TFEB and TFE3 (TFEB/3), key regulators of lysosomal biogenesis and autophagy, play diverse roles depending on cell type. This study highlights a hitherto unrecognized role of TFEB/3 crucial for peripheral nerve repair. Specifically, they promote the generation of progenitor-like repair Schwann cells after axonal injury. In Schwann cell-specific TFEB/3 double knock-out mice of either sex, the TFEB/3 loss disrupts the transcriptomic reprogramming that is essential for the formation of repair Schwann cells. Consequently, mutant mice fail to populate the injured nerve with repair Schwann cells and exhibit defects in axon regrowth, target reinnervation, and functional recovery. TFEB/3 deficiency inhibits the expression of injury-responsive repair Schwann cell genes, despite the continued expression of c-jun, a previously identified regulator of repair Schwann cell function. TFEB/3 binding motifs are enriched in the enhancer regions of injury-responsive genes, suggesting their role in repair gene activation. Autophagy-dependent myelin breakdown is not impaired despite TFEB/3 deficiency. These findings underscore a unique role of TFEB/3 in adult Schwann cells that is required for proper peripheral nerve regeneration. [ABSTRACT FROM AUTHOR]

19. Modelling and testing the definitions of teleworking within a local council environment

Teleworking was defined in terms of comprehension: root definition, conceptual definition and abstraction definition. The definitions were subsequently modelled in terms of four theories: socio-factors of teleworking (model 1 of 4), maturity model of teleworking (model 2 of 4), technical factors of teleworking (model 3 of 4) and taxonomy of teleworking (model 4 of 4). The modelling of the definitions of teleworking as stated adds further comprehension to the concept of teleworking. Teleworking is a socio and technical working practice and so, the research study turned to the socio aspect: there were a number of socio-factors (minor and major) identified as per existing literature. Subsequently, major socio-factors were mapped to a teleworking maturity model in terms of layers, resource, policy and connectivity. The technical aspect of the research study was able to identify and divide factors into dimensions, attributes and organisational roles. The three models (socio, technical and maturity) were brought together in terms of taxonomy of teleworking: an amalgamation of the socio and technical factors of teleworking in addition to three layers of a maturity model. The research methodology followed a positivist viewpoint with socio-factors measured using 7-point Likert scales. There were a large number of measures for socio-teleworking and so two research methods were adopted to reduce the number to a manageable amount namely: initial questionnaire design and Q-sort study. Following exclusions, a web-based survey was created with the remaining socio-measures of teleworking. The web-based survey was conducted in terms of a pilot study (at councils in the north of England) before surveying 264 employees at Council-Z (the primary study). Data collected from Council-Z was analysed in terms of confirmatory factor analysis. Theoretical models (factor structures) were created in terms of resource, policy and connectivity. The factor structures of each stated layer were tested for consistency to data. Four factor structures of resource were identified, A, B, C and D. Factor structure D showed the highest level of convergence of theory to observed data that is, the best-fitting model. Six factor structures of policy were identified, with factor structure C2 the most favourable in terms of exclusion of ambiguities and model-fit statistics. Three factor structures of connectivity were identified and for each of the absolute and incremental fit statistics factor structure B was consistently within the cut-off values for good model-fit, factor structure B was also the best fitting model. In terms of the utility of the study, definitions of teleworking and the modelling of the definitions have improved understanding of the research area. The extensive number of factors of teleworking identified through the theoretical modelling process and the measurements of these have demonstrated improved measurement techniques. The best-fitting models as per the confirmatory factor analyses have broad applicability to other similar organisations, and finally the data from the three best-fitting models can be utilised by Council-Z to introduce informed teleworking initiatives. In terms of limitations and future work, technical factors were out of scope in this research study. Hence, types of teleworking practices linked to technical factors of teleworking would be future work as would studies of the linkage between the socio-and technical factors. In terms of the taxonomical model empirical validation would be sought of each of the seven major socio-factors in terms of factor structures. This study empirically tested for each of the three layers of the maturity model, as opposed to each of the major socio-factors within the three layers. Furthermore, additional factors may be identifiable through future work, adding to the taxonomy and in turn, the comprehension of teleworking would be enhanced alongside further standardisation of teleworking definitions and measurements.

The Collaboratory for the Study of Earthquake Predictability (CSEP) is an open and global community whose mission is to accelerate earthquake predictability research through rigorous testing of probabilistic earthquake forecast models and prediction algorithms. pyCSEP supports this mission by providing open-source implementations of useful tools for evaluating earthquake forecasts. pyCSEP is a Python package that contains the following modules: (1) earthquake catalog access and processing, (2) representations of probabilistic earthquake forecasts, (3) statistical tests for evaluating earthquake forecasts, and (4) visualization routines and various other utilities. Most significantly, pyCSEP contains several statistical tests needed to evaluate earthquake forecasts, which can be forecasts expressed as expected earthquake rates in space–magnitude bins or specified as large sets of simulated catalogs (which includes candidate models for governmental operational earthquake forecasting). To showcase how pyCSEP can be used to evaluate earthquake forecasts, we have provided a reproducibility package that contains all the components required to re-create the figures published in this article. We recommend that interested readers work through the reproducibility package alongside this article. By providing useful tools to earthquake forecast modelers and facilitating an open-source software community, we hope to broaden the impact of the CSEP and further promote earthquake forecasting research.

