Abstract Exploring the mechanism of microbiota assembly and its ecological consequences is crucial for connecting microbiome variation to ecosystem function. However, the influencing factors underlying microbiota assembly in the host-microbe system and their impact on the host phenotype remain unclear. Through investigating the prevalent and worsening ecological phenomenon of insecticide resistance in global agriculture, we found that insecticide exposure significantly changed the gut microbiota assembly patterns of a major agricultural invasive insect pest, Spodoptera frugiperda. The relative importance of various microbiota assembly processes significantly varied with habitat heterogeneity and heterogeneous selection serving as a potential predictor of the host’s insecticide resistance in field populations. Moreover, disturbance of the gut microbiota assembly through antibiotics was revealed to significantly affect the rate and heritability of insecticide resistance evolution, leading to a delay in insecticide resistance evolution in this insect pest. These findings indicate that the gut microbiota assembly process of the insect host is influenced by persistent exposure to habitat conditions, particularly insecticides. This variation in insecticide exposure-related community assembly process subsequently influences the insect host’s insecticide resistance phenotype. This study provides insights into gut microbiota assembly processes from a symbiotic perspective and underscores the significant impact of symbiotic community changes on host phenotypic variation.

Abstract In higher plants, mature male gametophytes have distinct apertures. After pollination, pollen grains germinate, and a pollen tube grows from the aperture to deliver sperm cells to the embryo sac, completing fertilization. In rice, the pollen aperture has a single-pore structure with a collar-like annulus and a plug-like operculum. A crucial step in aperture development is the formation of aperture plasma membrane protrusion (APMP) at the distal polar region of the microspore during the late tetrad stage. Previous studies identified OsINP1 and OsDAF1 as essential regulators of APMP and pollen aperture formation in rice, but their precise molecular mechanisms remain unclear. We demonstrate that the Poaceae-specific OsSRF8 gene, encoding a STRUBBELIG-receptor family 8 protein, is essential for pollen aperture formation in Oryza sativa. Mutants lacking functional OsSRF8 exhibit defects in APMP and pollen aperture formation, like loss-of-function OsINP1 mutants. OsSRF8 is specifically expressed during early anther development and initially diffusely distributed in the microsporocytes. At the tetrad stage, OsSRF8 is recruited by OsINP1 to the pre-aperture region through direct protein-protein interaction, promoting APMP formation. The OsSRF8-OsINP1 complex then recruits OsDAF1 to the APMP site to co-regulate annulus formation. Our findings provide insights into the mechanisms controlling pollen aperture formation in cereal species.

Abstract Background Human hematopoietic organoids have a wide application value for modeling human bone marrow diseases, such as acute hematopoietic radiation injury. However, the manufacturing of human hematopoietic organoids is an unaddressed challenge because of the complexity of hematopoietic tissues. Methods To manufacture hematopoietic organoids, we obtained CD34+ hematopoietic stem and progenitor cells (HSPCs) from human embryonic stem cells (hESCs) using stepwise induction and immunomagnetic bead-sorting. We then mixed these CD34+ HSPCs with niche-related cells in Gelatin-methacryloyl (GelMA) to form a three-dimensional (3D) hematopoietic organoid. Additionally, we investigated the effects of radiation damage and response to granulocyte colony-stimulating factor (G-CSF) in hematopoietic organoids. Results The GelMA hydrogel maintained the undifferentiated state of hESCs-derived HSPCs by reducing intracellular reactive oxygen species (ROS) levels. The established hematopoietic organoids in GelMA with niche-related cells were composed of HSPCs and multilineage blood cells and demonstrated the adherence of hematopoietic cells to niche cells. Notably, these hematopoietic organoids exhibited radiation-induced hematopoietic cell injury effect, including increased intracellular ROS levels, γ-H2AX positive cell percentages, and hematopoietic cell apoptosis percentages. Moreover, G-CSF supplementation in the culture medium significantly improved the survival of HSPCs and enhanced myeloid cell regeneration in these hematopoietic organoids after radiation. Conclusions These findings substantiate the successful manufacture of a preliminary 3D hematopoietic organoid from hESCs-derived HSPCs, which was utilized for modeling hematopoietic radiation injury and assessing the radiation-mitigating effects of G-CSF in vitro. Our study provides opportunities to further aid in the standard and scalable production of hematopoietic organoids for disease modeling and drug testing.

