With the increasing prevalence of illicit drugs, social security and human health are at serious risk. However, traditional detection techniques require a lengthy pretreatment process to eliminate matrix interference and do not allow rapid, in situ analysis. Ambient ionization is a powerful tool for the rapid determination of illicit drugs in different matrices because it usually eliminates the need for sample pretreatment, and the portable mass spectrometer brings convenience to in situ detection. This study employed a thermal desorption-electrospray ionization (TD-ESI) source combined with a portable mass spectrometer to rapidly characterize illicit drugs in drinks, instant powders, and solid food surfaces. Air was utilized instead of nitrogen as the carrier gas for TD-ESI/MS in order to save space and cost together with rapid on-site analysis. A full-scan waveform combined with a qualitative pre-scan waveform (QPSW) was employed to form a two-step scanning mode to accurately identify the drugs. The results showed that the limits of detection (LODs) for the analytes were 100–200 ppb in drinks, 700–1000 ppb in instant powders, and 1 ng/cm2 on solid food surfaces. The complete TD-ESI/MS analysis time was less than 1 min.

A novel visible-light-catalyzed tandem radical addition/1,5-hydrogen atom transfer/cyclization cascade of 2-alkynylarylethers with sulfonyl chlorides in 2-methyltetrahydrofuran was developed under photocatalyst- and additive-free conditions. This reaction relies on unique energy transfer and solvent-radical relay strategies to generate sulfonyl radicals for the preparation of a series of sulfonyl-functionalized dihydrobenzofurans in moderate to high yields catalyzed by visible light or solar radiation.

A novel hydrophilic porous biocomposite was fabricated by incorporating graphene oxide (GO) @chitosan (CS) foam substrate (GO@CS@ZIF-8 foam) with ZIF-8 crystals in situ via a facile stirring method for simultaneous enrichment of glycopeptides and phosphopeptides from complex biological samples. The experimental results demonstrated that GO@CS@ZIF-8 foam exhibited favorable specificity for simultaneous enrichment of N-glycopeptides and phosphopeptides under the same condition for HRP and β-casein tryptic digest mixtures. The novel material was further applied to enriching both glycopeptides and phosphopeptides simultaneously from 4 μL complex human serum, and 423 N-glycopeptides and 40 phosphopeptides corresponding to 133 glycoproteins and 29 phosphoproteins were identified, respectively.

The existing particle swarm optimization (PSO) algorithms are only effective in deconvoluting the overlapping peaks in ion mobility spectrum with less than four component peaks, which limits the applicability of these algorithms.A high-performance two-step particle swarm optimization (TSPSO) algorithm was developed. As compared to the existing PSO algorithms, TSPSO can narrow the search ranges of all coefficients for the overlapping peaks through Gaussian model calculation, and thus, can deconvolute various overlapping peaks with high accuracy, even for 30 component overlapping peaks. In addition, the TSPSO could be further applied to enhance the resolution of the spectra by narrowing the peak widths after the peak deconvolution.Simulated overlapping peaks were first used to evaluate the performance of TSPSO as compared to the dynamic inertia weight particle swarm optimization (DIWPSO) algorithm. The results showed that the profiles of the peaks deconvoluted by using TSPSO were more consistent with the original ones. The fitness values and the standard deviations of the fitness values from TSPSO were also at least an order of magnitude less than those from DIWPSO. By applying TSPSO, the overlapping peaks from both MS and FAIMS spectra can also be well-deconvoluted. In addition, the resolutions of the MS and FAIMS spectra can be effectively enhanced after the peak deconvolution. The enhanced spectra matched excellently with the experimental ones acquired at high resolution modes.The experiment results convincingly demonstrated that TSPSO algorithm is capable of both deconvoluting complex overlapping peaks and enhancing the spectrum resolution with high accuracy.

