Embryonal carcinoma affects young males in the prime of life with a majority of these tumors already having metastasis at the time of diagnosis. Subcutaneous metastasis from embryonal carcinoma are very rare and often associated with wide spread disease and poor prognosis. We report a case of chest wall subcutaneous metastasis of embryonal carcinoma in a 27 year-old man that was the first presentation of the disease and was diagnosed on fine needle aspiration cytology (FNAC). Subsequent search led to the discovery of the primary in the testis. The cytomorphological features of embryonal carcinoma are quite distinctive and FNAC plays a vital role in early diagnosis. The criteria for diagnosis includes presence of cellular smears exhibiting disperse cells as well as cell aggregates forming microglandular patterns. Cells have large nuclei with prominent nucleoli and pale indistinct often vacuolated cytoplasm. Early diagnosis and treatment with platinum based chemotherapy in conjunction with radiotherapy and surgery have high cure rate.

Cutaneous metastasis is an uncommon manifestation of visceral malignancy and a grave prognostic indicator. In extremely rare cases it may be the presenting sign of underlying malignancy. Here we report two cases of carcinoma gall bladder that presented with skin nodule. Fine needle aspiration cytology of these nodules revealed metastatic deposits of adenocarcinoma and was instrumental in establishing the primaries.

Abstract Background Production of human monoclonal antibodies that exhibit broadly neutralizing activity is needed for preventing HIV-1 infection, however only a few such antibodies have been generated till date. Isolation of antibodies by the hybridoma technology is a cumbersome process with fewer yields. Further, the loss of unstable or slowly growing clones which may have unique binding specificities often occurs during cloning and propagation and the strongly positive clones are often lost. This has been avoided by the process described in this paper, wherein, by combining the strategy of EBV transformation and recombinant DNA technology, we constructed human single chain variable fragments (scFvs) against the third variable region (V3) of the clade C HIV-1 envelope. Results An antigen specific phage library of 7000 clones was constructed from the enriched V3- positive antibody secreting EBV transformed cells. By ligation of the digested scFv DNA into phagemid vector and bio panning against the HIV-1 consensus C and B V3 peptides followed by random selection of 40 clones, we identified 15 clones that showed V3 reactivity in phage ELISA. DNA fingerprinting analysis and sequencing showed that 13 out of the 15 clones were distinct. Expression of the positive clones was tested by SDS-PAGE and Western blot. All the 13 anti-V3 scFvs showed cross-reactivity against both the clade C and B V3 peptides and did not show any reactivity against other unrelated peptides in ELISA. Preliminary neutralization assays indicated varying degrees of neutralization of clade C and B viruses. EBV transformation, followed by antigen selection of lines to identify specific binders, enabled the selection of phage from un-cloned lines for scFv generation, thus avoiding the problems of hybridoma technology. Moreover, as the clones were pretested for antigen binding, a comparatively small library sufficed for the selection of a considerable number of unique antigen binding phage. After selection, the phage clones were propagated in a clonal manner. Conclusions This strategy can be efficiently used and is cost effective for the generation of diverse recombinant antibodies. This is the first study to generate anti-V3 scFvs against HIV-1 Clade C.

Class III peroxidases are plant-specific glycoproteins and widely distributed among plant species, that play a crucial role in plant resistance to different stresses, such as salt, heat, cold and metal toxicity. The present study is the first comprehensive and systematic report to characterize the NcPOD gene family in water lily ( Nymphaea colorata ). In this study, 94 NcPOD genes in water lily were identified, each possessing a conserved POD domain, which are dispersed unevenly across the genome. Through comparative maximum-likelihood phylogenetic analysis, these genes were categorized into 10 groups, along with two other species, Arabidopsis thaliana and Nymphaea thermarum . Notably, the largest group, group-c, comprised 32 distinct types of NcPOD proteins. These genes exhibited uneven distribution on 11 of the 14 chromosomes of water lily. Exon-intron and motif analyses exhibited the structural and functional diversity among the sub-groups. The Examination of duplication patterns suggests that tandem duplication has contributed to the expansion of NcPOD genes. The analysis of promoter cis-acting elements indicated the presence of regulatory elements associated with various responses such as ABA, MeJA, light responsiveness, anaerobic conditions, and drought inducibility. Finally, the RT-qPCR based expression and enzymes activity of ten NcPOD genes depicted the dynamically differential response to NaCl, heat, cold, and heavy metals (CuSO 4 and CdCl 2 ) stresses. The GDSL gene family plays diverse roles in plant growth and development. Despite its significance, the functions of the GDSL in the pitaya plant are still unknown. Pitaya ( Selenicereus undatus L.) also called Hylocereus undatus (Hu), belongs to the family Cactaceae and is an important tropical plant that contains high dietary fibers and antioxidants. In the present investigation, we screened 91 HuGDSL genes in the pitaya genome by conducting a comprehensive computational analysis. The phylogenetic tree categorized HuGDSL genes into 9 distinct clades in combination with four other species. Further, 29 duplicate events were identified of which 12 were tandem, and 17 were segmental. The synteny analysis revealed that segmental duplication was more prominent than tandem duplication among these genes. The majority of duplicated gene pairs (95%) indicate their Ka/Ks ratios ranging from 0.1 to 0.3, which shows that maximum HuGDSL genes were under purifying selection pressure. The cis -acting element in the promotor region contains phytohormones such as auxin, gibberellin, jasmonic acid, and abscisic acid abundantly. Finally, the HuGDSL gene expression pattern under single and multiple stresses was analyzed via; RNA-seq. We select ten stress-responsive HuGDSL genes for RT-qPCR validation. After careful investigation, we identified five HuGDSL candidate genes ( HuGDSL-1/3/55/59, and HuGDSL-78 ) based on RNA-seq, and RT-qPCR data that showed enhanced expression in stress and melatonin-applied seedlings. This work presents a comparative study on the structural, optical and electrochemical characteristics of visible light driven (VLD) reduced titanium dioxide (TiO 2- x ) nanocatalysts synthesized via inorganic and organic synthetic routes. X-ray diffraction (XRD) patterns, Raman spectra and X-ray absorption fine structure (XAFS) analyses reflected anatase phase titania. Whereas, the quantitative EXAFS fit and XANES analysis revealed structural distortion due to the presence of oxygen and titanium vacancies with low valent Ti states in anatase lattices of certain nanocatalysts, which subsequently leads to better electrochemical and photocatalytic activities. Moreover, owing to the large surface area and mesoporous structures, the Mg-TiO 2- x nanocatalysts exhibited enhanced water adsorption and ultimately increased overall water splitting with an OER overpotential equal to 420 mV vs. RHE at a current density of 10 mA cm -2 (Tafel slope = 62 mV dec -1 ), extended visible light absorbance, decreased photoluminescence (PL) intensity and increased carrier lifetime in comparison with commercial titania., Competing Interests: The authors declare that they have no known competing financial interests or personal relationships that could have appeared to influence the work reported in this paper., (This journal is © The Royal Society of Chemistry.)

