Abstract

This article investigates the effects of licorice root (Glycyrrhiza L.) syrup on laboratory rats exposed to ionizing gamma radiation, focusing on its potential radioprotective properties. Ionizing gamma radiation induces oxidative stress, characterized by an increase in free radicals and subsequent tissue damage, which can contribute to the development of various diseases such as neurodegenerative, cardiovascular, diabetes and kidney diseases. The body’s antioxidant defense mechanisms, including superoxide dismutase (SOD), catalase (CAT) and malondialdehyde (MDA), play crucial roles in combating oxidative stress. Historically recognized for its medicinal properties, licorice root may mitigate the harmful effects of radiation. The research employed Electron Paramagnetic Resonance (EPR) spectroscopy to analyze structural and functional changes in various biological systems under the influence of radiation and other stressors. This investigation aims to provide insights into the protective effects of licorice syrup, contributing to the understanding of potential therapeutic strategies against radiation-induced oxidative damage. As a continuation of the experiments, the levels of antioxidant enzymes in the rat’s erythrocytes were measured using various methods. Abstract

The emergence of antibiotic resistance, caused by the improper use of antibiotics, is a significant challenge in combating infectious diseases, leading to millions of annual fatalities. The occurrence of antimicrobial side effects catalyzes the investigation of novel antimicrobial compounds and sources of drugs. Consequently, the research on biological activity that is conducted on plants, plant extracts, and compounds that are produced from plant components is of utmost significance. In this study, CtAC/MNPs were obtained by the reaction of activated carbon (AC) obtained from the fruits of the Celtis tournefortii (Ct) plant and magnetic nanoparticles (MNPs), and a CtAC/MNPs-Ag nanocomposite was synthesized by the reduction in silver ions added to the reaction. The synthesized CtAC/MNPs and CtAC/MNPs-Ag nanocomposites were analyzed spectroscopically (FTIR, XRD), microscopically (SEM, EDX), optically (DLS), electrochemically (zeta potential) and magnetically (VSM). The antibacterial activities of CtAC/MNPs and CtAC/MNPs-Ag nanocomposites against S. aureus and E. coli were investigated by microdilution method using minimal inhibitory concentration (MIC) and disk diffusion methods. Antioxidant activity study, including total phenolic content and DPPH and cuprac assays, revealed the remarkable effect of the CtAC/MNPs-Ag nanocomposite. This study has the advantages of obtaining CtAC/MNPs and CtAC/MNPs-Ag nanocomposites in a short time without requiring energy, and most importantly, the reaction takes place without using any toxic substances. Abstract

Purpose: The phenolic composition, antioxidant, antimicrobial activity, enzyme inhibition activity, and cytotoxic activity potentials of the plant Rumex acetosellaL. (R. acetosella) were examined in this study. Materials and Methods: The chemical composition of R. acetosellamethanol extract was identified by the LC–MS/MS method. The antioxidant activity was tested using β-carotene/linoleic acid, DPPH free radical scavenging, ABTS cation radical scavenging, CUPRAC reducing power, and metal chelating activity methods. The cytotoxic activity was determined by the MTT assay using human ovarian adenocarcinoma (Skov-3), glioblastoma (U87), human dermal fibroblasts (HDF), and human colorectal adenocarcinoma (CaCo-2) cell lines. The antimicrobial activity of methanolic extracts was tested on gram-negative (Escherichia coliand Pseudomonas aeuriginosa) and gram-positive bacteria (Staphylococcus aureusand Bacillus subtilis) using the in vitro minimum inhibition concentration method (MIC). Enzyme inhibition activity of R. acetosellamethanol extract was measured spectrophotometrically against acetylcholinesterase (AChE) and glutathione S-transferase (GST) enzymes. Results: The findings showed that the major components of the methanol extract content were luteolin-7-O-glucoside (1.599 m/L), polydatin (91,024 m/L), and shikimic acid (0.773 m/L). It was determined that the extract and standard antioxidant (a-tocopherol) results in DPPH•, and ABTS• + tests performed to determine the antioxidant activity were close to each other, and this value was more effective than the standard antioxidant (α-tocopherol) in the CUPRAC test. These results suggested that the plant’s antioxidant potential was higher when compared with reference antioxidant compounds. It was determined that the methanol extract of R. acetosellahad a weaker effect on the growth of the tested microorganisms than the antibiotics used as standard. The activity of the GST and AChE enzymes was found to be severely inhibited by the methanol extract of R. acetosella. Conclusion: Based on these findings, R. acetosellaL. is a medicinal and commercially beneficial plant that warrants further investigation.

