The intensification of aquaculture has led to a rise in fish infections, necessitating the search for alternative antibiotics. In this context, our study investigated the effects of dietary supplementation with Chaetomorpha aerea, a filamentous green algae, on the immune health and resistance to infections in tilapia (Oreochromis mossambicus). Diets containing varying concentrations of C. aerea (0, 1, 2, 5, and 10 g/kg) were prepared and administered to the fish for 30 days, followed by a challenge with Edwardsiella tarda to evaluate survival rates. The results were significant. The diet containing 5 g/kg of C. aerea (group T3) brought about substantial improvements in hematological parameters, including increases in red blood cell count (RBC), hematocrit (Hct), and hemoglobin (Hb). The T3 group exhibited a robust immune response, with higher lysozyme and ceruloplasmin activity in immunological assays. LBP gene expression was significantly elevated in the spleen and thymus of fish in the T3 group, which correlated with higher survival after bacterial challenge compared to the control group. Principal Component Analysis (PCA) and cluster analysis confirmed that the 5 g/kg concentration stood out for maximizing immunological benefits without compromising the overall health of the fish. These findings highlight the robust immune response in the T3 group, a key finding of our study. We conclude that supplementation with C. aerea represents a promising and sustainable alternative in the formulation of diets for tilapia, contributing to improved health and resistance to diseases. This study develops environmentally benign capping technique to synthesize nanoparticles of Curcuma longa-coated titanium dioxide (CR-TiO 2 ) from titanium isopropoxide by utilizing the extract of Rosa rubiginosa flowers as reducing and chelating agent. The biogenically synthesized nanoparticles revealed excellent anti-bacterial, electrochemical, and photocatalytic properties due to the presence of porous TiO 2 nanostructures. The sharp peaks by XRD pattern showed the crystallinity and phase purity of TiO 2 nanoparticles. BET analysis proved mesoporous nature of the materials with specific surface area of 134 m 2  g -1 . The vibrational spectra suggest hydroxyl groups from flavonoids of Curcuma longa acting as functionalizing agent for TiO 2 nanoporous structures with visible luminescence, which is proven in fluorescence spectra and is applicable for photocatalytic studies. The anti-bacterial studies showed good inference on TiO 2 nanoparticles against Pseudomonas auruginosa and proved it to be an excellent antipseudomonal agent with the oxidative potential. The maximum degradation of phenol red dye in the presence of TiO 2 under visible light conditions was observed. The supercapacitor fabricated using the biogenic TiO 2 three-electrode system exhibited a specific capacitance of 128 Fg -1 (10 mV s -1 ), suggesting it as an excellent electrode material. The LSV curve at 50 mV s -1 scan rate showed that oxygen reduction potential (ORR) of CR-TiO 2 electrodes was 121 mV. The present study is a new application of nanoparticles in sustainability consideration of the environment as well as a solution to the power crisis with fewer limitations. The objective of this study was to explore a novel methodology for the synthesis of nanocoated probiotics following their collection and cultivation under optimized conditions, in light of their significant contribution to human health. Probiotics are instrumental in sustaining immune health by modulating the gastrointestinal microbiota and facilitating digestion. However, the equilibrium they maintain can be adversely affected by antibiotic treatments. It is critical to investigate the vulnerability of probiotics to antibiotics, considering the potential implications. This research aimed to assess whether nanoparticle coating could augment the probiotics' resistance to antibiotic influence. A strain of Lactococcus lactis ( L. lactis ) was isolated, cultured, and comprehensively characterized utilizing state-of-the-art methodologies, including the VITEK® 2 compact system, VITEK® MS, and 16S rRNA gene sequencing. The nanoparticle coating was performed using iron (III) chloride hexahydrate and tannic acid, followed by an evaluation of the probiotics' resistance to a range of antibiotics. The analysis through scanning electron microscopy (SEM) and atomic force microscopy (AFM) demonstrated a partial nanoparticle coating of the probiotics, which was further supported by UV/Vis spectroscopy findings, suggesting enhanced resistance to standard antibiotics. The results revealed that this strain possesses a unique protein profile and is genetically similar to strains identified in various other countries. Moreover, nano-encapsulation notably increased the strain's resistance to a spectrum of standard antibiotics, including Benzylpenicillin, Teicoplanin, Oxacillin, Vancomycin, Tetracycline, Rifampicin, Erythromycin, and Clindamycin. Cymodocea serrulata mediated titanium dioxide nanoparticles (TiO 2 NPs) were successfully synthesized. The XRD pattern and FTIR spectra demonstrated the crystalline structure of TiO 2 NPs and the presence of phenols, flavonoids and alkaloids in the extract. Further SEM revealed that TiO 2 NPs has uniform structure and spherical in shape with their size ranged from 58 to 117 nm. Antibacterial activity of TiO 2 NPs against methicillin-resistant Staphylococcus aureus (MRSA) and Vibrio cholerae (V. cholerae), provided the zone of inhibition of 33.9 ± 1.7 and 36.3 ± 1.9 mm, respectively at 100 μg/mL concentration. MIC of TiO 2 NPs against MRSA and V. cholerae showed 84% and 87% inhibition at 180 μg/mL and 160 μg/mL respectively. Subsequently, the sub-MIC of V. cholerae demonstrated minimal or no impact on bacterial growth at concentration of 42.5 μg/mL concentration. In addition, TiO 2 NPs exhibited their ability to inhibit the biofilm forming V. cholerae which caused distinct morphological and intercellular damages analysed using CLSM and TEM. The antioxidant properties of TiO 2 NPs were demonstrated through TAA and DPPH assays and exposed its scavenging activity with IC 50 value of 36.42 and 68.85 μg/mL which denotes its valuable antioxidant properties with potential health benefits. Importantly, the brine shrimp based lethality experiment yielded a low cytotoxic effect with 13% mortality at 100 μg/mL. The preservation of drug stability in biological evidence during the processes of collection and storage poses a substantial obstacle to the progress of forensic investigations. In conjunction with other constituents, the microorganisms present in the samples play a vital role in this investigation. The present investigation employed the high-performance liquid chromatography (HPLC) technique to assess the stability of (1R,2 S)-(-)-2-methylamino-1-phenyl-1-propanol hydrochloride in plasma and urine samples that were inoculated with Escherichia coli. These samples were subjected to storage conditions of 37 °C for 48 h and - 20 °C for a duration of 6 months. Minimal inhibitory concentration (MIC) and Minimal bactericidal concentration (MBC) of MPPH against E. coli were determined using microdilution method. The stability of MPPH in plasma and urine samples inoculated with E. coli was investigated using HPLC method. The results showed the MIC and MBC of MPPH were 87.5 ± 25 ppm and 175 ± 50 ppm, respectively. While MPPH remained stable in plasma for 48 h at 37 °C, it showed a notable decrease of about 11% in stability when stored in urine for the same period and temperature. From the beginning of the first month, a decrease in the stability of the compound appeared in all samples that were stored at - 20 °C, and the decrease reached 7% for plasma samples and about 11% for urine samples. The decrease in the stability reached its peak in the sixth month, reaching more than 30% and 70% of plasma and urine samples preserved at - 20 °C. This work concluded that E. coli can negatively affect the stability of MPPH in plasma and urine samples. Background: Small colony variants (SCVs) are biotypes of bacteria that have a size of approximately one-tenth or less of the wild types and has distinct characteristics comparing to the wild type strains. Clinical SCVs are usually associated with persistent infection and require a long-term treatment program with antibiotics. In Saudi Arabia, there are few studies about SCVs Escherichia coli for this reason, this study is aimed to investigate the ability of gentamicin to mutate E. coli ATCC 25922 to produce small SCVs and investigate the genotypes and phenotypes changes and stress tolerance comparing to clinical SCVs E. coli and normal clinical E. coli Isolated from blood samples., Methods: In this investigation, four clinical blood samples were collected ted from patients and the cultivation and isolation were carried out in KFMC between December 2019 and February 2021. The identification of positive blood culture samples was done using phoenix MD. Non-SCV E. coli ATCC25922 were mutated to SCV using exposure to increasing gradual concentrations of gentamicin at 100-generation intervals. Biochemical features and susceptibility to standard antibiotics using automated Phoenix MD 50 and. The survival assays were done using several stresses including heat shock, low pH, high osmotic pressure, and oxidative pressure. Virulence genes screening included the detection of genes that encoded to α-haemolysin, CS12 fimbriae, F17-like fimbrial adhesion, P-related fimbriae, yersiniabactin siderophore system, P-fimbriae, aerobactin, iron-regulated genes using PCR and gel electrophoresis., Results: The data from the mutating E. coli