Abstract In contemporary society, career readiness holds paramount significance for individual life, exerting a direct influence on initial employment, job satisfaction, and the sense of career identity. Framed within multidimensional item response theory and text mining, this study embarks on exploring assessment methodologies for high school students’ career readiness by revising the “Career Readiness Questionnaire – Adolescent Version” and employing text mining techniques. Study One collected 1261 valid data points through cluster sampling. With the aid of Bayesian multivariate item response theory parameter estimation procedures and R language, the career readiness measurement tool was revised, yielding a concise scale that aligns with psychometric requirements. The research findings indicated that the concept of “career readiness” is more suitable for the multidimensional graded response model than for the bifactor model. The dataset’s discrimination parameters fell within the range of [1.59, 3.84], the difficulty parameters fell between [−2.91, 2.24], and the peak values of the maximum information functions fell within [0.24, 2.35]. After six items with the lowest peaks were removed (Items 4, 5, 6, 31, 32, and 33), the remaining 30 items composed the Chinese concise version “Career Readiness Questionnaire – Adolescent Version,” with discrimination parameters ranging from [1.45, 3.38], difficulty parameters between [−3.31, 1.76], and maximum information function peaks within [0.50, 2.64]. Building upon the effective participants from Study One, Study Two matched questionnaire data with textual information, resulting in 1012 valid participants. Leveraging text mining, a machine learning model was constructed to predict high school students’ career readiness based on essay texts. The results of Study 2 prove that the revised lexicon was more accurate in feature extraction. Building upon this, the machine learning model for essay text demonstrated excellent performance in predicting career readiness, with random forest outperforming the other algorithms. This study provides a novel approach for schools and parents to comprehend the state of career readiness among high school students, offering a convenient and effective tool for educational activities related to students’ career development.

Abstract The dynamic surface self‐reconstruction behavior in local structure correlates with oxygen evolution reaction (OER) performance, which has become an effective strategy for constructing the catalytic active phase. However, it remains a challenge to understand the mechanisms of reconstruction and to accomplish it fast and deeply. Here, we reported a photo‐promoted rapid reconstruction (PRR) process on Ag nanoparticle‐loaded amorphous Ni‐Fe hydroxide nanosheets on carbon cloth for enhanced OER. The photogenerated holes generated by Ag in conjunction with the anodic potential contributed to a thorough reconstruction of the amorphous substrate. The valence state of unsaturated coordinated Fe atoms, which serve as active sites, is significantly increased, while the corresponding crystalline substrate shows little change. The different structural evolutions of amorphous and crystalline substrates during reconstruction lead to diverse pathways of OER. This PRR utilizing loaded noble metal nanoparticles can accelerate the generation of active species in the substrate and increase the electrical conductivity, which provides a new inspiration to develop efficient catalysts via reconstruction strategies.

An effective method for the construction of functionalized indolizines has been developed in which β,β-difluoro peroxides act as novel C2-building blocks to implement [3+2] annulation with pyridinium ylides under base-mediated conditions. With this protocol, a broad range of multisubstituted indolizines were prepared in moderate to good yields under mild conditions, and many useful functional groups were tolerated.

High cadmium (Cd, 50 µM) caused transcriptional upregulation of Ca2+-dependent protein kinase 21 (CPK21) in roots and leaves of Populus euphratica. The promoter region (1803 bp) of PeCPK21 was engineered to fuse PeCPK21-pro::GUS and transformed in Arabidopsis. The activity of GUS significantly increased in roots, cotyledons, and hypocotyls under 100 μM CdCl2 treatment. The 1599 bp coding sequence of PeCPK21 was transferred into Arabidopsis, and overexpression of PeCPK21 increased Cd tolerance as evidenced by less reduction in root length and plant growth under CdCl2 stress conditions (100 µM, 7 d). PeCPK21-interacting proteins were enriched by HaloTag pull-down and corresponding expression profiles after Cd exposure indicate that PeCPK21 interacts with heavy metal stress-associated proteins (HMAPs) to mediate Cd tolerance in transgenic Arabidopsis. PeCPK21 interacted with the Cd-regulated cation/heavy metal transport proteins annexin, OPT3, COPT5, and PDF2.2 to limit Cd uptake and accumulation in transgenic plants. PeCPK21 may also interact with the V-type proton ATPase subunits, VHA-B1, VHA-C, and AVA-P2, which were transcriptionally upregulated by Cd to accelerate vacuolar Cd compartmentalization. Cd-triggered H2O2 production was limited in plants overexpressing PeCPK21, which was due to the activated antioxidant enzymes. The interaction of PeCPK21 with the antioxidant enzymes TAPX, APX1, APX2, TRXM4, GPX3, CDSP32, and PRXQ, which showed transcriptional upregulation by Cd, could enhance antioxidant defense in transgenic Arabidopsis. It is also possible that PeCPK21 interacts with plasma membrane intrinsic proteins, PIP1–1, PIP2A, and PIP2–7, which had an Cd-enhanced transcript, to improve water status in transgenic Arabidopsis. In summary, we hypothesize that the calcium sensor PeCPK21 can attenuate Cd toxicity by interacting with HMAPs to effectively limit Cd accumulation, increase ROS scavenging, and improve water status in transgenic Arabidopsis.