DNA binding proteins with one finger (Dof ) transcription factors are essential for seed development and defence against various biotic and abiotic stresses in plants. Genomic analysis of Dof has not been determined yet in pitaya (Selenicereus undatus ). In this study, we have identified 26 Dof gene family members, renamed as HuDof-1 to HuDof-26 , and clustered them into seven subfamilies based on conserved motifs, domains, and phylogenetic analysis. The gene pairs of Dof family members were duplicated by segmental duplications that faced purifying selection, as indicated by the K a /K s ratio values. Promoter regions of HuDof genes contain many cis -acting elements related to phytohormones including abscisic acid, jasmonic acid, gibberellin, temperature, and light. We exposed pitaya plants to different environmental stresses and examined melatonin's influence on Dof gene expression levels. Signifcant expression of HuDof -2 and HuDof -6 were observed in different developmental stages of flower buds, flowers, pericarp, and pulp. Pitaya plants were subjected to abiotic stresses, and transcriptome analysis was carried out to identify the role of Dof gene family members. RNA-sequencing data and reverse transcription quantitative PCR-based expression analysis revealed three putative candidate genes (HuDof -1, HuDof -2, and HuDof -8), which might have diverse roles against the abiotic stresses. Our study provides a theoretical foundation for functional analysis through traditional and modern biotechnological tools for pitaya trait improvement.

Brassica napus ( B. napus ) is susceptible to multiple abiotic stresses that can affect plant growth and development, ultimately reducing crop yields. In the past, many genes that provide tolerance to abiotic stresses have been identified and characterized. Peroxidase (POD) proteins, members of the oxidoreductase enzyme family, play a critical role in protecting plants against abiotic stresses. This study demonstrated a comprehensive investigation of the POD gene family in B. napus . As a result, a total of 109 POD genes were identified across the 19 chromosomes and classified into five distinct subgroups. Further, 44 duplicate events were identified; of these, two gene pairs were tandem and 42 were segmental. Synteny analysis revealed that segmental duplication was more prominent than tandem duplication among POD genes. Expression pattern analysis based on the RNA-seq data of B. napus indicated that BnPOD genes were expressed differently in various tissues; most of them were expressed in roots rather than in other tissues. To validate these findings, we performed RT-qPCR analysis on ten genes; these genes showed various expression levels under abiotic stresses. Our findings not only furnish valuable insights into the evolutionary dynamics of the BnPOD gene family but also serve as a foundation for subsequent investigations into the functional roles of POD genes in B. napus .

