International audience; Ceres, the most water-rich body in the inner solar system after Earth, has recently been recognized to have astrobiological importance. Chemical and physical measurements obtained by the Dawn mission enabled the quantification of key parameters, which helped to constrain the habitability of the inner solar system's only dwarf planet. The surface chemistry and internal structure of Ceres testify to a protracted history of reactions between liquid water, rock, and likely organic compounds. We review the clues on chemical composition, temperature, and prospects for long-term occurrence of liquid and chemical gradients. Comparisons with giant planet satellites indicate similarities both from a chemical evolution standpoint and in the physical mechanisms driving Ceres' internal evolution.

Frogs are an ecologically diverse and phylogenetically ancient group of living amphibians that include important vertebrate cell and developmental model systems, notably the genus Xenopus. Here we report a high-quality reference genome sequence for the western clawed frog, Xenopus tropicalis, along with draft chromosome-scale sequences of three distantly related emerging model frog species, Eleutherodactylus coqui, Engystomops pustulosus and Hymenochirus boettgeri. Frog chromosomes have remained remarkably stable since the Mesozoic Era, with limited Robertsonian (i.e., centric) translocations and end-to-end fusions found among the smaller chromosomes. Conservation of synteny includes conservation of centromere locations, marked by centromeric tandem repeats associated with Cenp-a binding, surrounded by pericentromeric LINE/L1 elements. We explored chromosome structure across frogs, using a dense meiotic linkage map for X. tropicalis and chromatin conformation capture (HiC) data for all species. Abundant satellite repeats occupy the unusually long (∼20 megabase) terminal regions of each chromosome that coincide with high rates of recombination. Both embryonic and differentiated cells show reproducible association of centromeric chromatin, and of telomeres, reflecting a Rabl configuration similar to the “bouquet” structure of meiotic cells. Our comparative analyses reveal 13 conserved ancestral anuran chromosomes from which contemporary frog genomes were constructed.

We describe the AMBITION project, a mission to return the first-ever cryogenically-stored sample of a cometary nucleus, that has been proposed for the ESA Science Programme Voyage 2050. Comets are the leftover building blocks of giant planet cores and other planetary bodies, and fingerprints of Solar System’s formation processes. We summarise some of the most important questions still open in cometary science and Solar System formation after the successful Rosetta mission. We show that many of these scientific questions require sample analysis using techniques that are only possible in laboratories on Earth. We summarize measurements, instrumentation and mission scenarios that can address these questions. We emphasize the need for returning a sample collected at depth or, still more challenging, at cryogenic temperatures while preserving the stratigraphy of the comet nucleus surface layers. We provide requirements for the next generation of landers, for cryogenic sample acquisition and storage during the return to Earth. Rendezvous missions to the main belt comets and Centaurs, expanding our knowledge by exploring new classes of comets, are also discussed. The AMBITION project is discussed in the international context of comet and asteroid space exploration.

Centaurs, minor planets with a semi-major axis between the orbits of Jupiter and Neptune (5–30 AU), are thought to be among the most diverse small bodies in the solar system. These important targets for future missions may have recently been Kuiper Belt Objects (KBOs), which are thought to be chemically and physically primitive remnants of the early solar system. While the Kuiper Belt spans distances of 30–50 AU, making direct observations difficult, Centaurs' proximity to the Earth and Sun make them more accessible targets for robotic missions. Thus, we outline a mission concept designed to reconnoiter 10199 Chariklo, the largest Centaur and smallest ringed body yet discovered. Named for a legendary Centaur tamer, the conceptual Camilla mission is designed to fit under the cost cap of the National Aeronautics and Space Administration (NASA) New Frontiers program, leveraging a conservative payload to support a foundational scientific investigation to these primitive bodies. Specifically, the single flyby encounter utilizes a combined high-resolution camera/VIS-IR mapping spectrometer, a sub-mm point spectrometer, and a UV mapping spectrometer. In addition, the mission concept utilizes a kinetic impactor, which would provide the first opportunity to sample the composition of potentially primitive subsurface material beyond Saturn, thus providing key insights into solar system origins. Such a flyby of the Chariklo system would provide a linchpin in the understanding of small body composition, evolution, and transport of materials in the solar system.

