Federated Learning (FL) is a privacy-preserving distributed machine learning approach geared towards applications in edge devices. However, the problem of designing custom neural architectures in federated environments is not tackled from the perspective of overall system efficiency. In this paper, we propose DC-NAS—a divide-and-conquer approach that performs supernet-based Neural Architecture Search (NAS) in a federated system by systematically sampling the search space. We propose a novel diversified sampling strategy that balances exploration and exploitation of the search space by initially maximizing the distance between the samples and progressively shrinking this distance as the training progresses. We then perform channel pruning to reduce the training complexity at the devices further. We show that our approach outperforms several sampling strategies including Hadamard sampling, where the samples are maximally separated. We evaluate our method on the CIFAR10, CIFAR100, EMNIST, and TinyImagenet benchmarks and show a comprehensive analysis of different aspects of federated learning such as scalability, and non-IID data. Topical delivery to treat dermatological disease is constrained by low skin permeability to most drugs due to the stratum corneum barrier. STAR particles containing microneedle protrusions can be topically applied on the skin to create micropores that dramatically increase skin permeability, even to water‐soluble compounds and macromolecules. This study addresses the tolerability, acceptability, and reproducibility of STAR particles rubbed on the skin at multiple pressures and after multiple applications to human subjects. One‐time STAR particle application at pressures between 40 and 80 kPa showed that skin microporation and erythema directly correlated with increased pressure, and 83% of subjects reported STAR particles to be comfortable at all pressures. Repeated application of STAR particles for 10 consecutive days at 80 kPa showed that skin microporation (~0.5% of skin area), erythema (low‐to‐moderate), and comfort with self‐administration (75%) were similar over the course of the study. Comfort of sensations associated with STAR particles increased from 58% to 71% during the study, and familiarity with STAR particles increased from 12.5% to 50% of subjects reporting STAR particle application not feeling different from other skin products. This study demonstrates that topically applied STAR particles were well tolerated and highly acceptable after application at various pressures and repeated daily use. The field of Active Domain Adaptation (ADA) has been investigating ways to close the performance gap between supervised and unsupervised learning settings. Previous ADA research has primarily focused on query selection, but there has been little examination of how to effectively train newly labeled target samples using both labeled source samples and unlabeled target samples. In this study, we present a novel Transferable Loss-based ADA (TL-ADA) framework. Our approach is inspired by loss-based query selection, which has shown promising results in active learning. However, directly applying loss-based query selection to the ADA scenario leads to a buildup of high-loss samples that do not contribute to the model due to transferability issues and low diversity. To address these challenges, we propose a transferable doubly nested loss, which incorporates target pseudo labels and a domain adversarial loss. Our TL-ADA framework trains the model sequentially, considering both the domain type (source/target) and the availability of labels (labeled/unlabeled). Additionally, we encourage the pseudo labels to have low self-entropy and diverse class distributions to improve their reliability. Experiments on several benchmark datasets demonstrate that our TL-ADA model outperforms previous ADA methods, and in-depth analysis supports the effectiveness of our proposed approach. Due to increasing interest in adapting models on resource-constrained edges, parameter-efficient transfer learning has been widely explored. Among various methods, Visual Prompt Tuning (VPT), prepending learnable prompts to input space, shows competitive fine-tuning performance compared to training of full network parameters. However, VPT increases the number of input tokens, resulting in additional computational overhead. In this paper, we analyze the impact of the number of prompts on fine-tuning performance and self-attention operation in a vision transformer architecture. Through theoretical and empirical analysis we show that adding more prompts does not lead to linear performance improvement. Further, we propose a Prompt Condensation (PC) technique that aims to prevent performance degradation from using a small number of prompts. We validate our methods on FGVC and VTAB-1k tasks and show that our approach reduces the number of prompts by ∼ 70% while maintaining accuracy. [ABSTRACT FROM AUTHOR]