The intricate problems concerning energy require innovative solutions. Herein, we propose a smart composite nano system that can be used in a sustainable and dichotomous manner to resolve energy crises. The current study describes a new way to synthesize a pure spinel cobalt oxide (Co 3 O 4 ) and boron (B), nitrogen (N), and sulfur (S) tri-doped Co 3 O 4 -reduced graphite oxide (rGO) nanocomposite (CBNS). A hydrothermal method has been used for the synthesis of these nanomaterials. The synthesized nanocomposite was characterized by UV-visible spectroscopy, X-ray diffraction (XRD), Raman spectroscopy, scanning electron microscopy (SEM), X-ray absorption spectroscopy (XAS), and transmission electron microscopy (TEM). The XRD results showed the formation of Co 3 O 4 and B, N, S doped nanocomposite with high purity and crystallinity. XAS analysis elucidates the formation of spinel Co 3 O 4 with tetrahedral and octahedral arrangement of cobalt ions. The peaks at 2.50 Å and 3.07 Å are due to the Co-Co bonding. The electrocatalytic oxygen reduction (ORR) was successfully implemented using these nanocomposites. The electrochemical study exhibits the better activity of the B, N, and S tri-doped Co 3 O 4 -rGO nanocomposite due to the mutual effect of B, N and S. Singlet molecular oxygen ( 1 O 2 ) has been reported in wide arrays of applications ranging from optoelectronic to photooxygenation reactions and therapy in biomedical proposals. It is also considered a major determinant of photodynamic therapy (PDT) efficacy. Since the direct excitation from the triplet ground state ( 3 O 2 ) of oxygen to the singlet excited state 1 O 2 is spin forbidden; therefore, a rational design and development of heterogeneous sensitizers is remarkably important for the efficient production of 1 O 2 . For this purpose, quantum dots (QDs) have emerged as versatile candidates either by acting individually as sensitizers for 1 O 2 generation or by working in conjunction with other inorganic materials or organic sensitizers by providing them a vast platform. Thus, conjoining the photophysical properties of QDs with other materials, e.g., coupling/combining with other inorganic materials, doping with the transition metal ions or lanthanide ions, and conjugation with a molecular sensitizer provide the opportunity to achieve high-efficiency quantum yields of 1 O 2 which is not possible with either component separately. Rare earth(III) β-diketonates are highly remarkable luminophores in the visible spectral region among the rare earth compounds, owing to the efficient contribution from the 4f-4f intraconfigurational transitions. To get detailed structural insight into the RE 3+ sites (RE = Eu, Gd, and Sm), X-ray absorption near-edge spectroscopy (XANES) can be very potent in probing the local chemical environment around the RE 3+ ion. In this work, a PyFitIt machine learning approach was employed as a new strategy to simulate the Eu, Gd, and Sm L 3 -edge XANES and thereby determine the local atomic structure of the luminescence RE 3+ β-diketonate complexes, [Eu(tta) 3 (H 2 O) 2 ], [C 4 mim][Eu(dbm) 4 ], [Gd(tta) 3 (H 2 O) 2 ], and [Sm(dbm) 3 (phen)] (tta, 3-thenoyltrifluoroacetonate; dbm, dibenzoylmethane; phen, phenanthroline; and C 4 mim, 1-butyl-3-methylimidazolium bromide). Continuous Cauchy wavelet transform validated the PyFitIt calculated XANES by visualizing very efficiently the coordination geometries, composed of O and O/N backscatterers around the RE 3+ (RE = Eu and Gd) and Sm 3+ ions, respectively, as a pinkish-red color map in the two-dimensional images of the corresponding complexes. Extended X-ray absorption fine structure fit in Artemis also corroborated the three-dimensional structures generated by PyFitIt XANES simulation for all the compounds. Though, relatively slightly higher bond distance values for the Sm 3+ complex are due to the higher atomic radius of the Sm 3+ ion when compared to the Eu 3+ and Gd 3+ complexes. Meanwhile, higher Debye-Waller factor (σ 2 ) values for the [C 4 mim][Eu(dbm) 4 ] when compared to the [Eu(tta) 3 (H 2 O) 2 ] indicated the structure disorder, owing to the distortion in the local geometry. It is noteworthy that the optical properties, described mainly by the Ω λ (λ = 2 and 4) 4f-4f intensity parameters, are very sensitive to the local coordination environment around the Eu 3+ ion. Thus, a close agreement between the experimental and theoretically calculated Ω λ parameter values confirmed that the PyFitIt calculated square antiprismatic structures are precisely similar to the real structures of the Eu 3+ complexes.

The GRAS gene family is one of the most important families of transcriptional factors that have diverse functions in plant growth and developmental processes including axillary meristem patterning, signal-transduction, cell maintenance, phytohormone and light signaling. Despite their importance, the function of GRAS genes in pitaya fruit ( Selenicereus undatus L.) remains unknown. Here, 45 members of the HuGRAS gene family were identified in the pitaya genome, which was distributed on 11 chromosomes. All 45 members of HuGRAS were grouped into nine subfamilies using phylogenetic analysis with six other species: maize, rice, soybeans, tomatoes, Medicago truncatula and Arabidopsis. Among the 45 genes, 12 genes were selected from RNA-Seq data due to their higher expression in different plant tissues of pitaya. In order to verify the RNA-Seq data, these 12 HuGRAS genes were subjected for qRT-PCR validation. Nine HuGRAS genes exhibited higher relative expression in different tissues of the plant. These nine genes which were categorized into six subfamilies inlcuding DELLA ( HuGRAS-1 ), SCL-3 ( HuGRAS-7 ), PAT1 ( HuGRAS-34 , HuGRAS-35 , HuGRAS-41 ), HAM ( HuGRAS-37 ), SCR ( HuGRAS-12 ) and LISCL ( HuGRAS-18 , HuGRAS-25 ) might regulate growth and development in the pitaya plant. The results of the present study provide valuable information to improve tropical pitaya through a molecular and conventional breeding program.

Self-medication (SM) is characterized by the procurement and use of medicines by bypassing primary healthcare services and without consulting a physician, usually to manage acute symptoms of self-diagnosed illnesses. Due to the limited availability of primary healthcare services and the anxiety associated with the COVID-19 pandemic, the compulsion to SM by the public has increased considerably. The study aimed to assess the characteristics, practices, and associated factors of SM by the public during the COVID-19 pandemic in Sargodha, Pakistan. χ2-tests and univariable analyses were conducted to explore the identification of characteristics and the potential contributing factors for SM during COVID-19, while multivariable logistic regression models were run to study the effect of variables that maintained a significant association. The study was performed during July−September 2021, with n = 460 questionnaires returned overall (response rate: 99.5%). The majority of respondents were males (58.7%, n = 270) who live in the periphery of the town (63.9%, n = 294), and most of the respondents belonged to the age group of 18−28 years (73.3%, n = 339). A large number, 46.1% (n = 212), of the participants were tested for COVID-19 during the pandemic, and among them, 34.3% (n = 158) practiced SM during the pandemic; the most common source of obtaining medicines was requesting them directly from a pharmacy (25.0%; n = 127). The chances of practicing SM for medical health professionals were 1.482 (p-value = 0.046) times greater than for non-medical health personnel. The likelihood of practicing SM in participants whose COVID-19 test was positive was 7.688 (p-value < 0.001) times more than who did not test for COVID-19. Allopathic medicines, acetaminophen (23.6%), azithromycin (14,9%), and cough syrups (13%), and over the counter (OTC) pharmaceuticals, vitamin oral supplements, such as Vitamin C (39.1%), folic acid (23.5%), and calcium (22.6%), were the most commonly consumed medicines and supplements, respectively; being a healthcare professional or having a COVID-test prior showed a significant association with the usage of Vitamin C (p < 0.05 in all cases). Respondents who mentioned unavailability of the physician and difficulty in travelling/reaching healthcare professionals were found 2.062-times (p-value = 0.004) and 1.862-times (p-value = 0.021) more likely to practice SM, respectively; SM due to fear of COVID was more common in individuals who had received COVID-tests prior (p = 0.004). Practices of SM were observed at alarming levels among our participants. Consciousness and understanding about the possible adverse effects of SM must be established and validated on a continuous level; in addition, on a commercial level, collaboration from pharmacists not to sell products (especially prescription-only medicines) without a certified prescription must be developed and implemented.