Abstract Background Emerging data suggested a favorable outcome in advanced non-small cell lung cancer (NSCLC) with chronic obstructive pulmonary disease (COPD) patients treated by immunotherapy. The objective of this study was to investigate the effectiveness of neoadjuvant immunotherapy among NSCLC with COPD versus NSCLC without COPD and explore the potential mechanistic links. Patients and methods Patients with NSCLC receiving neoadjuvant immunotherapy and surgery at Shanghai Pulmonary Hospital between November 2020 and January 2023 were reviewed. The assessment of neoadjuvant immunotherapy’s effectiveness was conducted based on the major pathologic response (MPR). The gene expression profile was investigated by RNA sequencing data. Immune cell proportions were examined using flow cytometry. The association between gene expression, immune cells, and pathologic response was validated by immunohistochemistry and single-cell data. Results A total of 230 NSCLC patients who received neoadjuvant immunotherapy were analyzed, including 60 (26.1%) with COPD. Multivariate logistic regression demonstrated that COPD was a predictor for MPR after neoadjuvant immunotherapy [odds ratio (OR), 2.490; 95% confidence interval (CI), 1.295–4.912; P = 0.007]. NSCLC with COPD showed a down-regulation of HERV–H LTR-associating protein 2 (HHLA2), which was an immune checkpoint molecule, and the HHLA2low group demonstrated the enrichment of CD8+CD103+ tissue-resident memory T cells (TRM) compared to the HHLA2high group (11.9% vs. 4.2%, P = 0.013). Single-cell analysis revealed TRM enrichment in the MPR group. Similarly, NSCLC with COPD exhibited a higher proportion of CD8+CD103+TRM compared to NSCLC without COPD (11.9% vs. 4.6%, P = 0.040). Conclusions The study identified NSCLC with COPD as a favorable lung cancer type for neoadjuvant immunotherapy, offering a new perspective on the multimodality treatment of this patient population. Down-regulated HHLA2 in NSCLC with COPD might improve the MPR rate to neoadjuvant immunotherapy owing to the enrichment of CD8+CD103+TRM. Abstract As one of the most intractable neurological diseases, spinal cord injury (SCI) often leads to permanent neurological impairment in patients. Unfortunately, due to the complex pathological mechanisms and unique postinjury microenvironment, there is currently no way to completely repair the injured spinal cord. In recent years, with the rapid development of tissue engineering technology, the combination of biomaterials and medicine has provided a new idea for treating SCI. Here, we systematically summarize representative biomaterials, including natural, synthetic, nano, and hybrid materials, and their applications in SCI treatment. In addition, we describe several state-of-the-art fabrication techniques for tissue engineering. Importantly, we provide novel insights for the use of biomaterial-based therapeutic strategies to reduce secondary damage and promote repair. Finally, we discuss several biomaterial clinical studies. This review aims to provide a reference and new insights for the future exploration of spinal cord regeneration strategies.

Conventional oxidation processes for alkyl aromatics to ketones employ oxidants that tend to generate harmful byproducts and cause severe equipment corrosion, ultimately creating critical environmental problems. Thus, in this study, a practical, efficient, and green method was developed for the synthesis of aromatic ketones by applying a bis(2-butoxyethyl) ether/O2 system under external catalyst-, additive-, and base-free conditions. This O2-mediated oxidation system can tolerate various functional groups and is suitable for large-scale synthesis. Diverse target ketones were prepared under clean conditions in moderate-to-high yields. The late-stage functionalization of drug derivatives with the corresponding ketones and one-pot sequential chemical conversions to ketone downstream products further broaden the application prospects of this approach.