ATCC 25922 small colony test with gentamicin revealed that the first emergence of the multidrug resistance (MDR) SCV E. coli strain occurred at generation number 250, corresponding to a gentamicin concentration of 57 g/ml. Pathogenicity islands detection revealed that all tested E. coli strains have PAI IV 536 genes on their chromosomes furthermore, mutated SCV E. coli ATCC 25922 acquired PAII CFT 073 and PAI IV 536 . Survival tests showed no significant differences changes in tolerance of mutated SCVs comparing to parental strain., Conclusion: The present work concluded that gentamicin sub-MIC concentration gradual exposure can induce mutation responsible for SCV formation and evolving of MDR E. coli strains. Staphylococcus aureus in the blood of sickle cell disease (SCD) patients may result in a significant increase in morbidity and mortality. S. aureus strains contain various virulence characteristics, including the ability to create a variety of toxins and develop drug resistance. The current study sought to assess the prevalence of S. aureus in SCD patients and to identify the pathogen's virulence characteristics. Between 2017 and 2021, blood samples and data were collected at King Saud University Medical City (KSUMC) in Riyadh, Saudi Arabia. The Vitek system PCR and gene sequencing methods were used for identification, antibiotic resistance patterns, and genetic analysis. During the study period, 47 S. aureus blood isolates (methicillin-resistant S. aureus (MRSA) 41.6% and non-MRSA 58.4%) were isolated from 2406 SCD patients. The prevalence percentages of virulence genes ( finbB , sdrC , sdrD , icaA , coa , nuc , hlg , hla , finbA , clfA , efb , pvl , agr , spa , seb , sea , sec , tst , and sed ) among all the isolates from the SCD patients compared with non-SCD patients (control group) were as follows: (100% vs. 100%), (100% vs. 100%), (100% vs. 100%), (100% vs. 87.5%), (100% vs. 81.3%), (100% vs. 100%), (100% vs. 100%), (100% vs. 100%), (97.9% vs. 81.3%), (97.9% vs. 100%), (97.9% vs. 87.5%), (54.3% vs. 56.3%), (46.8% vs. 75%), (42.6% vs. 43.8%), (27.7% vs. 0%), (25.5% vs. 12.5%), (12.8% vs. 6.3%), (4.3% vs. 12.5%), and (4.3% vs. 0%). Regarding the resistance genes ( plaZ , mecA , ermA , ermC , tetK , tetM , and ermB ) of the S. aureus strains isolated from the SCD patients compared with non-SCD patients (control group), the prevalence percentages were as follows: (100% vs. 100%), (100% vs. 56.3%), (0% vs. 31.3%), (31.9% vs. 18.8%), (40.4% vs. 25%), (0% vs. 0%), and (0% vs. 0%). As for the antibiotic (ampicillin, penicillin, amoxicillin, cefazolin, imipenem, oxacillin, erythromycin, tetracycline, azithromycin, ciprofloxacin, moxifloxacin, and levofloxacin) resistance of the S. aureus strains isolated from the SCD patients compared with non-SCD patients (control group), the prevalence percentages were as follows: (100% vs. 100%), (97.9% vs. 100%), (72.3% vs. 25%), (68.1% vs. 37.5%), (68.1% vs. 25%), (66% vs. 25%), (36.2% vs. 18.8%), (23.4% vs. 12.5%), (19.1% vs. 12.5%), (17% vs. 12.5%), (14.9% vs. 25%), and (10.6% vs. 18.7%). This study concluded that several virulence genes were present in the S. aureus strains recovered from the SCD patients at KSUMC, with all the isolates containing the finbB , sdrC , sdrD , icaA , coa , nuc , hlg , and hla genes.

There is no doubt that the risk of drug-resistant pathogens and cancer diseases is on the rise. So, the goal of this study was to find out how effective silver nanoparticles (Ag-NPs) made by Senna alexandrina are at fighting these threats. In this work, S. alexandrina collected from Medina, Saudi Arabia was used and the biosynthesis method was applied to produce the Ag-NPs. The characterization of Ag-NPs was done using different analytical techniques, including UV spectroscopy, FT-IR, TEM, and XRD analysis. The MIC, MBC, and MTT protocols were applied to confirm the bioactivity of the Ag-NPs as antibacterial and anticancer bioagents. The findings reported indicating that the aqueous extract of S. alexandrina leaves, grown naturally in Saudi Arabia, is ideal for the production of bioactive Ag-NPs. The hydroxyl, aliphatic, alkene, N-H bend of primary amines, C-H bonds, and C-O bonds of alcohol were detected in this product. The small, sphere-shaped particles (4-7 nm) were the most prevalent among the bioactive Ag-NPs produced in this work. These nanoparticles inhibited some important multidrug-resistant pathogens (MDRPs) ( Escherichia coli , Acinetobacter baumanii/haemolyticus , Staphylococcus epidermidis , and Methicillin-resistant Staphylococcus aureus (MRSA)), as well as their ability to inhibit breast cancer cells (MCF-7 cells). The MIC of Ag-NPs ranged from 0.03 to 0.6 mg/mL, while their MBC ranged from 0.06 to 2.5 mg/mL. Anticancer activity test showed that IC 50 of the Ag-NPs against tested breast cancer cells was 61.9 ± 3.8 µg/mL. The stability of drugs in biological evidence during collection and storage is of particular concern in forensic investigations. Microbes in the samples and other elements are an essential component of these investigations. In the current study, the HPLC method was used to examine the stability of (1R, 2S)-(-)-Ephedrine hydrochloride in plasma and urine samples inoculated with C. albicans after storage at 37 °C for 48 h and -20 °C for six months. In the stability experiment, MIC 50% of (1R, 2S)-(-)-Ephedrine hydrochloride was applied according to MIC and MFC that were determined in this work. This drug had MIC and MFC of 50 and 100 ppm, respectively. In HPLC analysis, the standard (1R, 2S)-(-)-Ephedrine hydrochloride had a retention time of 1.63 and was used to identify this drug in samples that had or had not been exposed to C. albicans . The findings demonstrated that within 48 h at 37 °C, C. albicans had an impact on the drug concentration (10% and more than 15%, respectively, were lost in plasma and urine samples inoculated with C. albicans ). In plasma samples, the drug content remained stable at -20 °C for three months, although it degraded in urine samples after one month. In plasma and urine samples, the concentration reduction had surpassed 70% and 50% by the sixth month, respectively. Recent years, metal pollution is an alarming factor to know about protects the environmental ecosystem due to the toxic, persistent and abundant in nature. Metals are present everywhere in the biotic and abiotic samples including soil, water, and microbes. The rate of bioaccumulation and biotransformation are very high. The excess concentration of the metals causes heavy metal pollution or contamination. Due to these defects, the removal of metals using biological sources is heightened in the current research. In this current investigation, the biosorption potential ability of the metal tolerable Bacillus cereus on Pb and Cu rich environment was chosen and thoroughly monitored. The 16s rRNA of the Bacillus cereus was sequenced, and named as Bacillus cereus RMN 1 (MK521259). The various test concentration (10-60 mg/mL) of Pb and Cu was exhibited the maximum removal percentages of 85.2% and 60.2%. The result of bisorption factors exhibited, 300 mg/mL of the biosorbent potency, 60 min contact time and pH 7, and they found to be optimal to remove the maximum of Pb ion from the solution. In the regression coefficients, the Freundlich and Langmuir isotherm models were used to study the adsorption kinetics of metal ions. In addition, the isotherm model confirmed that the of B. cereus biomass medicated metal adsorption was more favourable reaction for metal degradation. With the above evidences, the results of the present investigation proved that B. cereus derived biomass was actively adsorbing the metals ions. This study looked at the development of effective biosorbents to recover the most toxic elements from industrial water. B. amyloliquefaciens was isolated from marine soils showing extreme resistance to Chromium (Cr(VI)) ions. During the 60 min of contact time, 79.90% Cr(VI) was adsorbed from the aqueous solution. The impact of important factors such as biomass concentration, pH of the medium, and initial metal ions concentration on biosorption rate was also examined. The desorption study indicated that 1 M HCl (91.24%) was superior to 0.5 M HCl (74.81%), 1 M NaOH (64.96%), and distilled water (3.66%). Based on the Langmuir model, the maximum adsorption capacity of the bio-absorbent was determined to be 48.44 mg/g. The absorption mechanism was identified as monolayer, and 1/n from the Freundlich model falls within 1, thus indicating favorable adsorption. Based on the findings of the present study, the soil bacterium B. amyloliquefaciens was found to be the best alternative and could be used to develop strategies for managing existing environmental pollution through biosorption., (Copyright © 2022 Elsevier Inc. In this study Pergularia daemia unripe fruits were used to synthesize zinc oxide nanoparticles (Pd-ZnONPs). UV-vis Spectroscopy detected the production of ZnONPs. XRD, FTIR, SEM, and TEM studies were used to characterize the synthesized Pd-ZnONPs. Aedes aegypti (Ae. aegypti) third instar larvae were analyzed to diverse concentrations of Pd-unripe fruit extract and Pd-ZnONPs for 24 hours to assess the larvicidal effect. Mortality was also detected in Ae. aegypti larvae under laboratory conditions, with corresponding LC 50 and LC 90 values of 11.11 and 21.20 µg/ml respectively. As a result of this study, the levels of total proteins, esterases, acetylcholine esterase, and phosphatase enzymes in the third instar larvae of Ae. aegypti were significantly lower than the control. Recent year, bacterial laccases are increasing interest in the field of industry and environmental applications