Here, we show that monocytes with a trained immunity phenotype, due to exposure to the Bacillus Calmette-Guérin (BCG) vaccine, are characterized by an increased biosynthesis of different lipid mediators (LM) derived from long-chain polyunsaturated fatty acids (PUFA). Pharmacological and genetic approaches show that long-chain PUFA synthesis and lipoxygenase-derived LM are essential for the BCG-induced trained immunity responses of human monocytes. Furthermore, products of 12-lipoxygenase activity increase in monocytes of healthy individuals after BCG vaccination. Grasping the underscoring lipid metabolic pathways contributes to our understanding of trained immunity and may help to identify therapeutic tools and targets for the modulation of innate immune responses.

Recent studies have shown that trained immunity is orchestrated by sustained changes in epigenetic marks and metabolic pathways, leading to an altered transcriptional response to a second challenge. However, the potential heterogeneity of trained-immunity induction in innate immune cells has not been explored. In this study, we demonstrate cellular transcriptional programs in response to 4 different inducers of trained immunity in monocyte populations at single-cell resolution. Specifically, we identified 3 monocyte subpopulations upon the induction of trained immunity, and replicated these findings in an in vivo study. In addition, we found gene signatures consistent with these functional programs in patients with ulcerative colitis, sepsis, and COVID-19, suggesting the impact of trained-immunity programs in immune-mediated diseases.

Thallium-activated sodium iodide (NaI(Tl)) detectors can be used in gamma cameras, environmental radiation assessments, including radiation emission levels from nuclear reactors, and radiation analysis equipment. This three-phase investigation aimed to model a standard NaI(Tl) detector using the Monte Carlo N-Particle eXtended (MCNPX) general-purpose Monte Carlo simulation techniques. Accordingly, a standard NaI(Tl) detector was designed along with the required properties. Next a validation study of the modelled NaI(Tl) detector has been performed based on the experimental results for absolute detector efficiency values obtained from 226Ra, 232Th, and 40K radioisotopes. Our findings indicate that the obtained absolute detector efficiency values are quite close to used experimental values. Finally, we used the modelled detector for determination of mass attenuation coefficients of Ordinary concrete, Lead, Hematite-serpentine concrete, and Steel-scrap concrete at 186.1, 295.22, 351.93, 609.31, 1120.29, 1764.49, 238.63, 911.2, 2614, and 1460.83 keV gamma-ray energies. Additionally, according to our findings, mass attenuation coefficients obtained from the newly designed detector are compatible with the standard NIST (XCOM) data. To conclude, continuous optimisation procedures are strongly suggested for sophisticated Monte Carlo simulations in order to maintain a high degree of simulation reliability. As a result, it can be concluded that the validation of the simulation model is necessary using measured data. Finally, it can also be concluded that the validated detector models are effective instruments for obtaining basic gamma-ray shielding parameters such as mass attenuation coefficients. © 2022 Huseyin Ozan Tekin et al., published by De Gruyter.