The Geostationary Interferometric InfraRed Sounder (GIIRS) provides a novel opportunity to acquire high-spatiotemporal-resolution atmospheric information. Previous studies have demonstrated the positive impacts of assimilating GIIRS radiances from either long-wave temperature or middle-wave water vapor bands on modeling high-impact weather processes. However, the impact of assimilating both bands on forecast skill has been less investigated, primarily due to the non-identical geolocations for both bands. In this study, a locally cloud-resolving global model is utilized to assess the impact of assimilating GIIRS observations from both long-wave and middle-wave bands. The findings indicate that the GIIRS observations exhibit distinct inter-channel error correlations. Proper inflation of these errors can compensate for inaccuracies arising from the treatment of the geolocation of the two bands, leading to a significant enhancement in the usage of GIIRS observations from both bands. The assimilation of GIIRS observations not only markedly reduces the normalized departure standard deviations for most channels of independent instruments, but also improves the atmospheric states, especially for temperature forecasting, with a maximum reduction of 42% in the root-mean-square error in the lower troposphere. These improvements contribute to better performance in predicting heavy rainfall.

While a multitude of opinion mining studies have examined the impact of attribute performance on customer satisfaction, they have offered limited insights into the role performance of service employees by investigating tourists’ experiences. Focusing on the hotel industry, this study investigated tourists’ evaluations of service personnel by scrutinizing 68,439 hotel reviews and explored the asymmetric effects of employee performance on customer satisfaction according to service employee functions and traveler types. The results indicate that different travelers have distinct expectations of different kinds of services, thereby moderating the asymmetric impact of role performance on tourist satisfaction. Manager employee, in particular, constitutes an excitement factor for family, solo, and couple travelers. The findings have implications for the formulation of operation strategies to promote tourists’ satisfaction rather than frustration. This study contributes to the literature by offering a novel understanding of the asymmetric effect of customer–personnel interactions on tourist satisfaction.

Abstract Background Patients with non‐small cell lung cancer (NSCLC) and chronic obstructive pulmonary disease (COPD) experience worse clinical outcomes but respond better to immunotherapy than patients with NSCLC without COPD. Mucosal‐associated invariant T (MAIT) cells, a versatile population of innate immune T lymphocytes, have a crucial function in the response to infection and tumors. This study investigated the distribution of MAIT cells in COPD‐associated NSCLC and their involvement in the immune response. Methods Flow cytometry, immunohistochemistry, and immunofluorescence were performed on tissue samples of patients with NSCLC, with or without COPD, treated with or without anti‐programmed death 1 (PD1) immunotherapy. MAIT cells were stimulated with 5‐OP‐RU using a mouse subcutaneous tumor model. Results Tumors contained significantly more MAIT cells than paraneoplastic tissues, and CD8+ MAIT cells accounted for more than 90% of these cells. Patients with NSCLC and COPD had higher CD8+ MAIT cell counts than those with NSCLC without COPD. Additionally, patients with NSCLC and COPD displayed reduced expression of the activation marker, CD69, and functional markers, granzyme B (GZMB) and interferon γ (IFNγ), and higher expression of the immune exhaustion marker, PD1. Among patients who received immunotherapy, the proportion with a complete or partial response was higher in those with COPD than in those without COPD. In patients with NSCLC and COPD, the major pathologic response (MPR) group had higher MAIT levels than those in the no major pathologic response (NPR) group. In the mouse subcutaneous tumor model stimulation of MAIT cells using 5‐OP‐RU enhanced the antitumor effects of anti‐PD1. Conclusions In patients with NSCLC and COPD, response to immunotherapy is associated with accumulation of CD8+ MAIT cells showing immune exhaustion. These findings may contribute to innovative approaches for immunotherapy targeting CD8+ MAIT cells.

