Microfluidic reagent-based nutrient sensors offer a promising technology to address the global undersampling of ocean chemistry but have so far not been shown to operate in the deep sea (>200 m). We report a new family of miniaturized lab-on-chip (LOC) colorimetric analyzers making in situ nitrate and phosphate measurements from the surface ocean to the deep sea (>4800 m). This new technology gives users a new low-cost, high-performance tool for measuring chemistry in hyperbaric environments. Using a combination of laboratory verification and field-based tests, we demonstrate that the analyzers are capable of in situ measurements during profiling that are comparable to laboratory-based analyses. The sensors feature a novel and efficient inertial-flow mixer that increases the mixing efficiency and reduces the back pressure and flushing time compared to a previously used serpentine mixing channel. Four separate replicate units of the nitrate and phosphate sensor were calibrated in the laboratory and showed an average limit of detection of 0.03 μM for nitrate and 0.016 μM for phosphate. Three on-chip optical absorption cell lengths provide a large linear range (to >750 μM (10.5 mg/L-N) for nitrate and >15 μM (0.47 mg/L-P) for phosphate), making the instruments suitable for typical concentrations in both ocean and freshwater aquatic environments. The LOC systems automatically collected a series of deep-sea nitrate and phosphate profiles in the northeast Atlantic while attached to a conductivity temperature depth (CTD) rosette, and the LOC nitrate sensor was attached to a PROVOR profiling float to conduct automated nitrate profiles in the Mediterranean Sea.

To inform cost-effective monitoring of offshore geological storage of carbon dioxide (CO 2), a unique field experiment, designed to simulate leakage of CO 2 from a sub-seafloor storage reservoir, was carried out in the central North Sea. A total of 675 kg of CO 2 were released into the shallow sediments (∼3 m below seafloor) for 11 days at flow rates between 6 and 143 kg d -1. A set of natural, inherent tracers ( 13C, 18O) of injected CO 2 and added, non-toxic tracer gases (octafluoropropane, sulfur hexafluoride, krypton, methane) were used to test their applicability for CO 2 leakage attribution and quantification in the marine environment. All tracers except 18O were capable of attributing the CO 2 source. Tracer analyses indicate that CO 2 dissolution in sediment pore waters ranged from 35 % at the lowest injection rate to 41% at the highest injection rate. Direct measurements of gas released from the sediment into the water column suggest that 22 % to 48 % of the injected CO 2 exited the seafloor at, respectively, the lowest and the highest injection rate. The remainder of injected CO 2 accumulated in gas pockets in the sediment. The methodologies can be used to rapidly confirm the source of leaking CO 2 once seabed samples are retrieved.

Carbon capture and storage (CCS) is a key technology to reduce carbon dioxide (CO2) emissions from industrial processes in a feasible, substantial, and timely manner. For geological CO2 storage to be safe, reliable, and accepted by society, robust strategies for CO2 leakage detection, quantification and management are crucial. The STEMM-CCS (Strategies for Environmental Monitoring of Marine Carbon Capture and Storage) project aimed to provide techniques and understanding to enable and inform cost-effective monitoring of CCS sites in the marine environment. A controlled CO2 release experiment was carried out in the central North Sea, designed to mimic an unintended emission of CO2 from a subsurface CO2 storage site to the seafloor. A total of 675 kg of CO2 were released into the shallow sediments (∼3 m below seafloor), at flow rates between 6 and 143 kg/d. A combination of novel techniques, adapted versions of existing techniques, and well-proven standard techniques were used to detect, characterise and quantify gaseous and dissolved CO2 in the sediments and the overlying seawater. This paper provides an overview of this ambitious field experiment. We describe the preparatory work prior to the release experiment, the experimental layout and procedures, the methods tested, and summarise the main results and the lessons learnt. publishedVersion

Recent interest in Antarctic subglacial lakes has seen the development of bespoke systems for sampling them. These systems are considered pristine environments potentially harboring undisturbed sedimentary sequences and ecosystems adapted to these cold oligotrophic environments in the absence of sunlight. The water/sediment interface is considered a prime location for the detection of microbial life and so is of particular interest. This article describes the development of a small corer to capture and retain a short core that includes the water/sediment interface specifically to address the question of whether life exists in these lakes. This apparatus was developed as part of the UK led project to access, measure, and sample subglacial Lake Ellsworth. In addition to addressing the constraints of coring in this difficult environment, the results of subsequent testing suggest that this corer can be applied to sampling sediments in other environments and would be particularly useful in low energy environments when the water‐sediment interface is indistinct or unconsolidated.

