1. Quasi-ergodic theorems for Feynman-Kac semigroups and large deviation for additive functionals

Abstract

We study the long-time behavior of an additive functional that takes into account the jumps of a symmetric Markov process. This process is assumed to be observed through a biased observation scheme that includes the survival to events of extinction and the Feynman-Kac weight by another similar additive functional. Under conditioning for the convergence to a quasi-stationary distribution and for two-sided estimates of the Feynmac-Kac semigroup to be obtained, we shall discuss general assumptions on the symmetric Markov process. For the law of additive functionals, we will prove a quasi-ergodic theorem, namely a conditional version of the ergodic theorem and a conditional functional weak law of large numbers. Abstract

In this paper, when a given symmetric Markov process X satisfies the stability of global heat kernel two-sided (upper) estimates by Markov perturbations, we give a necessary and sufficient condition on the stability of global two-sided (upper) estimates for fundamental solution of Feynman-Kac semigroup of X. As a corollary, under the same assumptions, a weak type global two-sided (upper) estimates holds for the fundamental solution of Feynman-Kac semigroup with (extended) Kato class conditions for measures. This generalizes all known results on the stability of global integral kernel estimates by symmetric Feynman-Kac perturbations with Kato class conditions in the framework of symmetric Markov processes.

Abstract

In this paper we study the scattering length for positive additive functionals of symmetric stable processes on ${\bf R}^d$. The additive functionals considered here are not necessarily continuous. We prove that the semi-classical limit of the scattering length equals the capacity of the support of a certain measure potential, thus extend previous results for the case of positive continuous additive functionals. We also give an equivalent criterion for the fractional Laplacian with a measure valued non-local operator as a perturbation to have purely discrete spectrum in terms of the scattering length, by considering the connection between scattering length and the bottom of the spectrum of Schr\"odinger operator in our settings., Comment: To appear in Mathematische Nachrichten

The index is given by the Dirichlet forms of the Markov processes. 34. Vertical load-carrying behavior and design models for micropiles considering foundation configuration conditions

Abstract

In the present study, the vertical load-carrying behavior of micropile foundations with various configuration conditions was investigated based on results from model load tests. Considered configuration conditions included micropile inclination angle, spacing of micropiles, and types of micropile foundations. The ultimate load capacity of micropiles varied with installation angle, showing an initial increase and peak followed by a gradual decrease with further increase in installation angle. The ultimate load capacity of micropiled rafts was affected by both installation angle and micropile spacing. The load-carrying mechanism of micropiles for the inclined condition was proposed based on the decomposed axial and lateral load and resistance components. Using the proposed load-carrying mechanism and test results obtained in this study, the load capacity ratio for an inclined micropile was proposed. The group effect and interaction effect factors for group micropiles and micropiled rafts were proposed, respectively, all of which can be used to estimate the load-carrying capacity of micropile foundations. Field load tests were conducted and it was seen that estimated results using the proposed method were in good agreement with measured results. Additional comparison with case examples from the literature also confirmed the validity of the proposed method. Dans la presente etude, la charge verticale portant le comportement des fondations de micropieux avec diverses conditions de configuration ont ete etudiees en fonction des resultats des essais de charge modele. Les conditions de configuration prise en compte ont inclus des angles d'inclinaison des micropieux et l'espacement des micropieux et les types de fondations des micropieux. La capacite de charge ultime des micropieux varie avec l'angle d'installation montrant l'augmentation initiale et le pic suivi par une diminution progressive avec augmentation de l'angle d'installation. La capacite de charge ultime de radeaux a micropieux a ete affectee a la fois par l'angle d'installation et l'espacement des micropieux. Le mecanisme de soutien de transport de charge de micropieux pour la condition inclinee a ete proposee sur la base de la charge et de la resistance des composants axiaux et lateraux decomposes. L'utilisation du mecanisme soutenant la charge proposee et les resultats d'essais obtenus dans cette etude, le rapport de capacite de charge pour un micropieux inclinee a ete propose. L'effet de groupe et les facteurs d'effet d'interaction pour les groupes micropieux et les radeaux de micropieux ont ete proposes, respectivement, qui peut etre utilise pour estimer la capacite de charge des fondations de micropieux. Les essais de charge sur le terrain menes et les resultats estimes a l'aide de la methode proposee etaient en bon accord avec les resultats mesures. 42. STAT3 activation in large granular lymphocyte leukemia is associated with cytokine signaling and DNA hypermethylation

Abstract

Large granular lymphocyte leukemia (LGLL) is characterized by somatic gain-of-function STAT3mutations. However, the functional effects of STAT3mutations on primary LGLL cells have not been studied in detail. In this study, we show that CD8+ T cells isolated from STAT3mutated LGLL patients have high protein levels of epigenetic regulators, such as DNMT1, and are characterized by global hypermethylation. Correspondingly, treatment of healthy CD8+ T cells with IL-6, IL-15, and/or MCP-1 cytokines resulted in STAT3 activation, increased DNMT1, EZH2, c-MYC, l-MYC, MAX, and NFκB levels, increased DNA methylation, and increased oxidative stress. Similar results were discovered in KAI3 NK cells overexpressing gain-of-function STAT3Y640Fand STAT3G618Rmutants compared to KAI3 NK cells overexpressing STAT3WT. Our results also confirm that STAT3 forms a direct complex with DNMT1, EZH2, and HDAC1. In STAT3mutated LGLL cells, DNA methyltransferase (DNMT) inhibitor azacitidine abrogated the activation of STAT3 via restored SHP1 expression. In conclusion, STAT3mutations cause DNA hypermethylation resulting in sensitivity to DNMT inhibitors, which could be considered as a novel treatment option for LGLL patients with resistance to standard treatments.

Abstract

Interleukin-23 (IL-23) is a proinflammatory cytokine mainly produced by myeloid cells that promotes tumor growth in various preclinical cancer models and correlates with adverse outcomes. However, as to how IL-23 fuels tumor growth is unclear. Here, we found tumor-associated macrophages to be the main source of IL-23 in mouse and human tumor microenvironments. Among IL-23-sensing cells, we identified a subset of tumor-infiltrating regulatory T (Treg) cells that display a highly suppressive phenotype across mouse and human tumors. The use of three preclinical models of solid cancer in combination with genetic ablation of Il23rin Tregcells revealed that they are responsible for the tumor-promoting effect of IL-23. Mechanistically, we found that IL-23 sensing represents a crucial signal driving the maintenance and stabilization of effector Tregcells involving the transcription factor Foxp3. Our data support that targeting the IL-23/IL-23R axis in cancer may represent a means of eliciting antitumor immunity.

Abstract

We study criticalities of generalized Schrödinger operators in terms of the Schrödinger forms induced by generalized Feynman-Kac perturbations of symmetric Markov processes, which extend earlier work due to Takeda [26]. The related functional inequalities and analytic characterizations of criticalities of Schrödinger forms are given. As applications, we establish some maximum principles via the analytic characterization.
