Screening of phytochemical Ephedra alte crude extract by GC–MS and HPLC analysis indicated the presence of alkaloids, tannins, flavonoids, terpenoids, and phenolic acid in the extract. The total phenolic content of E. alte methanol extract was 39.43 mg of Gallic acid eq/g, crude E. alte with 56.74, and 2.42 µg Trolox equivalent antioxidant capacity (TEAC)/g of plant extract according to DPPH and FRAP assay, respectively. The antimicrobial activity of E. alte against Staphylococcus aureus, staphylococcus epidermidis, Escherichia coli, and Klebsiellaoxytoca demonstrated a mean zone diameter of inhibition ranging from 0 to 17 mm. The MIC of the extracts ranged from 0.5 to 1.0 mg/mL. E. alte extract inhibits pepsin enzyme activity with IC50 values of 213.67 µg/ml. This study revealed that E. alte extract has pepsin enzyme inhibitory, antibacterial, antioxidant activities. The current outcomes indicate that E. alte might be employed as a natural agent for managing GERD and infectious diseases.

Bacterium Aeromonas hydrophila (A. hydrophila) was isolated from the Petra Wastewater Treatment Plant effluent in southern Jordan. It was identified by using morphological and biochemical characteristics. A. hydrophila was found to be able of using chlorobenzoate compounds as carbon and energy source. These capabilities were with different biodegradation rates (4- chlorobenzoic acid 5µM/hr, 3,4-dichlorobenzoic acid 15.5µM/hr, 2- chlorobenzoic acid 41µM/hr and 3- chlorobenzoic acid 65µM/hr). The degradation ability was monitored through the release of chloride, disappearance of the substrate and finally the growth of bacterial cells on these substrates. A. hydrophila dioxygenases physiologically induced by chlorobenzoic acid compounds, were analyzed for both ortho or meta ring-cleavage of these aromatic compounds. Only 1, 2-dioxygenase activity was detected which suggest that the cleavage is through the ortho pathway. The best results of degradation of 2-CBA compound were obtained with 3mM substrate concentration, 25ºC, pH 7and 200µl inoculum size. The carbon sources affected the 2-CBA degradation differently from that on chloride and cell mass production. Nitrogen sources used reduced the degradation activity of the 2-CBA as well as in the chlorine release from 2-CBA. However, the nitrogen source L-proline had a slight enhancement effect on the biodegradation between the 40-80h.

Jordan’s mining sector is considered the cornerstone to the Jordanian economy. It plays a vital role in growth of many other important sectors in the economy and contributes to the Jordan’s standard of living. The mining industry in Jordan is dominated by the production of phosphate, potash, building and decoration stones, glass sand and other nonmetallic resources. Recently, Jordan has been ranked as the sixth largest producer and the second largest exporter of phosphate, the fourth largest producer and the second largest exporter of potash, and has the 5th largest oil-shale reserves in the world. Also, Jordan has sustainable amounts of uranium around 3% of the world resources. The main objective of this research is to focus on the present status of mining industry in Jordan with the aim to emphasis the most important industries, and its effect on the economy of Jordan in order to generate new wealth depends on its mineral endowment. A literature review has been carried out by reviewing the most published articles and the publicly available reports published by various Jordanian Institutions as well as from the private sector. The author reveals that there are many positive outlooks which are looming with an increasing domestic added value along with investing in other areas, such as feldspar, clays, travertine, gypsum, zirconium, uranium, copper and rare earth elements (REE). This requires activating the exploitation of many other untapped mineral resources in the country to achieve a progress that works for meeting the global great demands worldwide. In this context, successive governments have put effort into policies that promote and facilitate investment in the mining sector in Jordan. A comprehensive outlook of the mining sector in Jordan has been discussed in this article. The author also highlighted the opportunities and threats in this sector and more important dilemmas focused on what needs to be carried out by the Jordanian government, including important recommendations to encourage investment in this field and inspire investors in the mining industry in Jordan.

