Acquired supracarinal tracheal stenosis can be defined as postintubation tracheal stenosis of the lower third of the trachea that cannot be bypassed by standard or adjustable flange tracheostomy tubes. Owing to difficulty of access in the deep trachea, the transoral laser-assisted endoscopic surgical management of such stenoses is a challenge both for the surgeon and the anaesthetist alike. It requires good rapport and efficient cooperation between the teams to ensure appropriate oxygenation and carbon-dioxide washout while providing intermittent apnoeic phases to the surgeon for clear visualisation of the target area and use of Diode laser which is otherwise hazardous in an oxygen/nitrous oxide rich environment. From our tertiary care referral centre with strong work load for such pathologies, we report one such case of acquired tracheal stenosis successfully managed with transoral Diode laser-assisted surgery. Schwannomas are benign peripheral nerve sheath tumours with a low risk of malignant transformation. About 25%–40% are in the head and neck region with the posterior third of the tongue being a rather rare site of its growth, and a lesion at this site is understandably difficult to approach and treat. Being benign and encapsulated, surgical excision is in the mainstay of treatment. Symptoms can range from a globus pharyngeus and dysphagia to airway compromise depending on the size and site. Traditionally, a paramedian lip split approach with paramedian mandibulotomy and mandible swing may be used. However, some recent reports of the use of carbon dioxide laser and robotic surgery for tongue base lesions are seen in the literature. Our case report is a unique addition to the management strategies for such tongue base lesions as we employed diode 980 nm laser using a minimally invasive transoral approach with a successful outcome. Introduction: Leptospirosis affects a wide range of mammals, humans, and even a few poikilothermic animal species. In Pakistan, serological studies of equine leptospirosis have reported a prevalence of over 40%, but no study has ever been conducted towards molecular detection of Leptospira in horses. Material and Methods: Blood samples from 128 horses were screened using ELISA and 41 positive samples were examined for the presence of leptospiral DNA using specific primers for 16S rRNA gene. Results: Out of 41 tested samples, 20 samples were found to be PCR-positive, revealing a fragment of 306 bp after gel electrophoresis. Sequencing and phylogenetic analysis of positive samples revealed circulation of pathogenic Leptospira spp. in Pakistani horses. No evidence of circulation of intermediate species was found in this study. Conclusion: This study reports the first molecular evidence of equine leptospirosis in Pakistan and lays ground for further research in this area. It also confirms the efficiency of 16S rRNA for the diagnosis of equine leptospirosis.

Androgenesis is reported in a few tree crops such as citrus and papaya; however, there is little information available about guava. Different modified Chu (N6) media and cold pretreatments of floral buds were tested to induce embryogenic calli in the anthers of two white flesh guava cultivars, 'Round' and 'Pyriform'. Four modified Chu (N6) media formulations, i.e., M1 (400 mgL-1 casein hydrolysate, 200 mgL-1 glutamine, 0.2 mgL-1NAA, 1.0 mgL-1 KIN, 0.5 mgL-1 6-BAP, 0.5 mgL-1 zeatin and 5% coconut water), M2 (400 mgL-1 casein hydrolysate, 200 mgL-1 glutamine, 0.2 mgL-1NAA, 1.0 mgL-1 KIN, 0.5 mgL-1 6-BAP), M10 (0.5 mgL-1 2,4-D), and M11 (0.5 mgL-1 2,4-D and 50 mgL-1 PVP) enhanced anther swelling and callus formation. Genotypic variability was found in pollen viability, phenolic exudation, and anther tissue browning after cold pretreatment for 0-72 hrs. intervals. Anthers of 'Round' showed more pollen viability across cold pretreatment intervals with less phenolic exudations and tissue browning than 'Pyriform'. On both M10 and M11 media, anthers with uni-nucleate microspores developed embryogenic calli; however, anthers of 'Pyriform' resulted in more calli induction (69.58%) on M10 media, whereas anthers of 'Round' developed more calli (66.60%) on M11 media. The proliferating calli in white flesh 'Round' and 'Pyriform' cultivars and pink flesh 'Round' cultivar formed more embryogenic masses upon transfer to growth hormone-free Chu (N6) media under light conditions; however, embryos showed delayed maturity with limited germination. More genotypes shall be explored to enhance embryo germination, and embryogenic media shall be further fine-tuned. We developed a molecular marker for MAS of mungbean resistant varieties against CLS from the consensus sequence (MB-CLsRG) of identified RGAs (MB-ClsRCaG1 and MB-ClsRCaG2). The MB-CLsRG sequence-specific primer pair was used to screen Cercospora leaf spot (CLS) resistant varieties of mungbean in genomic analysis that showed congruency with phenotypic screening. Validation of molecular marker linkage with CLS resistance was performed using rtPCR in transcriptomic analysis. The sequenced PCR products showed 100% homology with MB-CLsRG sequence and putative disease resistance proteins that confirmed the linkage of molecular marker with CLS resistance in mungbean. The antifungal potential of MB-CLsRG gene encoding protein was assessed. The MB-CLsRG gene sequence was cloned in the E. coli expression vector for recombinant protein production. The recombinant protein was then investigated for its in vitro antifungal potential against Cercospora canescens. The in vitro investigation showed strong antifungal activity of recombinant protein as it restricted the growth of fungal mycelial mass. The results validated the linkage of developed marker with CLS-resistant mungbean varieties; therefore, it can be used to screen resistant varieties from a large population in MAS. Moreover, the recombinant protein of the MB-CLsRG gene sequence revealed antifungal potential, which proved the gene sequence could be suitable to use in transgenic plants technology to develop fungal-resistant transgenic crops.
