Abstract Mudstones and shales serve as natural barrier rocks in various geoenergy applications. Although many studies have investigated their mechanical properties, characterizing these parameters at the microscale remains challenging due to their fine-grained nature and susceptibility to microstructural damage introduced during sample preparation. This study aims to investigate the micromechanical properties of clay matrix composite in mudstones by combining high-speed nanoindentation mapping and machine learning data analysis. The nanoindentation approach effectively captured the heterogeneity in high-resolution mechanical property maps. Utilizing machine learning-based k-means clustering, the mechanical characteristics of matrix clay, brittle minerals, as well as measurements on grain boundaries and structural discontinuities (e.g., cracks) were successfully distinguished. The classification results were validated through correlation with broad ion beam-scanning electron microscopy images. The resulting average reduced elastic modulus (E r ) and hardness (H) values for the clay matrix were determined to be 16.2 ± 6.2 and 0.5 ± 0.5 GPa, respectively, showing consistency across different test settings and indenter tips. Furthermore, the sensitivity of indentation measurements to various factors was investigated, revealing limited sensitivity to indentation depth and tip geometry (when comparing Cube corner and Berkovich tip in a small range of indentation depth variations), but decreased stability at lower loading rates. Box counting and bootstrapping methods were applied to assess the representativeness of parameters determined for the clay matrix. A relatively small dataset (indentation number = 60) is needed to achieve representativeness, while the main challenges is to cover a representative mapping area for clay matrix characterization. Overall, this study demonstrates the feasibility of high-speed nanoindentation mapping combined with data analysis for micromechanical characterization of the clay matrix in mudstones, paving the way for efficient analysis of similar fine-grained sedimentary rocks.

Spontaneously arisen hereditary diseases in domestic animals provide an excellent opportunity to study the physiological functions of the altered genes. We investigated two 4-month-old sibling domestic short haired kittens with dry dark debris around the eyes, nose, and ears, dark crusting on the legs and a thin poor hair coat. Skin biopsies revealed abnormal sebaceous gland morphology with lack of normal sebocyte arrangement and differentiation. Hair follicles had a distorted silhouette, interpreted as a change secondary to the observed sebaceous gland dysplasia. Whole genome sequencing on both affected kittens and 65 genetically diverse feline genomes was performed. Filtering for variants that were present in both kittens but absent from the control genomes revealed a homozygous missense variant in SOAT1, encoding sterol O-acyltransferase 1. The protein is localized in the endoplasmic reticulum and catalyzes the formation of cholesteryl esters, an essential component of sebum and meibum. The identified SOAT1:c.1531G > A variant is predicted to change a highly conserved glycine residue within the last transmembrane domain of SOAT1, p.Gly511Arg. In mice, variants in Soat1 or complete knockout of the gene lead to the "hair interior defect" (hid) or abnormal Meibomian glands, respectively. SOAT1:c.1531G > A represents a plausible candidate variant for the observed sebaceous gland dysplasia in both kittens of this study. The variant was not present in 10 additional cats with a similar clinical and histopathological phenotype suggesting genetic heterogeneity. SOAT1 variants should be considered as potential cause in hereditary sebaceous gland dysplasias of humans and domestic animals.

Introduction: This study was designed to determine the mineral composition of calculi in nephrocalcinosis with nephrolithiasis, diagnose the underlying disease, and monitor the course of renal function in patients with nephrocalcinosis-nephrolithiasis. Methods: Renal calculi extruded in a series of 8 patients with nephrocalcinosis were analysed using Fourier transmission infrared spectrometry. In 4 patients, next-generation sequencing using a nephrocalcinosis-nephrolithiasis panel was performed to determine the nature of the underlying disease. In addition, longitudinal analysis of renal function was performed in all patients. Results: Seven patients revealed carbonate apatite as the sole constituent of renal calculi. One patient showed a mixed composition of dicalcium phosphate dihydrate/carbonate apatite at first analysis yet in subsequent episodes also had calculi composed of pure carbonate apatite. Further molecular analysis displayed distal renal tubular acidosis in 2 of 4 patients who consented to sequencing. No known genetic defect could be found in the other two cases. In line with prior reports, decline of renal function was dependent on underlying disease. Distal renal tubular acidosis revealed a progressive course of renal failure, whereas other causes showed stable renal function in long term analysis. Conclusion: Nephrocalcinosis with nephrolithiasis is a rare condition with heterogeneous aetiology. Yet mineral composition of renal calculi predominantly consisted of pure carbonate apatite. This uniform finding is similar to subcutaneous calcifications of various origins and might propose a general principle of tissue calcification. Progressive decline of renal function was found in distal renal tubular acidosis, whereas other conditions remained stable over time.