especially decolorization of azo dyes. In industry, the dyes are present in stable nature including chemicals and lights. Due to these defects, the novel approaches are needed to removal of dyes before discharging into the environment. Among the various technologies, biological treatment methods and their strategies are very important, because of the decolorization and detoxification. Consecutively, biological mediated dyes removal are emerged with high potential especially microbes. Microbial laccases creates up new opportunities for their commercial applications. In this study, laccases were produced from Bacillus cereus (B. Cereus) and Pseudomonas parafulva (P. parafulva) by sub merged fermentation. For immobilization, the produced laccases were subjected to purify using 80% saturated ammonium sulphate and followed by dialysis. Then, crude laccases were immobilized through copper-alginate entrapment method. The maximum immobilized enzyme activity of the immobilized laccases were shown pH 8 at 50 °C and pH 7 at 40 °C for B. Cereus and P. parafulva respectively. In contrast, the normal enzyme activity was pH 10 at 40 °C and pH 8 at 40 °C were indicated for Bacillus cereus and P. parafulva respectively. Next, the free and immobilized laccases were performed the decolorization of three azo dyes T-blue, yellow GR and orange 3R, and exhibited that the 91.69 and 89.21% of Orange 3R were completely decolorized by both the B. Cereus and P. parafulva laccases when compared with free laccases enzymes. The confirmation of decolorization was monitored by UV-vis spectroscopy and FTIR spectroscopy, which clearly confirm the changes of peaks when compared with normal laccases. Finally, we have concluded that the B. Cereus and P. parafulva laccases are very important in azo dye decolorization and these used in future biological treatment of dyeing effluents., (Copyright © 2021 Elsevier Inc. Pseudomonas aeruginosa (PA) is an opportunistic pathogen that causes high mortality in cystic fibrosis patients. Treatment failures often occur due to the emergence of antibiotic resistance. Inhibition of virulence factors production without suppressing the growth of the pathogens is a potential alternative strategy to control the antibiotic resistance. In order to accomplish, three different interaction studies were performed using Bacillus subtilis BR4, PA and their extracellular contents. Firstly, co-cultivation was performed with different cell density of BR4 or PA. In co-culture setup (F), high cell density of BR4 significantly inhibits the biofilm formation of PA in a growth-independent manner (p < 0.01). To substantiate the biofilm inhibition, LC-MS/MS was performed and metabolic profile of monocultures and cocultures were compared. Multivariate analysis corroborated that metabolic profile of coculture setup (F) is drastically different from other coculture and monoculture setups. To check the effect of extracellular content of PA on BR4, supernatant of PA was extracted with ethyl acetate and different concentration of that extract (PA-EXT) was supplemented with BR4 culture. Exogenous supplementation PA-EXT (40 μg/mL) led to increased biofilm inhibitory activity (p < 0.01) in BR4. Further, to check the effect of extracellular content of BR4, PA was grown in the supernatant of BR4. PA survives in the spent media of BR4 without biofilm formation. Though 50% spent media of BR4 was replaced with fresh media, PA could not produce biofilm. In support of this, LC-MS/MS analysis has revealed that abundance of quorum sensing (QS) signals was reduced in the spent media grown PA than control. Furthermore, BR4 protects zebrafish larvae (Danio rerio) against PA infection and increases their survival rate (p < 0.05). We found that PA-induced oxidative stress and apoptosis were also significantly reduced in the BR4-pretreated larval group than control group. These results clearly indicate that BR4 exerts growth-independent QS inhibition in PA, suggesting that it could be used as a probiotic for future therapeutic interventions., (Copyright © 2022 Elsevier Ltd. In this work, graphene oxide-p-Phenylenediamine nanocomposites of two different ratios of Graphene oxide: p-Phenylenediamine (1:1 and 1:5) were prepared and characterized by using analytical, spectroscopic and microscopic studies (GO-pPD 11 and GO-pPD 15). These nanocomposites were employed as fluorescent chemosensors for sensing potential cations. Remarkably, graphene oxide-p-Phenylenediamine nanocomposites of ratio 1:1 (GO-pPD 15) was selective and sensitive to Ag + ions, whereas the graphene oxide-p-Phenylenediamine nanocomposites of ratio 1:5 (GO-pPD 15) was selective to Ce 3+ ions. A possible mechanism as switch "off-on" is proposed built on the inhibition of the photo induced electron transfer process in both the fluorescent probes in detecting the metal ions. In addition, interference studies were performed with the help of competitive complexation analysis and no significant interference were found by other potentially competing cations. The pH studies revealed that both the chemosensors can be used at the physiological pH for the ion detection and also the detection time was within 2-3 min. Both the chemosensors show good reversibility and hence the sensors can be used for multiple times. Red Sea represents one of the most remarkable marine ecosystems. However, it is also one of the world's least explored areas of marine biodiversity. The aims of this investigation were therefore, to isolate marine microorganisms from the seashore sediments and water in shallow region from west Yemen coast, to assess their antimicrobial potential, to identify the highly active isolate, and to purify and identify the bioactive compounds from it. In this regard, twenty-five bacterial strains have been isolated from twenty samples and tested for their antimicrobial ability against some pathogenic bacteria and yeast by using the agar disk diffusion and agar well diffusion assay. Out of the total 25 marine actinomycetes isolates only 13 exhibited interesting antimicrobial activity. The morphological, biochemical, and phylogenetic characteristics of the potential isolate 1S1 were compatible with their classification in the genus Streptomyces . The 16S rRNA gene sequences have shown that the isolate 1S1 clustered with Streptomyces longisporoflavus . The strain Streptomyces sp. 1S1 was cultivated and extracted with ethyl acetate. The GC-MS study of the extract indicated the presence of certain fatty acyl compounds e.g., tetradecanoic acid, 9-octadecenoic acid, hexadecanoic acid, and 9,12,15-octadecatrienoic acid. Using chromatographic techniques, three compounds were isolated and by spectroscopic methods e.g., IR, MS and NMR structurally elucidated. The three compounds were identified as a triacylglyceride, 9-octadecenoic acid, and hexadecanoic acid. Analyzing the health benefits of any two immunostimulants (synbiotics) in combined form and information on their interactions gain more visibility in the usage of synbiotics in aquafarms. With this intention, the current work explores the immunostimulant effect and structural interaction of synbiotic (β-1, 3 glucan binding protein from marine crab, Portunus pelagicus (Ppβ-GBP) and Bacillus licheniformis) on Oreochromis mossambicus. The experimental diet was prepared with Ppβ-GBP and B. licheniformis, and nourished to the fingerlings of O. mossambicus for 30 days. After the experimental trial, a higher growth rate and immune reactions (lysozyme, protease, myeloperoxidase and alkaline phosphatase activity) were noticed in the fish nourished with synbiotic (B. licheniformis and Ppβ-GBP) enriched diet. Moreover, the synbiotic enriched diet elevated the antioxidant responses like glutathione peroxidase (GSH-Px) and catalase (CAT) activity in the experimental diet-nurtured fish. At the end of the feed trial, synbiotic diet nourished fish shows an increased survival rate during Aeromonas hydrophila infection, reflecting the disease resistance potential of experimental fish. Also, the interaction between Ppβ-GBP and Bacillus licheniformis was analyzed through computational approaches. The results evidenced that, Ppβ-GBP interacts with the B. licheniformis through sugar-based ligand, β-glucan through a hydrogen bond with a good docking score. The edible fruiting bodies of desert truffles are seasonally collected and consumed in many regions of the world. Although they are very expensive, they are bought and sold as a result of considerable scientific reports confirming their health and nutritional benefits. This study aimed to conduct laboratory production of the fungal biomass of Tirmania nivea as a natural renewable resource of many active biological compounds using an artificial growth medium. The T. nivea collected from Hafar Al-Batin, which is north of Saudi Arabia, and their ascospores were harvested and used to produce fungal biomass in potato dextrose broth. The cultivation was conducted using a shaking incubator at 25 °C for two weeks at 200 rpm. The crud extracts of the fungal biomass and mycelium-free broth were prepared using ethyl acetate, methanol and hexane. Preliminary gas chromatography-mass spectrometry (GC-MS) analysis and their biological activity as antimicrobial agents were investigated. The results showed that the crude extracts have biological activity against mold, yeast and bacteria. The preliminary GC-MS analysis reported that the fungal biomass and extracellular metabolites in the growth medium are industrial renewable resources of several biological compounds that could be used as antifungal, antibacterial, antiviral, anticancer, antioxidant, anti-trypanosomal and anti-inflammatory agents.

