Abstract

When grapes are exposed to wildfire smoke, certain smoke-related volatile phenols (VPs) can be absorbed into the fruit, where they can be then converted into volatile-phenol (VP) glycosides through glycosylation. These volatile-phenol glycosides can be particularly problematic from a winemaking standpoint as they can be hydrolyzed, releasing volatile phenols, which can contribute to smoke-related off-flavors. Current methods for quantitating these volatile-phenol glycosides present several challenges, including the requirement of expensive capital equipment, limited accuracy due to the molecular complexity of the glycosides, and the utilization of harsh reagents. To address these challenges, we proposed an enzymatic hydrolysis method enabled by a tailored enzyme cocktail of novel glycosidases discovered through genome mining, and the generated VPs from VP glycosides can be quantitated by gas chromatography-mass spectrometry (GC-MS). The enzyme cocktails displayed high activities and a broad substrate scope when using commercially available VP glycosides as the substrates for testing. When evaluated in an industrially relevant matrix of Cabernet Sauvignon wine and grapes, this enzymatic cocktail consistently achieved a comparable efficacy of acid hydrolysis. The proposed method offers a simple, safe, and affordable option for smoke taint analysis.

2. Towards a theory of eigenvalue asymptotics on infinite metric graphs: the case of diagonal combs

Abstract

We examine diagonal combs, a recently identified class of infinite metric graphs whose properties depend on one parameter. These graphs exhibit a fascinating regime where they possess infinite volume while maintaining purely discrete spectrum for the Neumann Laplacian. In this regime, we establish polynomial upper and lower bounds on the $k$-th eigenvalue, revealing that the eigenvalues grow at a rate strictly slower than quadratic. However, once the diagonal combs transition to finite volume, their growth accelerates to a quadratic rate. 3. Optimizing the Fundamental Eigenvalue Gap of Quantum Graphs

Abstract

We study the problem of minimizing or maximizing the fundamental spectral gap of Schr\"odinger operators on metric graphs with either a convex potential or a ``single-well'' potential on an appropriate specified subset. (In the case of metric trees, such a subset can be the entire graph.) In the convex case we find that the minimizing and maximizing potentials are piecewise linear with only a finite number of points of non-smoothness, but give examples showing that the optimal potentials need not be constant. This is a significant departure from the usual scenarios on intervals and domains where the constant potential is typically minimizing. In the single-well case we show that the optimal potentials are piecewise constant with a finite number of jumps, and in both cases give an explicit estimate on the number of points of non-smoothness, respectively jumps, the minimizing potential can have. Furthermore, we show that, unlike on domains, it is not generally possible to find nontrivial bounds on the fundamental gap in terms of the diameter of the graph alone, within the given classes.

Abstract

We introduce a natural notion of mean (or average) distance in the context of compact metric graphs, and study its relation to geometric properties of the graph. We show that it exhibits a striking number of parallels to the reciprocal of the spectral gap of the graph Laplacian with standard vertex conditions: it is maximised among all graphs of fixed length by the path graph (interval), or by the loop in the restricted class of doubly connected graphs, and it is minimised among all graphs of fixed length and number of edges by the equilateral flower graph. We also establish bounds for the correctly scaled product of the spectral gap and the square of the mean distance which depend only on combinatorial, and not metric, features of the graph. This raises the open question whether this product admits absolute upper and lower bounds valid on all compact metric graphs.

6. Cheeger cuts and Robin spectral minimal partitions of metric graphs

Abstract

We study partition problems based on two ostensibly different kinds of energy functionals defined on $k$-partitions of metric graphs: Cheeger-type functionals whose minimisers are the $k$-Cheeger cuts of the graph, and the corresponding values are the $k$-Cheeger constants of the graph; and functionals built using the first eigenvalue of the Laplacian with positive, i.e. absorbing, Robin (delta) vertex conditions at the boundary of the partition elements. We prove existence of minimising $k$-partitions, $k \geq 2$, for both these functionals. We also show that, for each $k \geq 2$, as the Robin parameter $\alpha \to 0$, up to a renormalisation the spectral minimal Robin energy converges to the $k$-Cheeger constant. Moreover, up to a subsequence, the Robin spectral minimal $k$-partitions converge in a natural sense to a $k$-Cheeger cut of the graph. Finally, we show that as $\alpha \to \infty$ there is convergence in a similar sense to the corresponding Dirichlet minimal energy and partitions. Abstract

Inspired by a recent result of Funano's, we provide a sharp quantitative comparison result between the first nontrivial eigenvalues of the Neumann Laplacian on bounded convex domains $\Omega_{1} \subset \Omega_{2}$ in any dimension $d$ greater than or equal to two, recovering domain monotonicity up to an explicit multiplicative factor. We provide upper and lower bounds for such multiplicative factors for higher-order eigenvalues, and study their behaviour with respect to the dimension and order. We further consider different scenarios where convexity is no longer imposed. In a final section we formulate some related open problems.