Abstract Breakdown of reproductive isolation facilitates flow of useful trait genes into crop plants from their wild relatives. Hybrid sterility, a major form of reproductive isolation exists between cultivated rice (Oryza sativa) and wild rice (O. meridionalis, Mer). Here, we report the cloning of qHMS1, a quantitative trait locus controlling hybrid male sterility between these two species. Like qHMS7, another locus we cloned previously, qHMS1 encodes a toxin-antidote system, but differs in the encoded proteins, their evolutionary origin, and action time point during pollen development. In plants heterozygous at qHMS1, ~ 50% of pollens carrying qHMS1-D (an allele from cultivated rice) are selectively killed. In plants heterozygous at both qHMS1 and qHMS7, ~ 75% pollens without co-presence of qHMS1-Mer and qHMS7-D are selectively killed, indicating that the antidotes function in a toxin-dependent manner. Our results indicate that different toxin-antidote systems provide stacked reproductive isolation for maintaining species identity and shed light on breakdown of hybrid male sterility.

AimTo systematically appraise, compare, and summarize the measurement properties of intimate partner violence screening instruments for the general population and provide recommendations.MethodsWe searched PubMed, Embase, Web of Science, ProQuest Dissertations & Theses Global and EBSCO Psychology Behavioral Sciences Collection from their establishment to March 2024 using systematic search strategies. The methodological quality of the instruments that met the inclusion criteria and their measurement properties were assessed using the COSMIN methodology, and the COSMIN recommendations were followed. We reported this study using PRISMA 2020 checklist.ResultsA total of 23 studies were eventually included, and 18 instruments were identified. The evaluation of the methodological quality indicates poor content validity but good structural validity; however, that of the criterion validity in most of the studies was inadequate. Measurement error and responsiveness were not assessed. Four screening instruments could be used in the interim. The remaining 14 instruments were not recommended for use.ConclusionsThe overall methodological quality of most of the assessed instruments was insufficient. A rigorous intimate partner violence screening instrument with good measurement properties is urgently required to identify and screen for intimate partner violence in the general population.Prospero numberCRD42022365247.

ABSTRACTBackground Hb Chapel Hill [Alpha2 74(EF3) Asp > Gly] results from an GAC > GGC substitution at codon 74 of the HBA1 or HBA2 genes. Hb Chapel Hill has not been reported since 1986.Methods A heterozygous mutation, HBA2: c.224A > G, was identified in the proband, her father and sister. We compared the haematological and clinical data of this family with the data reported in the limited number of individuals.Results Having excluded iron deficiency, the Hb Chapel Hill was asymptomatic in heterozygous state. The cases presented here characterize cases in new techniques including capillary electrophoresis (CE). Two aberrant peaks were identified by CE, a major peak migrating in the zone 7 that correspond to Hb Chapel Hill (αChapel Hill 2β2) and a minor peak migrating in the zone 1 that correspond to Hb Chapel Hill2 (αChapel Hill 2δ2). Focusing on the variant expression, the Hb Chapel Hill plus Hb A2 variant were around 18.9–20.6% of total Hb in three members.Conclusion This data will be useful for providing up-to-date and high quality information on the Hb Chapel Hill.

Genogroup II genotype 4 (GII.4) norovirus causes acute gastroenteritis in children, and its infection is more severe than that of other genotypes. Early and precise detection and treatment are critical for controlling its spread and reducing the severity of infection. In this study, a rapid and efficient isothermal assay for the GII.4 norovirus detection (GII.4-CRISPR detection) was developed based on the CRISPR/Cas13a system. The assay can be applied without expensive instrumentation, and the results can be read via both fluorescence and lateral flow strip (LFS). The analytical sensitivity of this assay was 5 copies/reaction, and there was no cross-reaction with other genotypes of norovirus or other clinically common pathogens. There was a coincidence rate of 100% between our assay and commercial quantitative polymerase chain reaction. GII.4-CRISPR detection improves upon the shortcomings of some previously established molecular methods of detection, particularly with regard to accessibility. It provides an alternative tool for outbreak control and early diagnosis of GII.4 norovirus infection.