BackgroundGiven the lack of research on disulfidptosis, our study aimed to dissect its role in pan-cancer and explore the crosstalk between disulfidptosis and cancer immunity.MethodsBased on TCGA, ICGC, CGGA, GSE30219, GSE31210, GSE37745, GSE50081, GSE22138, GSE41613, univariate Cox regression, LASSO regression, and multivariate Cox regression were used to construct the rough gene signature based on disulfidptosis for each type of cancer. SsGSEA and Cibersort, followed by correlation analysis, were harnessed to explore the linkage between disulfidptosis and cancer immunity. Weighted correlation network analysis (WGCNA) and Machine learning were utilized to make a refined prognosis model for pan-cancer. In particular, a customized, enhanced prognosis model was made for glioma. The siRNA transfection, FACS, ELISA, etc., were employed to validate the function of c-MET.ResultsThe expression comparison of the disulfidptosis-related genes (DRGs) between tumor and nontumor tissues implied a significant difference in most cancers. The correlation between disulfidptosis and immune cell infiltration, including T cell exhaustion (Tex), was evident, especially in glioma. The 7-gene signature was constructed as the rough model for the glioma prognosis. A pan-cancer suitable DSP clustering was made and validated to predict the prognosis. Furthermore, two DSP groups were defined by machine learning to predict the survival and immune therapy response in glioma, which was validated in CGGA. PD-L1 and other immune pathways were highly enriched in the core blue gene module from WGCNA. Among them, c-MET was validated as a tumor driver gene and JAK3-STAT3-PD-L1/PD1 regulator in glioma and T cells. Specifically, the down-regulation of c-MET decreased the proportion of PD1+ CD8+ T cells.ConclusionTo summarize, we dissected the roles of DRGs in the prognosis and their relationship with immunity in pan-cancer. A general prognosis model based on machine learning was constructed for pan-cancer and validated by external datasets with a consistent result. In particular, a survival-predicting model was made specifically for patients with glioma to predict its survival and immune response to ICIs. C-MET was screened and validated for its tumor driver gene and immune regulation function (inducing t-cell exhaustion) in glioma.

Background: The work aimed to compare the pharmacokinetic (PK) profiles and other outcomes reported in observational studies in de novo kidney transplant recipients (KTRs) receiving novel once-daily extended-release tablet tacrolimus (LCPT; LCP-tacrolimus; Envarsus XR) or receiving standard-of-care capsule tacrolimus (PR-Tac; prolonged-release tacrolimus; Advagraf/IR-Tac; immediate-release tacrolimus; Prograf).Methods: A systematic review was conducted for all randomized controlled trials (RCTs) and cohort studies investigating the outcomes in KTRs receiving LCPT or PR-Tac/IR-Tac. We systematically searched PubMed, Web of Science, and EMBASE, with no language restriction. The registered trials and references listed in relevant studies were also searched. Data were extracted for the PK profile, tacrolimus trough level (TTL), and changes in the estimated glomerular filtration rate (eGFR) and serum creatinine (Scr), biopsy-proven acute rejection (BPAR) rate, delayed graft function (DGF) rate, post-transplant diabetes mellitus (PTDM) rate, tremor rate (TR), death rate (DR), and rate of infection by cytomegalovirus (CMV). This study was registered with PROSPERO (registration number: CRD42023403787).Results: A total of seven eligible articles including 1,428 patients with 712 in the LCPT group versus 716 in the PR-Tac/IR-Tac group were included in this study for evidence synthesis. The baseline characteristics of the LCPT, PR-Tac, and IR-Tac groups were similar. The pooled analysis showed a higher PK profile in the LCPT group, and this result was consistent with those of all the included studies. In addition, no significant difference was observed for other outcomes.Conclusion: Considering heterogeneity between studies and potential bias, care providers should select agents based on patient-specific factors and their clinical experience for the immunosuppressive treatment of de novo KTRs.