The innovative Saturn Ring Skimmer mission concept enables a wide range of investigations that address fundamental questions about Saturn and its rings, as well as giant planets and astrophysical disk systems in general. This mission would provide new insights into the dynamical processes that operate in astrophysical disk systems by observing individual particles in Saturn's rings for the first time. The Ring Skimmer would also constrain the origin, history, and fate of Saturn's rings by determining their compositional evolution and material transport rates. In addition, the Ring Skimmer would reveal how the rings, magnetosphere, and planet operate as an inter-connected system by making direct measurements of the ring's atmosphere, Saturn's inner magnetosphere and the material owing from the rings into the planet. At the same time, this mission would clarify the dynamical processes operating in the planet's visible atmosphere and deep interior by making extensive high-resolution observations of cloud features and repeated measurements of the planet's extremely dynamic gravitational field. Given the scientific potential of this basic mission concept, we advocate that it be studied in depth as a potential option for the New Frontiers program., Comment: White paper submitted to the Planetary Science and Astrobiology Decadal Survey (submission #420)

We outline specific steps that NASA and the space science community can take to advance collaboration and coordination between the communities represented by the four NASA Science Mission Directorate Divisions. It is important to note that the only way that this effort can succeed is if NASA initiates and supports it through directed resources.

International audience; Planetary rings are a multifaceted player in the system. Recent advances about Saturn’s rings argue that ring science at the ice giants, with magnetospheric and atmospheric sciences, are essential in advancing our knowledge about solar system evolution, the evolution of the moons and Ocean Worlds, and phenomena observed in the ice giant systems.

Context.Carbonaceous chondrites have undergone alteration in their parent bodies and display oxidized secondary phases, including sulfates in CI and CM chondrites. The cause of the formation of these sulfates is yet to be determined.Aims.This study investigates the potential of endogenous radiolysis of water (i.e., radiolysis caused by radionuclides present in the rock) on the parent bodies of carbonaceous chondrites. Radiolysis may have contributed to the enhanced degree of oxidation of CI and CM chondrites, and we also examined CV chondrites as a case with no measured sulfates.Methods.We quantified the oxidants produced by radiolysis and how much of the sulfur content could be oxidized to form sulfates by this method. The amount of oxidants was calculated using a radiolytic production model developed and used for Earth and planetary applications that takes into account relevant physical parameters (water-to-rock ratio, grain density) and composition (amount of radionuclides, sulfur content).Results.For CM and CI parent bodies, even using a very favorable set of assumptions, only slightly more than 1% of the available sulfur can be oxidized into sulfates by this process, significantly below the amount of sulfates observed in these chondrites.Conclusions.Endogenous radiolysis is unlikely to have significantly contributed to the abundance of sulfate in CI and CM meteorites. The hypothesis of oxidation of sulfur by large quantities of O2accreted with primitive ice, on the other hand, is quantitatively supported by measurements of O2in comet 67P/Churyumov-Gerasimenko.

Size is a fundamental feature of biology that affects physiology at all levels, from the organism to organs and tissues to cells and subcellular structures. How size is determined at these different levels, and how biological structures scale to fit together and function properly are important open questions. Historically, amphibian systems have been extremely valuable to describe scaling phenomena, as they occupy some of the extremes in biological size and are amenable to manipulations that alter genome and cell size. More recently, the application of biochemical, biophysical, and embryological techniques to amphibians has provided insight into the molecular mechanisms underlying scaling of subcellular structures to cell size, as well as how perturbation of normal size scaling impacts other aspects of cell and organism physiology.

International audience; During Cassini's Grand Finale, the spacecraft flew between Saturn's upper atmosphere and its innermost ring, the D ring. Throughout these final orbits, Cassini encountered material flowing from the D ring into Saturn's atmosphere. Here, we present a compositional analysis of this material using data collected by Cassini's Ion Neutral Mass Spectrometer. We confirm the identification of CH4, CO2, CO, N2, H2O, NH3, and organics in the ring material, and provide upper limits for organic compounds of interest. The H/C, O/C, N/C, and S/C ratios of ring material are constrained using three different kinds of model spectra: automated fits, hand fits of inorganics + hydrocarbons, and hand fits of inorganics + organics with priority given to O-, N-, and S-bearing organics. Additionally, we compare data from the final plunge to earlier orbits, and find that ring material can be divided into gas and dust constituents, with CO2 at the volatility boundary between the two phases. At increasing distance above the equatorial plane, the gas/dust molar ratio increases.