Objectives: Hypericum triquetrifolium Turra (HTT) has been traditionally used in medical treatments due to its sedative, antiseptic, antiinflammatory, and anthelmintic properties. The present study aims to investigate the lung-protective and antiapoptotic effects of HTT against cyclophosphamide (CP)-induced lung injury in rats. Methods: Thirty-five Sprague Dawley rats were categorized into 5 groups, each consisting of seven members. Phenolic acid and flavonoid contents of this plant were determined. The lung tissue samples cultivated from the rats were examined in histopathological and immunohistochemically for the apoptosis markers of Caspase-3, Bax, and Bcl-2. Results: Histopathological results indicated that structural defects, bleeding areas, and edema had occurred in the lungs of the CP-Alone Group. Besides, Caspase-3 and Bax positivity of the lung cells had also increased while Bcl-2 positivity had decreased. On the other hand, in the HTT+CP Group, HTT was shown to have reversed the aforementioned changes positively. Abstract

The current work's main objective was to determine the chemical composition of Amygdalus communis (AC) leaf extract and examine the antibacterial and cytotoxic properties of biosynthesized gold nanoparticles (AuNPs). The chemical composition of AC leaf extract was determined using LC-ESI/MS/MS to detect compounds that may be responsible for the reducing, stabilizing, and capping steps in the synthesis of nanoparticles and their biological activities. The AC-AuNPs were spherical, with a particle size lower than 100 nm and a face-centered cubic structure. The EDX spectrum confirmed the formation of AuNPs and a negative zeta potential value (-27.7 mV) suggested their physicochemical stability. The in vitro cytotoxic efficacy of the AC-AuNPs against colorectal adenocarcinoma (Caco-2), glioma (U118), and ovarian (Skov-3) cancer cell lines and human dermal fibroblasts (HDFs) was evaluated by MTT assay. CaCo-2 cell proliferation was effectively inhibited by the AC-AuNPs at concentrations between 25 and 100 g mL-1. The AC-AuNPs exerted preeminent antimicrobial activity against Bacillus subtilis with an MIC of 0.02 μg/mL, whilst good activity was shown against Staphylococcus aureus bacteria and Candida albicans yeast with an MIC of 0.12 μg/mL. Ultimately, the results support the high antibacterial and anticancer potential of biosynthesized AuNPs from AC leaf extract.

The aim of this study is to develop an easy and eco-friendly method for the synthesis of Au NPs using extracts from the Allium cepa(AC) red peel (skin) extract and investigate the antibacterial and antioxidant activity and also inhibitory effects on acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzymes. UV–Vis peak at ~ 564 nm confirmed the Au NPs absorbance. TEM images revealed the formation of Au NPs with mostly spherical shapes and sizes between 6.08 and 54.20 nm. FTIR analysis confirmed the important biological compounds responsible for the reduction of gold. The strong absorption property of Au NPs was studied by EDX. The produced Au NPs demonstrated significant antibacterial and antifungal activity against the bacterial and fungal strains tested, as well as efficient inhibitory activity against both AChE and BChE enzymes. The highest antimicrobial activities were found against Staphylococcus aureus(0.06° mg/ml) and Candida albicans(0.06° mg/ml). The antioxidant test findings revealed that AC-Au NPs had lesser activity when compared to normal antioxidants. The Au NPs showed excellent inhibitory efficacy against AChE and BChE. The proposed green technique could encourage the innocuous generation of Au NPs, implying therapeutic possibilities.

Abstract

Water is an important part of the ecosystem for life. With industrialization, pollution in water resources has reached a worrying level. Water pollution due to heavy metals and their increasing concentrations have caused researchers to increase their interest in the subject due to the damage they cause to water ecosystems. It requires serious cost and time to eliminate the pollution caused by heavy metals in water. In recent, years, the use of biosorption method using bacteria to remove heavy metals in water has become widespread. The main reason why this method is preferred is that gram-positive bacteria have a thick peptidoglycan layer on the cell wall and increases the adsorption capacity. In this study. in drinking, waste, river water and artificially prepared samples, batch method of heavy metal biosorption and biosorption competition in multiple prepared heavy metal solutions were investigated. For these processes. Bacillus licheniformth sp. Bacillus subtilis sp. and Bacillus subtilis (ATCC 6051) strains were used as a biosorbent. Biosorbtion of Cd (II), Cu (H), Pb (II), Fe(II), Ni (II) and Zn (II) metals from waters with these biosorbents at different pHs at. 25 degrees C with 0.25 mg LI It was investigated. Surface morphological structures of biosorbents were evaluated using SEM images and element compositions were evaluated using EDAX profile. Element content was determined using ICP-OES. It was found that heavy metal ions were removed up to 98% with maximum biosorption at pH 6.0.