The ocean’s biological carbon pump plays a central role in regulating atmospheric CO2 levels. In particular, the depth at which sinking organic carbon is broken down and respired in the mesopelagic zone is critical, with deeper remineralisation resulting in greater carbon storage. Until recently, however, a balanced budget of the supply and consumption of organic carbon in the mesopelagic had not been constructed in any region of the ocean, and the processes controlling organic carbon turnover are still poorly understood. Large-scale data syntheses suggest that a wide range of factors can influence remineralisation depth including upper-ocean ecological interactions, and interior dissolved oxygen concentration and temperature. However these analyses do not provide a mechanistic understanding of remineralisation, which increases the challenge of appropriately modelling the mesopelagic carbon dynamics. In light of this, the UK Natural Environment Research Council has funded a programme with this mechanistic understanding as its aim, drawing targeted fieldwork right through to implementation of a new parameterisation for mesopelagic remineralisation within an IPCC class global biogeochemical model. The Controls over Ocean Mesopelagic Interior Carbon Storage (COMICS) programme will deliver new insights into the processes of carbon cycling in the mesopelagic zone and how these influence ocean carbon storage. Here we outline the programme’s rationale, its goals, planned fieldwork and modelling activities, with the aim of stimulating international collaboration.

It is 4 years since the subglacial lake community published its plans for accessing, sampling, measuring and studying the pristine, and hitherto enigmatic and very different, Antarctic subglacial lakes, Vostok, Whillans and Ellsworth. This paper summarizes the contrasting probe technologies designed for each of these subglacial environments and briefly updates how these designs changed or were used differently when compared to previously published plans. A detailed update on the final engineering design and technical aspects of the probe for Subglacial Lake Ellsworth is presented. This probe is designed for clean access, is negatively buoyant (350 kg), 5.2 m long, 200 mm in diameter, approximately cylindrical and consists of five major units: (i) an upper power and communications unit attached to an optical and electrical conducting tether, (ii)–(iv) three water and particle samplers, and (v) a sensors, imaging and instrumentation pack tipped with a miniature sediment corer. To date, only in Subglacial Lake Whillans have instruments been successfully deployed. Probe technologies for Subglacial Lake Vostok (2014/15) and Lake Ellsworth (2012/13) were not deployed for technical reasons, in the case of Lake Ellsworth because hot-water drilling was unable to access the lake during the field season window. Lessons learned and opportunities for probe technologies in future subglacial access missions are discussed.

We describe the first results obtained using a novel free-drifting neutrally buoyant sediment trap called PELAGRA (Particle Export measurement using a LAGRAngian trap). The trap uses an APEX float (Autonomous Profiling EXplorer) to maintain its location at a predetermined depth or density horizon and is designed to be deployed in the depth range 100–600 m for periods of up to a week. PELAGRA was deployed into a well characterised region of the Northeast (NE) Atlantic over the Porcupine Abyssal Plain (PAP) during the summers of 2003, 2004, 2005 and 2006 in post-bloom conditions. In parallel, measurements were made of total and new primary production and estimates of particle export based on 234Th deficits in the upper water column. Samples of sinking material from PELAGRA were recovered almost uncontaminated by “swimmers”. The material collected differed from that obtained using water bottles and an in situ filtration system and this indicated selective export of centric diatoms. Fluxes of 234Th into the traps were less than that calculated from the deficit of this isotope in the upper water column. It is concluded that this reflects export events that had occurred prior to these deployments. Fluxes of organic carbon into the PELAGRA traps were similar to those expected from measurements of new production in the upper mixed layer. During 2006, simultaneous PELAGRA deployments at depths of 150 and 250 m provided a direct measurement of the decrease in flux with depth. This decrease was substantially more than that predicted by the often-used “Martin equation” and yielded a “b value” of 1.7. Using this value in the NE Atlantic about 14% of the total production reaches a depth of 150 m during the post-bloom period under nutrient limitation.

