2. Proposing a Continuous Professional Development Model to Support and Enhance Professional Learning of Teachers in Special Schools in South Africa

Abstract

A collaborative continuous professional (CPD) model was implemented and evaluated in six special schools in Gauteng, South Africa in order to support teachers in their professional capacity. The study which reports on a two year study aimed to establish the value of the CPD programme on teachers' learning, students' outcomes and whole school change. The focus during the study was to adapt learner activities and teacher support materials for the Learn Not to Burn (LNTB) fire safety programme for students with special needs. A qualitative, multi-phased case study was employed to understand teachers' views of the collaborative continuous professional development programme, which culminated in the development of Dynamic Collaborative Networking model. This study relied on current CPD models in order to develop this model which was suitable for special education. This research serves as an example of where valuable internal and external networks were formed for the benefit of all involved with the study.

Abstract

Het Groene Hart en andere veengebieden kampen al jaren met een zakkende bodem door zetting en het inklinken van veengrond, door ontwatering en droogte. Bij de landelijke overheid werd onvoldoende aandacht voor de effecten van bodemdaling ervaren. Dit terwijl bodemdaling een forse impact heeft, vanwege de CO 2 uitstoot vanuit het landelijk veengebied, en door de kosten die met name in het stedelijk gebied in de miljarden lopen. Binnen de regio werd de noodzaak gevoeld om tot vernieuwende invalshoeken te komen voor onder meer het wegenonderhoud, de agrarische sector en de fundering van woningen. Daarom dienden acht regionale overheidsorganisaties in 2018 gezamenlijk een aanvraag in bij het Rijk voor de Regio Deal bodemdaling Groene Hart. Want de aanpak van bodemdaling vraagt om samenwerking. De aanvraag werd gehonoreerd en regionale overheden, kennisinstellingen, agrarische sector, bewoners en bedrijfsleven ontwikkelden samen in de periode 2019 -2023 innovatieve ideeën en experimenten om tot een andere aanpak en nieuwe kennis te komen voor het omgaan met bodemdaling. Het resulteerde in meer dan 25 gerealiseerde projecten, verdeeld naar de rubrieken: stad, platteland en kennis. Deze staan beschreven in dit magazine.

Abstract

Aims Patients with cardiac disease are considered high risk for poor outcomes following hospitalization with COVID-19. The primary aim of this study was to evaluate heterogeneity in associations between various heart disease subtypes and in-hospital mortality. Methods and results We used data from the CAPACITY-COVID registry and LEOSS study. Multivariable Poisson regression models were fitted to assess the association between different types of pre-existing heart disease and in-hospital mortality. A total of 16 511 patients with COVID-19 were included (21.1% aged 66-75 years; 40.2% female) and 31.5% had a history of heart disease. Patients with heart disease were older, predominantly male, and often had other comorbid conditions when compared with those without. Mortality was higher in patients with cardiac disease (29.7%; n= 1545 vs. 15.9%; n= 1797). However, following multivariable adjustment, this difference was not significant [adjusted risk ratio (aRR) 1.08, 95% confidence interval (CI) 1.02-1.15; P = 0.12 (corrected for multiple testing)]. Associations with in-hospital mortality by heart disease subtypes differed considerably, with the strongest association for heart failure (aRR 1.19, 95% CI 1.10-1.30; P <0.018) particularly for severe (New York Heart Association class III/IV) heart failure (aRR 1.41, 95% CI 1.20-1.64; P < 0.018). None of the other heart disease subtypes, including ischaemic heart disease, remained significant after multivariable adjustment. Serious cardiac complications were diagnosed in <1% of patients. Conclusion Considerable heterogeneity exists in the strength of association between heart disease subtypes and in-hospital mortality. Of all patients with heart disease, those with heart failure are at greatest risk of death when hospitalized with COVID-19. Serious cardiac complications are rare during hospitalization. Abstract

Purpose: Pathogenic variants in GABRB3 have been associated with a spectrum of phenotypes from severe developmental disorders and epileptic encephalopathies to milder epilepsy syndromes and mild intellectual disability (ID). In this study, we analyzed a large cohort of individuals with GABRB3 variants to deepen the phenotypic understanding and investigate genotype–phenotype correlations. Methods: Through an international collaboration, we analyzed electro-clinical data of unpublished individuals with variants in GABRB3, and we reviewed previously published cases. All missense variants were mapped onto the 3-dimensional structure of the GABRB3 subunit, and clinical phenotypes associated with the different key structural domains were investigated. Results: We characterized 71 individuals with GABRB3 variants, including 22 novel subjects, expressing a wide spectrum of phenotypes. Interestingly, phenotypes correlated with structural locations of the variants. Generalized epilepsy, with a median age at onset of 12 months, and mild-to-moderate ID were associated with variants in the extracellular domain. Focal epilepsy with earlier onset (median: age 4 months) and severe ID were associated with variants in both the pore-lining helical transmembrane domain and the extracellular domain. Conclusion: These genotype–phenotype correlations will aid the genetic counseling and treatment of individuals affected by GABRB3-related disorders. Future studies may reveal whether functional differences underlie the phenotypic differences. © 2021 American College of Medical Genetics and Genomics