Background: The upgrowth and rapid prevalence of pandrug-resistant Acinetobacter baumannii strains that have a pathogenic activity to cause several infections are of considerable influence on the health of communities worldwide. No infections by these bacterial strains were recorded before 1998, and currently, the numbers are on the rise., Methods: The A. baumannii strains were isolated from male and female patients in Medical Microbiology Department, King Fahd Medical City (KFMC) in Riyadh, Saudi Arabia between 1/1/2020 to 29/12/2020. The statistical analysis was performed base on sex, age, source of samples, and response to commercially available antibiotics. The A. baumannii strains that resisted all the antibiotics including colistin and imipenem were selected for the synergic test., Results: The data showed that 62.28%, 77.07% of 342 A. baumannii strains were isolated from males and patients over 35 years of age. A. baumannii strains (pandrug-A. baumannii) that can resist all tested antibiotics were 8.19%. The major source of the A. baumannii isolates was the respiratory system (>50%). Among all isolates (N = 342), azidothymidine-resistant A. baumannii strains were more than 85%. There is a statistically significant difference (P < 0.05) in the number of colistin-resistant A. baumannii strains isolated from males comparing with the female. The combinations of colistin and silver nanoparticles or imipenem and silver nanoparticles resulted in synergistic action led to reduction of MICs of colistin, imipenem, and silver nanoparticles (more than four-fold reduction). Also, the combinations of colistin and imipenem had high synergistic action., Conclusion: The pandrug-resistant A. baumannii strains may represent a current and future threat that must be fought, and the synergy action of antibiotics and nanoparticles may be one of the available, rapid, and easy strategies to confront this global problem., (Copyright © 2021 The Author(s). Based on the excellent nutrient level, the current study was focused on isolation and anti-bacterial activity of the actinomycetes from marine mangrove soil samples. As result, 10 different strains of actinomycetes strains were identified on actinomycetes isolation agar plates. The identified strains were shown with white, clear, uncontaminated well matured spore producing ability. Based on the initial observation, the isolated colonies were actinomycetes . The partially extracted crude compound shown excellent anti-bacterial activity against P. aeruginosa and K. pneumoniae with 15 mm and 13 mm zone of inhibitions were observed at 500 μL concentrations. The minimum inhibition concentration result was also confirmed the 500 μL concentration against both the tested concentration with high inhibition rate. Then, the intracellular damages, decreased cell growth of the crude actinomycetes extract treated bacterial strains were clearly observed by confocal laser scanning electron microscope. The extracellular damages of bacterial cell wall and shape of the both the pathogens were clearly shown by scanning electron microscope. The removal of toxic heavy metal ions from contaminated environments is a great challenge and requires an alternative rapid, efficient, economical bioremediation approach. Henceforth, bioflocculant producing endophytic actinobacterial sp. was isolated from heavy metal contaminated marine environments for heavy metal biosorption process. After molecular characterization, the isolated actinomycete starin was Nocardiopsis sp. GRG 3 (KT235642). It was indicated that the maximum flocculating activity of 80.90% with glucose, and yield is 4.52 g L 1 . The optimum flocculating activity was reached at pH 7 in the presence of CaCl 2 ions. Further, the bioflocculent produced Nocardiopsis sp. GRG 3 (KT235642) was characterized by fourier transform infrared analysis spectra (FTIR) and displayed the presence of carboxyl, hydroxyl, amino groups and characteristic of more polysaccharide and protein. The heavy metal sorption by bioflocculant Nocardiopsis sp. GRG 3 (KT235642) was effectively removed 55.90% Cd, 85.90% Cr, 74.7% Pb, and 51.90% Hg. Therefore, this study was proved that the bioflocculant derived from endophytic actinobacteria, Nocardiopsis sp. Heavy metal pollution in the water bodies causes a serious threat to all living beings. Extended exposure of heavy metals such as nickel (Ni) ions causes cancer. Henceforth, the current study investigated the removal of Ni ions from the electroplating effluent using nanocomposites namely, Graphene Oxide (GO) and Reduced Graphene Oxide (rGO) in the presence of various factors such as contact time, pH, agitation speed and sorbent dosage. Further, it was determined the rate kinetic model and adsorption equilibrium isotherms. The study also focused on comparing the removal efficiency of two nanocomposites. The maximum sorption efficiency were found to be 90.8% and 84.4% at optimized pH (8), contact time (180-1440 m), RPM (250-300) and adsorbent dosage (0.2 mg/L) for GO and rGO respectively. Furthermore, toxicity of treated and untreated effluent were tested against Phosphobacter and Azospirillium using GO and rGO and found that the treated effluent was non-toxic. The contribution of this study to agriculture in using recycled effluent for crop cultivation was being verified by seed germination of Lablab purpureus seeds watered with treated and untreated effluent. Silica nanoparticles are generally mesoporous that are predominant in the sand and rocks. Silica nanoparticles have a wide range of applications in various fields such as medicine, waste management, effluent treatment and electronics. The present work has explored the synthesis of silica nanoparticles through acid and alkaline leaching method from Pedalium murex which is a common weed that is found in southern parts of Tamil Nadu. Silica nanoparticles (SiNps) and its functional groups were confirmed by EDX and FTIR analysis with their respective energy dispersion levels and wavenumbers. Size, shape and morphological features of SiNps were analysed by PSA, TEM and SAED analysis. Synthesised and characterized nanosilica was crosslinked over nylon-66 and cellulose nitrate membranes and were confirmed by FTIR analysis for their crosslinking with SiNps. Water retention activity of the crosslinked and non crosslinked membranes was analysed by contact angle measurement to ensure the receptability of the membranes to remove contaminants by the adsorption. The decolourisation efficiency of the crosslinked nylon 66 membrane was found as a potential source for the treatment with 65.5% colour reduction when compared with other membranes. The multi-drug resistant effect of the Gram negative bacteria K. pneumoniae was identified by disc diffusion method using specific UTI panel discs of Kleb 1 HX077 and Kleb 2 HX090 HEXA. Among the multi-drug resistant bacteria, the carbapenem resistant (CR) effect of the K. pneumoniae was screened by specific carbapenem detection antibiotics of HEXA HX066 and HX0103 HEXA by disc diffusion method. In addition, the effective antibiotics were further performed against K. pneumoniae by minimum inhibition concentration method. Further, the carbapenemase genes of VIM 1 and IMP 1 were detected from the isolated strains by multiplex PCR method. Furthermore, the biofilm forming ability of selected carbapenem resistant K. pneumoniae was initially identified by tissue culture plate method and confirmed by exopolysaccharide arrest ability of congo red agar assay. Urinary tract infections are second most important diseases worldwide due to the increased amount of antibiotic resistant microbes. Among the Gram negative bacteria, P. mirabilis is the dominant biofilm producer in urinary tract infections next to E. coli . Biofilm is a process that produced self-matrix of more virulence pathogens on colloidal surfaces. Based on the above fact, this study was concentrated to inhibit the P. mirabilis biofilm formation by various in-vitro experiments. In the current study, the anti-biofilm effect of essential oils was recovered from the medicinal plant of Solanum nigrum, and confirmed the available essential oils by liquid chromatography-mass spectroscopy analysis. The excellent anti-microbial activity and minimum biofilm inhibition concentration of the essential oils against P. mirabilis was indicated at 200 µg/mL. The absence of viability and altered exopolysaccharide structure of treated cells were showed by biofilm metabolic assay and phenol-sulphuric acid method. The fluorescence