- Abstract
Background and Objectives: Declines in stroke admission, IV thrombolysis (IVT), and mechanical thrombectomy volumes were reported during the first wave of the COVID-19 pandemic. There is a paucity of data on the longer-term effect of the pandemic on stroke volumes over the course of a year and through the second wave of the pandemic. We sought to measure the effect of the COVID-19 pandemic on the volumes of stroke admissions, intracranial hemorrhage (ICH), IVT, and mechanical thrombectomy over a 1-year period at the onset of the pandemic (March 1, 2020, to February 28, 2021) compared with the immediately preceding year (March 1, 2019, to February 29, 2020)., Methods: We conducted a longitudinal retrospective study across 6 continents, 56 countries, and 275 stroke centers. We collected volume data for COVID-19 admissions and 4 stroke metrics: ischemic stroke admissions, ICH admissions, IVT treatments, and mechanical thrombectomy procedures. Diagnoses were identified by their ICD-10 codes or classifications in stroke databases., Results: There were 148,895 stroke admissions in the 1 year immediately before compared with 138,453 admissions during the 1-year pandemic, representing a 7% decline (95% CI [95% CI 7.1–6.9]; p < 0.0001). ICH volumes declined from 29,585 to 28,156 (4.8% [5.1–4.6]; p < 0.0001) and IVT volume from 24,584 to 23,077 (6.1% [6.4–5.8]; p < 0.0001). Larger declines were observed at high-volume compared with low-volume centers (all p < 0.0001). There was no significant change in mechanical thrombectomy volumes (0.7% [0.6–0.9]; p = 0.49). Stroke was diagnosed in 1.3% [1.31–1.38] of 406,792 COVID-19 hospitalizations. SARS-CoV-2 infection was present in 2.9% ([2.82–2.97], 5,656/195,539) of all stroke hospitalizations., Discussion: There was a global decline and shift to lower-volume centers of stroke admission volumes, ICH volumes, and IVT volumes during the 1st year of the COVID-19 pandemic compared with the prior year. Mechanical thrombectomy volumes were preserved. These results suggest preservation in the stroke care of higher severity of disease through the first pandemic year., Trial Registration Information: This study is registered under NCT04934020.

