O Workshop da Pós-Graduação é um evento anual e tem como objetivo discutir temas pertinentes aos Pós-Graduandos, Docentes e Administração do Instituto deBiociências de Botucatu, promovendo o amadurecimento político-científico, análise do mercado atual para os egressos, aperfeiçoamento e conhecimento científico. Busca-se apresentar experiências externas e mostrar possibilidades diferentes da carreira acadêmica.Na oportunidade, facilita-se a formação de novas redes de trabalho e cooperação. Os contatos e experiências vivenciados durante o evento são de plena importância para vida profissional dos participantes. A organização do evento inclui a participação de docentes e de discentes que elaboraram a programação e participaram ativamente da revisão dos trabalhos aqui apresentados.A oitava edição do evento ocorreu no Instituto de Biociências durante os dias 23, 24 e 25 de abril no Instituto de Biociências de Botucatu e contou com a participação de 200 inscritos.A natureza do evento é interdisciplinar pois congrega os diferentes programas de pós-graduação do Instituto de Biociências de Botucatu: Farmacologia, Botânica, Genética, Zoologia, Biologia Geral e Aplicada e Biometria. O programa inclui mini-cursos ministrados pelos pós-graduandos tratando de temas atuais de pesquisa, estes eventos incluem além de exposições orais e oficinas, a produção de material de apoio que compõe este volume.Entre os temas tratados neste volume destacamos a biodiversidade, toxicologia aplicada a reprodução humana, biodiversidade, restauração ecológica, biologia floral, embriologia e farmacocinética aplicada a testes clínicos. A diversidade dos tópicos tratados ilustra a riqueza das atividades de pesquisa realizada no Instituto de Biociências de Botucatu.

This study evaluated the effects of peri-pubertal cisplatin administration on several reproductive end-points and the reversibility of these effects in adulthood. Peri-pubertal Wistar male rats (45 days old) were divided into two groups: control (saline 0.9%) and cisplatin (1 mg/kg/day, 5 days/week, for 3 weeks, i.p.). The study was conducted in two steps and evaluations were performed at ages of 66 (post-pubertal age) and 140 (adult age) days on: (i) organ weights, serum gonadotropins and testosterone levels, sperm counts, motility and morphology, testicular histomorphometry, spermatogenesis kinetics, Sertoli cell number and in situ detection of apoptotic germ cells and (ii) sexual behaviour, fertility and intratesticular testosterone. At the end of cisplatin therapy, rats showed reductions in sperm production and reserves, sperm with progressive movement, tubular diameter, intratesticular testosterone and fertility potential, but increased numbers of TUNEL-positive seminiferous tubules, immotile sperm and pre-implantation losses compared with control. Moreover, cisplatin-treated post-pubertal rats displayed impaired testicular histopathology and sexual behaviour. Serum gonadotropins and testosterone levels, sperm morphology, spermatogenesis kinetics and Sertoli cell number were comparable between experimental groups at both ages. Alterations found in post-puberty were recovered at adulthood, except for sperm motility and damage to testicular histology. The persistence of these cisplatin effects, despite the unaltered fertility after natural mating in rats, may have implications for reproductive function of young boys undergoing cancer therapy, given the lower reproductive efficiency in human beings compared with rats. This work employed pregnant rats treated with Solanum lycocarpum unripe fruits (10% in diet) from gestation day (GD) 06 to post-natal day (PND) 07, for the evaluation of the sperm number, daily sperm production and epididymal sperm transit time of the male offspring at PND 60 and PND 90. No differences were observed in the daily sperm production (DSP) and sperm number in the testis of the exposed males at PND 60 and PND 90. Also, no alterations were observed in sperm transit time in the caput epididymis of the exposed males at PND 60 and PND 90. However, a reduced sperm transit time was observed in the corpus/cauda epididymis of the experimental males at PND 90. The last data may explain the reduced sperm number observed in the corpus/cauda epididymis of the experimental male rats at PND 90. These data show that the male rats exposed to S. lycocarpum fruits during gestation did not present alterations in testis sperm production and number, however the sperm transit time through epididymis was impaired, resulting in a decreased number of spermatozoa in epididymis cauda. We conclude that S. lycocarpum may cause imbalance on hypothalamus-pituitary gland axis Ratas prenhes foram tratadas do dia 06 da gestação (GD 06) ao dia 07 pós-natal (PND 07) com frutos verdes secos e moídos da Solanum lycocarpum (10% na ração). Após nascimento das ninhadas, foi avaliado na prole masculina adulta aos 60 e 90 dias de vida, o número de espermátides e a produção espermática diária nos testículos e o tempo de trânsito espermático no epidídimo. A exposição não foi capaz de promover alterações na produção espermática diária (DSP) e no número de espermátides produzidas pelo testículo dos ratos expostos aos frutos verdes da S. lycocarpum durante a gestação e início da lactação. Não foram observadas alterações no tempo de trânsito espermático na cabeça do epidídimo, porém, foi constatado menor número de espermatozóides no corpo/cauda do epidídimo nos machos experimentais com 90 dias de vida, provavelmente resultante do menor tempo de trânsito espermático observado no corpo/cauda do epidídimo aos PND 90. Estes dados sugerem que a exposição de ratos aos frutos verdes da S. lycocarpum durante a gestação e início da lactação, não foi suficiente para promover alterações na produção mas sim no trânsito espermático, indicando possível alteração no eixo hormônio liberador das gonadotrofinas hipotálamo-hipófise-gônada

No. of bitstreams: 0 Previous issue date: 2002-01-01 Gossypol, a yellow pigment found in cottonseeds, well known for its antifertility properties in animals, has been used as a contraceptive by men. The aims of this work were to evaluate the effects of gossypol throughout sexual development of male rats and to provide additional data to clarify the target site or sites of this compound in the male reproductive system. Gossypol (15 mg/kg per day) was given to animals from weaning through prepuberty (41 days), early puberty (51 days), puberty (61 days), and sexual maturity (91 days). Ventral prostate weight and fructose levels were similar in control and treated rats, suggesting that androgen levels were normal. No histological effects on the testis were detected, but there was a significant decrease in the sperm concentration in the cauda epididymidis of gossypol-treated animals killed at 61 and 91 days, as well as a significant increase in abnormal sperm in the vas deferens of treated animals. Moreover, the histology of the cauda epididymidis of the rats treated throughout puberty (ie, until days 51 and 61) showed a great number of round bodies in the lumen of the epididymis. These structures stained for the epididymis-specific protein E. Collectively, the data demonstrate that the epididymis is a target of gossypol when postweaning exposure extends throughout pubertal development, and that whereas more subtle histological effects commence around puberty, indicators reproductive competence are compromised in adulthood.