Since groundwater is one of the main factors that affect the stability of highway slopes in mountainous regions, a smooth drainage is of importance for the safety of cut slopes. After years of service, the cut slopes of highway are often characterized with drainage problems or even drainage failures that threaten the stability and safety. In this paper, the clogging mechanism of drainage holes in highway cut slopes is firstly clarified through on-site survey. The soil and blockage samples are collected from typical slopes for laboratory tests and analysis, and the results reveal that all the blockages are made of fine-grained particles from the slopes. Scanning electron microscope (SEM) images indicate that the blockages penetrate the drainage pipe wrappings to form the clogging. The combined effect of clayey soil, chemicals, and biological clogging exacerbate the clogging process around the drainage pipes. Laboratory tests are also performed to simulate the CaCO3 crystal clogging around the drainage holes, and the microstructure of the clogged geotechnical screens and geotextiles is observed by scanning electron microscope as well. The results confirm that the amount of CaCO3 crystal attached to the drainage pipe surface increases with the time. The single-layer structure of screens does not facilitate the three-dimensional clogging as in the case of geotextiles. After soaking in diluted hydrochloric acid at a pH of 5.0, there is no significant decrease of CaCO3 crystals attached on the screens and geotextiles. However, the CaCO3 decrease is apparent after soaking in the hydrochloric acid at a pH of 3.0. The clogging of drainage holes can be classified into two stages according to the laboratory tests and acid soaking, and the relationship and characteristics between the two stages are summarized.

Natural soils are usually heterogeneous and characterized with complex microstructures. Sand–clay mixtures are often used as simplified soils to investigate the mechanical properties of soils with various compositions (from clayey to sandy soils) in the laboratory. Performing laboratory tests on a sand–clay mixture with definite clay fraction can provide information to understand the simplified soils’ mechanical behavior and better predict natural soils’ behavior at the engineering scale. This paper reviews previous investigations on sand–clay mixture and soil–structure interface direct shear test. It finds that even though there are many investigations on sand–clay mixtures and soil–structure interfaces that consider pure sand or pure clay, limited data on the mechanical behavior of the interface between sand–clay mixture and structure materials are available. Knowledge is missing on how the clay content influences the mechanical behavior of interface and how the soil particles’ arrangement changes as the clay content increases. Further study should be performed to investigate the interface in terms of a reconstituted sand–clay mixture and structure by interface direct shear test, to highlight the influence of clay fraction on the interface response, under various loading conditions.

Background: A key challenge in estimating epidemiological parameters for a pandemic such as the initial COVID-19 outbreak in Wuhan is the discrepancy between the officially reported number of infections and the true number of infections. A common approach to tackling the challenge is to use the number of infections exported from the originating city to infer the true number. This approach can only provide a static estimate of the epidemiological parameters before city lockdown because there are almost no exported cases thereafter. Methods: We propose a Bayesian estimation method that dynamically estimates the epidemiological parameters by recovering true numbers of infections from day-to-day official numbers. To illustrate the use of this method, we provide a comprehensive retrospection on how the COVID-19 had progressed in Wuhan from January 19 to March 5, 2020. Particularly, we estimate that the outbreak sizes by January 23 and March 5 were 11,239 [95% CI 4,794–22,372] and 124,506 [95% CI 69,526–265,113], respectively. Results: The effective reproduction number attained its maximum on January 24 (3.42 [95% CI 3.34–3.50]) and became less than 1 from February 7 (0.76 [95% CI 0.65–0.92]). We also estimate the effects of two major government interventions on the spread of COVID-19 in Wuhan. Conclusions: This case study by our proposed method affirms the believed importance and effectiveness of imposing tight non-essential travel restrictions and affirm the importance and effectiveness of government interventions (e.g., transportation suspension and large scale hospitalization) for effective mitigation of COVID-19 community spread.

The mixing order of silica sand, clay (kaolinite), and water controls the microstructure of resulting artificial soil samples. Most homogeneous microstructures can be achieved by applying the mixing order “sand-water-clay.” The following methods were used to validate this statement: (1) optical observation, (2) X-ray tomography, (3) scanning electron microscopy, and (4) Mercury intrusion porosimetry. For all samples, clays are mainly organized in a homogeneous matrix but are also dispersed heterogeneously in micrometer-sized layers surrounding sand particles, particularly where sand grains show a greater roughness. At water contents ≥1.5 wL, the microstructures are visually similar from the mm to μm scale whatever mixing order is used. However, for water contents lower than 1.5 wL, the mixing order controls the distribution of the clay particles. This paper proposes a motivated choice of a preparation protocol of artificial clayey materials to be used in laboratory experiments. It might contribute to better understanding and modeling grain movements and arrangements in artificial muds, used for instance in underground mining, foundation settlement, hydraulic containment, road construction, soil stabilization, and in natural soils in the occurrence of soil liquefaction, industrial brick manufacturing, and in studying shear processes in tectonic fault zones.