Abstract

Esophageal atresia (EA) and tracheoesophageal fistula (TEF) are relatively frequently occurring foregut malformations. EA/TEF is thought to have a strong genetic component. Not much is known regarding the biological processes disturbed or which cell type is affected in patients. This hampers the detection of the responsible culprits (genetic or environmental) for the origin of these congenital anatomical malformations. Therefore, we examined gene expression patterns in the TEF and compared them to the patterns in esophageal, tracheal and lung control samples. We studied tissue organization and key proteins using immunohistochemistry. There were clear differences between TEF and control samples. Based on the number of differentially expressed genes as well as histological characteristics, TEFs were most similar to normal esophagus. The BMP-signaling pathway, actin cytoskeleton and extracellular matrix pathways are downregulated in TEF. Genes involved in smooth muscle contraction are overexpressed in TEF compared to esophagus as well as trachea. These enriched pathways indicate myofibroblast activated fibrosis. TEF represents a specific tissue type with large contributions of intestinal smooth muscle cells and neurons. Abstract

Voor het opstellen van een gezamenlijke strategische agenda voor de Utrechtse jeugd is de methode van group concept mapping gebruikt. Dit is een beslissingsondersteunende methode voor planning en evaluatie bij samenwerkingsprojecten rondom een complex onderwerp. De methode van group concept mapping verloopt gestructureerd in zes vaste stappen: 1) bepalen brainstormfocus en deelnemers, 2) genereren opvattingen, 3) structureren opvattingen, 4) grafische representatie, 5) interpretatie, en 6) implementatie. De concept map geeft een overzicht van de thema’s, opgaven en kansen in hun onderlinge samenhang voor de zorg voor jeugd in Utrecht als het gaat om gezond, veilig en kansrijk opgroeien en opvoeden. Daarmee biedt de methode een basis om met elkaar tot een herijking van de huidige strategische agenda van de AWTJ Utrecht te komen.

Abstract

Sox2 is a Sry-box containing family member of related transcription factors sharing homology in their DNA binding domain. Sox2 is important during different stages of development, and previously we showed that Sox2 plays an important role in branching morphogenesis and epithelial cell differentiation in lung development. The transcriptional activity of Sox2 depends on its interaction with other proteins, leading to ‘complex-specific’ DNA binding and transcriptional regulation. In this study, we generated a mouse model containing a biotinylatable-tag targeted at the translational start site of the endogenous Sox2 gene (bioSox2). This tag was biotinylated by the bacterial birA protein and the resulting bioSox2 protein was used to identify associating partners of Sox2 at different phases of lung development in vivo (the Sox2 interactome). Homozygous bioSox2 mice are viable and fertile irrespective of the biotinylation of the bio tag, indicating that the bioSox2 gene is normally expressed and the protein is functional in all tissues. This suggests that partners of Sox2 are most likely able to associate with the bioSox2 protein. BioSox2 complexes were isolated with high affinity using streptavidin beads and analysed by MALDI-ToF mass spectrometry analysis. Several of the identified binding partners are already shown to have a respiratory phenotype. Two of these partners, Wdr5 and Tcf3, were validated to confirm their association in Sox2 complexes. This bioSox2 mouse model will be a valuable tool for isolating in vivo Sox2 complexes from different tissues.

De voorzitter van het Klimaatberaad heeft het CPB verzocht om, in samenspraak met het Planbureau voor de Leefomgeving (PBL), een beoordeling te geven van maatregelen op lasten voor burgers, bedrijven en de overheid, budgettaire effecten en inkomenseffecten. In het verzoek geeft de voorzitter aan dat “mogelijk niet van alle onderdelen een beoordeling op voornoemde drie onderdelen is te maken”, omdat het VHK nog niet op alle onderdelen concreet is uitgewerkt.