Yellow leaf disease (YLD) has been a major limiting factor threatening areca palm commonly known as betel palm ( Areca catechu L.) plantations in Hainan, China. The YLD disease is closely associated with areca palm velarivirus 1 (APV1), which belongs to the family Closteroviridae . YLD-affected betel palms show more serious yellowing symptoms in winter than in summer based on anecdotal observations. In the present work, the underlying mechanism was investigated. We first observed that the severity of YLD symptoms was closely related with the APV1 viral titer determined by qRT-PCR and ELISA under natural conditions. To further investigate whether temperature plays a key role in APV1 accumulation, the areca palm seedlings were artificially inoculated with APV1-positive mealybugs ( Ferrisia virgata ) and then cultivated under controlled conditions. According to our results, the YLD symptoms severity in inoculated seedlings were closely associated with temperature, e.g., severest symptoms at low temperature (16/22 ± 2°C, night/day), severer symptoms at room temperature (24/26 ± 2°C, night/day), while moderate symptoms at high temperature (27/34 ± 2°C, night/day). The qRT-PCR and ELISA results showed that APV1 titer accumulates significantly abundant at low temperature as compared to high and room temperatures. Although beneficial metalloid silicon (Si) has been proven to reduce the toxicity of several heavy metals, there is a lack of understanding regarding Si potential function in mitigating phytotoxicity induced by vanadium (V). In this study, effect of Si (1.5 mM) on growth, biomass production, V uptake, reactive oxygen species (ROS), methylglyoxal (MG) formation, selected antioxidants enzymes activities, glyoxalase enzymes under V stress (35 mg L -1 ) was investigated in hydroponic experiment. The results showed that V stress reduced rice growth, caused V accumulation in rice. Addition of Si to the nutritional medium increased plant growth, biomass yield, root length, root diameter, chlorophyll parameters, photosynthetic assimilation, ion leakage, antioxidant enzymes activities under V stress. Notably, Si sustained V-homeostasis and alleviated V caused oxidative stress by boosting ascorbate (AsA) levels and the activity of antioxidant enzymes in V stressed rice plants. Furthermore, Si protected rice seedlings against the harmful effects of methylglyoxal by increasing the activity of glyoxalase enzymes. Additionally, Si increased the expression of numerous genes involved in the detoxification of reactive oxygen species (e.g., OsCuZnSOD1, OsCaTB, OsGPX1, OsAPX1, OsGR2, and OsGSTU37) and methylglyoxal (e.g., OsGLYI-1 and OsGLYII-2). XAFS/XRF is a general-purpose absorption spectroscopy beamline at the Synchrotron-light for Experimental Science and Applications in the Middle East (SESAME), Jordan. Herein, its optical layout is presented along with its powerful capabilities in collecting absorption and fluorescence spectra within a wide energy range (4.7-30 keV). The beamline is equipped with a conventional fixed-exit double-crystal monochromator that allows the collection of an X-ray absorption spectrum within a few minutes in step-by-step mode. An on-the-fly scanning mode will be implemented shortly where the acquisition time will be reduced to less than a minute per scan. The full automation of the beamline allows performing successive measurements under different conditions. The different experimental setups and special features available to users are reported. Examples of XRF and XAFS measurements are presented, showing the performance of the beamline under different standard conditions., (open access.)

The goal of this work was to synthesize new cerium oxide-based nano-additives to minimise emissions from spark ignition (SI) engines fueled with gasoline blends, such as carbon monoxide (CO), unburned hydrocarbons (HC) and oxides of nitrogen (NO x ). To investigate the effect of transition metal dopants on their respective catalytic oxidation activity, nano-sized CeO 2 catalysts co-doped with Mn, Fe, Cu and Ag ions were successfully produced by a simple hydrothermal technique. The synthesis of nano-catalysts with cubic fluorite geometry was confirmed by XRD data. The addition of transition metal ions to the CeO 2 lattice increased the concentration of structural defects like oxygen vacancies and Ce 3+ ions, which are advantageous for the catalytic oxidation reaction, as also supported by XAFS and RAMAN analysis. Further, nano-gasoline fuel emission parameters are measured and compared to straight gasoline fuel. The results demonstrated that harmful exhaust pollutants such as CO, HC and NO x were significantly reduced. Yellow leaf disease (YLD) is the most destructive disease of betel palm ( Areca catechu ). A strong association between YLD and areca palm velarivirus 1 (APV1) has been observed. However, the causal relationship between APV1 and disease, and the transmission mode, warrant further investigation. This work showed that APV1 was transmitted by both Ferrisia virgata and Pseudococcus cryptus mealybugs and caused YLD symptoms in betel palm seedlings; therefore, we demonstrate that APV1 is a causal agent of YLD. APV1 was detected in the stylets, foreguts, midguts, and hindguts of the vectors via both immunocapture reverse transcription PCR and immunofluorescence assays. APV1 was not transmitted transovarially from viruliferous female F . virgata to their progeny. In summary, the transmission of APV1 by F . virgata may occur in a noncirculative, semipersistent manner. This study fills important gaps in our knowledge of velarivirus transmission, which is critical for developing YLD management practices.