During the austral summer of 2004–2005, a large multi-disciplinary research cruise investigated the development and fate of a naturally iron-fertilised phytoplankton bloom in the Southern Ocean (Crozet Plateau). As part of this extensive process study, a neutrally buoyant sediment trap (PELAGRA) was deployed to constrain the magnitude, composition, and variability of upper-ocean particle export. In the productive regime north of the plateau we observed depth-normalised (100-m) organic carbon fluxes between 11 and 440 mg C m −2 d −1 , and in the HNLC control region to the south similarly normalised fluxes between 28 and 46 mg C m −2 d −1 . Mass balance calculations indicate that the high levels of carbon export north of the plateau would need to be maintained for at least 30 days in order to account for estimated seasonal depletion of dissolved silicic acid in surface waters. This would imply that the flux of organic carbon is ≈15 g C m −2 for the period of the bloom north of the plateau. A range of export ratios (proportion of surface production lost to downward flux) was calculated using both contemporaneous and retrospective estimates of integrated production, and these highlight the temporal decoupling between production and export. Calculated export ratios were at their highest north of the plateau and correlate strongly with the selective export of large, heavily silicified diatoms, particularly Eucampia antarctica , relative to the surface community structure. By normalising the molar elemental ratios measured in the exported particles to the molar elemental ratios of the upper-ocean particle field we also observed a strong decoupling of Si:C and Si:N. This suggests that the decoupling of the global silica and carbon cycles, which is well known as a defining feature of the Southern Ocean, has its origins in the upper ocean.

Accumulations of sediment beneath the Antarctic Ice Sheet contain a range of physical and chemical proxies with the potential to document changes in ice sheet history and to identify and characterize life in subglacial settings. Retrieving subglacial sediments and sediment cores presents several unique challenges to existing technologies. This paper briefly reviews the history of sediment sampling in subglacial environments. It then outlines some of the technological challenges and constraints in developing the corers being used in sub-ice shelf settings (e.g. George VI Ice Shelf and Larsen Ice Shelf), under ice streams (e.g. Rutford Ice Stream), at or close to the grounding line (e.g. Whillans Ice Stream) and in subglacial lakes deep under the ice sheet (e.g. Lake Ellsworth). The key features of the corers designed to operate in each of these subglacial settings are described and illustrated together with comments on their deployment procedures.

Photosynthesis in the surface ocean produces approximately 100 gigatonnes of organic carbon per year, of which 5 to 15 per cent is exported to the deep ocean1, 2. The rate at which the sinking carbon is converted into carbon dioxide by heterotrophic organisms at depth is important in controlling oceanic carbon storage3. It remains uncertain, however, to what extent surface ocean carbon supply meets the demand of water-column biota; the discrepancy between known carbon sources and sinks is as much as two orders of magnitude4, 5, 6, 7, 8. Here we present field measurements, respiration rate estimates and a steady-state model that allow us to balance carbon sources and sinks to within observational uncertainties at the Porcupine Abyssal Plain site in the eastern North Atlantic Ocean. We find that prokaryotes are responsible for 70 to 92 per cent of the estimated remineralization in the twilight zone (depths of 50 to 1,000 metres) despite the fact that much of the organic carbon is exported in the form of large, fast-sinking particles accessible to larger zooplankton. We suggest that this occurs because zooplankton fragment and ingest half of the fast-sinking particles, of which more than 30 per cent may be released as suspended and slowly sinking matter, stimulating the deep-ocean microbial loop. The synergy between microbes and zooplankton in the twilight zone is important to our understanding of the processes controlling the oceanic carbon sink.

Optical holography provides non-intrusive, non-destructive recording of living, motile organisms and particles in their natural environment. High-resolution, three-dimensionality and `optical sectioning' of the image allow identification of species and enumeration of size, orientation and spatial distribution. We have developed a submersible holographic camera to simultaneously expose in-line and, uniquely, off-axis holograms to record organisms from a few micrometres upwards (in-line), in concentrations down to a few tens of particles per cubic metre (off-axis). It can record up to 50 000 cm3 of the water column in a single off-axis hologram and 3500 cm3 in an in-line hologram. We describe the first sea deployment of the camera and the successful recording of holograms of plankton and marine snow to a depth of 100 m water.

The cavities beneath Antarctic ice shelves are among the least studied regions of the World Ocean, yet they are sites of globally important water mass transformations. Here we report results from a mission beneath Fimbul Ice Shelf of an autonomous underwater vehicle. The data reveal a spatially complex oceanographic environment, an ice base with widely varying roughness, and a cavity periodically exposed to water with a temperature significantly above the surface freezing point. The results of this, the briefest of glimpses of conditions in this extraordinary environment, are already reforming our view of the topographic and oceanographic conditions beneath ice shelves, holding out great promises for future missions from similar platforms.

