Abstract

A timely and long-term programme of kaon decay measurements at a new level of precision is presented, leveraging the capabilities of the CERN Super Proton Synchrotron (SPS). The proposed programme is firmly anchored on the experience built up studying kaon decays at the SPS over the past four decades, and includes rare processes, CP violation, dark sectors, symmetry tests and other tests of the Standard Model. The experimental programme is based on a staged approach involving experiments with charged and neutral kaon beams, as well as operation in beam-dump mode. Abstract

A timely and long-term programme of kaon decay measurements at a new level of precision is presented, leveraging the capabilities of the CERN Super Proton Synchrotron (SPS). The proposed programme is firmly anchored on the experience built up studying kaon decays at the SPS over the past four decades, and includes rare processes, CP violation, dark sectors, symmetry tests and other tests of the Standard Model. The experimental programme is based on a staged approach involving experiments with charged and neutral kaon beams, as well as operation in beam-dump mode. The various phases will rely on a common infrastructure and set of detectors.

Abstract

Background\ud \ud The relative involvement of the large and small airways in asthma is not clear. Hyperpolarised gas MRI provides high resolution three-dimensional images of ventilation distribution which can be quantified by the ventilated volume percentage of the lungs (VV%).\ud \ud \ud Objective\ud \ud To (1) quantify the baseline reproducibility of VV%, (2) assess the ventilation distribution between the proximal and peripheral lungs and (3) investigate regional ventilation response to bronchodilator inhalation in a cohort of patients with asthma.\ud \ud \ud Methods\ud \ud 33 patients with poorly controlled, moderate-to-severe asthma were scanned with hyperpolarised 3He MRI. Two image datasets were acquired at baseline, and one image dataset was acquired after bronchodilator inhalation. Images were divided into proximal and peripheral regions for analysis.\ud \ud \ud Results\ud \ud Bland-Altman analysis showed strong reproducibility of VV% (bias = 0.12%, LOA = -1.86, 2.10%). VV% variation at baseline was greater in the periphery than in the proximal lung. The proximal lung was better ventilated than the peripheral lung. Ventilation increased significantly in response to bronchodilator, globally and regionally, and the ventilation increase in response to bronchodilator was greater in the peripheral lung than in the proximal lung. Hyperpolarised gas MRI was more sensitive to changes in response to bronchodilator (58%) than spirometry (33%).\ud \ud \ud Conclusion\ud \ud The peripheral lung showed reduced ventilation and a greater response to bronchodilator than the proximal lung. The high level of baseline reproducibility and sensitivity of hyperpolarised gas MRI to bronchodilator reversibility suggests that it is suitable for low subject number studies of therapy response.

Abstract

The NA62 experiment at CERN, configured in beam-dump mode, has searched for dark photon decays in flight to electron-positron pairs using a sample of 1.4×10^{17} protons on dump collected in 2021. No evidence for a dark photon signal is observed. The combined result for dark photon searches in lepton-antilepton final states is presented and a region of the parameter space is excluded at 90% confidence level, improving on previous experimental limits for dark photon mass values between 50 and 600  MeV/c^{2} and coupling values in the range 10^{-6} to 4×10^{-5}. An interpretation of the e^{+}e^{-} search result in terms of the emission and decay of an axionlike particle is also presented.

Abstract

The proportion of research outputs published in open access journals or made available on other freely-accessible platforms has increased over the past two decades, driven largely by funder mandates, institutional policies, grass-roots advocacy, and changing attitudes in the research community. However, the relative effectiveness of these different interventions has remained largely unexplored. Here we present a robust, transparent and updateable method for analysing how these interventions affect the open access performance of individual institutes. We studied 1,207 institutions from across the world, and found that, in 2017, the top-performing universities published around 80-90% of their research open access. Abstract