The cell wall (CW) is critical for the accumulation of heavy metals in metal-tolerant plants. Polysaccharides, the main component of the CW, contribute significantly to the immobilization of heavy metals. However, the mechanisms of rare earth elements (REEs) adsorption and detoxification by polysaccharides in the cell walls of Phytolacca americana L. (P. americana) remain unclear. In this work, we explored the binding sites of REEs and the modifications to polysaccharides in the cell walls of roots and leaves in P. americana, in order to elucidate the adsorption and fixation mechanism of REEs by the cell wall. Our findings indicated that up to 40.7% and 48.1% of cell-wall-bound REEs were present in the root and leaf pectin, respectively. The removal of pectin led to a 39.8% and 23.6% decrease in the maximum adsorption of REEs in the CW, suggesting that pectin was the main binding site for REEs in the cell walls of P. americana. Hydroxyl (-OH) and carboxyl (-COOH) groups in the cell wall interacted mainly with REEs ions under stress conditions, which played a key role in REEs binding. An obvious REEs fractionation was found during the various fractions of the CW, and all fractions of the root cell wall were enriched with HREEs, whereas all fractions of the leaf cell wall were enriched with LREEs. Moreover, P. americana modulated cell wall composition in reaction to REEs stress. In conclusion, cell wall pectin is the main binding site of REEs, and the functional groups on the cell wall play a significant role in the binding of REEs. At the same time, plants can control the selective adsorption and fixation of REEs by adjusting the composition of cell walls. This study offers valuable insights into the mechanisms of REEs adsorption and fixation in cell walls of P. americana, contributing to a theoretical basis for the bioremediation of REEs pollution.

With the Microwave Humidity Sounder-2 (MWHS-2)/Fengyun (FY)-3D in operation, this is the first study to evaluate the impact of a joint assimilation of MWHS-2 radiances under all-sky conditions from both the FY-3C and FY-3D satellites on typhoon forecasting within regional areas. In this study, Typhoon Hagupit in 2020 was chosen to investigate the impacts of assimilating MWHS-2 radiances; the forecasting performances of the joint assimilation method were slightly better than the experiments assimilating MWHS-2 observations from FY-3C or FY-3D only, and the results of the latter two experiments were comparable, especially in terms of the landfall location of Hagupit. With additional assimilated cloud- and precipitation-affected MWHS-2 observations, improved typhoon track and intensity forecasts as well as forecasts of the precipitation caused by Hagupit were achieved due to the improved analyses of relative humidity, temperature and wind fields around Hagupit compared to the clear-sky assimilation experiments. In addition, the channel-selection scheme evidently affected the forecasting performance; that is, the radiances from the MWHS-2 118 GHz and 183 GHz channels provided opposite results in terms of the Hagupit track, and this finding needs further investigation in the future.

Accurate remote sensing-based forest aboveground biomass (AGB) estimation is important for accurate understanding of carbon accounting and climate change at a large scale. However, over- and underestimation are common in the process, resulting in inaccurate AGB estimations. Here, the AGB was estimated and mapped by combining Landsat 8 images and forest inventory data in western Hunan Province, China. We used forest canopy density (FCD) mapper to quantify the forest canopy structure. The linear model (LR) and piecewise model with FCD gradients (classified by k-means clustering; sparse, medium, and dense) were developed to estimate AGB for each forest type (coniferous, broadleaf, mixed, and total forests). The piecewise model considered the following scenarios: piecewise model using the variables of LR model (PM), and piecewise model using the variables selected for different FCD gradients (PMV). The PM (R2:0.45–0.56) and PMV (R2:0.63–0.75) models showed better agreement between observed and predicted AGB than the LR (R2:0.18–0.27) models, and the PMV model was the most accurate for each forest type. The PM and PMV models performed better than LR models at different FCD gradients. The PM and PMV models can better alleviate the over- and underestimations of the LR models. At different FCD gradients, the PMV models had different variables, indicating that the correlation between the AGB and spectral variables was different. Overall, FCD is an important forest parameter that influences AGB estimation, and the piecewise model has potential to improve remote sensing-based AGB estimation.