differentiation of P. mirabilis treated cells was showed with more damages by confocal laser scanning electron microscope. Further, more morphological changes of essential oils treated cells were differentiated from normal cells by scanning electron microscope. In the present study, the chemical composition of Morinda citrifolia essential oils was determined by gas chromatography-mass spectrometry and was found to contain several anti-cancer compounds including L-scopoletin, nordamnacanthal, β-morindone, α-copaene, 9-H-pyrido[3,4-b]indole, β-thujene and terpinolene. The physico-chemical characterization of chitosan, chitosan nanoparticles and Morinda citrifolia essential oils loaded chitosan nanoparticles combination was carried out by Fourier transform infrared spectroscopy, powder X-ray diffraction and dynamic light scattering coupled with zeta potential. The morphological observation obtained by scanning electron microscopy and transmission electron microscopy provided clear indication that the immobile chitosan polymer formed a coating onto the Morinda citrifolia essential oils surface. The cytotoxic effect of Morinda citrifolia essential oils loaded chitosan nanoparticles against A549 cells were investigated, resulting in 54% inhibition at 40 μg/ml -1 . Information about in vitro morphological modification, nucleus damages, ROS generation and cell cycle arrest was obtained by fluorescence microscopy and flow cytometer analysis. The toxicity evaluation against human red blood cells suggested that the Morinda citrifolia essential oils loaded chitosan nanoparticles possess minimum cytotoxicity. Background: Antibiotic-resistant bacteria are one of the major global health issues that can affect humans, animals, and the environment. Antibiotic-resistant bacteria have emerged as opportunistic pathogenic bacteria that are frequently isolated from both clinical patients and healthy individuals. The aim of this study was to characterize the antibiotic-resistant bacteria isolated from powdered infant formulas marketed in Riyadh, Saudi Arabia., Methods: Infant powdered milk formulas were purchased from different pharmacies located within Riyadh, and ten products of powdered milk formulas designed for children of various ages were then transferred to the laboratory in the Department of Botany and Microbiology at King Saud University, Riyadh. Isolation and purification of Bacillus species were both performed according to standard protocols. The identification test was performed using the automated Vitek 2 system (BioMerieux, France), and antibiotic sensitivity tests were performed using the disk-diffusion method incorporating standard antibiotic disks foramikacin (30μg/disk), gentamicin (10μg/disk), imipenem (10μg/disk), moxifloxacin (5μg/disk), cefoperazone (75μg/disk), cefpodoxime (10μg/disk), ceftazidime (30μg/disk), and cefepime (30μg/disk). Statistical analysis was performed using Ward's method to obtain antibiotic resistance of the isolates., Results: The results obtained from the milk samples indicated that all isolates were sensitive to amikacin, gentamicin, and moxifloxacin. A group of isolates obtained from milk was resistant to cefoperazone by 6.49%, cefpodoxime by 25.9%, ceftazidime by 14.28%, and cefepime by 19.48%., Conclusions: Based on these findings, we concluded that the powdered infant formula marketed in Riyadh City may act as a source of bacterial isolates that are resistant to several standard antibiotics., (Copyright © 2019. Exotic woody weed plants are a very serious threat to seed dispersed by ungulate in the tropical forest of Asia. The ungulates in Point Calimere Wildlife Sanctuary (PCWS) are a significant role in native indigenous seed dispersal. The exotic woody weed tree Prosopis juliflora prevalence distributed in the PCWS and they might potentially alter the native medicinal plant species. In the present investigation, we have assessed the seed dispersal by ungulates in PCWS from January to March 2017. Four different ungulate species were selected to understand their seed dispersal rate of different plant species in selected sanctuary. This investigation was planned to confirm the seed dispersal by ungulates of blackbuck, spotted deer, wild boar and feral horse. Among the four different ungulates tested, the maximum numbers of pellets collected from blackbuck and no seed found in their pellets. The low quantities of pellets were collected from wild boar and this study has recorded medium-sized ungulates which dispersed variety of plant. However, the dispersal of the seed of medicinal plants were not considerably high and relatively moderate percentage of seeds dispersal occurred in medium-sized ungulates like wild boar and spotted deer. P. juliflora had 100% seed germination rate were observed from the faecal samples of wild boar and feral horse. Recent years Klebsiella pneumoniae ( K. pneumoniae ) biofilm formation (BF) is emerging thread worldwide. For tackling this problem, we have chosen Hibiscus rosa-. pneumoniae. The HPLC purified essential oils (EOs sinensis (H. rosa-sinensis) (HRS) to inhibit the BF K) of H. rosa-sinensis was performed against BF K. pneumoniae and showed concentration dependent biofilm inhibition. At the MBIC of EOs (90 µg/ml), the biofilm inhibition was showed at 92% against selected BF K. Pneumoniae . The biofilm metabolic assay, exopolysaccharide quantification and hydrophobicity index variation results exhibited with 88%, 92% and 89% reduction at 90 μg/mL was observed respectively. In addition, the morphological modification of MBIC treated K. pneumoniae was clearly viewed by scanning electron microscope (SEM). Bacterial infections are the most important problem of health care worldwide. The hemolymph antibacterial proteins of Mesocyclops leuckarti was isolated for the first time and its antibacterial efficacy was evaluated against four different human pathogenic microbes viz., Escherichia coli , Staphylococcus aureus , Klebsiella pneumonia and Shigella flexneri. The antibacterial potential of the antimicrobial proteins of hemolymph samples from plankton cultured in water enriched with Cow Urine Distillate (CUD) was compared with normal ones. The results indicated that the hemolymph proteins were more potential against Gram negative bacteria than Gram positive bacteria. Klebsiella pneumonia was more susceptible to the hemolymph proteins exhibiting a zone of inhibition measuring 27 mm. The supplement of CUD to the culture media further enriched the antibacterial activity of the hemolymph proteins (29 mm). The SDS-PAGE analysis indicated two different types of clear bands representing proteins of 53 kDa and 19 kDa. In this study, silver nanoparticles (Ag NPs) was eco-friendly synthesized using purified flavonoid rich content of Morinda citrifolia (M. citrifolia) extract. The synthesized Ag NPs was exhibited at 420 nm in UV-spectrometer, and surface morphology with available chemical composition, shape and size of the Ag NPs were confirmed by X-ray diffraction (XRD) variation, scanning electron microscope (SEM) with energy dispersive X-ray spectroscopy (EDX) and transmission electron microscope (TEM). In addition, the excellent phytochemicals and anti-oxidant activity of the Ag NPs were confirmed by total anti-oxidant and DPPH free radical scavenging assays. Further, the concentration dependent inhibition of synthesized Ag NPs against biofilm forming Staphylococcus aureus (S. aureus) was confirmed by minimum inhibition concentration (MIC). The growth cells were arrested in the log phase of the culture and detected by flow cytometry analysis. In addition, the bacterial viability, exopolysaccharide degradation, intracellular membrane damage, matured biofilm inhibition, architectural damage and morphological alteration were confirmed by confocal laser scanning electron microscope (CLSM) and SEM. Furthermore, the synthesized Ag NPs reacted with methylene blue (MB) dye molecules has 100% degradation at an irradiation time of 140 min. The objective of the present work was to observe and profile various antibiotic resistant strains of Staphylococcus aureus and highlight the need for continuous surveillance. Data regarding antibiotic-resistant S. aureus strains isolated and identified at the Medical Microbiology Department, King Khalid Hospital, Riyadh was obtained. Bacterial isolates were collected from several sites of infections in patients and an evaluation of susceptibility were carried out using a fully automated Vitek2 system. Relative frequency (%), odds ratios and Ward's minimum variance were calculated. The results showed that wounds were a source of more than 40% of the S. aureus (MRSA) strains that have ability to resist methicillin, and more than 45% of the methicillin-susceptible S. aureus (non-MRSA) strains. 