The tumor suppressor p53 is often inactivated via its interaction with endogenous inhibitors mouse double minute 4 homolog (MDM4 or MDMX) or mouse double minute 2 homolog (MDM2), which are frequently overexpressed in patients with acute myeloid leukemia (AML) and other cancers. Pharmacological disruption of both of these interactions has long been sought after as an attractive strategy to fully restore p53-dependent tumor suppressor activity in cancers with wild-type p53. Selective targeting of this pathway has thus far been limited to MDM2-only small-molecule inhibitors, which lack affinity for MDMX. We demonstrate that dual MDMX/MDM2 inhibition with a stapled α-helical peptide (ALRN-6924), which has recently entered phase I clinical testing, produces marked antileukemic effects. ALRN-6924 robustly activates p53-dependent transcription at the single-cell and single-molecule levels and exhibits biochemical and molecular biological on-target activity in leukemia cells in vitro and in vivo. Dual MDMX/MDM2 inhibition by ALRN-6924 inhibits cellular proliferation by inducing cell cycle arrest and apoptosis in cell lines and primary AML patient cells, including leukemic stem cell-enriched populations, and disrupts functional clonogenic and serial replating capacity. Furthermore, ALRN-6924 markedly improves survival in AML xenograft models. Abstract

Autism spectrum disorders (ASD) comprise an etiologically heterogeneous set of neurodevelopmental disorders. Neuroligin-3 (NL-3) is a cell adhesion protein that mediates synapse development and has been implicated in ASD. We performed ex-vivo high resolution magnetic resonance imaging (MRI), including diffusion tensor imaging (DTI) and behavioral (social approach and zero maze) tests at 3 different time points (30, 50 and 70 days-of-age) on NL-3 and wild-type littermates to assess developmental brain abnormalities in NL-3 mice. MRI data were segmented in 39 different gray and white matter regions. Volumetric measurements, along with DTI indices from these segmented regions were also performed. After controlling for age and gender, the NL-3 knock-in animals demonstrated significantly reduced sociability and lower anxiety-related behavior in comparison to their wild type littermates. Significantly reduced volume of several white and gray matter regions in the NL-3 knock-in mice were also observed after considering age, gender and time point as covariates. These findings suggest that structural changes in the brain of NL-3 mice are induced by the mutation in the NL-3 gene. No significant differences in DTI indices were observed, which suggests that the NL-3 mutation may not have a profound effect on water diffusion as detected by DTI. The volumetric and DTI studies aid in understanding the biology of disrupting function on an ASD risk model and may assist in the development of imaging biomarkers for ASD.

Abstract

Chronic exposure to stress is associated with a number of psychiatric disorders, but little is known about the epigenetic mechanisms that underlie the stress response or resilience to chronic stress. We investigated histone acetylation in seven different brain regions of rats exposed to chronic social defeat stress: the dorsal hippocampus (dHPC), ventral hippocampus (vHPC), medial prefrontal cortex (mPFC), basolateral amygdala (BLA), locus coeruleus (LC), paraventricular thalamus (PVT), and dorsal raphe (DR) nucleus. This stress paradigm was unique in that it allowed rats to display resilience in the form of an active coping mechanism. We found that there was an increase in acetylation of H3K9/14 (H3K9/14ac) and bulk acetylation of H4K5,8,12,16 (H4K5,8,12,16ac) in the DR nucleus of rats that were less resilient. Less resilient rats also displayed increased levels of H3K18 acetylation (H3K18ac) in the mPFC when compared to non-stressed controls. In the vHPC, there was an increase in H3K18ac and H4K12 (H4K12ac) in rats that were less resilient when compared to non-stressed control rats. In addition, there was a decrease in levels of H4K8 acetylation (H4K8ac) in both resilient and non-resilient rats as compared to controls. We assessed expression of histone modifying enzymes in the vHPC and the mPFC using quantitative real-time polymerase chain reaction (PCR) and found changes in expression of a number of targets. These included changes in Sirt1 and Sirt2 in the vHPC and changes in Kat5 in the mPFC. Overall, these results suggest that changes in histone acetylation and expression of histone modifying enzymes in these regions correlate with the behavioral response to stress in socially defeated rats., (Copyright © 2013 IBRO. Abstract

