Abstract

To study the increased resistance in fish against Vibrio vulnificus known as an important agent of vibrio septicemia in human, we analyzed specific and nonspecific immune response in flounder after administration of V. vulnificus bacterins by oral route. It contained the comparison of antibody concentrations in the sera of flounder after oral administration by two different protocols with uncoated heat killed bacterin of V vulnificus (UHKB, 20 mg/kg body weight), i.e., 4 weeks continuously (group 4W) and taking 2 weeks resting period between the 1st and last week of administration (group 1-2-1W). Even though, 1-2-1W group showed significantly increased level of specific antibody in serum, it did not reach to that of 4W group. Certainly, flounder vaccinated twice a week for four weeks (20 mg/kg b.w.) showed increased concentration of specific antibody against V. vulnificus at week 2 after last administration by oral route and maintained throughout the experimental period. It also was confirmed by the increased numbers of specific antibody secreting cells (SASC) in the leukocytes isolated from the splenocytes of the flounder of 4W group at week 1 after last administration until the end of experimental period. However, enteric, acid-resistant film coated heat killed bacterin (ECHB) did not show both greater immune reaction for antibody production and faster elimination of a challenge dose of V. vulnificus compared with those of the UHKB. These results suggested that UHKB administered by oral route was very effective method to prevent the contamination of V vulnificus in flounder, and did not show the increased antigenicity by coating the surface with acid-resistant film.

Abstract

This study has assessed the effects of vitamin E (−E, +LE, +HE; 0, 100, 1000 mg/kg, respectively) in fish diets containing high levels ( HL; 10%) of fish oil ( FO) or mixed vegetable oils ( VO) on the growth and inflammatory, oxidative stress, and apoptotic gene expression in the head kidney of olive flounder, Paralichthys olivaceus. Consequently, the highest weight gain was achieved in the FO group and the lowest in the HL-VO + LE group. The gene expression levels of each group were compared to the 5% FO group. The 5% VO group showed higher expression levels of tumor necrosis factor ( TNF) α, interleukin (IL)-1β, and scinderin-like ( ScinL) genes. Although lysozyme gene expression was higher in the HL-VO + LE group, the other gene expression levels of the HL-FO/ VO + LE groups were not different from those of the FO group. The HL-FO/ VO−E/+ HE groups showed a higher TNFα gene expression, but the cytochrome oxidase subunit III gene expression was higher in the HL-FO−E and HL-VO + HE groups. Lysozyme gene expression was higher in the HL-FO−E and HL-VO−E/+ HE groups. IL-6 and ScinL gene expression were higher in the HL-VO-E and HL-VO + HE groups, respectively. In conclusion, mixed VO and too high or too low vitamin E levels in fish diets may affect inflammatory, oxidative stress, and apoptotic gene expression in the head kidney of olive flounder. 44. Isolation and Structural Determination of the Antifouling Diketopiperazines from Marine-Derived Streptomyces praecox291-11

Abstract

Marine derived actinomycetes constituting 185 strains were screened for their antifouling activity against the marine seaweed, Ulva pertusa, and fouling diatom, Navicula annexa. Strain 291-11 isolated from the seaweed, Undaria pinnatifida, rhizosphere showed the highest antifouling activity and was identified as Streptomyces praecoxbased on a 16S rDNA sequence analysis. Strain 291-11 was therefore named S. praecox291-11. The antifouling compounds from S. praecox291-11 were isolated, and their structures were analyzed. The chemical constituents representing the antifouling activity were identified as (6S,3S)-6-benzyl-3-methyl-2,5-diketopiperazine (bmDKP) and (6S,3S)-6-isobutyl-3-methyl-2,5-diketopiperazine (imDKP) by interpreting the nuclear magnetic resonance and high-resolution mass spectroscopy data. Approximately 4.8 mg of bmDKP and 3.1 mg of imDKP were isolated from 1.2 g of the S. praecox291-11 crude extract. Eight different compositions of culture media were investigated for culture, the TBFeC medium being best for bmDKP and TCGC being the optimum for imDKP production. Two compounds respectively showed a 17.7 and 21 therapeutic ratio (LC50/EC50) to inhibit zoospores, and two compounds respectively showed a 263 and 120.2 therapeutic ratio to inhibit diatoms.

Abstract

An active antifouling diterpene was isolated from marine actinomycete strain PK209 and productivity was induced in a co-culture experiment. The active constituent was identified as the diterpene lobocompactol by interpretion of nuclear magnetic resonance and mass spectroscopy data. A PK209 co-culture was designed and a lobocompactol-resistant bacterium, KNS-16, was selected as co-culture competitor to induce lobocompactol production. Adding a small volume of 16-h-old KNS-16 culture to the 96-h-old PK209 culture caused rapid induction of lobocompactol production. The final yield was 2.7 mg/L, 10.4-fold higher than that collected from a single PK209 culture. The two bacteria, strains PK209 and KNS-16, were identified as Streptomyces cinnabarinus and Alteromonas sp. based on 16S rDNA sequencing. Lobocompactol showed significant antifouling activity, of 0.18 and 0.43 µg/mL, for EC₅₀ against the macroalga Ulva pertusa and the diatom Navicula annexa respectively. It showed activity with MIC of 61-112 µg/mL against fouling bacteria.

Abstract

The full length genes gyrB (2,415 bp), parC (2,277 bp), and parE (1,896 bp) in Edwardsiella tarda were cloned by PCR with degenerate primers based on the sequence of the respective quinolone resistance-determining region (QRDR), followed by elongation of 5' and 3' ends using cassette ligation-mediated PCR (CLMP). Analysis of the cloned genes revealed open reading frames (ORFs) encoding proteins of 804 (GyrB), 758 (ParC), and 631 (ParE) amino acids with conserved gyrase/topoisomerase features and motifs important for enzymatic function. The ORFs were preceded by putative promoters, ribosome binding sites, and inverted repeats with the potential to form cruciform structures for binding of DNA-binding proteins. When comparing the deduced amino acid sequences of E. tarda GyrB, ParC, and ParE with those of the corresponding proteins in other bacteria, they were found to be most closely related to Escherichia coli GyrB (87.6% identity), Klebsiella pneumoniae ParC (78.8% identity) and Salmonella typhimurium ParE (89.5% identity), respectively. The two topoisomerase genes, parC and parE, were found to be contiguous on the E. tarda chromosome. All 18 quinoloneresistant isolates obtained from Korea thus far did not contain subunit alternations apart from a substitution in GyrA (Ser83→Arg). However, an alteration in the QRDR of ParC (Ser84→Ile) following an amino acid substitution in GyrA (Asp87→Gly) was detected in E. tarda mutants selected in vitro at 8 microng/ml ciprofloxacin (CIP). A mutant with a GyrB (Ser464→Leu) and GyrA (Asp87→Gly) substitution did not show a significant increase in the minimum inhibitory concentration (MIC) of CIP. None of the in vitro mutants exhibited mutations in parE. Thus, gyrA and parC should be considered to be the primary and secondary targets, respectively, of quinolones in E. tarda.