Abstract

Cyclotrichium origanifolium is a medicinal plant belonging to the Lamiaceae family. In this study, phenolic content analysis, antimicrobial effects, and cytotoxic effects of extracts of C. origanifolium were investigated. In the extracts, phenolic compound analysis by the liquid chromatography–electrospray ionization–tandem mass spectrometry method, antimicrobial effect by the minimum inhibition concentration method, and cytotoxic effect on human dermal fibroblasts (HDF), glioblastoma cell (U87), ovarian adenocarcinoma cell (Skov‐3), and human colorectal adenocarcinoma cell (CaCo‐2) cancer cell lines were investigated. Cytotoxicity analyses were performed by the MTT method. In addition, the GST and AChE enzyme activities of the extracts were also measured. Around 18 compounds were detected in both the methanol and ethanol extract. It was found that the best antimicrobial effect on Gram‐negative Pseudomonas aeruginosa was on methanol extract, while the ethanol extract was on Candida albicans fungus (respectively, 2.50 mg/ml, 5.0 μg/ml). A 500 μg/ml of methanol extract has been shown to have cytotoxic activity high effect on HDF cells. GST and AChE activity were found to decrease in a concentration‐dependent manner. Abstract

Kurşun endüstrinin birçok alanınında kullanılmaktadır. Kullanım yaygınlığı düşünülürse buna bağlı olarak bu uygulamaların sonucunda ortaya çıkan atıklarda yoğun kurşun ağır metal içeriği bulunmakta ve büyük oranda en yakın su kaynağını doğrudan ya da dolaylı olarak etkilemektedir. Suda yüksek konsantrasyonlarda bulunması ile tüm yaşam formları için ciddi tehdit oluşturmaktadır. Bunların çevre dostu yöntemlerle iyileştirilmesi hem daha kolay hem de ekonomiktir. Çalışmamızda Dicle Nehri topraklarından izole edilen Bacillus licheniformis sp. kullanılarak sulardaki kurşun miktarı kesikli biyosorbsiyon yöntemi kullanılarak 25 oC de pH 5.5 da 120 dakikada 42.92 biyosorbsiyon kapasitesiyle % 98.4 oranında önemli ölçüde iyileştirildi. Biyosorbentin taramalı alan mikroskopisi (SEM) kullanılarak morfolojik görünümleri, fourier İnfrared spektroskopisi (FT-IR) dataları ile yüzey yapısında biyosorbsiyona etki eden fonksiyonel gruplar ve ısıl işlem karşısında gösterdiği direnç de termal gravimetrik analiz (TGA-DTA) verileri ile karakterize edildi. Sulu çözeltideki kurşun element içeriği İndüktif olarak eşleşmiş plazma optik emisyon spektroskopisi (ICP-OES) cihazı ile tespit edildi., Lead is used in many areas of industry. Considering the prevalence of use, there is an intense lead and heavy metal content in the wastes resulting from these applications, and the toxic pollution caused by these metals affects the nearest water source directly or indirectly. Lead poses a serious threat to all life forms in the ecosystem, even at low concentrations in water. Lead recovery with environmentally friendly methods is both easier and more economical. Bacillus licheniformis sp. type was used as a biosorbent in the study. Nearly 98.4% of lead was removed by using the batch biosorption method, at 25 oC, pH 5.5, with an adsorption capacity of 42.92 in 120 minutes. from the water. The properties of the biosorbent, such as its morphological appearance, were characterized by scanning electron microscopy (SEM). Besides, the functional groups affecting biosorption in the surface structure were investigated by fourier transform Infrared spectroscopy (FT-IR)while its resistance to heat treatment was measured by thermal gravimetric analysis (TGA-DTA). The lead(II) element content in the aqueous solution was also determined by inductively coupled plasma optical emission spectroscopy (ICP-OES).

Abstract

In this study, silver nanoparticles (AgNPs) were synthesized economically and simply using an environmentally friendly method with the extract obtained from agricultural waste olive leaves. AgNPs synthesized according to the analysis data were determined to have maximum absorbance at 433.5 nm wavelength, spherical appearance, 7.2 nm crystal nano size and −19.9 mV zeta potential. It was determined by the microdilution method with Minimum Inhibition Concentration (MIC) that AgNPs exert a suppressive effect on the growth of pathogen gram-negative, positive bacteria and yeast at very low concentrations. The cytotoxic effects of the particles were investigated on healthy cell lines (HDF) and cancerous cell lines (U118, CaCo-2, Skov-3). AgNPs showed up to 70% suppression in cancer cell lines. Abstract