This article tackles Partial Domain Adaptation (PDA) where the target label set is a subset of the source label set. A key challenging issue in PDA is to prevent negative transfer by isolating source-private classes. Since there is no label information for a target domain, PDA methods require to estimate a label commonness score between source and target domains. Existing approaches use either class-level or sample-level commonness to alleviate the negative transfer issue. However, class-level methods assign the same label commonness to all samples of the same class without considering each sample’s characteristics. Also, the recently introduced sample-level approaches show better performance but they still suffer from negative transfer due to non-trivial anomaly samples. To address these limitations, we propose Adaptive Graph Adversarial Networks (AGAN) consisting of two specialized modules. The adaptive class-relational graph module is designed to utilize the intra- and inter-domain structures through adaptive feature propagation. Complementarily, the sample-level commonness predictor computes a commonness score of each sample. Spiking Neural Networks (SNNs) have recently emerged as a new generation of low-power deep neural networks due to sparse, asynchronous, and binary event-driven processing. Most previous deep SNN optimization methods focus on static datasets (e.g., MNIST) from a conventional frame-based camera. On the other hand, optimization techniques for event data from Dynamic Vision Sensor (DVS) cameras are still at infancy. Most prior SNN techniques handling DVS data are limited to shallow networks and thus, show low performance. Generally, we observe that the integrate-and-fire behavior of spiking neurons diminishes spike activity in deeper layers. The sparse spike activity results in a sub-optimal solution during training (i. e., performance degradation). To address this limitation, we propose novel algorithmic and architectural advances to accelerate the training of very deep SNNs on DVS data. Specifically, we propose Spike Activation Lift Training (SALT) which increases spike activity across all layers by optimizing both weights and thresholds in convolutional layers. After applying SALT, we train the weights based on the cross-entropy loss. SALT helps the networks to convey ample information across all layers during training and therefore improves the performance. Furthermore, we propose a simple and effective architecture, called Switched-BN, which exploits Batch Normalization (BN). Previous methods show that the standard BN is incompatible with the temporal dynamics of SNNs. Therefore, in Switched-BN architecture, we apply BN to the last layer of an SNN after accumulating all the spikes from previous layer with a spike voltage accumulator (i. e., converting temporal spike information to float value). Even though we apply BN in just one layer of SNNs, our results demonstrate a considerable performance gain without any significant computational overhead. Through extensive experiments, we show the effectiveness of SALT and Switched-BN for training very deep SNNs from scratch on various benchmarks including, DVS-Cifar10, N-Caltech, DHP19, CIFAR10, and CIFAR100. By emulating biological features in brain, Spiking Neural Networks (SNNs) offer an energy-efficient alternative to conventional deep learning. To make SNNs ubiquitous, a 'visual explanation' technique for analysing and explaining the internal spike behavior of such temporal deep SNNs is crucial. Explaining SNNs visually will make the network more transparent giving the end-user a tool to understand how SNNs make temporal predictions and why they make a certain decision. In this paper, we propose a bio-plausible visual explanation tool for SNNs, called Spike Activation Map (SAM). SAM yields a heatmap (i.e., localization map) corresponding to each time-step of input data by highlighting neurons with short inter-spike interval activity. Interestingly, without the use of gradients and ground truth, SAM produces a temporal localization map highlighting the region of interest in an image attributed to an SNN's prediction at each time-step. Overall, SAM outsets the beginning of a new research area 'explainable neuromorphic computing' that will ultimately allow end-users to establish appropriate trust in predictions from SNNs. The extracellular vesicles (EVs) research