Gas chromatography-mass spectrometry (GC-MS) is an analytical technique widely used in materials science, biomedicine, and other fields. The target compound in the experiment is identified by searching for its mass spectrum in a large mass spectrum database using some algorithms. This work introduces the use of deep learning ranking for the identification of small molecules using low-resolution electron ionization MS. Because different spectra are often very similar, the algorithm produces wrong search results, and the search accuracy needs improvement. Due to the library's large amount of data, the algorithm sometimes requires a large amount of calculation and is very time consuming.Given these two problems, this work aims to develop a model for ranking based on mass-to-charge ratio (m/z) pre-retrieval method combined with deep learning to improve search accuracy and reduce the algorithm's computational time. The master spectral library maintained by the National Institute of Standards and Technology is used as the reference library for all the experiments, and the replicate library is used as the query library to evaluate the method's performance.Compared with non-machine learning algorithms, the combination of m/z matching pre-retrieval and deep learning significantly improves library retrieval accuracy by about 4%. Moreover, compared with the deep learning sorting algorithm that does not use the pre-retrieval process, it improves the accuracy of spectral library retrieval by about 0.1% and reduces the computational time of the algorithm by more than 2 h.This method identifies compounds more efficiently and accurately than non-machine learning and deep learning algorithms without a pre-retrieval process.

Because of its powerful analytical ability, ion mobility spectrometry (IMS) plays an important role in the field of mass spectrometry. However, one of the main defects of IMS is its low structural resolution, which leads to the phenomenon of peak overlap in the analysis of compounds with similar mass charge ratio.A multiobjective dynamic teaching-learning-based optimization (MDTLBO) method was proposed to separate IMS overlapping peaks. This method prevents local optimization and identifies peak model coefficients efficiently. In addition, the position information of particles largely reflects the half-peak width of IMS, which makes single peaks difficult to appear and coefficient identification easier.The performance comparison of MDTLBO with other deconvolution methods (genetic algorithm, improved particle swarm optimization algorithm, and dynamic inertia weight particle swarm optimization algorithm) shows that the maximum deconvolution error of MDTLBO is only 0.7%, which is much lower than that for the other three methods. In addition, robustness is a performance index that reflects the advantages and disadvantages of the algorithm.MBTLBO is more robust than other algorithms for separating overlapping peaks. The algorithm can separate the heavily overlapped mobility peaks, produce better analysis results, and improve the resolution of IMS.

Mass spectrometry (MS) data is susceptible to random noises and alternating baseline, posing great challenges to spectral peak detection, especially for weak peaks and overlapping peaks. Herein, an efficient peak detection algorithm combining continuous wavelet transform (CWT) and genetic algorithm-based threshold segmentation (denoted as WSTGA) for mass spectrometry was proposed. Firstly, Mexican Hat wavelet was selected as the mother wavelet by comparing the matching degree between the difference of Gaussian (DOG) and different wavelets. Subsequently, the ridges and valleys were identified from 2D wavelet coefficient matrix. Afterward, an improved threshold segmentation method, Otsu method based on genetic algorithm, was introduced to find optimal segmentation threshold and achieve better image segmentation, overcoming the deficiency of traditional Otsu method that cannot handle long-tailed unimodal histograms. Finally, the characteristic peaks were successfully identified by utilizing the ridge-valley lines in wavelet space and original spectrum. Receiver operating characteristic (ROC) curve, area under curve (AUC) and F₁ measure are used as criterions to evaluate performance of peak detection algorithms. Compared with multi-scale peak detection (MSPD) and CWT and image segmentation (CWT-IS) methods, all the results showed that WSTGA can achieve better peak detection. More importantly, the experimental results from MALDI-TOF spectra demonstrated that WSTGA can effectively detect more weak peaks and overlapping peaks while maintaining a lower false peak detection rate than MSPD and CWT-IS methods, indicating its great advantages in characteristic peak identification.