We describe the development, construction and sea testing of An underwater holographic camera (HoloCam) for in situ recording of marine organisms and particles in large volumes of sea water. HoloCam comprises a laser, power supply, holographic recording optics, and plate holders, a water- tight housing and a support frame. Added to this are control electronics such that the entire camera is remotely operable and controllable from ship or dock-side. Uniquely the camera can simultaneously record both in-line and off-axis holograms using a pulsed frequency double Nd:YAG laser. In- line holography is capable of producing images of organisms with a resolution of better than 10 micrometers . Off-axis holograms of aquatic systems of up to 50,000 cm3 volume, have been recorded. Following initial laboratory testing, the holo-camera was evaluated in an observation tank and ultimately was tested in Loch Etive, Scotland. In-line and off-axis holograms were recorded to a depth of 100 m. This illumination system is one component of a holographic camera, which is part of a multi-disciplinary, multi-national project (funded by the European Commission MAST III initiative) to use holography for the in situ recording and subsequent analysis of holograms of marine organisms and particles. Laboratory experiments have shown that the best results for off-axis recording are obtained using side illumination. Our design goal has been to provide even side illumination throughout the recording volume. This illumination system, the lightrod, is based on a cylinder made from a transparent material containing a series of partial reflectors inclined and spaced along its length. An unexpanded laser beam is fed into the cylinder along its axis. Each reflector diverts a fraction of the beam into the recording volume. Two versions of the lightrod have been designed and built. In one version, the cylinder uses solid Perspex spacers with a small air gap in between; in the other, the spacers are hollow with thin glass windows in between. Both lightrod designs are discussed together with an analysis of the illumination profile, and their practical implementation is outlined. Both lightrod prototypes have been tested in an observation tank. Results from these tests are presented.

The HoloCam system is a major component of a multi-national multi- discipline project known as HoloMar (funded by the European Commission under the MAST III initiative). The project is concerned with the development of pulsed laser holography to analyse and monitor the populations of living organisms and inanimate particles within the world's oceans. We describe here the development, construction and evaluation of a prototype underwater camera, the purpose of which is to record marine organisms and particles, in-situ. Recording using holography provides several advantages over conventional sampling methods in that it allows non-intrusive, non-destructive, high- resolution imaging of large volumes (up to 105 cm3) in three dimensions. The camera incorporates both in-line and off-axis holographic techniques, which allows particles from a few micrometres to tens of centimetres to be captured. In tandem with development of the HoloCam, a dedicated holographic replay system and an automated data extraction and image processing facility are being developed. We report here details of the design, development, initial testing and field-deployment of the HOLOMAR system for in-situ subsea holography and analysis of marine plankton and nonliving particles. HOLOMAR comprises a submersible holographic camera ("HoloCam") able to record in-line and off-axis holograms at depths down to 100 m, together with specialised reconstruction hardware ("HoloScan") linked to custom image processing and classification software. The HoloCam consists of a laser and power supply, holographic recording optics and holographic plate holders, a water-tight housing and a support frame. It utilises two basic holographic geometries, in-line and off-axis such that a wide range of species, sizes and concentrations can be recorded. After holograms have been recorded and processed they are reconstructed in full three-dimensional detail in air in a dedicated replay facility. A computer-controlled microscope, using video cameras to record the image at a given depth, is used to digitise the scene. Specially written software extracts a binarised image of an object in its true focal plane and is classified using a neural network. The HoloCam was deployed on two separate cruises in a Scottish sea loch (Loch Etive) to a depth of 100 m and over 300 holograms were recorded.

According to many prognostic scenarios by the Intergovernmental Panel on Climate Change (IPCC), a scaling-up of carbon dioxide (CO 2) capture and storage (CCS) by several orders-of-magnitude is necessary to meet the target of ≤2 °C global warming by 2100 relative to preindustrial levels. Since a large fraction of the predicted CO 2 storage capacity lies offshore, there is a pressing need to develop field-tested methods to detect and quantify potential leaks in the marine environment. Here, we combine field measurements with numerical models to determine the flow rate of a controlled release of CO 2 in a shallow marine setting at about 119 m water depth in the North Sea. In this experiment, CO 2 was injected into the sediment at 3 m depth at 143 kg d -1. The new leakage monitoring tool predicts that 91 kg d -1 of CO 2 escaped across the seafloor, and that 51 kg d -1 of CO 2 were retained in the sediment, in agreement with independent field estimates. The new approach relies mostly on field data collected from ship-deployed technology (towed sensors, Acoustic Doppler current profiler—ADCP), which makes it a promising tool to monitor existing and upcoming offshore CO 2 storage sites and to detect and quantify potential CO 2 leakage.