area has grown rapidly because of their pivotal roles in intercellular communications and maintaining homeostasis of individual organism. As a subtype of EVs, exosomes are made via unique biogenesis pathway and exhibit disparate functional and phenotypic characteristics. Functionally, exosomes transfer biological messages from donor cell to recipient cell, which makes exosomes as a novel therapeutic platform delivering therapeutic materials to the target tissue/cell. Currently, both academia and industry try to develop exosome platform‐based therapeutics for disease management, some of which are already in clinical trials. In this review, we will discuss focusing on therapeutic values of exosomes, recent advances in therapeutic exosome platform development, and late development of exosome therapeutics in diverse therapeutic areas. Various methods have been developed to stabilize DNA origami against low concentrations of divalent cations and the presence of nucleases. However, existing strategies typically require the complete encapsulation of nanostructures, which makes accessing the encased DNA strands difficult, or chemical modification, such as covalent crosslinking of DNA strands. We present a stabilization method involving the synthesis of DNA brick nanostructures with dendritic oligonucleotides attached to the outer surface. We find that nanostructures assembled from DNA brick motifs remain stable against denaturation without any chemical modifications. Understanding the spatiotemporal variability of extreme hydrologic events, especially those driven by typhoons over Asia, and their controlling factors is essential to testing and predicting their future changes under global warming. Here, we present a series of direct heavy-rainfall records over the past ca. 10,000 years based on lake sediments from a volcanic cone (Sara Oreum) on Jeju Island, Korea, where orographic amplification of precipitation is dominant and sensitive to present typhoon activity. The reconstructed heavy rainfall index based on the high-resolution elemental ratios (e.g., Ti/Al and Sr/Si) shows clear variability at centennial timescales and is similar to records of coastal flooding events along the southern coast of Korea, typhoon-driven overwash events along the south-western coast of Japan, indicating strong typhoon influence on the heavy rainfall events in Jeju Island during the Holocene. Furthermore, this heavy rainfall-typhoon linkage has been influenced by past El Niño–Southern Oscillation (ENSO) activity and sea surface temperature (SST) in the western Tropical Pacific, showing time-varying influences. This study suggests that on decadal to multi-centennial timescales, stronger ENSO activity is responsible for eastward shift in the location of typhoon genesis, resulting in more frequent recurving tracks of typhoon and increased landfalls in Korea and Japan, whereas SST affects the intensification rate of typhoon. Coastal evolution is influenced by past sea-level changes and resultant shifts from fluvial- to marine-dominant environments and the accompanying significant geochemical and isotopic changes in the water mass and sediments. We investigated the elemental and isotopic features of coastal sedimentary cores (27 m in length) from a small paleo-bay located on the southern coast of Korea to determine such geochemical variability and specify past changes in the bay environment and anoxic conditions and possible links to past climate changes. We analyzed total organic carbon (TOC), total sulfur (TS), their isotopes (δ13CTOC and δ34STS), and pyrite. The δ13CTOC values ranging from −25 to −19‰ (a proxy for terrestrial influence) were lower than average (−22.5‰) before 8300 cal yr BP and since 500 cal yr BP, while the intervening Early to Late Holocene showed higher δ13CTOC values, indicating a shallow coastal environment. The δ34STS values fluctuating between −35 and +5‰ resembled sedimentation rate change. Based on the changes in the ratios of TOC to TS (C/S ratios), sedimentation rate, and δ34STS, we found five possible periods with higher salinity and intensified anoxic conditions at millennial timescales: 8900–8200, 7950–6500, 5200–4300, 3500–2600, and 2000–1100 cal yr BP. These intensified anoxic conditions seem to have been influenced by increased air temperature and sea-surface temperature conditions, which could have intensified the intensity of thermal stratification (less ventilation and mixing) between surface and bottom waters and resultant anoxic conditions. Abstract: We synthesized five maleic acid amide derivatives (maleic, citraconic, cis-aconitic, 2-(2′-carboxyethyl) maleic, 1-methyl-2-(2′-carboxyethyl) maleic