The detection limit is one of the most important performance characteristics for portable mass spectrometers (MS). To significantly reduce the size and the weight, small vacuum pumps are used in portable MS which leads to the use of a discontinuous atmospheric pressure interface (DAPI). Multiple ion injections (M-DAPI), which accumulates ions through opening the DAPI pinch valve several times during the ion injection period while maintaining a radio frequency (RF) voltage continuously on the trap, was shown to improve the sensitivity of DAPI-based portable MS. A systematic investigation of the optimization of M-DAPI was performed. The primary parameters affecting the performance of M-DAPI were optimized and sensitivity and stability improvements are reported. An 11.6-fold improvement in sensitivity was achieved by M-DAPI compared to the single ion injection DAPI operation for 1 ��g/mL enrofloxacin. A more than 10-fold reduction in the limit of detection (LOD) to 0.01 ��g/mL was also obtained. In addition, the stability was improved by M-DAPI with a relative standard deviation of 14.4% compared to 4.6% for DAPI.

In this study, the effects of lead ions(Pb2+) on the conformations of leucine encephalin(LE) and methionine encephalin(ME) in gas phase were studied using hydrogen/deuterium exchange mass spectrometry(HDX-MS) and quantum chemistry theoretical calculations at the molecular level. The HDX-MS result revealed that the complexes with the monovalent compounds [LE+Pb−H]+ and [ME+Pb−H]+ had a 1:1 stoichiometric ratio, and different HDX reactivates were observed in a follow of [ME+H]+>[LE+H]+>[LE+Pb−H]+> [ME+Pb−H]+. Combining the collision-induced dissociation energies of the complexes and their HDX results, it was found that the more stable the complex, the harder it was for HDX. In addition, the favorable conformations of the complexes were obtained by theoretical calculations, revealing that the similar coordination type with different bond lengths was obtained. Then, the proton affinity(PA) values of the optimized complexes were calculated to interpret the HDX observations, indicating that the higher the PA values, the more difficult it was for HDX. Overall, the experiments and theoretical calculations revealed that Pb2+ could induce conformational changes of LE and ME, and generate ME into a more rigid conformation than LE. The results will prompt further fundamental investigations on the conformational properties of LE/ME in coordination with Pb2+.

In this work, a magnetic graphene material coated with mesoporous silica was selected as the substrate, 3-glycidoxypropyltrimethoxysilane and polyethyleneimine were sequentially bonded through chemical reactions, and then carrageenan was successfully introduced by electrostatic interaction; finally, hydrophilic nanocomposite material was prepared. Due to the large number of hydrophilic groups, and polyethyleneimine was connected by means of chemical bonds, this material exhibits good hydrophilicity and stability for glycopeptide enrichment. In the actual enrichment process, nanomaterial exhibits high selectivity (1:500), high sensitivity (2 fmol), and good repeatability (five cycles). In addition, the synthesized material also shows a good enrichment effect in the face of actual complex biological samples, which captured 40 N-glycopeptides from human saliva, indicating the application potential for enrichment of N-glycopeptides.

A flower-like nanostructured MnO2 with near-infrared (NIR) light-triggered high photothermal conversion capability of 30% and reactive oxygen species (ROS) generation ability was successfully developed. Different from the reported MnO2 nanomaterials those were used in the nanomedicine field for only relieving tumor hypoxia and/or imaging, the flower-like MnO2 inherently acts as a competent agent for simultaneously enhanced photothermal and photodynamic therapy. A flower-like nanostructured MnO2 with near-infrared (NIR) light triggered high photothermal conversion capability of 30% and reactive oxygen species (ROS) generation ability was successfully developed.