Klebsiella oxytoca was grown in batch cultures using a 2-chlorobenzoic acid (2-CBA) containing minimal salt medium. 2-CBA was the sole source for carbon and energy.1H NMR and electron-impact mass spectra results suggested an ortho-cleavage pathway of the 2-CBA degradation. An unstructured model with substrate inhibition was successfully applied to the 2-CBA experimental data at different concentrations. The maximum specific growth rate, saturation constant, and substrate inhibition constant were found to be 0.8573 h−1, 10.441 mM, and 50.95 mM, respectively. All carbon sources exhibited different growth kinetics and caused an increase in bacterial growth several times (2−4 fold). Glucose, sorbitol, and fructose at 0.2% concentration, as additional carbon sources, decreased the initial degradation rates of 2-CBA at ranges of 20−70% lower than that of the control. Enterobacter aerogenes is generally found in soil, sewage plants, and human gastrointestinal tract. Thus, this study was conducted to evaluate the ability of Enterobacter aerogenes to degrade four chlorobenzoic acid compounds (2-chlorobenzoic acid (2-CBA), 3-chlorobenzoic acid (3-CBA), 4-chlorobenzoic acid (4-CBA), and 3,4-dichlorobenzoic acid (3,4-dCBA)) in minimal salt medium. Enterobacter aerogenes was partially able to degrade and dechlorinate these CBAs at concentration of 3.5 mM within 72 h of incubation. According to Haldane single-substrate model, the values of maximum predicted growth rate (μmax), half saturation constant (K s), and inhibition constant (K i) fell in the range of 0.2–0.8 h−1, 8–41 mM, and 5–53 mM, respectively. Punica granatum L. (Punicaceae) peels extract had the highest free radical scavenging capacity among the tested medicinal plants which are being used traditionally for treatment of diabetes in Jordan. Accordingly, the present study aimed to investigate the antioxidant effect of P. granatum peel methanolic extract against oxidative damage in streptozotocin-induced diabetic rats. The antioxidant activity of P. granatum peel extract was investigated by examining the level of antioxidant enzymes, catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione-S-transferase (GST) and glutathione reductase (GR), the serum total antioxidant capacity and lipid peroxidation in the tissues of treated diabetic rates comparing with normal and untreated diabetic ones. The results revealed that intraperitoneal administration of 10 and 20 mg kg−1 (body weight) of P. granatum peel extract for 4 weeks significantly enhanced the activities of antioxidant enzymes in liver, kidney and RBC of STZ-induced diabetic rats. The extract also caused a significant reduction in malondialdehyde (MDA), a lipid peroxide's marker, in diabetic rat tissues and elevated the total serum antioxidant capacity in dose-dependent manner. In conclusion, this study clearly showed that P. granatum peel extract has protective role against the oxidative damage in STZ-induced diabetic rats.