This paper focuses on a numerical approach to finding the p–y curves for laterally loaded piles. The Drucker–Prager plastic model is employed and implemented within a finite element MATLAB code. The pre- and post-processing code for Gmsh and related numerical tools are established as well. The p–y curve results from this new approach have been validated and compared to the typical design equations of API (American Petroleum Institute) and Matlock. The validation reveals that the code leads to lower p–y curves than the API and Matlock equations when the horizontal displacement is less than 0.35 times the diameter of the pile (B). A sensitivity analysis of the number of elements and the interface thickness is presented. The results indicate that the obtained p–y curves are independent of the two factors. Finally, the influence of clay content on the p–y behavior is investigated by the implemented MATLAB code. When y < 0.15B, the same lateral capacity values are resulted at clay contents of 27.5% and 55%, and they are higher than the ones for 0% clay content. The p–y curves show a decreasing trend with increasing clay content after y > 0.15B.

The mechanical behaviors of Martian regolith-structure interfaces are of great significance for the design of rover, development of excavation tools, and construction of infrastructure in Mars exploration. This paper presents an experimental investigation on the properties of a Martian regolith simulant (JEZ-1) through one-dimensional oedometer test, direct shear test, and interface direct shear tests between JEZ-1 and steel plates with different roughness. Oedometer result reveals that the compression and swelling indexes of the JEZ-1 are quite low, thus it is a less compressible and lower swelling soil. The cohesion and adhesion of JEZ-1 are lower than 5 kPa. The values of the internal friction angle range from 39.7° to 40.6°, and the interface friction angles are 16.7° to 36.2° for the smooth and rough interface. Furthermore, the direct shear and interface direct shear results indicate that the interface friction angles are lower than the internal friction angles of JEZ-1 and increase close to the internal friction angles with increasing interface roughness.

A large number of experimental studies on sand–clay mixtures are well documented in the literature; however, the preparation protocol is rarely clearly detailed or varies a lot according to the authors. Variations in the preparation technique obviously increases the challenge of comparing different test results. As a consequence, sample preparation for sand–clay mixtures should be kept as constant as possible to ensure homogeneity and uniformity of samples and limit result variability. This paper develops a detailed procedure on how to prepare sand–clay samples for interface direct shear tests. Sand–clay mixtures are prepared with Fontainebleau sand, kaolinite clay and distilled water by the S1 (sand–water–clay) protocol. The uniformity of the reconstituted specimens is assessed by measuring the water content and density on three slices from the top to the bottom across the specimens. The repeatability of the samples is checked with oedometer and interface direct shear tests. This sample preparation procedure can be used for preparing sand–clay mixture for interface direct shear tests to investigate the influence of clay content or other effects (e.g., temperature) on the mechanical behavior of soil–structure interface. It has demonstrated great performance in preparing samples with good homogeneity and shape, compared to other traditional reconstitution techniques. With the sample preparation procedure, we can obtain repeatable test results as well.

The shaft lining failure which occurs in deep alluvium is a sudden coal mine hazard. The strata grouting is one of the treatment methods for the shaft lining failure. To investigate the impact of grout injection pressure on the shaft lining, the field measurement and the numerical analysis of the shaft lining stress variation during the grouting were conducted. To improve the strata grouting, the underground continuous impervious curtain (UCIC) is proposed as a new method by using the chain conveyor cutter technique without the impact on the shaft lining. The effects of the new method were also analyzed by means of the numerical methods. The results show that the strata grouting in the deep alluvium needs a high injection pressure, and in the horizontal direction, the shaft lining sustains the repeat tensile and compressive force during the grouting process. The negative influence of strata grouting on the stability of the shaft lining is obvious and serious. The UCIC built around the shaft lining can restrain the stress concentration induced by the aquifer drawdown. The triangular UCIC has a similar effect as that of the vertical one, and the small angle is better for preventing the shaft lining failure.