Limited sample loading capacity is one of the major reasons that prevents the utility of capillary electrophoresis (CE) as a routine separation method as compared to liquid chromatography (LC). In our previous study, separation voltage polarity switching transient capillary isotachophoresis (PS-tCITP) was proposed. Both sample loading capacity and separation resolution could be improved using a single PS-tCITP instead of routine transient capillary isotachophoresis (tCITP). In this study, a detailed investigation on the optimization strategy of the PS-tCITP method was performed systematically. A possible mechanism of sample preconcentration in multiple PS-tCITP was first proposed to better understand the multiple PS-tCITP process. Several optimization experiments were then performed, including single PS-tCITP, paused PS-tCITP and multiple PS-tCITP, sequentially using a mixture of five peptides. By selecting an optimum polarity switching time, sample loading capacity of 100% capillary volume could be achieved in a single PS-tCITP. Introducing an additional pause between each polarity switching in a single PS-tCITP further improved the separation resolution. Experimental results showed a baseline separation of five selected peptide standards at 100% sample loading volume using a 100 min pause in a single PS-tCITP. To further improve separation efficiency while still maintaining 100% sample loading volume, a multiple PS-tCITP technique was developed through this study. Compared to the separation performance of the optimal single PS-tCITP at 100% sample loading volume with a 10 min pause, the separation window was improved by 54% and the peak capacity was improved by 48% in the optimal four PS-tCITP with the same sample loading volume and pause.

3-Pyridinylboronate, a zwitterionic boronic acid, displayed effective in situ ESI for reversible covalent tagging of saccharides in both cation and anion modes. The ion mobilities of thus-generated ions were examined with the Bruker timsTOF fleX instrument. Nine disaccharides were examined using this method. They have identical mass-to-charge ratios, differing only in monomer compositions, regio-linkages, and anomeric configurations (α or β). The IMS separations of the disaccharides from this method were compared with those from sodium adducts reported in the literature. The differentiation effects of this method on the disaccharide isomers were increased on average by an order of magnitude. Using this method, all the pairs of disaccharides selected from nine isomers were completely identified by comparing the mobility spectra of single-tagged and double-tagged ions.

Chiral analysis is critical to many research fields due to different biological functions of enantiomers in living systems. Although the use of ion mobility spectrometry (IMS) has become an alternative technology in the area of chiral measurements, there is still a lack of a general chiral selector for IMS-based chiral recognition, especially for small chiral molecules. Here, a new method using oligosaccharides as the chiral selector has been developed to discriminate chiral amino acids (AAs) by trapped ion mobility spectrometry-mass spectrometry (TIMS-MS). We analyzed 21 chiral amino acids, including small molecules (e.g., alanine and cysteine). Our data showed that the use of nonreducing tetrasaccharides was effective for the separation of chiral AAs, which differentiated 21 chiral AAs without using metal ions. By further incorporating a copper ion, the separation resolution could be improved to 1.64 on average, which accounts for an additional 52% improvement on top of the already achieved separation in metal-free analysis. These results indicate that the use of tetrasaccharides is an effective strategy for the separation of AA enantiomers by TIMS. The method developed in this study may open up a new strategy for effective IMS-based chiral analysis.

Near-infrared (NIR) light-triggered pH-manipulation has been realized by utilizing upconversion nanoparticle assisted ring-closing reactions of the conventional photoacid merocyanine (MC). This pH manipulation behavior was then used to regulate the switch of an acid-labile cap for facilitating drug release on the basis of mesoporous silica coated upconversion nanoparticles, in which the drug release rate and amount and the cell killing ability have been greatly improved upon NIR light irradiation due to the locally high concentration of H+ within pore channels that is generated by upconversion assisted MC. This proof of concept may provide a way to utilize NIR light to regulate pH change for new drug delivery system designs and applications in biomedical field.

In this work, a novel, rapid, and simple analytical method was proposed for the detection of parabens in milk sample by gas chromatography coupled with mass spectrometry. At the same time, milk sample was pretreated by magnetic solid phase extraction, which detected up to five parabens. A series of important parameters of magnetic solid phase extraction were investigated and optimized, such as pH value of loading buffer, amount of material, adsorption time, ionic strength, eluting solvents, and eluting time. Under the optimized conditions, the corresponding values were more than 0.9991, limits of detection and the limit of quantification were 0.1 and 0.5 ng/mL, respectively. In addition, the recoveries were achieved in range of 95-105%, the liner range were within 0.1-600 ng/mL, and the relative standard deviations were even lower than 5%.