The characterization of materials in ever smaller dimensions is crucial for the growing demand for miniaturized devices. Hence, in situ fracture experiments are frequently performed at the micron to sub-micron scale. To evaluate fracture process of these experiments, knowledge of the crack length or the crack tip opening displacement is required. Acquired in situ frames provide a direct measurement of the crack length, crack tip opening displacement and -angle. An algorithm was developed to extract these parameters from the in situ frame sequences automatically. To verify the performance of the algorithm, fracture characteristics were measured manually for several frames of the available in situ experiments. The fracture behavior of these samples ranged from brittle over semi-brittle to ductile. The comparison between algorithmic results and manual measurements demonstrated the applicability of the algorithm to different fracture behaviors. Additionally, the fracture characteristics determined by the algorithm are in accordance with the fracture toughness data reported in literature. The crack tip opening displacement measurement gives thorough insight into the plastic deformation during fracture. The automatic extraction of the fracture characteristics allows a more detailed analysis of small-scale fracture processes and enables a reproducible, continuous evaluation of the fracture characteristics of all frames.

Thermal shock cracks are frequently observed in environments where extensive thermal cycling at high amplitudes is common. Typical cases for such are first wall materials in proposed fusion reactors and rotating anodes for computed tomography scanners. The formation of these cracks is driven by significant tensile stresses during the cooldown phase, following prior plastic deformation at elevated temperatures.This work proposes an experimental approach, where artificial thermal shock-like structures are generated via femtosecond laser ablation in WRe as a model material. Following a detailed laser parameter study in a W sheet material, patterns were introduced to WRe samples and investigated by different microscopy techniques. Their morphology as well as their response to thermo-cyclic loads was investigated and compared to conventionally induced thermal shock cracks.The introduction of artificial cracks by femtosecond laser ablation that mimic the morphology and behaviour of naturally induced thermal shock cracks is feasible within certain boundaries. Cuts, comparable to thermal shock cracks in width and depth, can be ablated. The cuts show an effect on the surrounding microstructure in line with the thermal shock cracks. Therefore, comparability could be proven concerning morphological and microstructural aspects. The conducted thermo-cyclic fatigue tests revealed an analogous effect on subsequent damage accumulation between the thermal shock cracks induced by thermal loading and artificial laser ablated cuts, proving the comparability under dynamic loading conditions. The extent of the effect is adjustable by tailoring the geometry of the lasered structures.The presented work suggests the possibility of studying the influence of thermal shock cracks on further fatigue mechanisms in exceptionally demanding applications, such as first wall materials or rotating anodes, by practical experimentation. Based on the resulting in-depth understanding of underlying interactions, the lifetime of these components might be prolonged by a deliberate introduction of crack networks or laser-ablated structures.

As multi-photon lithography technology matures, advanced integrated device design concepts become possible, positioned onto small surfaces, such as optical fibres. Only local adhesion anchors the printed object on the surface and ensure proper print-development without structure loss and long-term usability. Although surface treatments such as silanization are well known to considerably increase adhesion strength, quantification of adhesion is not straightforward and commonly assessed indirectly by surface characteristics and technological methods. The present work aims to quantify adhesion on a micrometer scale through SEM in situ tension experiments, that utilize a newly developed specimen geometry. Multiple specimens, with various footprints, were manufactured on both silanized and untreated fused silica substrates, allowing the comparable quantification of adhesion stresses between different surface conditions. Specimens on an untreated substrate failed with a high scatter in detaching force, whereas specimens on a silanized substrate tend to fail within the photoresist, either at the gauge section or just above the substrate, rather than at the interface itself. Thus, the adhesion stress was increased at least by a factor of 1.4 by the silanization treatment. Generally, a novel approach was developed, that can quantify local adhesions properties in realistic packaging or integrated circuits.