40% of the isolates were MRSA (N = 251), and all MRSA strains were sensitive to vancomycin, daptomycin, teicoplanin, tigecycline, nitrofurantoin, and itraconazole while all non-MRSA (N = 338) strains were sensitive to vancomycin, cefoxitin, daptomycin, gentamicin, oxacillin, teicoplanin, tigecycline, and mupirocin. Strength of association between antibiotic-resistant S. aureus strains and source of samples (site of infection) was established. The study concluded that S. aureus strains had developed resistance towards 20 (for non-MRSA) and 22 (for MRSA) of the antibiotics tested. Microbial enhanced oil recovery (MEOR) is a kind of enhanced oil recovery (EOR) development, often used as a tertiary stage where oil recovery is no longer possible utilizing primary and secondary conventional techniques. Among a few potential natural operators valuable for MEOR, biosurfactants, biopolymers and biosurfactant based nanoparticles assume key jobs. Biosurfactant which are produced by microorganisms' act as are surface active agents that can be used as an alternative to chemically synthesized surfactants. Pseudomonas aeruginosa TEN01, a gram-negative bacterium isolated from the petroleum industry is a potential biosurfactant (Rhamnolipid) producer using cassava waste as the substrate. This work focuses on production and characterization of rhamnolipid from P. aeruginosa TEN01 and its use in enhanced oil recovery. The effectiveness of Chitosan that is deacetylated form of chitin which is a biopolymer that provides density and viscosity to the fluids is not known in enhanced oil recovery yet and so it is studied. Moreover, the fabrication of biosurfactant-mediated silver nanocrystals and its application in enhanced oil recovery is also studied. Sand-Pack column was constructed and the mechanism of oil recovery in the column was studied. While incubating the crude oil containing sand packed column with Biosurfactant-biopolymer and brine flooding in the ratio of 1:2, and Biosurfactant incubation - flooding with 3 g/l of biopolymer was found to be 34.28% and 44.5% respectively. The biosurfactant based silver nanoparticles are non-toxic and have better stability when compared to chemically synthesized silver nanoparticles. This study emphasis the production of yellow pigment from endolichenic Bacillus sp. isolated from the lichen Dirinaria aegialita (Afzel. ex Ach.) B.J. Moore . Yellow pigment-producing twenty different strains were investigated. The hyperactive pigment-producing bacterial strain was identified as Bacillus gibsonii based on 99 % sequence similarity. Maximum bacterial pigment production appeared in Luria Bertani medium. Methanol extraction of the pigment and its partial purification using TLC was carried out. Furthermore, isolated pigments were characterized using UV-visible spectroscopy, FTIR spectroscopy, and GC-MS results related to the possibility of the carotenoid occurrence. The pigment also exhibited efficient antifungal activity against selected fungal pathogens of economic importance. Likewise, the pigment extract evaluated for the total antioxidant potential using Phosphomolybdenum and Ferric reducing antioxidant power assay and the results represented in Ascorbic Acid Equivalent (AAE)- 21.45 ± 1.212 mg/mL. β-Glucan-binding protein (βGBP) is important for the rational expansion of molecular biology. Here, zinc oxide nanoparticle (ZnONP) was synthesized using βGBP from the crab Scylla serrata (Ss-βGBP-ZnONP). Ss-βGBP-ZnONP was observed as a 100 kDa band on sodium dodecyl sulfate polyacrylamide gel and characterized with UV-vis spectroscopy at 350 nm. X-ray diffraction analysis displayed values consistent with those for zincite. Fourier transform infrared spectroscopy revealed the presence of functional groups, including amide, alcohol, alkane, alkyl halide, and alkene groups. The zeta potential (-5.36 mV) of these particles indicated their stability, and transmission electron microscopy revealed the presence of 50 nm nanocones. Ss-βGBP-ZnONPs were tested at 100 μg/mL against the gram-positive Enterococcus faecalis and gram-negative Pseudomanas aeruginosa using confocal laser scanning microscopy and the bacterial viability assay was also performed. The growth of MCF7 breast cancer cells was inhibited following treatment with 75 μg/mL Ss-βGBP-ZnONPs. Eleusine coracana (Finger millet) has high nutritional value with numerous health benefits and is of low cost. Isolation of beta-glucan (βG) from E. coracana (Ec-βG) has gained increasing research attention. UV-vis spectroscopy used to measure the surface plasmon resonance at 361 nm to confirm the presence of polysaccharides (glucan molecules) in Ec-βG. X-ray diffraction analysis of Ec-βG displayed a crystalline nature and confirmed the presence of the βG molecule. Further, the bioactive compounds of Ec-βG were screened using gas chromatography-mass spectrometry. The antibacterial activity of Ec-βG against both Gram-positive (Lysinibacillus fusiformis, Enterococcus faecalis) and Gram-negative (Proteus vulgaris, Shigella sonnei) bacteria were assessed through minimum inhibitory concentrations <70 μg/ml of Ec-βG. In addition, the antibiofilm activity and bacterial viability of Ec-βG at 100 μg/ml was confirmed by light and confocal laser scanning microscopy. Furthermore, Ec-βG inhibits α-amylase and α-glucosidase at an IC 50 -value of 1.23 and 1.42 μg/ml, respectively. Superoxide anion scavenging activity at IC 50 -1.4 μg/ml and DPPH radical scavenging activity at IC 50- 1.2 μg/ml showed that Ec-βG had potential antioxidant property. The in vitro hemolysis assay for biocompatibility of Ec-βG at 200 μg/ml showed 0.06 ± 0.09%. In this study graphene/chitosan nanoparticles (GR/CS NCs) were developed. The homogenous combination of GR and CS was confirmed by FTIR spectroscopy. The combination of CS with GR sheets reduced the XRD intensity of the GR peak in GR/CS NCs, while TEM images revealed the immobile CS coating of GR sheets. Further, the anti-biofilm activity of GR/CS NCs was tested. The tests showed that the formation of biofilm by Pseudomonas aeruginosa and Klebsiella pneumoniae was inhibited at 40□g/mL GR/CS NCs up to 94 and 92 %, respectively. The intracellular and cell surface damage of the bacteria was observed by CLSM and SEM. Also, GR/CS NCs produced a toxic effect of 90 % on Artemia franciscana at 70□g/mL upon 24 h incubation. Ammonia is a widespread pollutant that is toxic to living organisms in aquaculture. This study aimed to evaluate the effects of a diet supplemented with beta-glucan from yeast, Saccharomyces cerevisiae (Sc-βG), on the stress response of Oreochromis mossambicus (Tilapia) to ammonia. Fish were divided into four groups, including a control fed a basal diet and three experimental groups fed diets supplemented with Sc-βG at 2, 5 and 10 mg/g respectively. After 8 weeks, experimental groups were exposed to ammonia at 100 mg L -1 for 1 week. Growth was measured after the 8-week feeding trial and serum, mucus, and liver tissue were sampled before and after the ammonia challenge. Compared with the control diet, feed supplemented with Sc-βG at 10 mg/g significantly (p < 0.05) improved growth performance (7.8-9.9 g increase in weight). The cellular immune responses (myeloperoxidase, reactive oxygen species, and reactive nitrogen species), humoral immune responses (alkaline phosphatase, lysozyme, and peroxidase inhibition), and antioxidant response (catalase, superoxide dismutase, and glutathione) were tested in serum, mucus and liver tissue. Compared with the control, these responses were significantly (p < 0.05) enhanced at 10 mg/g supplementation with Sc-βG. Soil contamination has enlarged over the decades due to intensive use of pesticides and chemical fertilizers in agronomy. Earthworms are significant organisms in the soil community. Earthworms are the major role in soil fertility in most ecological system and the production of biogenic structures. Moreover, earthworm gut mucus enhances the beneficial soil microorganism potential biological activities. They are used as model organisms for assessing the ecological risks of chemicals. Enrichment of essential nutrients in soil through earthworm is a cost-effective and eco-friendly approach. In India, the organophosphorus pesticide monocrotophos is commonly