acid amide), and compared their degradability for the future development of pH-sensitive biomaterials with tailored kinetics of the release of drugs, the change of charge density, and the degradation of scaffolds. The degradation kinetics was highly dependent upon the substituents on the cis-double bond. Among the maleic acid amide derivatives, 2-(2′-carboxyethyl) maleic acid amide with one carboxyethyl and one hydrogen substituent showed appropriate degradability at weakly acidic pH, and the additional carboxyl group can be used as a pH-sensitive linker. Abstract: Eosinophilia is one of the characteristic signs of allergic inflammation. Massive migration of eosinophils to the airways can cause epithelial tissue injury, contraction of airway smooth muscle and increased bronchial responsiveness. Previously, we discovered a new compound, 1H,8H-pyrano[3,4-c]pyran-1,8-dione (PPY), derived from the fruit of Vitex rotundifolia L. and evaluated its anti-inflammatory and anti-asthmatic properties. In this study, we synthesized a new modified compound, 4-acetyl-3-methyl-6-(3,4,5-trimethoxyphenyl) pyrano[3,4-c]pyran-1,8-dione (PPY-345), which was based on the PPY skeleton, and we evaluated its anti-asthmatic effects. To evaluate the anti-asthmatic effect of PPY-345 in vitro, A549 lung epithelial cells were stimulated with TNF-alpha, IL-4 and IL-1-beta to induce the expression of CCL11 (Eotaxin), a chemokine involved in eosinophil chemotaxis. To characterize the anti-asthmatic properties of PPY-345 in vivo, we examined the influence of PPY-345 in an ovalbumin (OVA)-induced asthma model. PPY-345 treatments significantly reduced CCL11 secretion. PPY-345 treatment did not inhibit the translocation of NF-κB into the nucleus but suppressed the phosphorylation of signal transducers and activators of transcription 6 (STAT6). PPY-345 treatment significantly reduced airway hyperreactivity as measured by whole-body plethysmography. PPY-345 further reduced total cells, including eosinophil, macrophage and lymphocytes, in the BAL fluid, goblet cell hyperplasia and myosin light chain 2 positive smooth muscle cell area in the lung tissue. Additionally, PPY-345 significantly suppressed the levels of OVA–IgE present in the serum. These results suggested that PPY-345 could improve asthma symptoms in OVA-sensitized mice. A traditional medicine from Vitex rotundifolia ameliorates asthma symptoms.Vitex rotundifolia L. (VR) as long been used in China and Korea in traditional medicine. This study was conducted to evaluate the ability of Vitex rotundifolia L. to prevent airway inflammation and remodeling in an ovalbumin (OVA)-induced murine asthma model. The total cell number and number of inflammatory cells in the bronchoalveolar lavage (BAL) fluid were counted. The levels of cytokines in the BAL fluid and serum IgE levels were measured using an ELISA. For histological analysis, hematoxylin and eosin staining, periodic acid-Schiff staining and immunohistochemistry were evaluated. The release of total cells into the BAL fluid was significantly inhibited in OVA-induced asthmatic mice treated with VR extract. In addition, eosinophilia and lymphocytosis were reduced significantly in mice that received VR extract. Furthermore, levels of the Th2 cytokines IL-4 and IL-5 and pro-inflammatory cytokine TNF-α in the BAL fluid and total IgE in serum were markedly suppressed by VR extract. OVA-specific IgE in the serum and IL-13 in the BAL fluid were decreased, but not significantly. The allergic effects of VR extract were accompanied by a reduction in airway hyperresponsiveness. Additionally, morphologic findings demonstrated that VR extract substantially inhibited OVA-induced eosinophilia, goblet cell hyperplasia and smooth muscle mass production. This finding suggests that VR extract may have pharmacological effects that would be useful for the treatment of asthma via the inhibition of the Th2 response and airway remodeling. In this era of IoT, energy-efficient and adversarially secure implementation of deep neural networks (DNNs) on hardware has become imperative. Memristive crossbars have emerged as an energy-efficient component of deep learning hardware accelerators due to their compact and efficient matrix-vector multiplication (MVM) implementation. However, they suffer from nonidealities (such as, interconnect parasitics, device variations, and sneak paths) introduced by their circuit topology that degrades computational accuracy. A 1T-1R synapse, adding a transistor (1T) in series with the memristive synapse (1R), has been proposed to mitigate sneak paths in a crossbar. However, we observe that the nonlinear characteristics of the