A bacterial strain was isolated from Petra City Wastewater Treatment Plant. This isolate was identified as Klebsiella oxytoca based on 16S rDNA analysis. A single plasmid (> 23 kb) was detected in this strain and transformed into Esherichia coli JM83. The transformed E. coli cells exhibited elevated resistance to cadmium as compared to parental plasmid-free cells. The sodium dodecyl sulfate (SDS)-treated cells showed higher efficiency in plasmid curing than the ethidium bromide–treated cells. The ethidium bromide–cured cells grew only in a 10 μ g/ml Cd+ 2 minimal tolerable concentration, whereas the SDS-treated cells had no growth in any of the Cd concentrations tested (2, 5, 10, 20, 30, 40, and 50 ppm). Contrary to the Freundlich model, the Langmuir model gave a good fit to the Cd biosorption data by K. oxytoca cells. This study was conducted to determine the prevalence rate of VTEC in slaughtered sheep and goats and to evaluate the contamination rate of VTEC in slaughterhouses and butchers' shops in southern Jordan. 201 E. coli isolates from animals' faecal samples and 33 E. coli isolates from slaughterhouse/butcher shop samples were characterized by multiplex PCR (mPCR) reaction for detection of stx1, stx2, eae A and E-hly A virulent genes. Twenty-six virulent E. coli isolates were characterized by mPCR to seven different virulent patterns: stx1, stx1+stx2, stx1+eae A, stx1+E-hly A, stx1+eae A+E-hly A, eae A and E-hly A. It was found that VTEC comprised 6.4% and 21% of the total E. coli isolates from slaughtered small ruminants and slaughterhouses/ butchers' shops, respectively. The VTEC comprised 76.2% of the virulent isolates. The proportion of stx1:stx1+stx2 patterns was 19:1. It was found that the characterized complex VTEC (containing eae A and/or E-hly A) possessed three virulence patterns, including (VTEC) stx1 +eae A, (VTEC/EHEC) stx1 +E-hly A and (VTEC/EHEC) stx1 +eae A +E-hly A in percentages of 30%, 25% and 10%, respectively, in relation to the total VTEC isolates. Only two VTEC isolates were characterized as E. coli O157 and O26 serotypes, as highly pathogenic strains. Each of the O157 and O26 VTEC isolates was in a percentage of 0.4% in relation to the total E. coli isolates with virulent patterns stx1, eae A and E-hly A. The rest of the VTEC isolates were non-O157 VTEC. The antibiotic sensitivity test showed that the isolated VTEC was highly sensitive to gentamicin and co-trimoxazole and highly resistant to tetracycline and ampicillin. The anionic surfactants linear alkylbenzosulfonate (LAS) and sodium dodecyl sulfate (SDS) were degraded by a consortium of the mixed facultative anaerobes Acinetobacter calcoaceticus and Pantoea agglomerans which were isolated from wastewater. The growth of this consortium in nutrient broth medium at temperature of 30 °C; pH 8.5; and agitation rates of 250 rpm, was able to degrade high biomass of the two surfactants. Under these growth conditions a complete degradation of 4000 ppm SDS biomass was achieved within 120 h incubation time while a longer period (150 h) was capable of degrading only 60% of 300 ppm LAS biomass. However, a full degradation of LAS was accomplished by additional supplementation of the mixed culture with some carbon sources. Also, an alternative supplementation with nitrogen nutrients has increased the biodegradation extent of LAS from 60% to 90%. In contrast, such supplementation of the mixed culture with carbon and nitrogen nutrients had adverse effects on the induction of SDS degradation. Both surfactants at higher concentrations became toxic to the mixed co-culture bacteria particularly affecting the A. calcoaceticus strain of the consortium. The supplementation of either surfactant as sole carbon source to the mixed culture growing in minimal medium produced 10% degradation of SDS and 60% degradation of LAS, respectively. These data are discussed in terms of differences between the enzymic induction of LAS and SDS bacterial biodegradation.

The rate of biodegradation of phenol by Klebsiella oxytoca strain was studied in the nutrient broth and M9 minimal medium. It was found that K. oxytoca degrade phenol at elevated phenol concentration where 75% of initial phenol concentration of 100 ppm will degrade within 72 h. This rate was increased with increasing the initial cell densities, increasing the aeration rate and increasing the time required for complete degradation. At phenol concentration above 400 ppm, the cells were unable to degrade the substrate efficiently due to the increasing concentration of phenol in the medium. The culture conditions were also showed a significant impact on the ability of these cells to remove phenol. The optimum solution pH and temperature were 6.8 and 37°C, respectively. Chlorinated benzoic acids (CBA) have been intentionally released to the environment due to their use in agriculture as herbicides or pesticides, or unintentionally because they are common metabolites in the aerobic transformation of many chlorinated pollutants. The biodegradation of 3,4-dichlorobenzoic acid compound was investigated by using Corynebacterium jeikeium bacteria, which was isolated from Petra wastewater plant in Jordan. 3,4-dichlorobenzoic acid compound (3,4-DCBA) was used as a sole carbon and energy source in minimal salt media (MSM). 3,4-DCBA was the most degradable compound among the Chlorobenzoic acid compounds tested by this bacteria. Different conditions such as substrate concentration, temperature, pH, agitation rate, carbon starvation and carbon adaptation were used to obtain optimal Biodegradation. The optimal conditions for the biodegradation were 3mM as substrate concentration and the culture conditions were also showed a significant impact on the ability of these cells to remove 3,4-DCBA. The optimum solution, temperature and agitation rate were 7.5, 37 °C and 150 rpm, respectively. Adaptation of the cells to 3,4-DCBA for 24 hr and 48 hr and cells starvation for 24 hr and 48 hr increased the initial degradation rate. The degradation ability was monitored through disappearance of the substrate from the medium and the measuring the accompanying chloride release. Corynebacterium jeikeium dioxygenases were physiologically induced by 3,4-DCBA compound. They were analyzed for both ortho or meta ring-cleavage of this aromatic compound. Only 1,2-dioxygenase activity was detected which mean that the cleavage is through the ortho pathway. This microorganism can be a valuable and promising candidate for use in the biotreatment of wastewater samples contaminated with 3,4-DCBA.