As a member of MOFs, Zn-MOFs (ZIF-8) are seldom used in phosphopeptide enrichment because ZIF-8 is soluble in acid solutions. Therefore, properly designing a novel strategy to overcome the defect of ZIF-8 is necessary. In this study, a novel multifunctional nanoprobe was designed by uniting magnetic core, titania shell and hydrophilic metal-organic frameworks (named as Fe3O4@PDA@mTiO2@PEI-g-ZIF-8). Integrating the strategies of hydrophilic interaction affinity chromatography (HILIC), immobilized metal ion affinity chromatography (IMAC) and metal oxide affinity chromatography (MOAC), the Fe3O4@PDA@mTiO2@PEI-g-ZIF-8 mesoporous microspheres can enrich phosphorylated peptides and glycosylated peptides simultaneously. Fe3O4@PDA@mTiO2@PEI-g-ZIF-8 has high selectivity (maximum molar ratio β-casein/HRP : BSA = 1 : 1000), low detection limit (2 fmol) towards phosphopeptides and glycopeptides. Besides, the Fe3O4@PDA@mTiO2@PEI-g-ZIF-8 also exhibited a fine performance in the actual sample detection. In the experiment, taking saliva as a sample, 16 phosphorylated peptides were identified, and from a human serum sample, 4 phosphorylated peptides were selectively identified. All in all, the materials show great potential in the future study of phosphoproteomics and glycoproteomics.

Capillary electrophoresis (CE) coupled with mass spectrometry (MS) through electrospray ionization (ESI) is a promising alternative technique to liquid chromatography-ESI-MS (LC-ESI-MS) due to its high separation efficiency and high detection sensitivity. A sensitive and robust interface is essential in CE-ESI-MS. Continued development of CE-ESI-MS interfaces in the last decade, including junction-at-the-tip interfaces and sheathless interfaces, has improved the sensitivity and robustness of CE-ESI-MS significantly. The limited loading capacity of CE, one of major reasons that limits the utility of CE as a routine separation method, has also been addressed effectively by the use of in-capillary sample preconcentration techniques, such as transient CITP/CZE and dynamic pH junction. CE-ESI-MS could yield higher sensitivity as compared with the conventional LC-ESI-MS, and, therefore, is capable of identifying more proteins and peptides when the sample amount is very limited, such as single cell analysis. To improve the protein sequence coverage, CE-ESI-MS can also be used as a complementary technique to LC-ESI-MS, or combined with reversed phase LC to form a two dimensional separation technique. CE-ESI-MS is also effective in quantifying targeted peptides/proteins in complex bio-matrix.

Tumor-derived exosomes, which contain RNA, DNA, and proteins, are a potentially rich non-invasive source of biomarkers. However, no efficient isolation or detection methods are yet available. Here, we developed a microfluidic Raman biochip designed to isolate and analyze exosomes in situ. Anti-CD63 magnetic nanoparticles were used to enrich exosomes through mixing channels of a staggered triangular pillar array. EpCAM-functionalized Raman-active polymeric nanomaterials (Raman beads) allow rapid analysis of exosome samples within 1 h, with a quantitative signal at 2230 cm-1. The limit of detection of this biochip approaches 1.6 × 102 particles per mL with 20 μL samples. The newly developed biochip assay was successfully applied in the determination of exosomes from clinical serum samples. Thus, this novel device may have potential as a clinical exosome analysis tool for prostate cancer.