We present observations of five stellar occultations for (11351) Leucus and reports from two efforts on (21900) Orus. Both objects are prime mission candidate targets for the Lucy Discovery mission. Combined results for Leucus indicate a very dark surface with p V = 0.037 ± 0.001, which is derived from the average of the multichord occultations. Our estimate of the triaxial ellipsoidal shape is for axial diameters of 63.8 × 36.6 × 29.6 km assuming that the spin pole is normal to the line of sight. The actual shape of the object is only roughly elliptical in profile at each epoch. Significant topography is seen with horizontal scales up to 30 km and vertical scales up to 5 km. The most significant feature is a large depression on the southern end of the object as seen from a terrestrial viewpoint. For this work we developed a method to correct for differential refraction, accounting for the difference in color between the target object and the reference stars for astrometry derived from ground-based images.

Sulfide assemblages are commonly found in chondritic meteorites as small inclusions in the matrix or in association with chondrules. These assemblages are widely hypothesized to form through pre-accretionary corrosion of metal by H2S gas or through parent body processes. We report here on two unequilibrated R chondrite samples that contain large, chondrule-sized sulfide nodules in the matrix. Both samples are from Mount Prestrud (PRE) 95404. Chemical maps and spot and broad-beam electron microprobe analyses (EMPA) were used to assess the distribution, stoichiometry, and bulk composition of sulfide nodules and silicate chondrules in the clasts. Oxygen isotope data were collected via secondary ion mass spectrometry (SIMS) to assess the relationship of chondrules to other chondrite groups. Scanning electron microscopy (SEM), focused ion beam (FIB), and transmission electron microscopy (TEM) analyses were used to assess fine-scale features and identify crystal structures in sulfide assemblages. Thermodynamic models were used to assess the temperature, sulfur fugacity (fS2), total pressure, dust-to-gas ratio, and oxygen fugacity (fO2) conditions during sulfide nodule and chondrule formation. The unequilibrated clasts include a mixture of type I and type II chondrules, as well as non-porphyritic chondrules. Chondrule oxygen isotopes overlap with ordinary-chondrite chondrules. Sulfide nodules average 200 µm in diameter, have rounded shapes, and are primarily composed of pyrrhotite, pentlandite, and magnetite. Some are deformed around chondrules in a petrologic relationship similar in appearance to compound chondrules. Both nodules and sulfides in chondrules include phosphate inclusions and Cu-rich lamellae, which suggests a genetic relationship between sulfides in chondrules and in the matrix. Ni/Co ratios for matrix and chondrule sulfides are solar, while Fe and Ni are non-solar and inversely related. We hypothesize that sulfide nodules formed via pre-accretionary melt processes. During chondrule formation, precursors composed of a mixture of silicate and sulfide material were heated to form immiscible melt droplets, which separated and cooled to form Si-rich chondrules and S-rich nodules. Sulfide melt was stabilized by a high total pressure (∼1 atm) in a dust- or ice-enriched environment. Heating of this material contributed to a high fS2 (2 × 10−3 atm at 1138 °C), and high fO2 (IW – 1 to IW – 4), in an environment with peak temperatures between 1539 °C and 1750 °C. Oxygen isotopic compositions in this region were similar to those recorded by the LL-chondrite chondrules.