Tomato is an important vegetable that is highly sensitive to drought (DR) stress which impairs the development of tomato seedlings. Recently, melatonin (ME) has emerged as a nontoxic, regulatory biomolecule that regulates plant growth and enhances the DR tolerance mechanism in plants. The present study was conducted to examine the defensive role of ME in photosynthesis, root architecture, and the antioxidant enzymes' activities of tomato seedlings subjected to DR stress. Our results indicated that DR stress strongly suppressed growth and biomass production, inhibited photosynthesis, negatively affected root morphology, and reduced photosynthetic pigments in tomato seedlings. Per contra, soluble sugars, proline, and ROS (reactive oxygen species) were suggested to be improved in seedlings under DR stress. Conversely, ME (100 µM) pretreatment improved the detrimental-effect of DR by restoring chlorophyll content, root architecture, gas exchange parameters and plant growth attributes compared with DR-group only. Moreover, ME supplementation also mitigated the antioxidant enzymes [APX (ascorbate peroxidase), CAT (catalase), DHAR (dehydroascorbate reductase), GST (glutathione S-transferase), GR (glutathione reductase), MDHAR (monodehydroascorbate reductase), POD (peroxidase), and SOD (superoxide dismutase)], non-enzymatic antioxidant [AsA (ascorbate), DHA (dehydroascorbic acid), GSH (glutathione), and GSSG, (oxidized glutathione)] activities, reduced oxidative damage [EL (electrolyte leakage), H 2 O 2 (hydrogen peroxide), MDA (malondialdehyde), and O 2 •- (superoxide ion)] and osmoregulation (soluble sugars and proline) of tomato seedlings, by regulating gene expression for SOD , CAT , APX , GR , POD , GST , DHAR , and MDHAR . These findings determine that ME pretreatment could efficiently improve the seedlings growth, root characteristics, leaf photosynthesis and antioxidant machinery under DR stress and thereby increasing the seedlings' adaptability to DR stress.

The development of QDs-based fluorescent bionanoprobe for cellular imaging fundamentally relies upon the precise knowledge of particle-cell interaction, optical properties of QDs inside and outside of the cell, movement of a particle in and out of the cell, and the fate of particle. We reported engineering and physicochemical characterization of water-dispersible Eu 3+ /Mn 2+ co-doped ZnSe@ZnS core/shell QDs and studied their potential as a bionanoprobe for biomedical applications, evaluating their biocompatibility, fluorescence behaviour by CytoViva dual mode fluorescence imaging, time-dependent uptake, endocytosis and exocytosis in RAW 264.7 macrophages. The oxidation state and local atomic structure of the Eu dopant studied by X-ray absorption fine structure (XAFS) analysis manifested that the Eu 3+ ions occupied sites in both ZnSe and ZnS lattices for the core/shell QDs. A novel approach was developed to relieve the excitation constraint of wide bandgap ZnSe by co-incorporation of Eu 3+ /Mn 2+ codopants, enabling the QDs to be excited at a wide UV-visible range. The QDs displayed tunable emission colors by a gradual increase in Eu 3+ concentration at a fixed amount of Mn 2+ , systematically enhancing the Mn 2+ emission intensity via energy transfer from the Eu 3+ to Mn 2+ ion. The ZnSe:Eu 3+ /Mn 2+ @ZnS QDs presented high cell viability above 85% and induced no cell activation. The detailed analyses of QDs-treated cells by dual mode fluorescence CytoViva microscopy confirmed the systematic color-tunable fluorescence and its intensity enhances as a function of incubation time. The QDs were internalized by the cells predominantly via macropinocytosis and other lipid raft-mediated endocytic pathways, retaining an efficient amount for 24 h. The unique color tunability and consistent high intensity emission make these QDs useful for developing a multiplex fluorescent bionanoprobe, activatable in wide-visible region.

Background: Areca palm (Areca catechu L.) is an important commercial crop in southeast Asia, but its cultivation is threatened by yellowing leaf disease (YLD). Areca palm velarivirus 1 (APV1) was recently associated with YLD, but little is known regarding its population and genetic diversity. To assess the diversity of YLD, the APV1 genome was sequenced in YLD samples collected from different sites in Hainan., Results: Twenty new and complete APV1 genomes were identified. The APV1 isolates had highly conserved sequences in seven open reading frames (ORFs; > 95% nucleotide [nt] identity) at the 3' terminal, but there was diversity (81-87% nt identity) in three ORFs at the 5' terminal. Phylogenetic analysis divided the APV1 isolates into three phylogroups, with 16 isolates (> 70%) in phylogroup A. Mixed infections with different genotypes in the same tree were identified; this was closely correlated with higher levels of genetic recombination., Conclusions: Phylogroup A is the most prevalent APV1 genotype in areca palm plantations in Hainan, China. Mixed infection with different genotypes can lead to genomic recombination of APV1. Our data provide a foundation for accurate diagnostics, characterization of etiology, and elucidation of the evolutionary relationships of APV1 populations., (© 2021. The Author(s).)

