Background: Thioacetamide (TAA) has been used extensively in the development of suitable animal models of acute and chronic liver injury employing various doses, times and routes of its administration, particularly in drinking water due to its resemblance with human liver fibrosis and cirrhosis. The aim of this study was to investigate and compare hematological alteration during the acute and chronic liver inflammation. Methods: Acute Liver inflammation was induced in Wistar rats via intraperitoneal injection of thioacetamide and the animals were sacrificed 12 h after the TAA administration. Induction of chronic liver inflammation was performed by continuous administration of TAA in the drinking water (200 mg/L) during 18 weeks of experiment. After that all animals were sacrificed and Blood samples were collected for further analysis. Results: Single intra peritoneal injection of TAA (300 mg/kg B.W.) induced an acute condition with hematological changes including leukocytosis with marked neutrophilia (P = 0.0429), lymphopenia, thrombocytosis as well as increased hemoglobin concentration (P 0.05) and decline of erythrocytic count (P = 0.0009). Eighteen weeks of uninterrupted supply of TAA (200 mg/L) in drinking water lead to chronic inflammation and the hematological alterations were leucopenia (P = 0.0197) accompanied with neutropenia and thrombocytopenia. Increase in RBCs (P = 0.0073) and Hb contents was also observed with a decline of red cell indices. Conclusion: Taken together these findings we can conclude that the animals respond differently under acute and chronic inflammatory condition with TAA administration. Leukocytosis with marked neutrophilia, thrombocytosis as well as increased hemoglobin concentration and decline of erythrocytic count were observed in acute while leucopenia accompanied with neutropenia and thrombocytopenia and increase in RBCs, Hb and Hct was also observed with a decline of other red cell indices during chronic phase.

Interleukin-22 (IL-22) is a pluripotent T cell-derived cytokine which is a member of IL-10 cytokine family. It is the only interleukin produced by immune cells but does not target immune system components. IL-22 is mainly produced by dendritic cells (DCs) and TH17, TH22, NK, and NKT cells and targets a number of body tissues including liver, pancreas, and other epithelial tissues. It provokes a series of downstream signaling pathways upon binding with IL-22R complex which protects liver damage through STAT3 activation. IL-22BP is an inhibitor of IL-22 which has 20–1000x more affinity to bind with IL-22 compared to IL-22R1 that inhibits IL-22 activity. Its level was found to be positively correlated with the severity of liver damage and fibrosis. So, the present review is an effort to reveal the exact mechanism lying in the hepatoprotective activity of IL-22 and some of its future therapeutic implications.

Acetylcholinesterase (AChE), arylesterase (AE), carboxylesterase (CE), and cholinesterase (ChE) activities were determined in six adult populations of the lesser grain borer, Rhyzopertha dominica (Fabricius) (Coleoptera: Bostrichidae) collected from grain warehouses in various areas of Punjab, namely Lahore (L), Sialkot (S), Wazirabad (W), Multan (M), Chichawatni (C), and Karachi (K). The multiple forms of esterases were resolved by polyacrylamide gel electrophoresis (PAGE) and activities determined biochemically, using a spectrophotometer. All the strains had the same AChE activity except Sialkot strain, which had 46% more activity than the susceptible K strain. Similarly K, S, W, L, and C strains exhibited almost the same AE level, whereas the M strain showed 29% lower activity than K strain. The CE activity of L (352%), S (198%) and C (214%) strains and ChE activity of L (178%) and C (317%) strains were significantly higher than that of K strain. The total esterases of L, S and C strains showed respectively 152%, 63% and 73% higher activity than the susceptible K strain. The PAGE pattern of various esterases coincided with the biochemical analysis. The variable thickness of the bands in the gel indicated relative esterase induction that could be correlated with the development of resistance in R. dominica.