- Abstract
Background and Objectives COVID-19–related inflammation, endothelial dysfunction, and coagulopathy may increase the bleeding risk and lower the efficacy of revascularization treatments in patients with acute ischemic stroke (AIS). We aimed to evaluate the safety and outcomes of revascularization treatments in patients with AIS and COVID-19. Methods This was a retrospective multicenter cohort study of consecutive patients with AIS receiving intravenous thrombolysis (IVT) and/or endovascular treatment (EVT) between March 2020 and June 2021 tested for severe acute respiratory syndrome coronavirus 2 infection. With a doubly robust model combining propensity score weighting and multivariate regression, we studied the association of COVID-19 with intracranial bleeding complications and clinical outcomes. Subgroup analyses were performed according to treatment groups (IVT-only and EVT). Results Of a total of 15,128 included patients from 105 centers, 853 (5.6%) were diagnosed with COVID-19; of those, 5,848 (38.7%) patients received IVT-only and 9,280 (61.3%) EVT (with or without IVT). Patients with COVID-19 had a higher rate of symptomatic intracerebral hemorrhage (SICH) (adjusted OR 1.53; 95% CI 1.16–2.01), symptomatic subarachnoid hemorrhage (SSAH) (OR 1.80; 95% CI 1.20–2.69), SICH and/or SSAH combined (OR 1.56; 95% CI 1.23–1.99), 24-hour mortality (OR 2.47; 95% CI 1.58–3.86), and 3-month mortality (OR 1.88; 95% CI 1.52–2.33). Patients with COVID-19 also had an unfavorable shift in the distribution of the modified Rankin score at 3 months (OR 1.42; 95% CI 1.26–1.60). Discussion Patients with AIS and COVID-19 showed higher rates of intracranial bleeding complications and worse clinical outcomes after revascularization treatments than contemporaneous non–COVID-19 patients receiving treatment. Sudden infant death syndrome (SIDS) remains the leading cause of infant mortality beyond the neonatal period. An increase in body temperature as a result of high environmental temperature, overwrapping of infants, and (or) infection are associated with SIDS. Endotoxins such as lipopolysaccharide (LPS) and heat stress may perturb cardiorespiratory function and thermoregulation. Although LPS-mediated body temperature and cytokine responses are well documented in older animals, the capacity of LPS to induce fever and cytokine response in young rats remains unclear. Therefore, we sought to investigate the acute effects of LPS on body temperature and cytokine concentrations in rat pups. Postnatal day 7 rat pups were divided into 3 groups: Group 1, rats were administered LPS intraperitoneally (200 [micro]g/kg); Group 2, rats received saline at volume equal to that administered in the LPS group; Group 3, rats received no treatment. Pups were placed in custom-made chambers maintained at ambient temperature of 33[degrees]C. Body surface temperature was continuously monitored for 4 h. Thereafter, the rats were euthanized and serum was collected for cytokine analysis. We demonstrate that LPS treatment increased MIP-1[alpha], IL-10, MCP-1, IP-10, fractalkine, and TNF-[alpha] with no concurrent rise in body surface temperature. Although neonatal rats produced an array of cytokines in response to LPS, there was no evidence of fever. Key words: inflammation, fever, sudden infant death syndrome, neonatal, cytokines. Les endotoxines comme les lipopolysaccharides (LPS) et le stress thermique pourraient perturber la fonction cardiorespiratoire et la thermoregulation. Bien que la temperature corporelle et les reactions par les cytokines mediees par les LPS soient bien documentees chez les animaux plus ages, la capacite des LPS a provoquer de la fievre ainsi et une reaction par les cytokines demeurent a preciser chez les rats en bas age. Par consequent, nous avons tente d'etudier les effets aigus des LPS sur la temperature corporelle et les concentrations de cytokines chez des ratons. Nous avons reparti des ratons de 7 jours dans 3 groupes: Groupe 1, administration intraperitoneale de LPS (200 [micro]g/kg); Groupe 2, administration d'un volume de solution saline identique au volume administre dans le groupe LPS; Groupe 3, aucune intervention. Nous avons place les ratons dans des chambres sur mesure maintenues a une temperature ambiante de 33[grados]C. Nous avons surveille la temperature corporelle de surface en continu pendant 4 h. Par la suite, nous avons euthanasie les rats et preleve du serum en vue du dosage de cytokines. Nous montrons que l'administration de LPS entramait une augmentation des taux de MIP-1[alfa], d'IL-10, de MCP-1, d'IP-10, de fractalkine et de TNF-[alfa], sans hausse concomitante de la temperature corporelle de surface. Meme si les rats nouveau-nes produisaient une gamme de cytokines en reaction aux LPS, nous n'avons observe aucun indice de fievre. The devastating 2023 M7.8 Türkiye–Syria earthquake sequence once again highlighted the gap between (seismological) knowledge and action: Although the impacted region is known to be at high seismic risk (i.e., highly seismically active and densely populated), the political and societal conditions have complicated and delayed protective measures. To reduce the seismic risk and prepare local communities, experts from different disciplines must collaborate effectively in redesigning the built environment and engaging the community in risk education and management (Comfort et al., 2023). In recent years, three subjects have become increasingly relevant to build that needed bridge between scientific knowledge and societal action: open science, transdisciplinarity, and ethics (see Figure 1). They have influenced scientific discussions on how to transition from purely scientific research to practical and societally relevant applications that increase societies’ resilience to disasters (e.g., Marti et al., 2022) – just as envisioned by the European Union’s Horizon 2020 project ‘Real-time earthquake rIsk reduction for a reSilient Europe’ (RISE) concerning earthquake risk. We and other early career scientists of RISE identified these three subjects in several virtual discussions while reflecting on our needs to better understand how we can make our research efforts more societally meaningful and effective. Eventually, together with several senior scientists of RISE, we discussed and evaluated these subjects during a three-day workshop under the theme “Bringing research to practical applications that increase society’s earthquake resilience” (Supplement 1). This theme resembled RISE’s overall goal to advance the scientific and societal knowledge on dynamic seismic risk, the overarching topic we additionally wanted to explore. First, keynotes from experts gave us a background on the three subjects, and lightning talks by all participants revealed the range of our expertise. Second, we drew a rich picture for the overarching topic and each subject following the Soft Systems Methodology (Pohl, 2020): separate groups sketch and express their ideas as mental models, receive feedback from the other groups, and revise it accordingly (see Supplement S2a-d for the evolution of the rich pictures). This approach allowed us to integrate all our expertise on the topic and subjects. We provide the conceptual background of the three main subjects, stress their relevance in current research, and illustrate their link to dynamic seismic risk (Figure 1). We believe that these reflections can be transferred to any other research field since the subjects affect various disciplines.