transistor affect the overall conductance of the 1T-1R cell which in turn affects the MVM operation. This 1T-1R nonlinearity arising from the input voltage-dependent nonlinearity is not only difficult to model or formulate, but also causes a drastic performance degradation of DNNs when mapped to such crossbars. In this article, we first analyses the nonlinearity in ideal 1T-1R crossbars (excluding nonidealities, such as device variations and interconnect parasitics) and propose a novel nonlinearity aware training (NEAT) method to address the nonlinearities. Specifically, we first identify the range of network weights, which can be mapped into the 1T-1R cell within the linear operating region of the transistor. After that, we regularize the weights of neural networks to exist within the linear operating range by using an iterative training algorithm. Our iterative training significantly recovers the classification accuracy drop caused by the nonlinearity. Moreover, we find that each layer has a different weight distribution and in turn requires different gate voltage of transistor to guarantee linear operation. Based on this observation, we achieve energy efficiency while preserving classification accuracy by applying heterogeneous gate-voltage control to the 1T-1R cells across different layers. Finally, we conduct various experiments on CIFAR10 and CIFAR100 benchmark datasets to demonstrate the effectiveness of our NEAT. Overall, NEAT yields $\sim 20\%$ energy gain with less than 1% accuracy loss (with homogeneous gate control) when mapping ResNet18 networks on 1T-1R crossbars. Thereafter, we integrate the 1T-1R crossbars with various nonidealities. We show that NEAT leads to more adversarially robust mappings of DNNs onto nonideal 1T-1R crossbars than standard DNNs mapped directly onto 1R crossbars. In case of a VGG11 network on CIFAR100 dataset, we obtain $\sim 17$ % improvement in clean accuracy and $\sim 2$ %–8% & $\sim 5$ %–6% improvements in adversarial accuracies, respectively, for fast gradient sign method (FGSM) and projected gradient descent (PGD)-based adversarial attacks via NEAT on nonideal 64 $\times $ 64 crossbars, in comparison to standard DNNs. In East Asia, freshwater inputs in coastal areas are mainly controlled by rainfall occurring during the East Asian summer monsoon (EASM), which is linked to seasonal latitudinal shifts in the intertropical convergence zone (ITCZ). To investigate the centennial-time-scale relationship between those inputs and the seasonal shifts in the ITCZ, we examined the behaviors of terrestrial input proxies in coastal sediments of western Korea, including grain size, elemental ratios from XRF core scanning (e.g., Zr/Ti and Si/Al), and C/N ratios. Correlation tests linked these proxies to past freshwater input variability, suggesting that elemental ratios can serve as a proxy for past EASM changes. During the period 8500–7800 cal yr BP, the stronger EASMs described by high-resolution peaks of Zr/Ti ratios corresponded to periods of intensified Indian summer monsoons (ISMs) and weakened South American summer monsoon (SASMs), supporting the existence of synchronous global monsoon (GM) events induced by a northward ITCZ shift. This study also shows that the 8.2-ka cooling event recorded in Greenland ice cores synchronously influenced the GM through a southward shift in the ITCZ, resulting in a weakened EASM and ISM in the Northern Hemisphere and an intensified SASM in the Southern Hemisphere. The ongoing pandemic and increasing frequency of infectious disease outbreaks due to climate change, urbanization, and global human migration have put great focus on nucleic acid amplification‐based diagnostic technologies (NAAT). These methods can provide gold standard accuracy and sensitivity, and their widespread availability at the point‐of‐care is crucial for managing the spread of pathogens in human and animal populations, as well as for environmental surveillance. However, so far the reach of NAAT‐based platforms has mostly remained limited to centralized facilities in the hand of trained workers, causing great distress in times of need. In this review, the current state‐of‐the‐art research, as well as, commercial diagnostic products, is highlighted, their performances are discussed, and the role of academic‐industrial collaborations in developing the next generation of breakthrough technologies is emphasized. It is envisioned that with these collaborations, the next generation of autonomous, affordable, and mobile NAAT devices can be developed, for a broad range of targets, and for providing truly democratized access to NAAT diagnostics for the global population in the