Abstract

Overlimiting current (OLC) through electrolytes interfaced with perm-selective membranes has been extensively researched in recent years for understanding the fundamental mechanisms of transport and developing efficient applications from electrochemistry to sample analysis and separation. Predominant mechanisms responsible for OLC include surface conduction, convection by electro-osmotic flow, and electro-osmotic instability depending on input parameters such as surface charge and geometric constrictions. This work studies how a network of microchannels in a non-uniform array, which mimicks a natural pore configuration, can contribute to OLC. To this end, micro/nanofluidic devices are fabricated with arrays of parallel microchannels with non-uniform size distributions. All cases maintain the same surface and bulk conduction to allow probing the sensitivity only by the non-uniformity of the channels. Both experimental and theoretical current-voltage relations demonstrate that OLCs increase with increasing non-uniformity. Furthermore, the visualization of internal recirculating flows indicates that the non-uniform arrays induce flow loops across the network enhancing advective transport. These evidences confirm a new driving mechanism of OLC, inspired by natural micro/nanoporous materials with random geometric structure. Therefore, this result can advance not only the fundamental understanding of nanoelectrokinetics but also the design rule of engineering applications of electrochemical membrane.

29. The Economic Impact of the Logistics Industry in South Korea: Based on an Input-Output Approach

Abstract

Using input-output analysis, this study investigates the economic impact of the logistics industry and its four sub-sectors: land freight, water freight, air freight and logistics facilities. The inter-industry linkage model shows that land freight and logistics facilities have large pull effects on the economy. Estimates from the production-inducing model indicate that investments in the logistics industry have considerable effects on both upstream and downstream industries. The employment-inducing model suggests that although employment-inducing effects have fallen over time, total induced employment has risen, particularly for permanent jobs. Lastly, estimates from the supply-shortage model indicate large effects for land freight which highlights the need for greater diversity in transportation modes. The findings and their implications may be useful to formulate effective policies in Korea.

Abstract

Protein-based biosensors or activators have been engineered to visualize molecular signals or manipulate cellular functions. Here we integrate these two functionalities into one protein molecule, an integrated sensing and activating protein (iSNAP). A prototype that can detect tyrosine phosphorylation and immediately activate auto-inhibited Shp2 phosphatase, Shp2-iSNAP, is designed through modular assembly. When Shp2-iSNAP is fused to the SIRPα receptor which typically transduces anti-phagocytic signals from the 'don't eat me' CD47 ligand through negative Shp1 signaling, the engineered macrophages not only allow visualization of SIRPα phosphorylation upon CD47 engagement but also rewire the CD47-SIRPα axis into the positive Shp2 signaling, which enhances phagocytosis of opsonized tumor cells. A second SIRPα Syk-iSNAP with redesigned sensor and activator modules can likewise rewire the CD47-SIRPα axis to the pro-phagocytic Syk kinase activation. Thus, our approach can be extended to execute a broad range of sensing and automated reprogramming actions for directed therapeutics.Protein-based biosensors have been engineered to interrogate cellular signaling and manipulate function. Here the authors demonstrate iSNAP, a tool to detect tyrosine phosphorylation and activate desired protein enzymes allowing the control of phagocytosis in macrophages.