Unilateral posterior crossbite is a common finding in patients seeking orthodontic treatment. When treating this condition with a rapid maxillary expander, clinicians occasionally observe no resolution of the crossbite and document an unnecessary overexpansion of the contralateral dentition. This article describes a method for treating unilateral posterior crossbite and, specifically, the lack of crossbite correction after expansion with a rapid maxillary expander. A composite ramp is bonded to the mesial buccal cusp of the maxillary first molar in crossbite. The necessary materials and instruments are typically found in an orthodontic office. This adjunctive procedure requires no more than 5 extra minutes during an examination, and no additional appointments are necessary.

Statement of Problem: Soft and hard stabilizing appliances have been used to treat temporomandibular disorders. No data exist to suggest whether a hard or soft appliance is beneficial., Purpose: This study compared soft and hard acrylic resin stabilizing appliances in the reduction of masticatory muscle pain in patients with temporomandibular disorders., Materials and Methods: Twenty-three patients with at least one clinical sign from the list of diagnostic subgroups of temporomandibular disorders were alternately assigned a hard or soft appliance for temporomandibular disorder treatment. No other temporomandibular disorder treatment (self-care, physical therapy, biofeedback, or muscle or joint injections) was rendered. Each patient was seen by two dentists at each visit. One dentist initially fabricated the appliance and adjusted the appliance on each visit and an examining dentist examined the patient each visit and recorded signs of temporomandibular disorders. The appliance material (soft or hard) was not disclosed to the examining dentist, only to the dentist who fabricated and adjusted the appliance. Patients were examined and appliances were adjusted every 2 to 3 weeks for a minimum of 10 weeks. Masticatory muscles were palpated and charted on each visit. Data were analyzed and subjected to nonparametric Mann-Whitney test., Results: Eighteen of the initial 23 patients, 7 in the hard appliance group and 11 in the soft appliance group finished the study over 10- to 15-week period. Soft and hard appliances performed the same in reduction of masticatory muscle pain., Conclusion: This study suggests, based on the limited number of participants, that soft and hard stabilizing appliances may be equally useful in reducing masticatory muscle pain in short-term appliance therapy.

Abstract

The purpose of this study was to determine if there was a difference between the temporomandibular condylar movement patterns of a symptomatic adult population and those of an asymptomatic adult population. Thirty-five volunteers who were not seeking treatment for TMD underwent two different assessments for TMD signs and symptoms: (1) a self-administered questionnaire and (2) a clinical examination. Based on the information obtained from the questionnaires, subjects were divided into "reported-symptomatic" and "reported-asymptomatic" groups. Based on the investigator's clinically evaluation of the same subjects, subjects were divided into "clinically symptomatic" and "clinically asymptomatic" groups. To compare condylar movement patterns, both groups of subjects then had their mandibular border condylar movements measured bilaterally using a sagittal recording device during maximum opening, maximum protrusion, and maximum left and right excursion movements. The patterns were separated into two broad groups, "symmetric" and "asymmetric." Symmetric gliding movements were defined as uninterrupted bilaterally mirror-like patterns of each condyle with a difference between left and right total length excursion not exceeding 2 mm during opening in the sagittal plane or horizontal plane. Our results show that 63% of the subjects who reported clinically asymptomatic for TMD demonstrated asymmetric condylar movements. However, 100% of the patients (n = 5) who reported clinically symptomatic for TMD exhibited asymmetric condylar movements. This finding suggests that, while a very high percentage of TMD subjects will have asymmetric condylar movements, condylar movements alone are not necessarily diagnostic of TMD, and the sagittal recording device may alert the clinician to abnormal movements.