- Abstract
© 2022 American Academy of Neurology, Background and objectives: Declines in stroke admission, IV thrombolysis (IVT), and mechanical thrombectomy volumes were reported during the first wave of the COVID-19 pandemic. There is a paucity of data on the longer-term effect of the pandemic on stroke volumes over the course of a year and through the second wave of the pandemic. We sought to measure the effect of the COVID-19 pandemic on the volumes of stroke admissions, intracranial hemorrhage (ICH), IVT, and mechanical thrombectomy over a 1-year period at the onset of the pandemic (March 1, 2020, to February 28, 2021) compared with the immediately preceding year (March 1, 2019, to February 29, 2020). Methods: We conducted a longitudinal retrospective study across 6 continents, 56 countries, and 275 stroke centers. We collected volume data for COVID-19 admissions and 4 stroke metrics: ischemic stroke admissions, ICH admissions, IVT treatments, and mechanical thrombectomy procedures. Diagnoses were identified by their ICD-10 codes or classifications in stroke databases. Results: There were 148,895 stroke admissions in the 1 year immediately before compared with 138,453 admissions during the 1-year pandemic, representing a 7% decline (95% CI [95% CI 7.1-6.9]; p < 0.0001). ICH volumes declined from 29,585 to 28,156 (4.8% [5.1-4.6]; p < 0.0001) and IVT volume from 24,584 to 23,077 (6.1% [6.4-5.8]; p < 0.0001). Larger declines were observed at high-volume compared with low-volume centers (all p < 0.0001). There was no significant change in mechanical thrombectomy volumes (0.7% [0.6-0.9]; p = 0.49). Stroke was diagnosed in 1.3% [1.31-1.38] of 406,792 COVID-19 hospitalizations. SARS-CoV-2 infection was present in 2.9% ([2.82-2.97], 5,656/195,539) of all stroke hospitalizations. Discussion: There was a global decline and shift to lower-volume centers of stroke admission volumes, ICH volumes, and IVT volumes during the 1st year of the COVID-19 pandemic compared with the prior year. Mechanical thrombectomy volumes were preserved. These results suggest preservation in the stroke care of higher severity of disease through the first pandemic year.

Background and ObjectivesCOVID-19–related inflammation, endothelial dysfunction, and coagulopathy may increase the bleeding risk and lower the efficacy of revascularization treatments in patients with acute ischemic stroke (AIS). We aimed to evaluate the safety and outcomes of revascularization treatments in patients with AIS and COVID-19.MethodsThis was a retrospective multicenter cohort study of consecutive patients with AIS receiving intravenous thrombolysis (IVT) and/or endovascular treatment (EVT) between March 2020 and June 2021 tested for severe acute respiratory syndrome coronavirus 2 infection. With a doubly robust model combining propensity score weighting and multivariate regression, we studied the association of COVID-19 with intracranial bleeding complications and clinical outcomes. Subgroup analyses were performed according to treatment groups (IVT-only and EVT).ResultsOf a total of 15,128 included patients from 105 centers, 853 (5.6%) were diagnosed with COVID-19; of those, 5,848 (38.7%) patients received IVT-only and 9,280 (61.3%) EVT (with or without IVT). Patients with COVID-19 had a higher rate of symptomatic intracerebral hemorrhage (SICH) (adjusted OR 1.53; 95% CI 1.16–2.01), symptomatic subarachnoid hemorrhage (SSAH) (OR 1.80; 95% CI 1.20–2.69), SICH and/or SSAH combined (OR 1.56; 95% CI 1.23–1.99), 24-hour mortality (OR 2.47; 95% CI 1.58–3.86), and 3-month mortality (OR 1.88; 95% CI 1.52–2.33). Patients with COVID-19 also had an unfavorable shift in the distribution of the modified Rankin score at 3 months (OR 1.42; 95% CI 1.26–1.60).DiscussionPatients with AIS and COVID-19 showed higher rates of intracranial bleeding complications and worse clinical outcomes after revascularization treatments than contemporaneous non–COVID-19 patients receiving treatment. Current available data do not allow direct conclusions to be drawn on the effectiveness of revascularization treatments in patients with COVID-19 or to establish different treatment recommendations in this subgroup of patients with ischemic stroke. Our findings can be taken into consideration for treatment decisions, patient monitoring, and establishing prognosis.Trial Registration InformationThe study was registered under ClinicalTrials.gov identifierNCT04895462.

OBJECTIVE The objectives of this study were to measure the global impact of the pandemic on the volumes for intravenous thrombolysis (IVT), IVT transfers, and stroke hospitalizations over 4 months at the height of the pandemic (March 1 to June 30, 2020) compared with two control 4-month periods. METHODS We conducted a cross-sectional, observational, retrospective study across 6 continents, 70 countries, and 457 stroke centers. Diagnoses were identified by their ICD-10 codes and/or classifications in stroke databases.