Hydrothermal processes on Enceladus Saturn's moon Enceladus has a subsurface ocean covered by a layer of ice. Some liquid escapes into space through cracks in the ice, which is the source of one of Saturn's rings. In October 2015, the Cassini spacecraft flew directly through the plume of escaping material and sampled its chemical composition. Waite et al. found that the plume contains molecular hydrogen, H 2 , a sign that the water in Enceladus' ocean is reacting with rocks through hydrothermal processes (see the Perspective by Seewald). This drives the ocean out of chemical equilibrium, in a similar way to water around Earth's hydrothermal vents, potentially providing a source of chemical energy. Cassini's final phase of exploration The Cassini spacecraft spent 13 years orbiting Saturn; as it ran low on fuel, the trajectory was changed to sample regions it had not yet visited. A series of orbits close to the rings was followed by a Grand Finale orbit, which took the spacecraft through the gap between Saturn and its rings before the spacecraft was destroyed when it entered the planet's upper atmosphere. Six papers in this issue report results from these final phases of the Cassini mission. Dougherty et al. measured the magnetic field close to Saturn, which implies a complex multilayer dynamo process inside the planet. Roussos et al. detected an additional radiation belt trapped within the rings, sustained by the radioactive decay of free neutrons. Lamy et al. present plasma measurements taken as Cassini flew through regions emitting kilometric radiation, connected to the planet's aurorae. Hsu et al. determined the composition of large, solid dust particles falling from the rings into the planet, whereas Mitchell et al. investigated the smaller dust nanograins and show how they interact with the planet's upper atmosphere. Finally, Waite et al. identified molecules in the infalling material and directly measured the composition of Saturn's atmosphere. Determining how size is controlled is a fundamental question in biology that is poorly understood at the organismal, cellular and subcellular levels. The Xenopus species, X. laevis and X. tropicalis differ in size at all three of these levels. Despite these differences, fertilization of X. laevis eggs with X. tropicalis sperm gives rise to viable hybrid animals that are intermediate in size. We observed that although hybrid and X. laevis embryogenesis initiates from the same sized zygote and proceeds synchronously through development, hybrid animals were smaller by the tailbud stage, and a change in the ratio of nuclear size to cell size was observed shortly after zygotic genome activation (ZGA), suggesting that differential gene expression contributes to size differences. Transcriptome analysis at the onset of ZGA identified twelve transcription factors paternally expressed in hybrids. A screen of these X. tropicalis factors by expression in X. laevis embryos revealed that Hes7 and Ventx2 significantly reduced X. laevis body length size by the tailbud stage, although nuclear to cell size scaling relationships were not affected as in the hybrid. Together, these results suggest that transcriptional regulation contributes to biological size control in Xenopus.

Exogenous delivery of amino acids and other organic molecules to planetary surfaces may have played an important role in the origins of life on Earth and other solar system bodies. Previous studies have revealed the presence of indigenous amino acids in a wide range of carbon-rich meteorites, with the abundances and structural distributions differing significantly depending on parent body mineralogy and alteration conditions. Here we report on the amino acid abundances of seven type 3–6 CK chondrites and two Rumuruti (R) chondrites. Amino acid measurements were made on hot water extracts from these meteorites by ultrahigh-performance liquid chromatography with fluorescence detection and time-of-flight mass spectrometry. Of the nine meteorites analyzed, four were depleted in amino acids, and one had experienced significant amino acid contamination by terrestrial biology. The remaining four, comprised of two R and two CK chondrites, contained low levels of amino acids that were predominantly the straight chain, amino-terminal (n-ω-amino) acids β-alanine, and γ-amino-n-butyric acid. This amino acid distribution is similar to what we reported previously for thermally altered ureilites and CV and CO chondrites, and these n-ω-amino acids appear to be indigenous to the meteorites and not the result of terrestrial contamination. The amino acids may have been formed by Fischer–Tropsch-type reactions, although this hypothesis needs further testing.

In the future, what role will the academic research library play in achieving the mission of higher education? This essay describes seven strategies that academic research libraries can adopt to become future-present libraries—libraries that foster what Douglas Thomas and John Seely Brown have called "a new culture of learning." Written in light of Johanna Drucker's vision of the next university described in her article "The University as a Fully Integrated and Distributed Platform" in this edition of portal, the author argues that, to transform successfully, academic research libraries must ask themselves: How can we cultivate the imagination of students? What are we enabling students to do and become? The essay offers examples of how one academic research library, UCLA Library, is applying these strategies within its own institutional context, while learning from other forward-thinking organizations.