Summary: Background: Interstitial lung disease (ILD) is a common manifestation of idiopathic inflammatory myopathies (IIM) and a substantial contributor to hospitalisation, increased morbidity, and mortality. In-vivo evidence of ongoing tissue remodelling in IIM-ILD is scarce. We aimed to evaluate fibroblast activation in lungs of IIM-patients and control individuals using ⁶⁸Ga-labelled inhibitor of Fibroblast-Activation-Protein (FAPi) based positronic emission tomography and computed tomography imaging (PET/CT). Methods: In this prospective observational pilot study, consecutive patients with IIM and participants without rheumatic conditions or ILD serving as a control group were recruited at the Medical University of Vienna, Austria, and underwent FAPi PET/CT imaging. Standard-of-care procedures including clinical examination, assessment of severity of dyspnoea, high-resolution computed tomography (HR-CT), and pulmonary function testing (PFT) were performed on all patients with IIM at baseline and for patients with IIM-ILD at follow-up of 12 months. Baseline pulmonary FAPi-uptake was assessed by the maximum (SUVmax) and mean (SUVmean) standardized uptake values (SUV) over the whole lung (wl). SUV was corrected for blood pool background activity and target-to-background ratios (TBR) were calculated. We compared pulmonary FAPi-uptake between patients with IIM-ILD and those without ILD, as well as controls, and correlated baseline FAP-uptake with standard diagnostic tools such as HR-CT and PFT. For predictive implications, we investigated whether patients with IIM and progressive ILD exhibited higher baseline FAPi-uptake compared to those with stable ILD. Metrics are reported as mean with standard deviation (±SD). Findings: Between November 16, 2021 and October 10, 2022, a total of 32 patients were enrolled in the study. Three participants from the control group were excluded due to cardiopulmonary disease. In individuals with IIM-ILD (n = 14), wlTBRmax and wlTBRmean were significantly increased as compared with both non-ILD-IIM patients (n = 5) and the control group (n = 16): wlTBRmax: 2.06 ± 1.04 vs. 1.04 ± 0.22 (p = 0.019) and 1.08 ± 0.19 (p = 0.0012) and wlTBRmean: 0.45 ± 0.19 vs. 0.26 ± 0.06 (p = 0.025) and 0.27 ± 0.07 (p = 0.0024). Similar values were observed in wlTBRmax or wlTBRmean between non-ILD IIM patients and the control group. Patients with progressive ILD displayed significantly enhanced wlTBRmax and wlTBRmean values at baseline compared to patients with stable ILD: wlTBRmax: 1.30 ± 0.31 vs. 2.63 ± 1.04 (p = 0.0084) and wlTBRmean: 0.32 ± 0.08 vs. 0.55 ± 0.19 (p = 0.021). Strong correlations were found between FAPi-uptake and disease extent on HR-CT (wlTBRmax: R = 0.42, p = 0.07; wlTBRmean: R = 0.56, p = 0.013) and severity of respiratory symptoms determined by the New York Heart Association (NYHA) classification tool (wlTBRmax: R = 0.52, p = 0.022; wlTBRmean: R = 0.59, p = 0.0073). Further, pulmonary FAPi-uptake showed inverse correlation with forced vital capacity (FVC) (wlTBRmax: R = −0.56, p = 0.012; wlTBRmean: R = −0.64, p = 0.0033) and diffusing capacity of the lungs for carbon monoxide (DLCO) (wlTBRmax: R = −0.52, p = 0.028; wlTBRmean: R = −0.68, p = 0.0017). Interpretation: Our study demonstrates higher fibroblast activation in patients with IIM-ILD compared to non-ILD patients and controls. Intensity of pulmonary FAPi accumulation was associated with progression of ILD. Considering that this study was carried out on a small population, FAPi PET/CT may serve as a useful non-invasive tool for risk stratification of lung disease in IIM. A 3-months old Chinese shar-pei puppy with ichthyosis was investigated. The dog showed generalized scaling, alopecia and footpad lesions. Histopathological examinations demonstrated a non-epidermolytic hyperkeratosis. The parents of the affected puppy did not show any skin lesions. A trio whole genome sequencing analysis identified a heterozygous de novo 3 bp deletion in the KRT1 gene in the affected dog. This variant, NM_001003392.1:c.567_569del, is predicted to delete a single asparagine from the conserved coil 1A motif within the rod domain of KRT1, NP_001003392.1:p.(Asn190del). Immunohistochemistry demonstrated normal levels of KRT1 expression in the epidermis and follicular epithelia. This might indicate that the variant possibly interferes with keratin dimerization or another function of KRT1. Missense variants affecting the homologous asparagine residue of the human KRT1 cause epidermolytic hyperkeratosis. Histologically, the investigated Chinese shar-pei showed a non-epidermolytic ichthyosis. The finding of a de novo variant in an excellent functional candidate gene strongly suggests that KRT1:p.Asn190del caused the ichthyosis phenotype in the affected Chinese shar-pei. To the best of our knowledge, this is the first description of a KRT1-related non-epidermolytic ichthyosis in domestic animals.