Abstract

OBJECTIVE: Suicidal behavior is heritable and is a major cause of death worldwide. Two large-scale genome-wide association studies (GWASs) recently discovered and cross-validated genome-wide significant (GWS) loci for suicide attempt (SA). The present study leveraged the genetic cohorts from both studies to conduct the largest GWAS meta-analysis of SA to date. Multi-ancestry and admixture-specific meta-analyses were conducted within groups of significant African, East Asian, and European ancestry admixtures. METHODS: This study comprised 22 cohorts, including 43,871 SA cases and 915,025 ancestry-matched controls. Analytical methods across multi-ancestry and individual ancestry admixtures included inverse variance-weighted fixed-effects meta-analyses, followed by gene, gene-set, tissue-set, and drug-target enrichment, as well as summary-data-based Mendelian randomization with brain expression quantitative trait loci data, phenome-wide genetic correlation, and genetic causal proportion analyses. Abstract

Cerebral edema (CE) and hemorrhagic transformation (HT) are frequent and unpredictable events in patients with acute ischemic stroke (AIS), even when an effective vessel recanalization has been achieved. These complications, related to blood-brain barrier (BBB) disruption, remain difficult to prevent or treat and may offset the beneficial effect of recanalization, and lead to poor outcomes. The aim of this translational study is to evaluate the association of circulating and imaging biomarkers with subsequent CE and HT in stroke patients with the dual purpose of investigating possible predictors as well as molecular dynamics underpinning those events and functional outcomes. Concurrently, the preclinical study will develop a new mouse model of middle cerebral artery (MCA) occlusion and recanalization to explore BBB alterations and their potentially harmful effects on tissue. The clinical section of the study is based on a single-center observational design enrolling consecutive patients with AIS in the anterior circulation territory, treated with recanalization therapies from October 1, 2015 to May 31, 2020. The study will employ an innovative evaluation of routine CT scans: in fact, we will assess and quantify the presence of CE and HT after stroke in CT scans at 24 h, through the quantification of anatomical distortion (AD), a measure of CE and HT. We will investigate the relationship of AD and several blood biomarkers of inflammation and extracellular matrix, with functional outcomes at 3 months. In parallel, we will employ a newly developed mouse model of stroke and recanalization, to investigate the emergence of BBB changes 24 h after the stroke onset. The close interaction between clinical and preclinical research can enhance our understanding of findings from each branch of research, enabling a deeper interpretation of the underlying mechanisms of reperfusion injury following recanalization treatment for AIS.

29. Spectral minimal partitions of unbounded metric graphs

Abstract

We investigate the existence or non-existence of spectral minimal partitions of unbounded metric graphs, where the operator applied to each of the partition elements is a Schr\"odinger operator of the form $-\Delta + V$ with suitable (electric) potential $V$, which is taken as a fixed, underlying ``landscape'' on the whole graph. We show that there is a strong link between spectral minimal partitions and infimal partition energies on the one hand, and the infimum $\Sigma$ of the essential spectrum of the corresponding Schr\"odinger operator on the whole graph on the other. Namely, we show that for any $k\in\mathbb{N}$, the infimal energy among all admissible $k$-partitions is bounded from above by $\Sigma$, and if it is strictly below $\Sigma$, then a spectral minimal $k$-partition exists. We illustrate our results with several examples of existence and non-existence of minimal partitions of unbounded and infinite graphs, with and without potentials. The nature of the proofs, a key ingredient of which is a version of Persson's theorem for quantum graphs, strongly suggests that corresponding results should hold for Schr\"odinger operator-based partitions of unbounded domains in Euclidean space., Comment: Revised, shortened version, a number of proofs and examples have been tightened. We introduce a new criterion for compactness of the resolvent and apply this to identify a transition from purely discrete to non-empty essential spectrum among a class of infinite metric graphs, a phenomenon that seems to have no known counterpart for Laplacians on Euclidean domains of infinite volume. In the case of discrete spectrum we then prove upper and lower bounds on eigenvalues, thus extending a number of bounds previously only known in the compact setting to infinite graphs. Abstract