Mercury is known to be one of the most toxic heavy metals in the environment and is released into the water systems in significant quantities through natural events and industrial process activities. Many chemical materials are used as adsorbents in the removal of toxic metals from the environment and wastewaters. However, using microorganisms as bio-sorbents instead of chemical materials has become common recently due to their low cost, easy availability and presence in nature. In this study, Bacillus licheniformis in the soil isolated from the Tigris River was used as bio-sorbent. The mercury (Hg(II)) absorption behaviour of Bacillus licheniformis bacteria (BLB) was investigated using inductively coupled plasma mass spectrometry. The effects of equilibrium of adsorption time, temperature, adsorbent dosage and pH on the adsorption of Hg (II) onto BLB were determined. The maximum adsorption capacity of Hg (II) onto BLB was determined as 82.12 mg/g (T = 25°C, pH 5, Co = 50 mg/L, m = 25 mg). The BLB was characterised using Fourier transform infrared spectroscopy analysis, thermal gravimetric analysis/differential thermal analysis, scanning electron microscopy analysis and energy-dispersive X-ray spectroscopy analysis. In addition, pseudo-first-order and pseudo-second-order kinetic models were applied. The equilibrium data for the adsorption of Hg(II) onto BLB were examined by the Langmuir and Freundlich isotherm models. The activation energy was calculated using the pseudo-second-order rate constant. These results suggested the BLB can be used as an efficient adsorbent for the removal of Hg(II) metal ions from wastewater. When the results of bio-sorption studies were examined, it was found that the bio-sorbent could be reused easily. Abstract

Bu çalışma Pleurotus eryngii'nin farklı çözücü ekstraktlarının besinsel içerikleri, yağ asidi bileşimleri, in vitro antioksidan, antikanser ve antimikrobiyal aktiviteleri ile ilgilidir. P. eryngii'nin besinsel içeriği ayrıntılı olarak araştırıldı. Meyve gövdesinin ham protein (g 100 kg-1), karbonhidrat (g 100 kg-1), lipit (g 100 kg-1), kül (g 100 kg-1), diyet lifi (g 100 kg-1) ve enerji değeri (100 g-1) analiz edildi. Pleurotus eryngii'nin toplam lipit, fosfolipid ve triaçilgliserol fraksiyonlarının yağ asidi bileşimleri Gaz kromatografisi ile araştırıldı. Toplam 13 farklı yağ asidi bileşeni belirlendi. Toplam antioksidan aktivite, serbest radikal süpürme aktivitesi, indirgeme gücü aktivitesi, metal şelatlama aktivitesi ve süperoksit anyon radikali süpürme aktivitesini içeren antioksidan özellikler farklı testler kullanılarak değerlendirildi. En yüksek toplam antioksidan, DPPH radikali süpürme, metal şelatlama ve süperoksit anyon radikal süpürme aktiviteleri etil asetat özütünde belirlendi. PC-3 (prostat kanseri) hücre hattı üzerindeki sitotoksik aktivite potansiyelinin, test edilen özüt fraksiyonlarının konsantrasyonuna ve tipine bağlı olduğu bulundu. Pleurotus eryngii'nin heksan, etil asetat ve metanol ekstraktlarının, test edilen mikroorganizmaların büyümesini inhibe edici etkiye sahip olduğu belirlendi. Mevcut sonuçlar, günlük besin olarak kullanılan Pleurotus eryngii'nin yeni ilaç gelişmeleri ve bazı kanser tedavilerinde umut vaat eden yeni bir kaynak olabileceğini düşündürmektedir., The present study related the nutritional contents, fatty acid compositions, in vitro antioxidant, anticancer and antimicrobial activities of different solvents extracts of Pleurotus eryngii. The nutritional contents of the P. eryngii were investigated in detail. Crude protein (g 100 kg-1), carbohydrate (g 100 kg-1), lipid (g 100 kg-1), ash (g 100 kg-1), dietary fiber (g 100 kg-1) and energy value (cal 100 g-1) of fruiting bodies were analysed. Fatty acid compositions of total lipid, phospholipid and triacylglycerol fractions of Pleurotus eryngii were investigated by Gas chromatography. Totally 13 different fatty acids constituents were identified. Antioxidant properties including total antioxidant activity, free radical scavenging activity, reducing power activity, metal chelating activity, and superoxide anion radical scavenging activity were evaluated using different tests. The highest total antioxidant, DPPH scavenging, metal chelating, and superoxide anion radical scavenging activities were determined in ethyl acetate extracts. The potential of cytotoxic activity on PC-3 (prostate cancer) cell lines was found to depend on the concentration and the type of the tested extracts fractions. It was determined that hexane, ethyl acetate, and methanol extracts of Pleurotus eryngii have inhibition effect on the growth of tested microorganisms. The present results suggested that used as a daily nutrient, the Pleurotus eryngii could be a novel source for new drug developments and promising in some cancer treatment.