used to control agricultural pests. Hence, it is important to study the effect of monocrotophos on the gut microbiota in Lampito mauritii. A 15-day exposure to a low (1/10th of the LC 50 after 96 h i.e., 0.093 ppm kg -1 ) and high sublethal concentration (1/3rd of the LC 50 after 96 h i.e., 0.311 ppm kg -1 ) of monocrotophos led to reduced proliferation of the gut microbiota in L. mauritii. However, exposure for 30 days led to a recuperation of the microbial populations to near control values. Among the eight bacterial and five fungal species that inhabit the gut of L. mauritii, only six bacterial and three fungal species were able to survive after exposure to monocrotophos. In addition to the study, histopathological changes were observed in the intestine of L.mauritii after application of lower sublethal concentration of monocrotophos. Severe pathological changes such as vacuolization, degenerated nuclei, damaged villi and congestion of the blood sinuses were noticed in the intestine on 1st and, 5th day of the experiment. But in 30th day the damages were slowly recovered due to degradation of monocrotophos by the presence of some pesticides degrading bacterial and fungal species and regenerative capability of chloragogen cells in the intestine. The results suggested that reduced microbial populations and pathological damages in intestine were observed during the application of monocrotophos. The albumen plays a major role in the protection of eggs against microorganisms. It contains an arsenal of natural antimicrobial molecules and antibacterial proteins, including the well-known ovotransferrin and lysozyme, which exert their activities against a range of bacteria. In the present study, the hen's albumen extract treated with the dried insect body of blister beetle M. pustulata was assessed for antibacterial, antibiofilm, anti-inflammatory and anti-proliferative activity. The zone of inhibition against Gram positive E. faecalis and S. aureus was 10.8 mm and 12.1 mm respectively at 100 μg mL -1 . However, it was 13.6 mm and 15.3 mm for Gram negative P. aeruginosa and P. vulgaris respectively. The biofilm of tested bacteria was significantly inhibited at 100 μg mL -1 . The hydrophobicity of bacterial biofilms was considerably condensed after treatment with the hen's albumen extracts at 100 μg mL -1 . The anti-inflammatory activity of hen's albumen extracts was confirmed by the inhibition of cyclooxygenase (COX) enzyme to 84.91% at 100 μg mL -1 with the relative IC 50 of 8.26 μg mL -1 . The albumen extract effectively inhibited the viability (23.61%) of HepG2 hepatic cancer cells at 100 μg mL -1 . The anti-proliferative activity of the albumen extracts was further revealed by the induction of HepG2 apoptotic cell morphology. Antibiotic-resistant Escherichia coli strains including extended-spectrum β-lactamase (ESBL) isolates are globally widespread in medical, food, and environmental sources. Some of these strains are considered the most pathogenic bacteria in humans. The present work examined the predominance of antibiotic resistance in E. coli strains in wound infections comparing with E. coli strains isolated from a raw milk as a potential source of those strains. The wound infections included abdomen, anus, arm, back, buttock, chest, foot, hand, head, leg, lung, mouth, neck, penis, thigh, toe, and vagina infections. In total, 161 and 153 isolates identified as E. coli were obtained from wound infections and raw milk, respectively. A Vitek 2 system innovated by bioMérieux, France was applied to perform the identification and susceptibility tests. The E. coli isolates that have ability to produce ESBL were detected by an ESBL panel and NO45 card (bioMérieux). Over half of the E. coli were from abdomen, back, and buttock wound infections. More than 50%of the E. coli isolates obtained from wound infections were resistant to cefazolin, ampicillin, cefuroxime, ciprofloxacin, mezlocillin, moxifloxacin, piperacillin, and tetracycline; 70% of the isolates from wound infections and 0% of the isolates from raw milk were E. coli isolates produced ESBL. ZnO nanoparticles (NPs) synthesized using haemocyanin (Hc-ZnONPs) purified from haemolymph of Penaeus semisulcatus were characterized using various techniques. HR-TEM and SEM microscopy indicated Hc-ZnONPs had a typical size of 20-50 nm and were spherical. The objective of current investigation was to assess the effects of dietary supplementation of Hc-ZnONPs on the development and activity of digestive and metabolic enzymes, as well as the antioxidant levels in P. semisulcatus. Trial basal diets were supplemented with Hc-ZnONPs at rates of 0, 10, 20, 40, 60, and 80 mg kg -1 (dry feed weight) and were fed to P. semisulcatus for 30 d. For 60 mg kg -1 Hc-ZnONPs-supplemented feed, significantly (P < 0.05) enhanced endurance, development, and activity of the digestive enzyme were observed. The enzymatic antioxidants and metabolic enzymes activities in the muscle exhibited no significant changes when 10-60 mg kg -1 Hc-ZnONPs-supplemented feed was fed to P. semisulcatus. Conversely, feeding the P. semisulcatus with 80 mg kg -1 Hc-ZnONPs produced a harmful outcome, with significant increase in the enzymatic antioxidants and metabolic enzymes. Consequently, 80 mg kg -1 Hc-ZnONPs was identified as lethal to P. semisulcatus. Current scenario of bio-nanotechnology, successfully fabrication of ultrafine titanium dioxide nanoparticles (TiO 2 NPs) using various biological protein sources for the multipurpose targets. The present research report involves synthesis of TiO 2 NPs using antimicrobial peptide (AMP) crustin (Cr). Crustin previously purified from the blue crab, Portunus pelagicus haemolymph, by blue Sepharose CL-6B matrix assisted affinity column chromatography. Synthesized Cr-TiO 2 NPs was physico-chemically characterized by UV-Visible spectroscopy (UV-Visible), X-ray Diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), High-resolution transmission electron microscopy (HR-TEM) and zeta potential examination. X-ray diffraction analysis for crystalline nature and phase identification of titanium dioxide nanoparticles was absorbed. Functional groups were found through FTIR ranges between 1620 and 1700 cm -1 . HR-TEM analysis showed that the synthesized Cr-TiO 2 NPs tetragonal shape and sizes ranging from 10 to 50 nm. Finally, the surface charge of the Cr-TiO 2 NPs was confirmed through zeta potential analysis. Furthermore, the characterized Cr-TiO 2 NPs exhibited good biofilm inhibition against GPB - S. mutans (Gram Positive Bacteria- Streptococcus mutans), GNB - P. vulgaris (Gram Negative Bacteria- Proteus vulgaris) and fungal Candida albicans. Moreover, photocatalysis demonstrated that the Cr-TiO 2 NPs was effectively explored the degradation of dyes. Background: Investigation for a naturally occurring anti-obesity drug has become the need of society all over the world. Betulinic acid (BA) is a lupane-type pentacyclic triterpene and is sourced from various organisms. This high potential biologically active molecule is reported to have anti-obesity effect. In this study, we report the molecular mechanism of action of BA that underlies anti-obesity activity and also an improved method of its isolation common teak tree., Methods: Mouse pre-adipocyte cells were used to develop hyperlipidemic conditions in vitro. Change in expression of genes associated to adipogenesis was checked using quantitative real-time PCR (qPCR). Co-factor specificity of PPAR gamma was analyzed through immune precipitation and immunoblot., Results: Betulinic acid was found to be effective in reducing the lipid content in 3T3L1 cells. Level of PPAR gamma and LXR alpha was reduced in connection to reduced adipogenesis. Change in steroid responsive co-activators (SRCs) during BA treatment proved that the compound can impart profound change in co-factor selectivity, which is crucial in determining the activity profile of PPAR gamma. Background: Trace elements of copper (Cu) are one of the main forms of ecological noxious waste in freshwater systems that affect the survival and development of organisms. The objective of the current study was to investigate the effects of chronic exposure to Cu on the growth, oxidative stress, immune and biochemical response in the Nile tilapia, Oreochromis niloticus., Methods: Three groups of O. niloticus were tested as follows; the first group was used as the control (not treated with Cu in water), while the 2nd and 3rd groups were exposed to (low) 40 μg L -1 and (high) 400 μg L -1 