We derive several upper bounds on the spectral gap of the Laplacian with standard or Dirichlet vertex conditions on compact metric graphs. In particular, we obtain estimates based on the length of a shortest cycle (girth), diameter, total length of the graph, as well as further metric quantities introduced here for the first time, such as the avoidance diameter. Abstract

Trait stability of measures is an essential requirement for individual differences research. Functional MRI has been increasingly used in studies that rely on the assumption of trait stability, such as attempts to relate task related brain activation to individual differences in behavior and psychopathology. However, recent research using adult samples has questioned the trait stability of task-fMRI measures, as assessed by test-retest correlations. To date, little is known about trait stability of task fMRI in children. Here, we examined within-session reliability and long-term stability of individual differences in task-fMRI measures using fMRI measures of brain activation provided by the adolescent brain cognitive development (ABCD) Study Release v4.0 as an individual's average regional activity, using its tasks focused on reward processing, response inhibition, and working memory. We also evaluated the effects of factors potentially affecting reliability and stability. Reliability and stability (quantified as the ratio of non-scanner related stable variance to all variances) was poor in virtually all brain regions, with an average value of 0.088 and 0.072 for short term (within-session) reliability and long-term (between-session) stability, respectively, in regions of interest (ROIs) historically-recruited by the tasks. Only one reliability or stability value in ROIs exceeded the 'poor' cut-off of 0.4, and in fact rarely exceeded 0.2 (only 4.9%). Motion had a pronounced effect on estimated reliability/stability, with the lowest motion quartile of participants having a mean reliability/stability 2.5 times higher (albeit still 'poor') than the highest motion quartile. Poor reliability and stability of task-fMRI, particularly in children, diminishes potential utility of fMRI data due to a drastic reduction of effect sizes and, consequently, statistical power for the detection of brain-behavior associations. This essential issue urgently needs to be addressed through optimization of task design, scanning parameters, data acquisition protocols, preprocessing pipelines, and data denoising methods.

Abstract

Temporal stability of individual differences is an important prerequisite for accurate tracking of prospective relationships between neurocognition and real-world behavioral outcomes such as substance abuse and psychopathology. Here we report age-related changes and longitudinal test-retest stability (TRS) for the Neurocognition battery of the Adolescent Brain and Cognitive Development (ABCD) study, which included the NIH Toolbox (TB) Cognitive Domain and additional memory and visuospatial processing tests administered at baseline (ages 9-11) and two-year follow-up. As expected, performance improved significantly with age, but the effect size varied broadly, with Pattern Comparison and the Crystallized Cognition Composite showing the largest age-related gain (Cohen's d:.99 and.97, respectively). TRS ranged from fair (Flanker test: r = 0.44) to excellent (Crystallized Cognition Composite: r = 0.82). A comparison of longitudinal changes and cross-sectional age-related differences within baseline and follow-up assessments suggested that, for some measures, longitudinal changes may be confounded by practice effects and differences in task stimuli or procedure between baseline and follow-up. In conclusion, a subset of measures showed good stability of individual differences despite significant age-related changes, warranting their use as prospective predictors. However, caution is needed in the interpretation of observed longitudinal changes as indicators of neurocognitive development.

Abstract

Gene expression profiling and proteome analysis of normal and malignant hematopoietic stem cells (HSCs) point to shared core stemness properties. However, discordance between mRNA and protein signatures highlights an important role for post-transcriptional regulation by microRNAs (miRNAs) in governing this critical nexus. Here, we identify miR-130a as a regulator of HSC self-renewal and differentiation. Enforced expression of miR-130a impairs B lymphoid differentiation and expands long-term HSCs. Integration of protein mass spectrometry and chimeric AGO2 crosslinking and immunoprecipitation (CLIP) identifies TBL1XR1 as a primary miR-130a target, whose loss of function phenocopies miR-130a overexpression. Moreover, we report that miR-130a is highly expressed in t(8;21) acute myeloid leukemia (AML), where it is critical for maintaining the oncogenic molecular program mediated by the AML1-ETO complex. Our study establishes that identification of the comprehensive miRNA targetome within primary cells enables discovery of genes and molecular networks underpinning stemness properties of normal and leukemic cells.
