Abstract

Histamine H4 receptor (H4R) has immune-modulatory and chemotaxic effects in various immune cells. This study aimed to determine the osteoclastogenic role of H4R in rheumatoid arthritis (RA). The concentration of histamine in synovial fluid (SF) and sera in patients with RA was measured using ELISA. After RA SF and peripheral blood (PB) CD14+ monocytes were treated with histamine, IL-17, IL-21 and IL-22, and a H4R antagonist (JNJ7777120), the gene expression H4R and RANKL was determined by real-time PCR. Osteoclastogenesis was assessed by counting TRAP-positive multinucleated cells in PB CD14+ monocytes cultured with histamine, Th17 cytokines and JNJ7777120. SF and serum concentration of histamine was higher in RA, compared with osteoarthritis and healthy controls. The expression of H4R was increased in PB monocytes in RA patients. Histamine, IL-6, IL-17, IL-21 and IL-22 induced the expression of H4R in monocytes. Histamine, IL-17, and IL-22 stimulated RANKL expression in RA monocytes and JNJ7777120 reduced the RANKL expression. Histamine and Th17 cytokines induced the osteoclast differentiation from monocytes and JNJ7777120 decreased the osteoclastogenesis. H4R mediates RANKL expression and osteoclast differentiation induced by histamine and Th17 cytokines. 19. Mechanism for Site-Selective Hydroboration of C70Fullerene with Borane by DFT-D3 Study

Abstract

We studied the hydroboration of the C70fullerene using both B3LYP-D3(BJ)/6-311G(d,p) and M06-2X-D3/6-311G(d,p) levels of theory, incorporating the empirical dispersion interaction, and Fukui index calculations. Potential energy surfaces (PESs) and Gibbs free energy surfaces (GFESs) were calculated for the pathways from four BH3adducts (located at the AB, CC, D, and Esites) on the C70to eight products formed by the 1,2-addition of BH3across the four [6,6]-ring fused bonds (AB, CC, DE, and EE) and across the two [5,6]-ring fused bonds (AAand DD). These pathways are two-step consecutive reactions. We denoted the positions on the fullerene cage as Athrough E, from the pole to the equator, based on the D5hsymmetry of the C70fullerene. In the first step reaction, the product ratios for the four adduct intermediates should be as the primary intermediate BH3(D), the secondary intermediate BH3(AB), the tertiary intermediate BH3(CC), and the minor intermediate BH3(E), based on the Fukui indices. In addition, in the second step reaction, transition states (TSs) from four adduct intermediates to eight product isomers, namely, BH2(A)H (B) to BH2(E)H (E), were obtained using the QST2 method. The calculated reaction coordinates showed exothermic reactions for all bonds except the EEbond. We also confirmed the transition states by frequency calculations and intrinsic reaction coordinate (IRC) analyses. The PESs and GFESs suggest spontaneous processes for the four isomers, of which the primary products are BH2(A)H (B) and its isomer BH2(B)H (A), the secondary product is BH2(C)H (C), and the tertiary product is BH2(D)H (D), all formed through adduct intermediates. Therefore, through the hydroboration reaction of C70, we could predict and design the site selectivity of C70by controlling the energy barrier of the transition state in the second step of the reaction. This implies that we could selectively synthesize mainly BH2(B)H (A) isomers across the AB-[6,6]-ring fused bond and also design BH2(D)H(D) isomers across the DD-[5,6]-ring fused bond. Abstract

Microcystes (MC) produced by several cyanobacteria are potent hepatotoxins, whereas the toxicokinetics and subsequent detrimental effects of MC are not fully understood in marine bivalves. In this study, the accumulation and depuration pattern of MC-Leucine-Arginine (MC-LR) along with the antioxidant response were investigated in the gill tissue of Pacific oyster Crassostrea gigas and blue mussel Mytilus edulis exposed to 0.1, 1, 10, and 20 [micro]g [L.sup.-1] MC-LR for 7 days. Time- and concentration-dependent accumulation and depuration of MC-LR were observed in the gill tissue during the exposure and depuration phases. The values for maximum accumulation of MC-LR and malondialdehyde content were statistically significant (P < 0.05) at the end of exposure (day 7) to 20 [micro]g [L.sup.-1] MC-LR for both the bivalves, and at the beginning of the depuration period when M. edulis and C. gigas were exposed to 10 and 20 [micro]g [L.sup.-1], respectively. A significant level of glutathione (GSH) was detected in M. edulis in response to 20 [micro]g [L.sup.-1] MC-LR during the exposure and depuration periods, whereas no significant levels of GSH were detected in C. gigas. The MC-LR accumulation induced oxidative stress in the gill tissue through modulation of the activities of antioxidant enzymes, the antioxidant response being more significant in the mussel than in the oyster. The enzymatic activities of glutathione S-transferase, superoxide dismutase (SOD), glutathione peroxidase, and glutathione reductase (GR) increased significantly in M. edulis, whereas in C. gigas, significant increase in the activities of SOD, catalase, and GR were observed at the end of exposure (day 7) to 20 [micro]g [L.sup.-1] MC-LR. Similarly, during the depuration period, significant increases in the antioxidant activities were observed in M. edulis mostly at the beginning (day I and day 5) of exposure compared with the levels in C. gigas. The results obtained in this study will be useful in the understanding of MC-LR accumulation in bivalve gill tissues and the subsequent antioxidant defense response. 47. Role of vitamin E on bovine skeletal-muscle-derived cells from Korean native cattle under heat treatment.

Abstract

Our study aimed to evaluate the dualistic effect of heat stress on muscle differentiation at different temperatures, and whether vitamin E, a powerful antioxidant, could offset any negative effects, using bovine skeletal-muscle-derived cells (BSMCs) with myogenic properties. The BSMCs were extracted from the skeletal muscle of 30-mo-old Korean native cattle and subjected to myogenic differentiation under 3 heat exposure conditions: 37 °C (control; CON), 39 °C (mild heat stress; MHS), and 41 °C (severe heat stress; SHS) for 24 h with or without vitamin E treatment (NE or VE). After 24 h treatments, the cells were returned to 37 °C incubators and differentiated until day 6. On day 1, because of the heat exposure, the gene expression of MYOG was the highest in MHS (P  = 0.047), suggesting a promotive effect of mild heat stress on myogenic differentiation, while on day 6, compared with CON and MHS, MYOD (P  = 0.013) and MYOG (P  = 0.029) were the lowest in SHS. Vitamin E treatment also lowered MYOG (P  = 0.097), regardless of heat exposure. On day 1, HSPB1 (P  = 0.001) and HSP70 (P  < 0.001) were the highest in SHS, and an interaction between heat exposure and vitamin E treatment was found on day 6 (P  < 0.027). BCL-2 was also the highest on day 1 in SHS (P  = 0.05), and an interaction of heat exposure and vitamin E treatment was found on day 1 on BAX expression (P  = 0.038). For antioxidant genes, SOD1 (P  = 0.002) and GPX1 (P  < 0.001) were affected by heat exposure, with the highest levels being observed in SHS, and on day 6, GPX1 was still the highest in SHS (P  = 0.027). The fusion index was also affected by heat exposure, showing a decrease in SHS and an increase in MHS compared with CON (P  < 0.001). Significant effects were noted from heat exposure (P  < 0.001), vitamin E treatment (P  < 0.001), and the interaction of heat exposure and vitamin E treatment (P  = 0.002) on the protein content. Taken together, our findings provide evidence that vitamin E could ameliorate the harmful effects of heat exposure by modulating heat shock proteins and apoptosis regulators, improving the protein synthesis of BSMCs during myogenic differentiation.
