The current study explored the differential interaction between ethylene (ET) and abscisic acid (ABA) in relation to salt stress in mustard (Brassica juncea L.) plants. Significant reductions in seed germination, growth, and photosynthesis were observed with 100 mmol NaCl. Among the cultivars tested, the Pusa Vijay cultivar was noted as ET-sensitive. Pusa Vijay responded maximally to an application of 2.0 mmol ethephon (Eth; 2-chloethyl phosphonic acid-ethylene source), and exhibited the greatest growth, photosynthesis, activity of 1-aminocyclopropane carboxylic acid (ACC) synthase (ACS), and ET evolution. Notably, Eth (2.0 mmol) more significantly improved the seed germination percentage, germination and vigor index, amylase activity, and reduced H2O2 content under salt stress, while ABA (25 µmol) had negative effects. Moreover, the individual application of Eth and ABA on Pusa Vijay under both optimal and salt-stressed conditions increased the growth and photosynthetic attributes, nitrogen (N) and sulfur (S) assimilation, and antioxidant defense machinery. The addition of aminoethoxyvinylglycine (0.01 µmol AVG, ET biosynthesis inhibitor) to ABA + NaCl-treated plants further added to the effects of ABA on parameters related to seed germination and resulted in less effectiveness of growth and photosynthesis. In contrast, the effects of Eth were seen with the addition of fluoridone (25 µmol Flu, ABA biosynthesis inhibitor) to Eth + NaCl. Thus, it can be suggested that ET is crucial for alleviating salt-induced inhibition in seed germination, growth, and photosynthesis, while ABA collaborated with ET to offer protection by regulating nutrient assimilation and enhancing antioxidant metabolism. These findings provide insight into the complex regulatory processes involved in ET–ABA interaction, enhancing our understanding of plant growth and development and the mitigation of salt stress in mustard. It opens pathways for developing hormonal-based strategies to improve crop productivity and resilience, ultimately benefiting agricultural practices amidst a challenging environment. Soil salinity poses a significant threat to agricultural productivity, impacting the growth and yield of wheat (Triticum aestivum L.) plants. This study investigates the potential of melatonin (MT; 100 µM) and hydrogen sulfide (H2S; 200 µM sodium hydrosulfide, NaHS) to confer the tolerance of wheat plants to 100 mM NaCl. Salinity stress induced the outburst of reactive oxygen species (ROS) resulting in damage to the chloroplast structure, growth, photosynthesis, and yield. Application of either MT or NaHS augmented the activity of antioxidant enzymes, superoxide dismutase, ascorbate peroxidase, glutathione reductase, and reduced glutathione (GSH) levels, upregulated the expression of Na+ transport genes (SOS1, SOS2, SOS3, NHX1), resulting in mitigation of salinity stress. Thus, improved stomatal behavior, gas-exchange parameters, and maintenance of chloroplast structure resulted in enhanced activity of the Calvin cycle enzymes and overall enhancement of growth, photosynthetic, and yield performance of plants under salinity stress. The use of DL-propargylglycine (PAG, an inhibitor of hydrogen sulfide biosynthesis) and p-chlorophenyl alanine (p-CPA, an inhibitor of melatonin biosynthesis) to plants under salt stress showed the comparative necessity of MT and H2S in mitigation of salinity stress. In the presence of PAG, more pronounced detrimental effects were observed than in the presence of p-CPA, emphasizing that MT was involved in mitigating salinity through various potential pathways, one of which was through H2S. PurposeTo define the molecular basis of retinal degeneration in consanguineous Pakistani pedigrees with early onset retinal degeneration.MethodsA cohort of 277 individuals representing 26 pedigrees from the Punjab province of Pakistan was analyzed. Exomes were captured with commercial kits and sequenced on an Illumina HiSeq 2500. Candidate variants were identified using standard tools and analyzed using exomeSuite to detect all potentially pathogenic changes in genes implicated in retinal degeneration. Segregation analysis was performed by dideoxy sequencing and novel variants were additionally investigated for their presence in ethnicity-matched controls.ResultsWe identified a total of nine causal mutations, including six novel variants in RPE65, LCA5, USH2A, CNGB1, FAM161A, CERKL and GUCY2D as the underlying cause of inherited retinal degenerations in 13 of 26 pedigrees. In addition to the causal variants, a total of 200 variants each observed in five or more unrelated pedigrees investigated in this study that were absent from the dbSNP, HapMap, 1000 Genomes, NHLBI ESP6500, and ExAC databases were identified, suggesting that they are common in, and unique to the Pakistani population.ConclusionsWe identified causal mutations associated with retinal degeneration in nearly half of the pedigrees investigated in this study through next generation whole exome sequencing. All novel variants detected in this study through exome sequencing have been cataloged providing a reference database of variants common in, and unique to the Pakistani population.

