10. Identity of tumorigenic human urothelial cell lines and 'spontaneously' transformed sublines

Abstract

Restriction fragment length polymorphism (RFLP) analysis, comparative marker chromosome analysis, and polymorphic enzyme analysis was carried out on a total of eight human urothelial cell lines and sublines selected according to our knowledge of their HLA-A,B phenotype. RFLP analysis and cytogenetic analysis showed that the cell lines Hu1703He, Hu1922, and T24 are genuine cell lines of different origin. The identity of Hu1703He could not be confirmed by its isozyme phenotype which was identical to the T24 phenotype. RFLP analysis and isozyme analysis revealed that three cell lines, Hu456, Hu549, and Hu961a, and two transformed sublines, HCV-29Tmv and Hu609Tmv, are sublines of T24. A common origin of Hu456, Hu549, Hu961a, HCV-29Tmv, and Hu609Tmv was confirmed by marker chromosome analysis. However, the T24 origin of these cytogenetically related cell lines was not supported by chromosome analysis of T24. RFLP analysis and HLA phenotyping of two tumorigenic and invasive sublines isolated from a culture of non-tumorigenic Hu609 cells showed that non-tumorigenic Hu609 cells can transform 'spontaneously' in vitro into tumorigenic Hu609T cells. The results emphasise the need for careful monitoring and screening of cell lines for their identity using more than one identification parameter.

Abstract

Mouse monoclonal antibodies to human urinary bladder carcinoma cells have been examined by indirect membrane immunofluorescence using a panel of 27 human cell lines. Two of the monoclonal antibodies, 7E9 (IgG3) and S2C6 (IgG1), were found to distinguish between urinary bladder carcinoma cells and normal urothelium. The third monoclonal antibody, T24.05(IgG1), discriminated among cell lines of urothelial and non-urothelial origin but did not distinguish between urinary bladder carcinoma and normal urothelial cells. None of the of the antibodies was found to be strictly selective, and occasional cross-reactions with unrelated cell types were observed. The monoclonal antibody 7E9, showing the highest degree of selectivity, was further examined by an indirect immunoperoxidase technique on frozen tissue sections from 19 patients. The antibody reacted with all ( 7 7 ) bladder carcinomas examined and gave negative results with control normal bladder mucosa ( 0 8 ) and unrelated tumor tissue ( 0 4 ) sections. The 7E9 antibody was purified by protein A affinity chromatography, labeled with 131 I and used for gamma-scintigraphy in nude mice xenografted with human urinary bladder carcinoma T24. The 7E9 antibody was capable of locating the T24 xenografts in nude mice; it localized preferentially in the T24 tissue compared to normal mouse tissues. The T24 xenografts could not be detected by gamma-scintigraphy with 131 I -labeled monoclonal antibody against human mammary carcinoma cells and two other control antibodies. Likewise the 131 I -labeled 7E9 antibody was not capable of locating human mammary carcinoma xenografts in nude mice.

Abstract

SummaryThe aminoacylation of tRNA is an important step in the regulation of protein synthesis. The aminoacyl-tRNA synthetases may have a regulatory function in the transformation of normal cells to neoplastic cells.Seryl and phenylalanyl synthetase have been isolated from ascitic tumor cells which were previously treated with 20-methyl-cholanthrene. The different steps of preparation are (NH4)2SO4 precipitation of the 105,000g supernatant (between 50 and 65% saturation), chromatography on DEAE cellulose, CM Sephadex C-50 and hydroxy-apatite.The purification fold of the seryl-tRNA synthetase was 450 times with the molecular weight of 120,000. The present studies have shown that the Km for serine was about 20 times higher in MCA-treated cells than in the controls, while the Km for ATP was only increased 3–4 times. These values indicate that the synthetases possibly have a regulatory role in the protein synthesis of the cancer cells with and without treatment with MCA. Abstract

a. MIT used in the present work was found to be a useful method for the evaluation of sensitization occurring in oncological experimental systems, where transplantation tests may fail due to high virulence of the tumor.b. Both cell lines used, L-1 and L-2c, which are established tumorogenic cell lines derived from the C3H strain, sensitize isogenic hosts when injected sc.c. No cross-reaction between the two cell lines could be demonstrated by the MIT as used in this work.d. Both cell lines sensitize hosts of DBA strain across H-2. However, only slight signs of cross-reaction were seen, when DBA spleen sensitized this way was challenged by C3H spleen cell homogenate.e. Experiments on tumors derived from the tumorogenic cell lines show cross-reactions with the original cell lines.f. Slight signs of autoimmunization were observed in C3H mice after sensitization by the transformed cell lines, when challenged by the normal C3H spleen homogenate.

Abstract

Udgivelsesdato: 1979-Feb, ST-L1 is a cell line established from lung explants from a normal ST/a mouse. The ST-L1 cells have undergone spontaneous malignant alteration in vitro. The cells were rejected after inoculation into syngeneic immunocompetent hosts, and a syngeneic humoral immuneresponse against the ST-L1 cells has been detected. The specificity of this humoral response was investigated. The syngeneic response to ST-L1 was characterized by indirect immunofluorescence tests and by immunoprecipitation of radiolabelled cells and of a C-type virus produced by the cell lines. The specific anti-ST-L1 reactivities were found to be directed against the envelope glycoprotein of an endogenous C-type virus expressed by the antigenic cell line.
