Abstract

BACKGROUND: Sleep quality in patients with chronic low back pain (CLBP) may affect quality of life (QoL), possibly due to worsening pain, central sensitization (CS), and cognitive factors. However, causal relationship among the factors has not been confirmed yet. OBJECTIVE: The purpose of this study was to test the hypothesis that sleep quality in patients with CLBP is attributable to pain, cognitive factors, and CS, and influences QoL, by structural covariance analysis. METHODS: This is a cross-sectional study. Participants were recruited from six health care facilities and 101 patients with CLBP were included. Structural covariance analysis assessed the fit of data to the model using goodness of fit index (GFI), adjusted goodness of fit index (AGFI), comparative fit index (CFI), and mean squared approximation error (RMSEA). RESULTS: The structural covariance analysis showed that the goodness-of-fit indices were high (GFI = 0.993, AGFI = 0.964, CFI = 1.00, RMSEA < 0.01). Sleep quality was not directly influenced by QoL but rather by CS and cognitive factors. CONCLUSION: This study suggests that sleep quality in patients with CLBP is indirectly mediated through multiple pathways, including cognitive factors and CS, which may influence QoL. 23. Spatiotemporal change in duration of households with every member out-of-home: a case in Kumamoto, Japan

Abstract

ABSTRACTNobody-at-home situations can cause several problems, such as home-delivery failures and burglaries. A recent study demonstrated temporal profiles of households with every member out-of-home (HEMO) situation by using household travel surveys. However, the spatial distribution of HEMO and its transition were not examined, and the effect of household attributes on HEMO was not analyzed statistically. In this study, the spatiotemporal variation in HEMO duration was investigated to address this gap, and the duration was analyzed using two econometric models. The 1984, 1997, and 2012 household travel surveys from Kumamoto, Japan, were used for the spatiotemporal visualization. In addition, the Tobit model and time allocation model were developed to statistically determine the reason for the variation in duration. The average HEMO duration increased by more than 1 h between 1984 and 2012. The downtown area revealed a longer HEMO duration, and the area with a longer duration expanded to rural areas between 1984 and 2012. The estimated econometric models revealed the statistical impacts of household attributes on HEMO duration. The HEMO duration of single-person households with a worker or student was long, and that of households with a working husband and homemaker wife was short. The spatiotemporal distribution of HEMO durations presented in this paper has the potential to be used in future urban studies, including those on the logistics of home-delivery, home-visiting survey design, crime prevention, and energy research.

Abstract

Ly6Chi monocytes migrate to injured sites and induce inflammation in the acute phase of tissue injury. However, once the causes of tissue injury are eliminated, monocyte-derived macrophages contribute to the resolution of inflammation and tissue repair. It remains unclear whether the emergence of these immunoregulatory macrophages is attributed to the phenotypic conversion of inflammatory monocytes in situ or to the recruitment of bone marrow-derived regulatory cells de novo. Here, we identified a subpopulation of Ym1+Ly6Chi monocytes that contribute to the resolution of inflammation and tissue repair. Ym1+Ly6Chi monocytes greatly expanded in bone marrow during the recovery phase of systemic inflammation or tissue injury. Ym1+Ly6Chi monocytes infiltrating into an injured site exhibited immunoregulatory and tissue-reparative phenotypes. Deletion of Ym1+Ly6Chi monocytes resulted in delayed recovery from colitis. These results demonstrate that a distinct monocyte subpopulation destined to act in immunoregulation is generated in bone marrow and participates in resolution of inflammation and tissue repair. Abstract