Summary Size is a fundamental feature of biological systems that affects physiology at all levels. For example, the dynamic, microtubule-based spindle that mediates chromosome segregation scales to a wide range of cell sizes across different organisms and cell types. Xenopus frog species possess a variety of egg and meiotic spindle sizes, and differences in activities or levels of microtubule-associated proteins in the egg cytoplasm between Xenopus laevis and Xenopus tropicalis have been shown to account for spindle scaling [ 1 ]. Increased activity of the microtubule severing protein katanin scales the X. tropicalis spindle smaller compared to X. laevis [ 2 ], as do elevated levels of TPX2, a protein that enriches the cross-linking kinesin-5 motor Eg5 at spindle poles [ 3 ]. To examine the conservation of spindle scaling mechanisms more broadly across frog species, we have utilized the tiny, distantly related Pipid frog Hymenochirus boettgeri. We find that egg extracts from H. boettgeri form meiotic spindles similar in size to X. tropicalis but that TPX2 and katanin-mediated scaling is not conserved. Instead, the microtubule depolymerizing motor protein kif2a functions to modulate spindle size. H. boettgeri kif2a possesses an activating phosphorylation site that is absent from X. laevis. Comparison of katanin and kif2a phosphorylation sites across a variety of species revealed strong evolutionary conservation, with X. laevis and X. tropicalis possessing distinct and unique alterations. Our study highlights the diversity and complexity of spindle assembly and scaling mechanisms, indicating that there is more than one way to assemble a spindle of a particular size.

To explore the origins and consequences of tetraploidy in the African clawed frog, we sequenced the Xenopus laevis genome and compared it to the related diploid X. tropicalis genome. We characterize the allotetraploid origin of X. laevis by partitioning its genome into two homoeologous subgenomes, marked by distinct families of 'fossil' transposable elements. On the basis of the activity of these elements and the age of hundreds of unitary pseudogenes, we estimate that the two diploid progenitor species diverged around 34 million years ago (Ma) and combined to form an allotetraploid around 17-18 Ma. More than 56% of all genes were retained in two homoeologous copies. Protein function, gene expression, and the amount of conserved flanking sequence all correlate with retention rates. The subgenomes have evolved asymmetrically, with one chromosome set more often preserving the ancestral state and the other experiencing more gene loss, deletion, rearrangement, and reduced gene expression.

This is a brief sampling of Slavic visual sources in the University of Virginia Library and Art Museum. Sources available for study include printed material, such as early twentieth-century Russian art and architectural history monographs, eighteenth- and nineteenth-century histories of Russian explorations, and English-language travel literature on Russia. Rare and unique materials include an illustrated fair copy manuscript by Russian writer and illustrator Aleksei Remizov, collections of photographs of Russia taken during the Civil War and Stalinist eras, and works on paper by Slavic artists Marc Chagall, Alexander Brodsky, Ilya Utkin, Jiři Kolař, and others.

This article offers a brief discussion of two projects—the creation of a set of digital images of Russian art and an exhibit of modern Russian children's books—developed in collaborative partnership between a Slavist and librarians at the University of Virginia. Both of these projects were designed to create shareable, flexible, and sustainable resources for research and educational use. The challenges and benefits of collaborative partnerships between scholars and librarians are discussed, and models for conceptualizing the relationships are also proposed. This article is adapted from a paper presented at the annual meeting of the American Association for the Advancement of Slavic Studies in November 2006.

Nap1 is required for linker histone H1M-mediated mitotic chromosome condensation in Xenopus egg extracts, and glutamylation of Nap1 is required for proper deposition and turnover of H1M on chromatin during both interphase and mitosis., Linker histone H1 is required for mitotic chromosome architecture in Xenopus laevis egg extracts and, unlike core histones, exhibits rapid turnover on chromatin. Mechanisms regulating the recruitment, deposition, and dynamics of linker histones in mitosis are largely unknown. We found that the cytoplasmic histone chaperone nucleosome assembly protein 1 (Nap1) associates with the embryonic isoform of linker histone H1 (H1M) in egg extracts. Immunodepletion of Nap1 decreased H1M binding to mitotic chromosomes by nearly 50%, reduced H1M dynamics as measured by fluorescence recovery after photobleaching and caused chromosome decondensation similar to the effects of H1M depletion. Defects in H1M dynamics and chromosome condensation were rescued by adding back wild-type Nap1 but not a mutant lacking sites subject to posttranslational modification by glutamylation. Nap1 glutamylation increased the deposition of H1M on sperm nuclei and chromatin-coated beads, indicating that charge-shifting posttranslational modification of Nap1 contributes to H1M dynamics that are essential for higher order chromosome architecture.