The effect of temperature stresses on Cefaclor suspensions under different storage conditions for a duration of 14 days was tested. The degradation of Cefaclor was determined on the 2nd, 7th and 14th day after reconstitution using a sensitive and precise Reversed phase High Performance Liquid Chromatographic (RP-HPLC) method. The RSD values for Forticef, Midocef, Ceclor, Cefabac and Cloracef, indicated a good precision of the RP-HPLC method. The limit of detection (LOD) and the limit of quantification (LOQ) were found 0.008 mg/ml and 0.03mg/ml respectively. The antimicrobial effect of Cefaclor suspension was also tested against pathogenic bacteria using the cylinder diffusion method. The RSD values range of the antimicrobial assay for all the Cefaclor compounds were 1.47-3.7%. The LOD and LOQ were 0.2mg/ml and 1mg/ml respectively. During the normal use of Ceclor, Midocef, and Forticef the loss of activity and the degradation were less than 5% on the 14th day of preservation at 4°C. However, the percentage of degradation for Cefabac and Cloracef on the 14th day reached 5 and 6%, respectively. Statistical multiple comparison between the effect of 4°C and 25°C indicated non significant mean differences (P0.05) for Forticef, Cefabac, Ceclor and Cloraf and significant effect for Midocef (P0.05). Significant effects were observed between (4oC and 37°C) and (25°C and 37°C) for all except Ceclor. Multiple comparisons between days of storage showed non significant mean difference values at 4°C except Cefabac. However significant results between days were found at 25°C and 37°C except for Midocef between (7th and 14th day). It was found that the pediatric suspensions of Cefaclor in the Jordanian market were stable and contained the amount of active ingredient specified by the United States pharmacopoeias specification (USP) and the British Pharmacopoeias specifications (BP).

A Neurospora crassa cytoplasmic ribosomal protein gene, crp-5, has been isolated and characterized. The cDNA was isolated by a differential screening of a cDNA library for glucose-inducible genes. The cDNA was subsequently used to identify and isolate crp-5 genomic sequences. Computer analysis of the DNA sequences showed that they contain an open reading frame which encodes a protein homologous to the rat ribosomal protein S26. The crp-5 mRNA levels are regulated in a carbon-source-dependent manner. The organization of the gene and the region upstream of the coding sequences are discussed.

This study was carried out to evaluate the antibacterial activity of aqueous and organic extracts of Thymus capitatus L. (Lamiaceae) leaves and stems. Dried ground powder leaves and stems were extracted with water (aqueous extracts), ethanol, dichloromethane and hexane (Soxhlet extracts). The antibacterial activity of these extracts was evaluated against bacteria using disc diffusion method. The result obtained showed that the leaves had stronger antibacterial activity than the stems extracts. The ethanolic extract had the highest yield products and the high antibacterial activity than all other solvents. The results suggest that essential oil as non-polar organic compounds could be the main active compounds in this plant. Therefore the antibacterial activity of leaves ethanol extracts (LEE) was compared with essential oils leaves extracts (LEO) of T. capitatus. The LEO showed greater antibacterial activity than LEE. The LEO showed a broad spectrum of antibacterial activity and the Pseudomonas aeruginosa was the most sensitive bacteria.