A significant threat to the ongoing rise in temperature caused by global warming. Plants have many stress-resistance mechanisms, which is responsible for maintaining plant homeostasis. Abiotic stresses largely increase gaseous molecules' synthesis in plants. The study of gaseous signaling molecules has gained attention in recent years. The role of gaseous molecules, such as nitric oxide (NO), hydrogen sulfide (H2S), carbon dioxide (CO2), carbon monoxide (CO), methane (CH4), and ethylene, in plants under temperature high-temperature stress are discussed in the current review. Recent studies revealed the critical function that gaseous molecules play in controlling plant growth and development and their ability to respond to various abiotic stresses. Here, we provide a thorough overview of current advancements that prevent heat stress-related plant damage via gaseous molecules. We also explored and discussed the interaction of gaseous molecules. In addition, we provided an overview of the role played by gaseous molecules in high-temperature stress responses, along with a discussion of the knowledge gaps and how this may affect the development of high-temperature-resistant plant species. Ethylene is an essential plant hormone, critical in various physiological processes. These processes include seed germination, leaf senescence, fruit ripening, and the plant's response to environmental stressors. Ethylene biosynthesis is tightly regulated by two key enzymes, namely 1-aminocyclopropane-1-carboxylate synthase (ACS) and 1-aminocyclopropane-1-carboxylate oxidase (ACO). Initially, the prevailing hypothesis suggested that ACS is the limiting factor in the ethylene biosynthesis pathway. Nevertheless, accumulating evidence from various studies has demonstrated that ACO, under specific circumstances, acts as the rate-limiting enzyme in ethylene production. Under normal developmental processes, ACS and ACO collaborate to maintain balanced ethylene production, ensuring proper plant growth and physiology. However, under abiotic stress conditions, such as drought, salinity, extreme temperatures, or pathogen attack, the regulation of ethylene biosynthesis becomes critical for plants' survival. This review highlights the structural characteristics and examines the transcriptional, post-transcriptional, and post-translational regulation of ACS and ACO and their role under abiotic stress conditions. Reviews on the role of ethylene signaling in abiotic stress adaptation are available. However, a review delineating the role of ACS and ACO in abiotic stress acclimation is unavailable. Exploring how particular ACS and ACO isoforms contribute to a specific plant's response to various abiotic stresses and understanding how they are regulated can guide the development of focused strategies. Salinity is a significant environmental stressor that impacts plant growth and development, leading to severe loss of agricultural output across the world. One way to reduce salt-induced detrimental effects is with the use of hormones. Through synergistic or antagonistic crosstalk with other hormones, phytohormones regulate multiple signals required for growth and development under normal and environmental stress conditions. Ethylene is a well-known phytohormone involved in salt tolerance. Its signaling component EIN3/EIL has a positive regulatory effect in salinity tolerance that is mediated by the AP2/ERF transcription factor, through which ethylene maintains ionic and osmotic homeostasis, fine-tunes photosynthesis, and reduces salt-induced reactive oxygen species production in stressful environments. Furthermore, a negative regulator of ethylene signaling, CTR1, was found to have a positive role in stabilizing EIN3 and improving resistance to salinity. However, certain studies have demonstrated overproduction of ethylene and its signaling to have negative effects on plant growth and development, contradictory to its established positive role. Therefore, it can be considered that ethylene is involved in salinity tolerance and its interplay with other hormones enables it to finely regulate plant salinity response. This review focuses on a) a general introduction of salt stress, its impact on plants, and the responses of plants under salinity stress; b) the role of ethylene in salinity tolerance, along with its biosynthesis and signaling; c) an overview of isoprenoids and the synthesis of their derived phytohormones brassinosteroids, gibberellic acid, abscisic acid, cytokinins, and strigolactones; and d) crosstalk of ethylene with isoprenoid-derived phytohormones under salt stress. • Ethylene signaling in salt tolerance. • Isoprenoid-derived signaling molecules in salt tolerance. • Ethylene crosstalk with isoprenoid-derived hormones.