Graphene oxide (GO) is a promising and strategic carbon-based nanomaterial for innovative and disruptive technologies. It is therefore essential to address its environmental health and safety aspects. In this work, we evaluated the chemical degradation of graphene oxide by sodium hypochlorite (NaClO, bleach water) and its consequences over toxicity, on the nematode Caenorhabditis elegans. The morphological, chemical, and structural properties of GO and its degraded product, termed NaClO-GO, were characterized, exploring an integrated approach. After the chemical degradation of GO at room temperature, its flake size was reduced from 156 to 29 nm, while NaClO-GO showed changes in UV-vis absorption, and an increase in the amount of oxygenated surface groups, which dramatically improved its colloidal stability in moderately hard reconstituted water (EPA medium). Acute and chronic exposure endpoints (survival, growth, fertility, and reproduction) were monitored to evaluate material toxicities. NaClO-GO presented lower toxicity at all endpoints. For example, an increase of over 100% in nematode survival was verified for the degraded material when compared to GO at 10 mg L -1 . Additionally, enhanced dark-field hyperspectral microscopy confirmed the oral uptake of both materials by C. elegans. Finally, this work represents a new contribution toward a better understanding of the links between the transformation of graphene-based materials and nanotoxicity effects (mitigation), which is mandatory for the safety improvements that are required to maximize nanotechnological benefits to society., (Copyright © 2021 Elsevier Ltd. This work reports an integrated nanosafety study including the synthesis and characterization of the graphene oxide-silver nanoparticle hybrid material (GO-AgNPs) and its nano-ecotoxicity evaluation in the zebrafish embryo model. The influences of natural organic matter (NOM) and a chorion embryo membrane were considered in this study, looking towards more environmentally realistic scenarios and standardized nanotoxicity testing. The nanohybrid was successfully synthesized using the NaBH 4 aqueous method, and AgNPs (~ 5.8 nm) were evenly distributed over the GO surface. GO-AgNPs showed a dose-response acute toxicity: the LC 50 was 1.5 mg L -1 for chorionated embryos. The removal of chorion, however, increased this toxic effect by 50%. Furthermore, the presence of NOM mitigated mortality, and LC 50 for GO-AgNPs changed respectively from 2.3 to 1.2 mg L -1 for chorionated and de-chorionated embryos. Raman spectroscopy confirmed the ingestion of GO by embryos; but without displaying acute toxicity up to 100 mg L -1 , indicating that the silver drove toxicity down. Additionally, it was observed that silver nanoparticle dissolution has a minimal effect on these observed toxicity results. Finally, understanding the influence of chorion membranes and NOM is a critical step towards the standardization of testing for zebrafish embryo toxicity in safety assessments and regulatory issues., (Copyright © 2020 The Authors. The use of hybrid nanostructures based on magneto-luminescent properties is a promising strategy for nano-bio applications and theranostics platforms. In this work, we carried out the synthesis and functionalization of iron oxide nanocubes (IONCs) to obtain multifunctional hybrid nanostructures towards biomedical applications. The IONCs were functionalized with tetraethylorthosilicate, thenoyltrifluoroacetone-propyl-triethoxysilane and europium(iii)-dibenzoylmethane complexes to obtain the materials termed as IOCNCs@SiO 2 , IONCs@SiO 2 TTA, IONCs@SiO 2 TTA-Eu and IONCs@SiO 2 -TTA-Eu-DBM, respectively. Then, the biological interactions of these nanostructures with red blood cells - RBCs (hemolysis) and human blood plasma (protein corona formation) were evaluated. The XPS spectrocopy and EDS chemical mapping analysis showed that each domain is homogeneously occupied in the hybrid material, with the magnetic core at the center and the luminescent domain on the surface of the hybrid nanomaterial with a core@shell like structure. Futhermore, after each functionalization step, the nanomaterial surface charge drastically changed, with critical impact on RBC lysis and corona formation. While IONCs@SiO 2 and IONCs@SiO 2 -TTA-Eu-DBM showed hemolytic properties in a dose-dependent manner, the IONCs@SiO 2 TTA-Eu did not present any hemolytic effect up to 300 μg mL -1 . Protein corona results showed a pattern of selective adsorption of proteins with each surface of the synthesized hybrid materials. However, as a general result, a suppression of hemolysis after protein corona formation in all tests was verified. Finally, this study provides a solid background for further applications of these hybrid magneto-luminescent materials containing new surface functionalities in the emerging field of medical nanobiotechnology.

Gadolinium oxysulfate doped with terbium (Gd 2 O 2 SO 4 :Tb 3+ ; 0.1, 1.0, and 10.0 mol%) materials were obtained using thermal decomposition from sulfate hydrate under a dynamic air atmosphere and between 1320-1400 K. The materials were characterized using Fourier transform infrared spectroscopy, thermogravimetric/derivative thermogravimetric investigations and X-ray powder diffraction patterns. The Tb 2 O 2 SO 4 compound was obtained at 1300 K and was used to compare thermal stability and photoluminescence behaviour with that of Gd 2 O 2 SO 4 :Tb 3+ (0.1, 1.0, and 10.0 mol%). Magnetic susceptibility measurements indicated the presence of 15% Tb 4+ phases within Tb 2 O 2 SO 4 . The materials were excited at 377 nm and displayed green narrow lines with the strongest emission peak at 545.5 nm due to the 5 D 4 → 7 F 5 transition of Tb 3+ ions. Brightness of terbium-activated gadolinium oxysulfate phosphors was enhanced with increase in the concentration of Tb 3+ . Detailed analysis of spectroscopic properties of materials under investigations revealed efficient Gd 2 O 2 SO 4 to Tb 3+ and Tb 3+ to Tb 3+ energy transfers. Increase in dopant concentration led to the enhancement of 5 D 4 → 7 F J emission intensity and reduction of 5 D 3 → 7 F J emission intensity via cross-relaxation mechanisms. Distribution of particle size was increased by controlling dopant concentration in the host lattice. In this work, we evaluated the effect of protein corona formation on graphene oxide (GO) mixture toxicity testing (i.e., co-exposure) using the Daphnia magna model and assessing acute toxicity determined as immobilisation. Cadmium (Cd 2+ ) and bovine serum albumin (BSA) were selected as co-pollutant and protein model system, respectively. Albumin corona formation on GO dramatically increased its colloidal stability (ca. 60%) and Cd 2+ adsorption capacity (ca. 4.5 times) in reconstituted water ( Daphnia medium). The acute toxicity values (48 h-EC 50 ) observed were 0.18 mg L -1 for Cd 2+ -only and 0.29 and 0.61 mg L -1 following co-exposure of Cd 2+ with GO and BSA@GO materials, respectively, at a fixed non-toxic concentration of 1.0 mg L -1 . After coronation of GO with BSA, a reduction in cadmium toxicity of 110 % and 238% was achieved when compared to bare GO and Cd 2+ -only, respectively. Integration of datasets associated with graphene-based materials, heavy metals and mixture toxicity is essential to enable re-use of the data and facilitate nanoinformatics approaches for design of safer nanomaterials for water quality monitoring and remediation technologies. Hence, all data from this work were annotated and integrated into the NanoCommons Knowledge Base, connecting the experimental data to nanoinformatics platforms under the FAIR data principles and making them interoperable with similar datasets.

