Abstract

In the present study, the possible anthelminitic effects of different extracts (petroleumether, chloroform, and ethanol) and two isolated compounds β-stigmasterol (PI) andSitosterol (PII) from the leaves of Memecylon umbellatum on Indian earthwormsPheretima Posthuma was investigated by in vitro experiments. Various concentrations(25, 50, 100mg/ml) of all extracts were tested and results were expressed in terms oftime for paralysis and time for death of worms. From the activity results it was found thatthe ethanolic extract and pure compound PI having the equipotent activity with standarddrug albendazole. Form the observations made higher concentration of extract producedparalytic effect much earlier and the time to death was shorter for all worms.

Abstract

Five species of Memecylon were investigated for proximate analysis and elemental composition. These species include M. umbellatum M. malabaricum, M. talbotianum, M. wightii and M. lushingtonii. Among the proximates M. wightii showed highest percentage of ash, crude fiber and carbohydrate. M. umbellatum showed high percentage of moisture and crude fat. Crude protein was more in M. malabaricum Nutritive value was highest in M. lushingtonii among all the species. Among the macronutrients Na and K, Ca and Mg P and N were highest in M. malabaricum, M. talbotianum and M. umbellatum respectively. M. umbellatum recorded highest values of Cu and Mn where as M. lushingtonii and M. wightii recorded highest values of Zn and Fe. Among the heavy metals Pb and Cd M. umbellatum recorded highest values of Pb and M. talbotianum recorded highest values of Cd but both the heavy metals are absent in M. malabaricum

Abstract

NRAGE (also known as Maged1, Dlxin) is a member of the MAGE gene family that may play a role in the neuronal apoptosis that is regulated by the p75 neurotrophin receptor (p75NTR). To test this hypothesis in vivo, we generated NRAGE knockout mice and found that NRAGE deletion caused a defect in developmental apoptosis of sympathetic neurons of the superior cervical ganglia, similar to that observed in p75NTR knockout mice. Primary sympathetic neurons derived from NRAGE knockout mice were resistant to apoptosis induced by brain-derived neurotrophic factor (BDNF), a pro-apoptotic p75NTR ligand, and NRAGE-deficient sympathetic neurons show attenuated BDNF-dependent JNK activation. Hair follicle catagen is an apoptosis-like process that is dependent on p75NTR signaling; we show that NRAGE and p75NTR show regulated co-expression in the hair follicle and that identical defects in hair follicle catagen are present in NRAGE and p75NTR knockout mice. Interestingly, NRAGE knockout mice have severe defects in motoneuron apoptosis that are not observed in p75NTR knockout animals, raising the possibility that NRAGE may facilitate apoptosis induced by receptors other than p75NTR. 27. MT-125 Inhibits Non-Muscle Myosin IIA and IIB, Synergizes with Oncogenic Kinase Inhibitors, and Prolongs Survival in Glioblastoma.

Abstract

We have identified a NMIIA and IIB-specific small molecule inhibitor, MT-125, and have studied its effects in GBM. MT-125 has high brain penetrance and retention and an excellent safety profile; blocks GBM invasion and cytokinesis, consistent with the known roles of NMII; and prolongs survival as a single agent in murine GBM models. MT-125 increases signaling along both the PDGFR- and MAPK-driven pathways through a mechanism that involves the upregulation of reactive oxygen species, and it synergizes with FDA-approved PDGFR and mTOR inhibitors in vitro . Combining MT-125 with sunitinib, a PDGFR inhibitor, or paxalisib, a combined PI3 Kinase/mTOR inhibitor significantly improves survival in orthotopic GBM models over either drug alone, and in the case of sunitinib, markedly prolongs survival in ∼40% of mice. Our results provide a powerful rationale for developing NMII targeting strategies to treat cancer and demonstrate that MT-125 has strong clinical potential for the treatment of GBM., Highlights: MT-125 is a highly specific small molecule inhibitor of non-muscle myosin IIA and IIB, is well-tolerated, and achieves therapeutic concentrations in the brain with systemic dosing.Treating preclinical models of glioblastoma with MT-125 produces durable improvements in survival.MT-125 stimulates PDGFR- and MAPK-driven signaling in glioblastoma and increases dependency on these pathways.Combining MT-125 with an FDA-approved PDGFR inhibitor in a mouse GBM model synergizes to improve median survival over either drug alone, and produces tumor free, prolonged survival in over 40% of mice.

Abstract

Cortical vein thrombosis (CVT) is rare and is most common in the third decade of life. Cerebral venous thrombosis may be due to a variety of pathologic conditions like deficiencies of protein S (PS), antithrombin III, protein C, factor V Leiden, prothrombin gene mutations and hyperhomocysteinemia. Protein S is a vitamin K-dependent anticoagulant present in plasma and prevent thrombosis in association with protein C. Lack of it results in venous thromboembolism (VTE) rarely causing thrombosis of cerebral venous sinuses. Our patient is a 35-year-old male who presented with focal seizures. MRI brain showed venous infarcts, and MR venogram showed extensive thrombosis of superior sagittal sinus. Later work up for hypercoagulable state showed significant Protein S deficiency.