The unspecified visual art referent in Akhmatova’s enigmatic poem, “An Old Portrait” (Staryi portret, 1910), dedicated to the painter Aleksandra Ekster, has long inspired critical speculation. scholars (superfin and Timenchik, Rosslyn, Rubins) have proposed numerous readings, but none has entirely resolved the poem’s mysteries. I will argue that the ballets Le Pavillon d’Armide and Cleopâtre, performed in the first Paris season of the Ballets Russes, provide hitherto unexplored subtexts for the spare but dramatic poem. Considered within the context of the ballets, the poem’s characters and details take on more specific and meaningful contours. By analyzing these ballets’ respective libretti, costume designs and stage sets, it becomes possible to detect in the poem references to the Gobelins portrait depicting the deadly enchantress in Le Pavillon d’Armide, the poisoning of Amoun at the conclusion of Cleopâtre, and Vaclav Nijinsky’s performances as Armida’s servant in Le Pavillon d’Armide and Cleop...

Histone H1 induces bending and looping of single DNA molecules at nanomolar concentrations. H1 increases the rate of assembly for single chromatin fibers under force in Xenopus egg extracts and protects decondensing sperm nuclei from stretching and fragmenting in egg cytoplasm., Histone H1 binds to linker DNA between nucleosomes, but the dynamics and biological ramifications of this interaction remain poorly understood. We performed single-molecule experiments using magnetic tweezers to determine the effects of H1 on naked DNA in buffer or during chromatin assembly in Xenopus egg extracts. In buffer, nanomolar concentrations of H1 induce bending and looping of naked DNA at stretching forces below 0.6 pN, effects that can be reversed with 2.7-pN force or in 200 mM monovalent salt concentrations. Consecutive tens-of-nanometer bending events suggest that H1 binds to naked DNA in buffer at high stoichiometries. In egg extracts, single DNA molecules assemble into nucleosomes and undergo rapid compaction. Histone H1 at endogenous physiological concentrations increases the DNA compaction rate during chromatin assembly under 2-pN force and decreases it during disassembly under 5-pN force. In egg cytoplasm, histone H1 protects sperm nuclei undergoing genome-wide decondensation and chromatin assembly from becoming abnormally stretched or fragmented due to astral microtubule pulling forces. These results reveal functional ramifications of H1 binding to DNA at the single-molecule level and suggest an important physiological role for H1 in compacting DNA under force and during chromatin assembly.

Background Linker histone H1 has been studied in vivo and using reconstituted chromatin, but there have been few systematic studies of the effects of the cellular environment on its function. Due to the presence of many other chromatin factors and specific chaperones such as RanBP7/importin beta that regulate histone H1, linker histones likely function differently in vivo than in purified systems. Methodology/Principal Findings We have directly compared H1 binding to sperm nuclei in buffer versus Xenopus egg extract cytoplasm, and monitored the effects of adding nuclear import chaperones. In buffer, RanBP7 decondenses sperm nuclei, while H1 binds tightly to the chromatin and rescues RanBP7-mediated decondensation. H1 binding is reduced in cytoplasm, and H1 exhibits rapid FRAP dynamics in cytoplasm but not in buffer. RanBP7 decreases H1 binding to chromatin in both buffer and extract but does not significantly affect H1 dynamics in either condition. Importin beta has a lesser effect than RanBP7 on sperm chromatin decondensation and H1 binding, while a combination of RanBP7/importin beta is no more effective than RanBP7 alone. In extracts supplemented with RanBP7, H1 localizes to chromosomal foci, which increase after DNA damage. Unlike somatic H1, the embryonic linker histone H1M binds equally well to chromatin in cytoplasm compared to buffer. Amino-globular and carboxyl terminal domains of H1M bind chromatin comparably to the full-length protein in buffer, but are inhibited ∼10-fold in cytoplasm. High levels of H1 or its truncations distort mitotic chromosomes and block their segregation during anaphase. Conclusion/Significance RanBP7 can decondense sperm nuclei and decrease H1 binding, but the rapid dynamics of H1 on chromatin depend on other cytoplasmic factors. Cytoplasm greatly impairs the activity of individual H1 domains, and only the full-length protein can condense chromatin properly.
