19. The Role of Activator Protein-1 Complex in Diabetes-Associated Atherosclerosis: Insights From Single-Cell RNA Sequencing.

Abstract

Despite advances in treatment, atherosclerotic cardiovascular disease remains the leading cause of death in patients with diabetes. Even when risk factors are mitigated, the disease progresses, and thus, newer targets need to be identified that directly inhibit the underlying pathobiology of atherosclerosis in diabetes. A single-cell sequencing approach was used to distinguish the proatherogenic transcriptional profile in aortic cells in diabetes using a streptozotocin-induced diabetic Apoe −/− mouse model. Human carotid endarterectomy specimens from individuals with and without diabetes were also evaluated via immunohistochemical analysis. Further mechanistic studies were performed in human aortic endothelial cells (HAECs) and human THP-1–derived macrophages. We then performed a preclinical study using an activator protein-1 (AP-1) inhibitor in a diabetic Apoe −/− mouse model. Single-cell RNA sequencing analysis identified the AP-1 complex as a novel target in diabetes-associated atherosclerosis. AP-1 levels were elevated in carotid endarterectomy specimens from individuals with diabetes compared with those without diabetes. AP-1 was validated as a mechanosensitive transcription factor via immunofluorescence staining for regional heterogeneity of endothelial cells of the aortic region exposed to turbulent blood flow and by performing microfluidics experiments in HAECs. AP-1 inhibition with T-5224 blunted endothelial cell activation as assessed by a monocyte adhesion assay and expression of genes relevant to endothelial function. Furthermore, AP-1 inhibition attenuated foam cell formation. Critically, treatment with T-5224 attenuated atherosclerosis development in diabetic Apoe −/− mice. This study has identified the AP-1 complex as a novel target, the inhibition of which treats the underlying pathobiology of atherosclerosis in diabetes. Article Highlights: The cell-specific transcriptional profile of vascular cells in diabetes-associated atherosclerosis is not known. Single-cell RNA sequencing analysis not only defined the transcriptional profile of vascular cells in diabetes but also identified the activator protein-1 (AP-1) complex as one of the important transcription factor complexes in flow-mediated endothelial cell activation and foam cell formation. Although previous studies have implicated AP-1 in atherosclerosis, our study has defined the role of AP-1 as a central regulator of a gene expression program linked to endothelial dysfunction and foam cell formation in diabetes. Abstract

Wireless Sensor Networks are basically used for gathering information needed by smart environments but they are particularly useful in unattended situations where terrain, climate and other environmental constraints may hinder in the deployment of wired/conventional networks. Unlike traditional networks, these sensor networks do not have a continuous power supply at their disposal. Rather the individual sensors are battery operated and the lifetime of the individual sensors and thus the overall network depend heavily on duty cycle of these sensors. Analysis on WSNs shows that communication module is the main part which consumes most of the sensor energy and that is why energy conservation is the major optimization goal. Since routing protocols and MAC protocols directly access the communication module therefore the design of protocols in these two domains should take into account the energy conservation goal. In this paper, we discuss different state-of-the-art protocols both in MAC and routing domains that have been proposed for WSNs to achieve the overall goal of prolonging the network lifetime. The routing protocols in WSNs are generally categorized into three groups - data centric, hierarchical and location-based but we focus on only the first two categories because location-based routing protocols generally require a prior knowledge about sensors location which most of the times is not available due to random deployment of the sensors. We then discuss how schedule-based and contention-based MAC protocols can contribute to achieve optimal utilization of the limited energy resource by avoiding or reducing the chances of collisions and thus the need for retransmission., Comment: http://www.journalofcomputing.org

Abstract

This letter aims to explore the potential impact of the Turkey-Syria earthquake on the psychological well-being of the affected children and adolescents. It emphasises the crucial importance of prompt identification and management of emerging mental health disorders in this vulnerable population. The letter draws on existing research evidence to highlight the need for suitable mental health interventions to mitigate the anticipated suffering of many children and adolescents affected by the earthquake. A comprehensive overview of the mediating factors which may play a role in the extent of the impact of an earthquake on the mental health of children is discussed. A range of appropriate strategies and interventions are recommended and the call for continued global support is renewed. We have concluded that major earthquakes can potentially contribute to the development of mental health disorders among children and adolescents. However, by providing timely and effective support, it is possible to prevent long-term psychological consequences and facilitate early recovery. We propose the urgent implementation of effective mental health interventions in the aftermath of the Turkey-Syria earthquake to foster the recovery and resilience of the affected young population. Mental health disorders are more prevalent in children affected by earthquakes. Prompt identification of emerging mental health disorders among this population is crucial. Management of the psychological impact in this context must be practical and needs-based. A focused and continued global support response is needed to manage the long-term challenges.