Myelodysplastic syndrome (MDS) is a poor prognosis cancer that predominantly affects the elderly, arising from hematopoietic stem cell (HSC), resulting in bone marrow failure and predisposition to acute myelogenous leukemia (AML). It has long been known that numerical chromosome anomalies such as trisomy 8 are critical for the development of MDS. Trisomy 8 is known to be associated with relatively poor prognosis, and is a criterion for the diagnosis of MDS in patients. Systemic inflammation has been implicated in the selective advantage for clonal hematopoietic stem cells and the progression of MDS, such as trisomy 8 MDS associated with Behcet's disease. Next generation sequencing studies revealed that trisomy 8 MDS cells highly mutated RUNX1transcriptional factor (TF) and ASXL1, an epigenetic modifier, both of which regulate HSC function and the differentiation, relative to MDS cells without trisomy 8, suggesting that those mutations may help trisomy 8 to drive the development of MDS. However, previous clinical studies failed to identify genes on the chromosome 8 commonly amplified among trisomy 8 MDS patients, which may account for the pathogenesis of MDS, the molecular mechanism of development of trisomy 8 MDS remains unclear. Thus, we hypothesized that trisomy 8 initiates the transformation by multiple malfunctions of epigenetic and transcriptional regulators in HSC other than a gene dosage effect due to the extra chromosome 8.

49. Fundamental Study on Ammonia Low-NOx Combustion Using Two-Stage Combustion by Parallel Air Jets.

Abstract

Ammonia, which has advantages over hydrogen in terms of storage and transportation, is increasingly expected to become a carbon-free fuel. However, the reduction of fuel NOx emitted from ammonia combustion is an unavoidable challenge. There is the report that two-stage combustion with parallel independent jets could achieve Low-NOx combustion under ammonia/methane co-firing conditions. In order to further improve NOx reduction, we experimentally evaluated the effects of secondary air nozzle parameters, such as nozzle diameter and nozzle locations, on combustion characteristics in two-stage combustion of ammonia/natural gas co-firing using parallel independent jets. As a result of the experiments under various secondary air nozzle conditions, it was found that under the conditions where NOx was significantly reduced, the peak temperature in the furnace was observed at 300–500 mm in the axial direction from the burner, and then the temperature decreased toward the downstream of the furnace. We assumed that this temperature distribution reflected the mixing conditions of the fuel and secondary air and estimated the combustion conditions in the furnace. It was confirmed that the two-stage combustion was effective in reducing NOx by forming a fuel rich region near the downstream of the burner, and the lean combustion of the unburned portion of the first stage combustion with secondary air. We confirmed that the low NOx effects could be achieved by two-stage combustion using independent jets from the same wall under appropriate combustion and air nozzle conditions. Abstract

Inositol pyrophosphates regulate diverse physiological processes; to better understand their functional roles, assessing their tissue-specific distribution is important. Here, we profiled inositol pyrophosphate levels in mammalian organs using an originally designed liquid chromatography-mass spectrometry (LC-MS) protocol and discovered that the gastrointestinal tract (GIT) contained the highest levels of diphosphoinositol pentakisphosphate (IP7) and its precursor inositol hexakisphosphate (IP6). Although their absolute levels in the GIT are diet dependent, elevated IP7 metabolism still exists under dietary regimens devoid of exogenous IP7. Of the major GIT cells, enteric neurons selectively express the IP7-synthesizing enzyme IP6K2. We found that IP6K2-knockout mice exhibited significantly impaired IP7 metabolism in the various organs including the proximal GIT. In addition, our LC-MS analysis displayed that genetic ablation of IP6K2 significantly impaired IP7 metabolism in the gut and duodenal muscularis externa containing myenteric plexus. Whole transcriptome analysis of duodenal muscularis externa further suggested that IP6K2 inhibition significantly altered expression levels of the gene sets associated with mature neurons, neural progenitor/stem cells, and glial cells, as well as of certain genes modulating neuronal differentiation and functioning, implying critical roles of the IP6K2-IP7 axis in developmental and functional regulation of the enteric nervous system. These results collectively reveal an unexpected role of mammalian IP7--a highly active IP6K2-IP7 pathway is conducive to the enteric nervous system. [ABSTRACT FROM AUTHOR]