RATIONALE FAIMS has a great potential to become a portable technology for rapid detection of chemical and biological agents. However the ion current signals, measured at the exit of the planar FAIMS directly, may contain different types of noises. The peak information in the FAIMS spectrum, such as the compensation voltage value at the maximum peak intensity (CVP ) and the peak width at half maximum (Wh ), could not be accurately determined under the weak signal condition which significantly limits the achievable instrument sensitivity, and there are no existing solutions to the problem. METHODS This study analyzed the noise type of FAIMS signal in detail, and three different signal processing algorithms, including median filtering (MF), discrete wavelet transform (DWT) and zero-phase digital filtering (ZDF), were evaluated for their performance in denoising the FAIMS signal. RESULTS The results show that the standard deviation of CVp obtained from the signal denoised by using ZDF algorithm is at least 31.82% smaller as compared to using MF and DWT algorithms. The standard deviation of Wh is at least 45.45% smaller by using ZDF algorithm. Moreover, only ZDF algorithm can keep the percentage error for the CV value of the denoised signal to be within 0.50±0.47% of the true CV value, implying the effectiveness of ZDF algorithm in denoising while retaining the integrity of the signal. CONCLUSIONS The ZDF algorithm greatly reduces the analyte peak extraction error and improves the limit of detection in FAIMS measurements.

Background GC–MS is a powerful tool for component analysis of unknown compounds, especially in the fields of analytical chemistry and detection of biological samples. To effectively identify compounds in GC–MS, one of the most important ways is to use a matching algorithm to compare the similarity between the reference spectrum and the query spectrum. Objective To propose a novel way to improve compound identification accuracy. Methods This article proposes a method based on an m/z set match. First of all, select the maximum m/z and the m/z corresponding to the highest peak intensity in a pre-search. Next, employ the space vector model to carry on a refined search in the remaining spectra after the pre-search. Then, distinguish stereoisomers according to the order of the G value. Results Compared with the 10 peaks and the method based on an m/z number matching pre-search, the method based on m/z set matching showed higher accuracy and fewer remaining (missing) spectra. Furthermore, the refined search which is based on the m/z set matching method possesses shorter calculation time compared with no pre-search. Conclusions The method can reduce the remaining spectra and speed up the identification of compounds. Highlights The accuracy is higher, the number of remaining spectra is less, and the computational time is shorter.

A novel type of boric acid–functionalized magnetic covalent organic framework (mCOF) with polyethyleneimine (PEI) as a linker (denoted as mCOF@PEI@B(OH)2) has been prepared through a post-synthesis strategy, which points out an achievable path for the construction of boronic acid–functionalized COFs. Based on the boric acid chemistry, the obtained core-shell structured mCOF@PEI@B(OH)2 can selectively isolate glycopeptides through the modified boronic acid groups. The mCOF@PEI@B(OH)2 exhibits excellent performance with good reusability (ten cycles), low detection limit (0.5 fmol·μL−1), size-exclusion effect, and relatively high loading capacity (80 μg·mg−1), recovery yield (94.9 ± 2.8%), and selectivity (HRP digests:BSA digests = 1:500). Detection is done by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). In addition, 37 endogenous glycopeptides are captured from human saliva with mCOF@PEI@B(OH)2, providing effective proofs for its capability to capture low-abundance glycopeptides from actual biological samples.

Rationale Gas Chromatography Mass Spectrometry (GC-MS) with electron ionization (EI)is the most widely used analysis technique of gaseous samples, but it may be time-consuming for on-line monitoring of mixtures whose concentrations relatively change rapidly. On the other hand, current ionization methods, such as chemical ionization (CI) and proton transfer reaction (PTR), also have some disadvantages such as selectivity. Appropriate soft ionization sources therefore are searched for rapid on-line detection. Methods Hollow Electrode Capillary Plasma Ionization (HECPI) is based on single electrode plasma. A hollow capillary was placed as both the electrode and the inlet of the gaseous samples. The ionization source is coupled with a mass spectrometer for performance evaluation. Results Several typical compounds have been tested with HECPI-mass spectrometer. In this process, the dominant ion peaks of all compounds can be indexed as molecular ion peaks, and the product ions of HECPI are less than that of dielectric barrier discharge ionization (DBDI). Three gaseous samples (linalool, triethylamine and styrene) with various concentrations have been employed to further confirm the performance of this source, and the detection limit of linalool is as low as 10 ppb. Conclusions HECPI is simple in structure and shows good performance. The results also show that HECPI has the potential to be an effective tool for detecting on-line gaseous samples rapidly.
