistically to specify posterior mesoderm. The implications of these data for mesodermal morphogenesis are discussed.

46. The homeobox genes vox and vent are redundant repressors of dorsal fates in zebrafish.

Abstract

Ventralizing transcriptional repressors in the Vox/Vent family have been proposed to be important regulators of dorsoventral patterning in the early embryo. While the zebrafish genes vox (vega1) and vent (vega2) both have ventralizing activity in overexpression assays, loss-of-function studies are needed to determine whether these genes have distinct or redundant functions in dorsoventral patterning and to provide critical tests of the proposed regulatory interactions among vox, vent and other genes that act to establish the dorsoventral axis. We show that vox and vent are redundant repressors of dorsal fates in zebrafish. Mutants that lack vox function have little or no dorsoventral patterning defect, and inactivation of either vox or vent by injection of antisense morpholino oligonucleotides has little or no effect on the embryo. In contrast, embryos that lack both vox and vent function have a dorsalized phenotype. Expression of dorsal mesodermal genes, including chordin, goosecoid and bozozok, is strongly expanded in embryos that lack vox and vent function, indicating that the redundant action of vox and vent is required to restrict dorsal genes to their appropriate territories. Our genetic analysis indicates that the dorsalizing transcription factor Bozozok promotes dorsal fates indirectly, by antagonizing the expression of vox and vent. In turn, vox and vent repress chordin expression, restricting its function as an antagonist of ventral fates to the dorsal side of the embryo. Our results support a model in which BMP signaling induces the expression of ventral genes, while vox and vent act redundantly to prevent the expression of chordin, goosecoid and other dorsal genes in the lateral and ventral mesendoderm.

48. A Conserved Role for H15-Related T-Box Transcription Factors in Zebrafish and DrosophilaHeart Formation

Abstract

T-box transcription factors are critical regulators of early embryonic development. We have characterized a novel zebrafish T-box transcription factor, hrT(H15-related T box) that is a close relative of Drosophila H15and a recently identified human gene. We show that Drosophila H15and zebrafish hrTare both expressed early during heart formation, in strong support of previous work postulating that vertebrate and arthropod hearts are homologous structures with conserved regulatory mechanisms. The timing and regulation of zebrafish hrTexpression in anterior lateral plate mesoderm suggest a very early role for hrTin the differentiation of the cardiac precursors. hrTis coexpressed with gata4and nkx2.5not only in anterior lateral plate mesoderm but also in noncardiac mesoderm adjacent to the tail bud, suggesting that a conserved regulatory pathway links expression of these three genes in cardiac and noncardiac tissues. Finally, we analyzed hrTexpression in pandoramutant embryos, since these have defects in many of the tissues that express hrT,including the heart. hrTexpression is much reduced in the early heart fields of pandoramutants, whereas it is ectopically expressed subsequently. Using hrTexpression as a marker, we describe a midline patterning defect in pandoraaffecting the anterior hindbrain and associated midline mesendodermal derivatives. We discuss the possibility that the cardiac ventricular defect previously described in pandoraand the midline defects described here are related.

49. Expression of the human fibroblast interferon gene in Escherichia coli.

Abstract

We applied the method of Guarente et al. [Guarente, L., Lauer, G., Roberts, T.M. & Ptashne, M. (1980) Cell 20, 543-553] to construct plasmids that direct expression in Escherichia coli of the human fibroblast interferon (F-IF) gene. Two plasmids were recovered. One directs efficient synthesis of a protein whose primary sequence is that of pre-F-IF and the other, that of mature F-IF. Extracts of bacteria synthesizing mature F-IF display antiviral activity characteristic of human F-IF. This activity is lower than that expected from the differential rate of synthesis of the protein. We have detected no such activity in extracts of bacteria synthesizing pre-F-IF.