The increasing production and use of nanomaterials is causing serious concerns about their safety to human and environmental health. However, the applications of titanium dioxide nanoparticles (TiO 2 NP) and multiwalled carbon nanotubes (MWCNT) hybrids has grown considerably, due to their enhanced photocatalytic efficiency. To our knowledge, there are no reports available to the scientific community about their toxicity. In this work, we perform a toxicity assessment of TiO 2 NP and TiO 2 -MWCNT nanohybrid materials using Zebrafish embryos standardized 96 h early life stage assay, under different exposure conditions (with and without UV light exposure). After exposure the parameters assessed were acute toxicity, hatching rate, growth, yolk sac size, and sarcomere length. In addition, μ-probe X-ray fluorescence spectroscopy (µ-XRF) was employed to observe if nanoparticles were uptaken by zebrafish embryos and consequently accumulated in their organisms. Neither TiO 2 NP nor TiO 2 -MWCNT nanohybrids presented acute toxicity to the zebrafish embryos. Moreover, TiO 2 NP presents sublethal effects for total length (with and without UV light exposure) on the embryos. Luminescence-tunable multicolored LaF 3 :xCe 3+ ,xGd 3+ ,yEu 3+ (x = 5; y = 1, 5, 10, and 15 mol%) nanoparticles have been synthesized via a low cost polyol method. Powder X-ray diffraction and high-resolution transmission electron microscopy studies confirm the hexagonal phase of the LaF 3 :xCe 3+ ,xGd 3+ ,yEu 3+ nanophosphors with average sizes (oval shape) ranging from 5 to 7 nm. Energy-dispersive X-ray spectroscopy analyses show the uniform distribution of Ce 3+ , Gd 3+ , and Eu 3+ dopants in the LaF 3 host matrix. The photoluminescence spectra and electron paramagnetic resonance measurements guarantee the presence of Eu 2+ , corroborated through DC susceptibility measurements of the samples displaying paramagnetic behavior at 300 K, whereas weak ferromagnetic ordering is shown at 2 K. The non-radiative energy transfer processes from the 4f( 2 F 5/2 ) → 5d state (Ce 3+ ) to the intraconfigurational 4f excited levels of rare earth ions and simultaneous emissions in the visible region from the 4f 6 5d 1 (Eu 2+ ) and 5 D 0 (Eu 3+ ) emitting levels, leading to overlapped broad and narrow emission bands, have been proclaimed. The energy transfer mechanism proposes involvement of the Gd 3+ ion sub-lattice as the bridge and finally trapping by Eu 2+/3+ , upon excitation of the Ce 3+ ion. The calculation of experimental intensity parameters (Ω 2,4 ) has been discussed and the highest emission quantum efficiency (η = 85%) of the Eu 3+ ion for the y = 10 mol% sample is reported. The advantageous existence of the Eu 2+ /Eu 3+ ratio along with variously doped nanomaterials described in this work, results in tunable emission color in the blue-white-red regions, highlighting the potential application of the samples in solid-state lighting devices, scintillation devices, and multiplex detection.

The design of bifunctional magnetic luminescent nanomaterials containing Fe3O4 functionalized with rare earth ion complexes of calixarene and β-diketonate ligands is reported. Their preparation is accessible through a facile one-pot method. These novel Fe3O4@calix-Eu(TTA) (TTA = thenoyltrifluoroacetonate) and Fe3O4@calix-Tb(ACAC) (ACAC = acetylacetonate) magnetic luminescent nanomaterials show interesting superparamagnetic and photonic properties. The magnetic properties (M-H and ZFC/FC measurements) at temperatures of 5 and 300 K were explored to investigate the extent of coating and the crystallinity effect on the saturation magnetization values and blocking temperatures. Even though magnetite is a strong luminescence quencher, the coating of the Fe3O4 nanoparticles with synthetically functionalized rare earth complexes has overcome this difficulty. The intramolecular energy transfer from the T1 excited triplet states of TTA and ACAC ligands to the emitting levels of Eu(3+) and Tb(3+) in the nanomaterials and emission efficiencies are presented and discussed, as well as the structural conclusions from the values of the 4f-4f intensity parameters in the case of the Eu(3+) ion. These novel nanomaterials may act as the emitting layer for the red and green light for magnetic light-converting molecular devices (MLCMDs).