future. Understanding the natural variability of the East Asian monsoon (EAM) system, mainly consisting of summer and winter monsoons, is essential to predict future climate changes that will influence agricultural and industrial activities in the highly populated region. To date, the variability of the East Asian winter monsoon (EAWM) and its controlling factors is still unclear in downwind areas that experience the Siberian High, mostly due to the lack of a high-resolution winter monsoon index. Based on the correlation between diatom variations in a volcanic cone of Jeju Island, South Korea, with past regional climate changes (e.g., temperature, dust, and sea ice) and solar activity, in this study, we evaluated the changes in the climate from the sum of acidophilous species (SAS), known to be mainly controlled by the pH of the prevalent water pool, and inferred that the SAS increased during the cold and dry periods (e.g., Little Ice Age and Dark Age Cold Period). Furthermore, millennial-timescale fluctuation in the detrended SAS during the past 8000 years, with peaks at ~500, 1500, 3000, 5600, 6600, and 7600 cal yr B·P., corresponded to the ice-rafted events reported from studies conducted on subpolar ocean sediments and solar activity. We developed a new pH-sensitive drug delivery carrier based on β-cyclodextrin (β-CD) and 1-methyl-2-(2′-carboxyethyl) maleic anhydrides (MCM). The primary hydroxyl groups of β-CD were successfully attached to MCM residues to produce a medusa-like β-CD-MCM. The MCM residue was conjugated with cephradine (CP) with high efficiency ( > 90%). More importantly, β-CD-MCM-CP responded to the small pH drop from 7.4 to 5.5 and released greater than 80% of the drugs within 0.5 h at pH 5.5. In addition, the inclusion complex between β-CD-MCM-CP and the adamantane derivative was formed by simple mixing to show the possibility of introducing multi-functionality. Large herbivores can disperse seeds over long distances through endozoochory. The Korean water deer (Hydropotes inermis argyropus), an internationally vulnerable species but locally considered a vermin, is a potential endozoochorous seed dispersal vector. In this study, feeding experiments were conducted to test the efficiency of seed dispersal through gut ingestion by the Korean water deer, its temporal pattern and the effect of gut passage on seed recovery and germination rate. Eight plant species, including species that formerly germinated from its faeces, were used to feed three Korean water deer. Once the deer had consumed all the provided seeds, their faeces were collected after 24, 48, 72 and 96 h. The collected faeces were air-dried, and the number of seeds retrieved from the faeces was counted every 24 h (0–24, 24–48, 48–72 and 72–96 h). Among the eight plant species, six species were retrieved with intact seeds. Panicum bisulcatum had the highest recovery rate of 33.7%, followed by Amaranthus mangostanus (24.5%) and Chenopodium album (14.4%). Most of the seeds were recovered within the 24–48 h time interval. Germination tests were conducted on the ingested and uningested seeds for the four species which had a sufficient recovery rate. The effects of gut passage on seed germination differed according to plant species. The germination rate substantially decreased after gut passage. The results suggest that the Korean water deer can disperse seeds, potentially over long distances albeit at a high cost of low seed recovery and germination rate. [ABSTRACT FROM AUTHOR]

Abstract: Ethnopharmacological relevance: Schisandra chinensis Baillon (Sc), an anti-inflammatory herb that has been used in traditional Chinese medicine for thousands of years, is frequently used to treat upper respiratory tract infections. Aim of the study: This study was conducted to evaluate the ability of a water extract of Sc to prevent airway inflammation both in vitro and in vivo. Materials and methods: Human lung alveolar epithelial-derived A549 cells were stimulated with to interleukin-1β, tumor necrosis factor-α, and interferon-γ (IL-1β, TNF-α, and INF-γ; cytokine mixture; CM) and treated with Sc extracts. They were then evaluated using nitric oxide (NO), IL-8 and monocyte chemotactic protein-1 (MCP-1) secretions. In the in vivo study, BALB/c mice were challenged with lipopolysaccharide (LPS) to induce acute airway inflammation. After this challenge, the mice were treated with Sc extracts (10, 50 and 100mg/kg) by oral administration, and inflammatory cells in the bronchoalveolar lavage (BAL) fluid were counted. IL-6 and TNF-α secretions were measured using an enzyme-linked immunosorbent assay. Lung tissues of the LPS treated mice were prepared and stained with hematoxylin and eosin (HE) for histological examination. Results: In the A549 cells, Sc extracts dose-dependently and significantly inhibited CM-induced NO production and reduced IL-8 and MCP-1 secretions. Sc extracts efficiently suppressed neutrophil and macrophage infiltrations of lung tissues and increased IL-6 and TNF-α levels in BAL fluid in LPS-instilled BALB/c mice. In addition, Sc extracts treatment inhibited pathologic progress in the lung tissues, as confirmed by H&E staining. These findings indicate that Sc extracts could be potentially useful for the treatment of acute lung inflammation and acute lung injury. [Copyright &y& Elsevier]