Pseudomonas aeruginosa was isolated from infected burn patients and characterized by standard biochemical tests. The in vitro copper uptake was compared between this isolated pathogenic strain and two non-pathogenic control strains of Gram positive bacteria Bacillusthuringiensis strain Israelis as well as Gram negative bacteria Enterobacter aerogenes. Maximum copper uptake of 470 ppm/g biomass was obtained by P. aeruginosa strain, while the control strains B. thuringiensis and Enterobacter aerogenes had copper uptake of 350 and 383 ppm/g biomass, respectively. However, the lowest copper uptake (60 ppm/g biomass) was observed with another control the saprophytic strain Pseudomonas (Shewanella) putrefaciens. A further investigation regarding the effect of copper toxicity on bacterial growth, gave an MIC score of 600 ppm for P. aeruginosa strain compared to 460 and 300 ppm for the two Gram positive and Gram negative control strains, respectively. In tandem with these in vitro findings, blood analysis on burn patients infected with P. aeruginosa has indicated a selective decrease of copper (hypocupremia) and ceruloplasmin plasma levels. The iron metabolism was also affected by this copper deprivation leading to a similar decrease in plasma levels of PCV, iron, total iron binding capacity, and transferrin. A successful attempt was made to isolate linear alkylbenzene sulfonate (LAS)-degrading bacteria from soil irrigated with wastewater. The isolated bacteria were able to use LAS as sole carbon and energy source. Maximum growth rates on LAS reached only 0.27 h(-1). 16S-rRNA sequencing and fatty-acid analysis placed the bacteria in the genus Enterobacter cloacae. The growth curves of E. cloacae both in the presence of and the absence of LAS were monitored using measurements of optical density at 600 nm in two different media, nutrient broth and M9 minimal medium, and were modeled mathematically. Growth in NB fit the Riccati and Voltera models, indicating that LAS is not toxic to E. cloacae cells. However, growth of E. cloacae in LAS-containing MM fit the Riccati and Voltera models, whereas growth in LAS-free MM fit the Riccati model only. Furthermore, the kinetic data shown were modeled by Monod's, Andrew's, and Tessier's specific growth rate equations, coupled with the rate of consumption of different concentrations of LAS as sole carbon and energy source, and we determined that Andrew's model best fit these data adequately as a result of the cell-inhibitory effect.

The anti-fertility, anti-implantation, and ovarian histological alterations of the ethanolic extract of Ferula hormonis have been investigated in female mice. The intragastric application of 3 mg/kg per day of such extract for 6 weeks resulted in a significant reduction in female mice fertility. Furthermore, it caused a decrease in the number of mated females, the total number of implantations, and the number of viable fetuses. These changes were also associated with ovarian atrophy and a concomitant increase in the connective tissue. The ova showed degeneration while most of the ovarian follicles suffered follicular atresia.

The effects of an aqueous extract of Ferula hormonis on social aggression, fertility and some physiological and biochemical parameters were investigated in male mice. The ingestion of 3 mg/kg of aqueous extract of F. hormonis for six weeks clearly inhibited social aggression. Body wet weight and other sex accessory organ weights were significantly reduced by this treatment. The ingestion of this extract by male mice resulted in a significant reduction of their fertility. This treatment caused a significant decrease in the number of pregnant females, number of implantations and viable fetuses in females impregnated by males that ingested this extract. Additionally, the numbers of epididymal sperm and their motility were dramatically reduced in F. hormonis-treated mice. Concomitant increases in sperm abnormalities were also observed when compared with control. These data indicate that F. hormonis exposure during this period puts the exposed animals at significant risk for reduced reproductive capacity in adulthood.

We have isolated and sequenced a Neurospora crassa gene encoding a single copy of ubiquitin (UBI) fused to the S27a ribosomal (r) protein. We have opted to name this gene the ubiquitin/cytoplasmic r-protein gene 6 (ubi::crp-6). The ubi::crp-6 gene generates a 700-nucleotide (nt) transcript. It shares a 700-bp regulatory region with the cytoplasmic r-protein gene 5 (crp-5), a gene encoding the N. crassa S26 r-protein. The two genes are transcribed divergently from the common regulatory region and their mRNA levels are regulated in parallel during growth on a variety of carbon sources.