Atmospheric motion vectors (AMVs), known as cloud track winds, have positive impacts on global numerical weather forecasts (NWP). In this study, AMVs that were retrieved from Fengyun-2G and Fengyun-4A were compared in their data quality and impacts on the typhoon forecasts in order to investigate the differences between the first and second generation of the geostationary meteorological satellites of China. This report conducted data evaluation and assimilation-forecasting experiments on FY-2G and FY-4A atmospheric motion vectors (AMVs), respectively. The results showed that the AMVs data of FY-4A are of better quality than those of FY-2G and assimilating the AMVs of FY-2G and FY-4A have a neutral to slightly positive impacts on typhoon forecasts, which is quite encouraging for their operational use in the future.

Abstract Atmospheric motion vectors (AMVs) have produced positive impacts on global weather forecasts, but few studies have evaluated the impacts of AMV data from the Fengyun (FY) geostationary satellite series, especially from FY‐2G and FY‐4, on typhoon forecasts in a regional model. In this study, the qualities of FY‐2G and Himawari‐8 AMV data are comparatively evaluated with several preprocesses (e.g., height assignment, quality control, channel merging, thinning, and observation error assignment) employed, and a super typhoon (Typhoon Hato, which occurred in 2017) is selected to assess the forecasting performances with the AMV data assimilated in the Weather Research and Forecasting data assimilation (WRFDA) system. The results show that the AMV data from Himawari‐8 are better overall than those from FY‐2G, with smaller RMSEs and biases for full wind speeds, and the preprocesses improve the qualities of these two geostationary satellite AMV data sets, as they both depict the same general atmospheric circulations. On the other hand, assimilation of AMV data improves the forecasts of environmental fields, resulting in the simulated track and intensity being closer to the observations. Generally, assimilating FY‐2G AMV data has comparably positive impacts with Himawari‐8 AMVs, and the AMV data retrieved from multiple channels provide even better forecasts. Experiments involving another typhoon case are also performed to further provide evidence that the derived AMV data from the Fengyun geostationary satellite series have value in the operational applications.

Hand, foot, and mouth disease (HFMD) is a common infectious disease affecting mainly children under 5 years of age. Coxsackievirus A6 (CVA-6), a major causative pathogen of HFMD, has caused outbreaks in recent years. Currently, no effective vaccine or antiviral treatments are available. In this study, one-step reverse-transcription recombinase polymerase amplification (RT-RPA), combined with a disposable lateral flow strip (LFS) assay, was developed to detect CVA-6. This assay can be performed in less than 35 min at 37°C without expensive instruments, and the result can be observed directly with the naked eye. The sensitivity of the RT-RPA-LFS was 10 copies per reaction, which was comparable to that of the conventional real-time quantitative polymerase chain reaction (qPCR) assays. Moreover, the assay specificity was 100%. The clinical performance of the RT-RPA-LFS assay was evaluated using 142 clinical samples, and the coincidence rate between RT-RPA-LFS and qPCR was 100%. Therefore, our RT-RPA-LFS assay provides a simple and rapid approach for point-of-care CVA-6 diagnosis.