We present the findings of an extensive examination on newly designed CdO-rich and transparent glass shields for nuclear medicine facilities in lieu of traditional and unfavorable materials, such as lead and concrete. Gamma-ray transmission factors of newly designed glass shields are determined using a variety of diagnostic, therapeutic, and research radioisotopes, including 67Ga, 57Co, 111In, 201Tl, 99mTc, 51Cr, 131I, 58Co, 137Cs, 133Ba, and 60Co. A general-purpose Monte Carlo code MCNPX (version 2.7.0) is used to determine the attenuation parameters of different material thicknesses. Next, the findings are compared using a standard concrete shielding material. The results indicate that adding more CdO to the glass composition improves the overall gamma-ray attenuation properties. As a result, among the heavy and transparent glasses developed, the C40 sample containing 40% CdO exhibited the best gamma-ray absorption properties against all radioisotopes. Furthermore, the gamma-ray absorption characteristics of this created high-density glass were shown to be better to those of a standard and heavy concrete sample. It can be concluded that the newly developed CdO-rich and transparent glass sample may be used in medical radiation fields where the radioisotopes examined are used in daily clinical and research applications. © 2022 De Gruyter. The purpose of this study is to conduct a thorough examination of the direct and indirect impacts of increasing the quantity of heavy WO3 on gamma-ray transmission, shielding and mechanical properties for some selected barium-phosphate-tungsten glasses. Accordingly, mechanical properties of barium-phosphate-tungsten oxides with chemical formula (50-x)P2O5-50BaO-xWO3 (x = 0.0(S1), 5.0(S2), 10(S3), and 15(S4)) mol% was evaluated using Makishima-Mackenzie model. Next, newly online Phy-X/PSD software and Monte Carlo code were used to examine the gamma radiation characteristics. Gamma-ray transmission factor (TF) values were calculated for S1, S2, S3 and, S4 glass samples for a range of well-known radioisotope energies such for 67Ga, 57Co-57, 111In-111, 133Ba, 201Tl, 99 mTc, 51Cr, 131I, 58Co, 137Cs, 60Co. The total packing density (Vt) was enhanced from 0.589 for S1 glass sample (free with WO3) to 0.605 for S4 glass sample (with highest WO3 =15 mol%). The total energy dissociation (Gt) of the investigated glasses was increased with increasing the WO3 content: from 51.7 (kJ/cm3) for S1 glasses to 52.45 (kJ/cm3) for S4 glasses. All mechanical moduli were improved with increasing the tungsten trioxide concentration in the studied glasses. Poisson's ratios were increased with increasing the WO3 concentration. The trend of linear (LAC) and mass attenuation (MAC) coefficients were followed as: (LAC, MAC) S1 < (LAC, MAC) S2 < (LAC, MAC) S3 < (LAC, MAC) S4. Half (HVL) and tenth (TVL) value layers have the trend as (HVL, TVL) S1 > (HVL, TVL) S2 > (HVL, TVL) S3 > (HVL, TVL) S4. The effective atomic number (Zeff) and electron density (Neff) have the same trend. The lowest transmission Factor (TF) values for all glass specimens were examined at a thickness of 3 cm. The purpose of this study was to characterize the structural, optical, and physical properties of various kinds of glasses based on the 50TeO2–30B2O3-(20-x)Li2O-xCeO2 system (x = 0, 0.5, 1, 2, 3, 4, 5, 10, 15, 20). Consequently, ten glass samples were produced by melting-annealing. Calculations of the densities of the synthesized glasses were performed using the Archimedes technique. The sample's structural, optical, physical, and radiation interaction properties were determined using XRD analysis, Raman spectroscopy, and advanced modelling techniques with FLUKA code, yielding optical band gap, refractive index, and Urbach energy values. By increasing the CeO2 reinforcement from 0 to 20 mol %, the glass densities rose from 4.0614 to 4.7519 g cm−3. The transmittance spectra of TBLC glasses were found in the range of 200–1100 nm. Our findings showed that the lowest Urbach energy belonged to the TBLC1 sample, and the highest Urbach energy belonged to the TBLC20 sample. When the CeO2 ratio was raised, the optical transmittance and absorption characteristics changed nearly monotonically, suggesting that these qualities may be calculated and controlled using the CeO2 additive, as shown in this experiment. By substituting CeO2 for Li2O inside the structure, it was possible to substantially enhance the optical band gap. Additionally, at simulated energies greater than 0.02 MeV, the gamma-ray linear attenuation coefficient rises monotonically with CeO2 reinforcement. Consequently, linear attenuation coefficients were reported as 125.843 cm−1, 127.601 cm−1, 129.211 cm−1, 132.312 cm−1, 135.166 cm−1, 138.705 cm−1, 141.288 cm−1, 156.690 cm−1, 172.393 cm−1, 186.811 cm−1 for TBLC0, TBLC0.5, TBLC1, TBLC2, TBLC3, TBLC4, TBLC5, TBL10, TBLC15 and TBLC20 at 0.015 MeV, respectively. Trained immunity describes the capacity of innate immune cells to develop heterologous memory in response to certain exogenous exposures. This phenomenon mediates, at least in part, the beneficial off-target effects of the BCG vaccine. Using an in vitro model of trained immunity, we show that BCG exposure induces a persistent change in active histone modifications, DNA methylation, transcription, and adenosine-to-inosine RNA modification in human monocytes. By profiling DNA methylation of circulating monocytes from infants in the MIS BAIR clinical trial, we identify a BCG-associated DNA methylation signature that persisted more than 12 months after neonatal BCG vaccination. Genes associated with this epigenetic signature are involved in viral response pathways, consistent with the reported off-target protection against viral infections in neonates, adults, and the elderly. Our findings indicate that the off-target effects of BCG in infants are accompanied by epigenetic remodeling of circulating monocytes that lasts more than 1 year.