Item does not contain fulltext Most patients with Dravet syndrome have de novo mutations in the neuronal voltage-gated sodium channel type 1 (SCN1A) gene. We report on two unrelated fathers with severe childhood epilepsy compatible with a possible diagnosis of Dravet syndrome, who both have a child with Dravet syndrome. Analysis of the SCN1A gene revealed a pathogenic mutation in both children. One father exhibited somatic mosaicism for the mutation detected in his son. A relatively favorable cognitive outcome in patients with Dravet syndrome patients may be explained by somatic mosaicism for the SCN1A mutation in brain tissue. A mild form of Dravet syndrome in adult patients is associated with a high recurrence risk and possibly a more severe epilepsy phenotype in their offspring.

The EU cross-compliance (CC) instrument implies that farmers receive payments subject to meeting Statutory Management Requirements (SMRs) related to environment, food safety, animal and plant health and animal welfare, as well as standards of good agricultural and environmental conditions (GAECs). This report describes the approach and application methodology of an environmental impact tool to evaluate cross compliance measures on environmental effect indicators. It consists of a selection of the effect indicators to be applied and assessment tools to be used with a review of the CC measures to be evaluated in assessing the environmental impacts. Furthermore, it includes an evaluation of CC measures related to the Nitrates Directive and selected GAECs with the coupled CAPRI-MITERRA model for the prototype of the environmental impact tool.

Abstract

Na zijn eerste bundel, 'Yanacuna' in 1962, heeft de Surinaamse dichter Ashetu (ps. van H.G. van Ommeren, 1929-1982) niets meer zelf gepubliceerd. Kenner van de Surinaamse literatuur Michiel van Kempen heeft nu een bloemlezing samengesteld van honderd gedichten. De samensteller heeft zich als een pionier een weg moeten banen in de nalatenschap van meer dan duizend ongepubliceerde gedichten. Ashetu ontwikkelde zich als scheepsmarconist op de grote vaart tot schrijver. Zijn taalgebruik blijft gecodeerd, er valt geen eenduidige betekenis te ontlenen aan zijn woorden. Rusteloos verlangen blijkt ook een halve eeuw aan wal (in Den Haag) niet te stelpen - ' Moeder waar ben je / want ik kantelde zo / op de trap / Er is iets gebroken maar / ik weet niet wat.' De striemen van slavernij blijven branden op zijn afkomst, het uitzicht buiten zijn meeuwen. Een bundel waarin het Surinaamse gemoed geen voltooide tijd kent. Met een substantieel informatief nawoord.

Item does not contain fulltext, Lung development is determined by the coordinated expression of several key genes. Previously, we and others have shown the importance of the sex determining region Y-box 2 (Sox2) gene in lung development. Transgenic expression of Sox2 during lung development resulted in cystic airways, and here we show that modulating the timing of ectopic Sox2 expression in the branching regions of the developing lung results in variable cystic lesions resembling the spectrum of the human congenital disorder congenital cystic adenomatoid malformation (CCAM). Sox2 dominantly differentiated naive epithelial cells into the proximal lineage irrespective of the presence of Fgf10. Sox2 directly induced the expression of Trp63, the master switch toward the basal cell lineage and induced the expression of Gata6, a factor involved in the emergence of bronchoalveolar stem cells. We showed that SOX2 and TRP63 are coexpressed in the lungs of human patients with type II CCAM. The combination of premature differentiation toward the proximal cell lineage and the induction of proliferation resulted in the cyst-like structures. Thus, we show that Sox2 is directly responsible for the emergence of two lung progenitor cells: basal cells by regulating the master gene Trp63 and bronchoalveolar stem cells by regulating Gata6.

Abstract

The adult lung contains several distinct stem cells, although their properties and full potential are still being sorted out. We previously showed that ectopic Sox2 expression in the developing lung manipulated the fate of differentiating cells. Here, we addressed the question whether fully differentiated cells could be redirected towards another cell type. Therefore, we used transgenic mice to express an inducible Sox2 construct in type II pneumocytes, which are situated in the distal, respiratory areas of the lung. Within three days after the induction of the transgene, the type II cells start to proliferate and form clusters of cuboidal cells. Prolonged Sox2 expression resulted in the reversal of the type II cell towards a more embryonic, precursor-like cell, being positive for the stem cell markers Sca1 and Ssea1. Moreover, the cells started to co-express Spc and Cc10, characteristics of bronchioalveolar stem cells. We demonstrated that Sox2 directly regulates the expression of Sca1. Subsequently, these cells expressed Trp63, a marker for basal cells of the trachea. So, we show that the expression of one transcription factor in fully differentiated, distal lung cells changes their fate towards proximal cells through intermediate cell types. This may have implications for regenerative medicine, and repair of diseased and damaged lungs.