Introduction: Change in junior doctors working pattern has brought effective and safe clinical handover into a central role to ensure the patient safety and high quality care. We investigated whether the compliance and quality of clinical handover could be improved through the use of a standardised and structured handover template., Methods: A computerised template was developed in accordance with handover guidelines provided by the Royal College of Surgeons of England. Pre- and post-intervention audits against an eleven-point dataset pertaining to the handover of acute surgical admissions were undertaken. The results from the two discrete audits periods were compared to examine the impact of intervention., Results: There were 137 acute surgical admissions during pre-intervention and 155 admissions in post-intervention audit period. A significant improvement in overall handover practice was observed in post-intervention period. The documentation of patient hospital number (84 (61%) vs. 132 (85%) p<0.001), past medical history (39 (28%) vs. 75 (48%) p<0.001) and patient assessment by a senior member of the on-call team (3 (2%) vs. 125 (85%) p<0.001) all demonstrated significant improvements upon use of structured template. Compliance to effective handover improved following increased awareness of the importance of safe clinical handover among the junior doctors., Conclusion: Implementation of a standardised guideline-based structured handover template and training of junior doctors are likely to improve compliance to agreed standards, promote quality of care, and protect patient safety., (Copyright © 2012 Surgical Associates Ltd. The authors compared calculations of sunlamp maximum exposure times following current USFDA Guidance Policy on the Maximum Timer Interval and Exposure Schedule, with USFDA/CDRH proposals revising these to equivalent erythemal exposures of ISO/CIE Standard Erythema Dose (SED). In 2003, [USFDA/CDRH proposed replacing their unique CDRH/Lytle] erythema action spectrum with the ISO/CIE erythema action spectrum and revising the sunlamp maximum exposure timer to 600 J m(-2) ISO/CIE effective dose, presented as being biologically equivalent. Preliminary analysis failed to confirm said equivalence, indicating instead ∼38% increased exposure when applying these proposed revisions. To confirm and refine this finding, a collaboration of tanning bed and UV lamp manufacturers compiled 89 UV spectra representing a broad sampling of U.S. indoor tanning equipment. USFDA maximum recommended exposure time (Te) per current sunlamp guidance and CIE erythemal effectiveness per ISO/CIE standard were calculated. The CIE effective dose delivered per Te averaged 456 J(CIE) m(-2) (SD = 0.17) or ∼4.5 SED. The authors found that CDRH's proposed 600 J(CIE) m(-2) recommended maximum sunlamp exposure exceeds current Te erythemal dose by ∼33%. The current USFDA 0.75 MED initial exposure was ∼0.9 SED, consistent with 1.0 SED initial dose in existing international sunlamp standards. As no sunlamps analyzed exceeded 5 SED, a revised maximum exposure of 500 J(CIE) m(-2) (∼80% of CDRH's proposal) should be compatible with existing tanning equipment. A tanning acclimatization schedule is proposed beginning at 1 SED thrice-weekly, increasing uniformly stepwise over 4 wk to a 5 SED maximum exposure in conjunction with a tan maintenance schedule of twice-weekly 5 SED sessions, as biologically equivalent to current USFDA sunlamp policy.

Refeeding syndrome (RFS) describes the biochemical changes, clinical manifestations, and complications that can occur as a consequence of feeding a malnourished catabolic individual. RFS has been recognised in the literature for over fifty years and can result in serious harm and death. Crude estimates of incidence, morbidity, and mortality are available for specific populations. RFS can occur in any individual but more commonly occurs in at-risk populations. Increased awareness amongst healthcare professionals is likely to reduce morbidity and mortality. This review examines the physiology of RFS and describes the clinical manifestations. A management strategy is described. The importance of a multidisciplinary approach is emphasized.

Nonuniformity in the spatial patterning of gap junctions between heart muscle cells is now recognized as an important determinant of electromechanical function in working myocardium. Breakdown of the normal geometry of electrical intercellular connectivity in diseased myocardium correlates with reentry, arrhythmia, and conduction disturbance. The developmental mechanism(s) that determines this precise spatial order in gap junction organization in normal myocardium is at present unknown. To examine this question, we have used immunoelectron and immunoconfocal microscopy to analyze the spatial distributions of gap junctional (connexin43), desmosomal (desmoplakin), and adherens junctional (N-cadherin) components during maturation of rodent and canine left ventricular myocardium. In rats, a striking divergence in the distribution of gap junctions and cell adhesion junctions emerged within the first 20 days of postnatal life. It was found that although gap junctions initially demonstrated dispersed distributions across myocyte cell membranes, desmosomes and adherens junctions showed more rapid polarization toward cell termini (ie, nascent intercalated disks) after birth. Over subsequent postnatal development (20 to 90 postnatal days), gap junctions became progressively concentrated in these cell adhesion junction-rich zones of membrane. Quantitative analyses of this process in a series of rats aged 15 embryonic and 1, 5, 10, 20, 40, 70, and 90 postnatal days indicated that significantly higher levels (P < .01) of N-cadherin and desmoplakin than of connexin43 were immunolocalized to cell termini by as early as postnatal day 5. Although all three junctions types showed increasing polarization to myocyte termini with development, variation between junctions remained significant (P < .05) at all times points between 5 and 70 postnatal days. Only at 90 postnatal days, when the animals were nearly full grown, did the proportions of gap junction, desmosome, and adherens junction at intercalated disks become statistically similar (P > .05). Examination of myocardium from 1- and 3-month-old canines revealed that related differential changes to the spatiotemporal distribution of intercellular junctions occurred during postnatal maturation of the dog heart, suggesting that the process was not rodent specific. It is concluded that this progressive change in the organization and pattern of association between gap junctions and cell adhesion junctions is likely to be an important factor in maturation of electromechanical function within the mammalian heart.