[Display omitted] Arsenic (As)-contaminated soil inevitably exists in nature and has become a global challenge for a sustainable future. Current processes for As capture using natural and structurally engineered nanomaterials are neither scientifically nor economically viable. Here, we established a feasible strategy to enhance As-capture efficiency and ecosystem health by structurally reorganizing iron oxyhydroxide, a natural As stabilizer. We propose crystallization to reorganize FeOOH-acetate nanoplatelets (r-FAN), which is universal for either scalable chemical synthesis or reproduction from natural iron oxyhydroxide phases. The r-FAN with wide interlayer spacing immobilizes As species through a synergistic mechanism of electrostatic intercalation and surface chemisorption. The r-FAN rehabilitates the ecological fitness of As-contaminated artificial and mine soils, as manifested by the integrated bioassay results of collembolan and plants. Our findings will serve as a cornerstone for crystallization-based material engineering for sustainable environmental applications and for understanding the interactions between soil, nanoparticles, and contaminants. In this study, LPS-induced inflammatory responses in BEAS-2B human bronchial epithelial cells and human umbilical vein endothelial cell (HUVEC)s were found to be prevented by Dickkopf-1 (DKK-1), a secreted Wnt antagonist, and LGK974, a small molecular inhibitor of the Wnt secretion. LPS-induced IκB degradation and NF-κB nuclear translocation as well as the expressions of pro-inflammatory genes including IL-6, IL-8, TNF- α, IL-1β, MCP-1, MMP-9, COX-2 and iNOS, were all suppressed by DKK-1 and LGK974 in a dose-dependent manner. The suppressive effects of LGK974 on NF-κB, IκB, and pro-inflammatory gene expression were rescued by ectopic expression of β-catenin, suggesting that the anti-inflammatory activity of LGK974 is mediated by modulation of the Wnt/β-catenin pathway and not by unrelated side effects. When Wnt recombinant proteins were treated to cells, Wnt3a and Wnt5a significantly induced pro-inflammatory gene expressions, while Wnt7a and Wnt10b showed little effects. It was also found that Wnt3a and Wnt5a expressions were significantly induced by LPS treatment. Consistently, knockdown of Wnt3a and Wnt5a blocked LPS-induced inflammatory responses, while treatment of recombinant Wnt3a and Wnt5a proteins rescued the inhibition of inflammatory responses by LGK974. Malignant tumors develop through multiple steps of initiation and progression, and tumor initiation is of singular importance in tumor prevention, diagnosis, and treatment. However, the molecular mechanism whereby a signaling network of interacting pathways restrains proliferation in normal cells and prevents tumor initiation is still poorly understood. Here, we have reported that the Hippo, Wnt/β-catenin, and Notch pathways form an interacting network to maintain liver size and suppress hepatocellular carcinoma (HCC). Ablation of the mammalian Hippo kinases Mst1 and Mst2 in liver led to rapid HCC formation and activated Yes-associated protein/WW domain containing transcription regulator 1 (YAP/TAZ), STAT3, Wnt/β-catenin, and Notch signaling. Previous work has shown that abnormal activation of these downstream pathways can lead to HCC. Rigorous genetic experiments revealed that Notch signaling forms a positive feedback loop with the Hippo signaling effector YAP/TAZ to promote severe hepatomegaly and rapid HCC initiation and progression. Surprisingly, we found that Wnt/β-catenin signaling activation suppressed HCC formation by inhibiting the positive feedback loop between YAP/TAZ and Notch signaling. Furthermore, we found that STAT3 in hepatocytes is dispensable for HCC formation when mammalian sterile 20-like kinase 1 and 2 (Mst1 and Mst2) were removed. The molecular network we have identified provides insights into HCC molecular classifications and therapeutic developments for the treatment of liver tumors caused by distinct genetic mutations.