Introduction: While most thyroid cancer patients have a favorable prognosis, anaplastic thyroid carcinoma (ATC) remains a particularly aggressive form with a median survival time of just five months. Conventional therapies offer limited benefits for this type of thyroid cancer. Our study aims to identify ATC patients who might bene t from immunotherapy. Methods: Our study uses multiple algorithms by R4.2.0, and gene expression and clinical data are collected from TCGA, GEO and local cohort. In vitro experiments, such as western blot and immunofluorescence staining, are performed. Results: Using a set of five genes uniquely expressed across various types of thyroid cancer, we developed a machine-learning model to distinguish each type within the GEO dataset of thyroid cancer patients (GSE60542, GSE76039, GSE33630, GSE53157, GSE65144, GSE29265, GSE82208, GSE27155, GSE58545, GSE54958, and GSE32662). These genes allowed us to stratify ATC into three distinct groups, each exhibiting significantly different responses to anti-PD1 therapy as determined by consensus clustering. Through weighted gene co-expression network analysis (WGCNA), we identified 12 differentially expressed genes closely associated with immunotherapy outcomes. This led to the creation of a refined signature for predicting ATC's immune responsiveness to anti-PD1 therapy, which was further validated using thyroid cancer cohorts from TCGA and nine melanoma cohorts from clinical trials. Among the 12 genes, HLF stood out due to its strong association with various cancer hallmarks. Discussion: Our study revealed that HLF impedes ATC progression by down-regulating the epithelial-to-mesenchymal transition (EMT) pathway, reducing T cell exhaustion, and increasing sensitivity to sorafenib, as demonstrated through our in-vitro experiments. The key to extractive distillation separation lies in screening a suitable entrainer. Based on chemical process system engineering (PSE) concepts, a novel heuristic process prediction model is proposed with a minimized process economic cost index (PECI) for the efficient screening of an optimal green entrainer with the advantages of cost-effectiveness, environmental friendliness, and sustainability, followed by outlining the principles of characterizations and mathematical modeling in chemical ED processes. The relationships of αHC/SOL and NTR with QR, TAC, PECI, and LTEDI indicated that the heuristic process prediction model was scientifically valid as well as efficient. Moreover, the proposed model progresses from binomial intersecting influencing factors to trinomial juxtaposing influencing factors; summarizes and discloses the system’s theoretical rules and influencing elements of the changes in parameters and their relative volatility, avoiding the new azeotropes in the solvent recovery column; reduces the workloads of process simulation and optimization of distillation processes from (M × N) times to (1 + N) times; can be extended efficiently and accurately to provide the targeted prediction and systematic theoretical principles for separating different feed azeotropes, predicting and screening organic solvents, ionic liquids, deep eutectic solvents, and mixed solvents with the optimal TAC and environmental friendliness (AP and GWP) and sustainability (FETP, HTP, and TETP) as LCA impacts; has broad predictability and applicability combined with different process optimization algorithms; and provides infinite composite patterns and opportunities for integrating and developing innovative optimization module algorithms for future green chemical separation processes. The studies on the origin of versatile oxidation pathways toward targeted pollutants in the single-atom catalysts (SACs)/peroxymonosulfate (PMS) systems were always associated with the coordination structures rather than the perspective of pollutant characteristics, and the analysis of mechanism commonality is lacking. In this work, a variety of single-atom catalysts (M-SACs, M: Fe, Co, and Cu) were fabricated via a pyrolysis process using lignin as the complexation agent and substrate precursor. Sixteen kinds of commonly detected pollutants in various references were selected, and their lnkobs values in M-SACs/PMS systems correlated well (R² = 0.832 to 0.883) with their electrophilic indexes (reflecting the electron accepting/donating ability of the pollutants) as well as the energy gap (R² = 0.801 to 0.840) between the pollutants and M-SACs/PMS complexes. Both the electron transfer process (ETP) and radical pathways can be significantly enhanced in the M-SACs/PMS systems, while radical oxidation was overwhelmed by the ETP oxidation toward the pollutants with lower electrophilic indexes. In contrast, pollutants with higher electrophilic indexes represented the weaker electron-donatingcapacity to the M-SACs/PMS complexes, which resulted in the weaker ETP oxidation accompanied with noticeable radical oxidation. In addition, the ETP oxidation in different M-SACs/PMS systems can be regulated via the energy gaps between the M-SACs/PMS complexes and pollutants. As a result, the Fenton-like activities in the M-SACs/PMS systems could be well modulated by the reaction pathways, which were determined by both electrophilic indexes of pollutants and single-atom sites. This work provided a strategy to establish PMS-based AOP systems with tunable oxidation capacities and pathways for high-efficiency organic decontamination. [ABSTRACT FROM AUTHOR]