concentrations of Cu added to water, respectively. The duration of the experiment, which was conducted in triplicate, was 60 d. End points were evaluated on days 30 and 60. Following 30 d and 60 d of exposure to Cu, the fish were removed from experimental tanks to determine growth. Consequently, blood samples were collected from caudal veins at the end of the trial period (30 d and 60 d) and serum was separated to evaluate different immunological parameters, such as lysozymes (LYZ), respiratory burst activity (RBA) and myeloperoxidase (MPO). Gill and liver tissues were collected for evaluation of Cu and certain biochemical parameters as follows: antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione-S-transferase (GST); non-enzymatic antioxidants such as glutathione (GSH) and metallothionein (MT), and oxidative stress indicators such as malondialdehyde (MDA) and protein carbonyl (PCO). The results pertaining to treatments and the control were compared using two-way ANOVA and Tukey's HSD test. The level of significance was set at P ≤ 0.05. Data were expressed as mean ± SD., Results: Chronic exposure to Cu did not induce any mortality in fish during the test period. However, following exposure to Cu, growth of fish in the exposed groups was affected more than that in the control group (unexposed to Cu). In addition, accumulation of Cu in the liver tissue was higher than that in the gill tissues of fish exposed to Cu, compared to that in the control. Gill and liver tissues of Cu-exposed fish showed a significant (P ≤ 0.05) reduction in the activities of the antioxidant enzymes, SOD, CAT, GPx, and GST, compared to those of unexposed fish. Non-enzymatic antioxidants, GSH and MT, in gill and liver tissues were significantly increased (P ≤ 0.05) in fish exposed to both concentrations of Cu, compared to those in unexposed fish. Oxidative stress indicators, MDA and PCO in gills and liver of Cu-exposed fish was significantly (P ≤ 0.05) at both tested concentrations, when compared to control group. Non-specific immune response of LYZ, RBA, and MPO activity in serum decreased significantly (P ≤ 0.05) in Cu-exposed fish, compared with that of unexposed fish., Conclusion: Overall, the present results highlighted that chronic exposure to Cu ions may exert a strong effect on the antioxidant and immune responses of O. niloticus. Antibiotic-resistant Staphylococci are a global issue affecting humans, animals, and numerous natural environments. Antibiotic-resistant Staphylococcus epidermidis is an opportunistic pathogen frequently isolated from patients and healthy individuals. This study aimed to examine the antibiotic resistance of S. epidermidis isolated from patients, healthy students and compare the results with antibiotic-resistant bacteria isolated from pasteurized milk. Clinical strain isolation was performed in several hospitals in the Riyadh. Skin swabs from 100 healthy undergraduate candidate students were obtained at King Saud University. The pasteurized milk samples were obtained from local market (company, X). After isolation, identification and susceptibility tests were performed using an automated system. A multiplex tuf gene-based PCR assay was used to confirm identification. Biofilm production and biofilm-related gene expression were studied. S. epidermidis represented 17% of clinical bacterial isolates, and 1.7% of isolates obtained from healthy students were multiantibiotic-resistant. All patient strains were teicoplanin- and vancomycin-susceptible, while all student strains were gentamicin-, levofloxacin-, moxifloxacin-, and trimethoprim/sulfamethoxazole-susceptible. All the bacteria isolated from pasteurized milk were benzylpenicillin and oxacillin-resistant strains. Of the S. epidermidis strains, 91% could produce biofilms, and mecA, icaADBR, ica-ADB, ica-AD, ica-A only , and ica-C only were expressed in 83, 17.1, 25.7, 37.1, 20, and 0% of the strains, respectively. This work demonstrates that S. epidermidis can be accurately identified using a multiplex tuf -based assay, and that multiantibiotic-resistant S. epidermidis strains are widespread amongst patients and healthy students.

Here, we report the novel fabrication of ZnO nanoparticles using the Costus igneus leaf extract. Gas chromatography-mass spectrometry (GC-MS) and proton nuclear magnetic resonance ( 1 H NMR) spectroscopy to determine the bioactive components present in the plant extract. The synthesis of Ci-ZnO NPs (C. igneus- coated zinc oxide nanoparticles) was accomplished using a cost-effective and simple technique. Ci-ZnO NPs were specified using UV-visible spectroscopy, FTIR, XRD, and TEM. Ci-ZnO NPs was authenticated by UV-Vis and exhibited a peak at 365 nm. The XRD spectra proved the crystalline character of the Ci-ZnO NPs synthesized as hexagonal wurtzite. The FTIR spectrum illustrated the presence of possible functional groups present in Ci-ZnO NPs. The TEM micrograph showed evidence of the presence of a hexagonal organization with a size of 26.55 nm typical of Ci-ZnO NPs. The α-amylase and α-glucosidase inhibition assays demonstrated antidiabetic activity of Ci-ZnO NPs (74 % and 82 %, respectively), and the DPPH [2,2-diphenyl-1-picrylhydrazyl hydrate] assay demonstrated the antioxidant activity of the nanoparticles (75%) at a concentration of 100 μg/ml. The Ci-ZnO NPs exhibited promising antibacterial and biofilm inhibition activity against the pathogenic bacteria Streptococcus mutans, Lysinibacillus fusiformis, Proteus vulgaris, and Vibrio parahaemolyticus. The essential oil of Meriandra dianthera (Konig ex Roxb.) Benth. (Synonym: Meriandra bengalensis , Lamiaceae) collected from Saudi Arabia was studied utilizing GC and GC/MS. Forty four constituents were identified, representing 96.8% of the total oil. The M. dianthera essential oil (MDEO) was characterized by a high content of oxygenated monoterpenes (76.2%). Camphor (54.3%) was the major compound in MDEO followed by 1,8-cineole (12.2%) and camphene (10.4%). Moreover, MDEO was assessed for its cytotoxic, antimicrobial, and antioxidant activities. MDEO demonstrated an interesting cytotoxic activity against all cancer cell lines with IC 50 values of 83.6 to 91.2 μg/mL, especially against MCF-7 cancer cells. Using labeling with annexin VFITC and/or propidium iodide (PI) dyes and flow cytometer analysis, the apoptosis induction was quantitatively confirmed for MCF-7 cells. The MDEO exhibited a considerable antimicrobial activity against all bacterial and fungal strains with minimum inhibitory concentration (MIC)-values of 0.07 to 1.25 mg/mL. The most sensitive microbial strain was Staphylococcus aureus (MIC: 0.07 mg/mL). Minimum bactericidal concentration (MBC) or minimum fungicidal concentration (MFC) values were determined one time higher than that of MIC's. Additionally, the MDEO revealed a strong activity for reducing β-carotene bleaching with a total antioxidant value of 72.6% and significant DPPH free radical scavenging activity (78.4%) at the concentration 1000 μg/mL.

Recently, biogenic nanoparticles have been considered promising candidates for manufacturing antibacterial nanodrugs. Here, we synthesized AgNPs using the crab-borne antibacterial agent hemocyanin and assessed the antibacterial action against several pathogenic bacteria. In this study, the crustacean immune protein hemocyanin (Pp-Hc, 78 kDa) purified from Portunus pelagicus hemolymph was used to fabricate silver nanoparticles. Characterization of hemocyanin-fabricated AgNPs (Pp-Hc AgNPs) were achieved using ultraviolet-visible spectrophotometer, X-ray powder diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), high-resolution-transmission electron microscopy (HR-TEM), and energy-dispersive X-ray spectroscopy. The antibacterial efficacy of AgNO 3, Pp-Hc and Pp-Hc AgNPs was compared by growth inhibition, antibiofilm and live and dead analyses. Based on the results, Pp-Hc AgNPs was more efficient than Pp-Hc and AgNO 3 against pathogenic bacteria. Mechanistic analysis revealed membrane damage and reactive oxygen species (ROS) generation, suggesting that Pp-Hc and Pp-Hc AgNPs rely to similar modes of action. Intracellular protein molecules and nucleic acid leakage confirmed that Pp-Hc AgNPs increase membrane permeability, leading to cell death. Based on our results, capping of the exterior surface of nanoparticles with antimicrobial crab-borne peptides, such as Pp-Hc, improves their functions as potential agents against bacterial diseases, which may be useful in clinical applications., (Copyright © 2019 Elsevier GmbH.