The dynamic emissivity retrieved from window channels of the microwave humidity sounder II (MWHS-2) onboard the China Meteorological Administration’s FengYun (FY)-3C polar orbiting satellite can provide more realistic emissivity over lands and potentially improve the numerical weather prediction (NWP) forecasts. However, whether the assimilation with the dynamic emissivity works for the precipitation forecasts over the complex geography is less investigated. In this paper, a typical precipitating case generated by the Southwest Vortex is selected and the Weather Research and Forecasting data assimilation (WRFDA) system is applied to examine the impacts of assimilating MWHS-2/FY-3C with the uses of the emissivity atlas and the dynamic emissivity on the forecasts. The results indicate that the use of the dynamic emissivity retrieved from the 89 GHz channel of MWHS-2/FY-3C apparently increases the used data number for assimilation and does improve the initial fields and the 24-hour forecasts (from 0000 UTC 24 June 2016 to 0000 UTC 25 June 2016) of precipitation distribution and intensity except for the rainfall over 100 mm. But these positive impacts are not evidently better than those with the emissivity atlas. In general, these results still suggest that the future use of the dynamic emissivity in the assimilation over the complex terrain is promising.

The binary interaction is one of the most challenging factors to improve the forecast accuracy of multiple tropical cyclones (TCs) in close vicinity. The effect of binary interaction usually results in anomalous track and variable intensity of TCs. A typical interaction type, one-way influence mode, has been investigated by many studies which mainly focused on the anomalous track and record-breaking precipitation, such as typhoons Morakot and Goni. In this paper, a typical case of this type, typhoons Tembin and Bolaven, occurred in the western North Pacific in August 2012, was selected to study how one typhoon impacts the track and intensity of the other one. The vortex of Tembin or Bolaven and the monsoon circulation were removed by a TC bogus scheme and a low-pass Lanczos filter, respectively, to carry out the numerical experiments. The results show that the presence of monsoon made the binary interaction more complex by affecting the tracks and the translation speeds of the TCs. The influence of Bolaven on the track of Tembin was more significant than the influence of Tembin on the track of Bolaven, and the looping track of Tembin was also affected by the external surrounding circulation associated with Bolaven. The absence of Tembin was not conducive to the development of Bolaven due to stronger vertical wind shear condition and the less kinetic energy being transported to upper troposphere. Note that the above analysis demonstrates the interacting processes between TCs and sheds some light on the prediction of binary TCs.

The aim of this study was to determine whether the concentration of disintegrin and metalloprotease protein12 (ADAM12) in first trimester maternal serum can be used as a marker for first-trimester complete spontaneous abortions, missed abortions, ectopic pregnancies and hydatidiform moles.The maternal serum concentrations of ADAM12 were measured in the range of 5-9+6 weeks of gestation using an automated AutoDelfia immunoassay platform in 9 cases of complete spontaneous abortion, 27 cases of missed abortions, 56 cases of ectopic pregnancies, 12 cases of hydatidiform moles, and 100 controls. Logistic regression analysis was used to determine significant factors for predicting adverse pregnancy outcomes in early pregnancy. Screening performance was assessed using receiver operating characteristic curves.Two hundred and four women were enrolled in the study. In the control group, the level of ADAM12 increased with gestational age. The median ADAM12 levels in the spontaneous abortion (0.430 MoM), ectopic pregnancy (0.460 MoM) and hydatidiform mole (0.037 MoM) groups were lower than that in the control group, while the median ADAM12 level in the missed abortion group (1.062 MoM) was not significant from the controls (1.002 MoM). Logistic regression analysis demonstrated that the level of ADAM12 in maternal serum facilitated the detection of ectopic pregnancies (OR = 0.909; 95% CI = 0.841 ∼ 0.982) and complete spontaneous abortion (OR = 0.863; 95% CI = 0.787 ∼ 0.946).In complete spontaneous abortion and ectopic pregnancy, ADAM12 maintained at low levels in early pregnancies, and there were significant differences compared to normal pregnancies. ADAM12 is a promising marker for the diagnosis of complete spontaneous abortion and ectopic pregnancy in symptomatic women, and under certain conditions, ADAM12 can diagnose ectopic pregnancy and spontaneous abortion before an ultrasonographic detection of the conditions.
