1. Evolution of Globus Pallidus Targeting for Parkinson's and Dystonia Deep Brain Stimulation: A 15-Year Experience

Abstract: Objective: The aim of this study is to evaluate the evolution of GPi DBS targeting.Methods: This retrospective, single-center study included patients implanted with GPi DBS leads for dystonia or PD during the years 2004 to 2018 at the University of Florida Fixel Institute for Neurological Diseases. Each patient underwent a high-resolution targeting study on the day prior to the surgery, which was fused with a high resolution CT scan that was acquired on the day of the procedure. Intraoperative target location was selected using a digitized 3D Schaltenbrand-Bailey atlas. All patients underwent a high-resolution head CT scan without contrast approximately one month after lead implantation and accurate measurement of neuroanatomical lead position was acquired after fusion of pre-operative and post-operative image studies.Results: We analyzed 253 PD patients with 352 leads and 80 dystonia patients with 141 leads. During 15 years of follow-up, lead locations in the PD group migrated more laterally (β = 0.09, p < 0.0001), posteriorly [slope (β) = 0.04, p < 0.05], and dorsally (β = 0.07, p < 0.001), whereas leads in the dystonia group did not significantly change position aside from a trend in the dorsal direction (β = 0.06, p = 0.053).Conclusion: The evolving target likely results from multiple factors including improvements in targeting techniques and clinical feedback intraoperatively and post-operatively. Our demonstrates the potential importance of a systematic post-operative DBS lead measurement protocol to ensure quality control and to inform and optimize DBS programming.

Abstract: The size of the optics used in observatories is often limited by fabrication, metrology, and handling technology, but having a large primary mirror provides significant benefits for scientific research. The evolution of rocket launch options enables heavy payload carrying on orbit and outstretching the telescope's form-factor choices. Moreover, cost per launch is lower than the traditional flight method, which is obviously advantageous for various novel space observatory concepts. The University of Arizona has successfully fabricated many large-scale primary optics for ground-based observatories including the Large Binocular Telescope (LBT, 8.4 meter diameter two primary mirrors), Large Synoptic Survey Telescope (now renamed to Vera C. Rubin Observatory, 8.4 meter diameter monolithic primary and tertiary mirror), and the Giant Magellan Telescope (GMT, 8.4 meter diameter primary mirror seven segments). Launching a monolithic primary mirror into space could bypass many of the difficulties encountered during the assembly and deployment of the segmented primary mirrors. However, it might bring up unprecedented challenges and hurdles, also. We explore and foresee the expected challenges and evaluate them. Abstract: New development approaches, including launch vehicles and advances in sensors, computing, and software, have lowered the cost of entry into space, and have enabled a revolution in low-cost, high-risk Small Satellite (SmallSat) missions. To bring about a similar transformation in larger space telescopes, it is necessary to reconsider the full paradigm of space observatories. Here we will review the history of space telescope development and cost drivers, and describe an example conceptual design for a low cost 6.5 m optical telescope to enable new science when operated in space at room temperature. It uses a monolithic primary mirror of borosilicate glass, drawing on lessons and tools from decades of experience with ground-based observatories and instruments, as well as flagship space missions. It takes advantage, as do large launch vehicles, of increased computing power and space-worthy commercial electronics in low-cost active predictive control systems to maintain stability. We will describe an approach that incorporates science and trade study results that address driving requirements such as integration and testing costs, reliability, spacecraft jitter, and wavefront stability in this new risk-tolerant "LargeSat" context., Comment: Presented at SPIE, Optics+Photonics 2023, Astronomical Optics: Design, Manufacture, and Test of Space and Ground Systems IV in San Diego, CA, USA. Abstract: Twenty-six years ago a small committee report, building on earlier studies, expounded a compelling and poetic vision for the future of astronomy, calling for an infrared-optimized space telescope with an aperture of at least $4m$. With the support of their governments in the US, Europe, and Canada, 20,000 people realized that vision as the $6.5m$ James Webb Space Telescope. A generation of astronomers will celebrate their accomplishments for the life of the mission, potentially as long as 20 years, and beyond. This report and the scientific discoveries that follow are extended thank-you notes to the 20,000 team members. The telescope is working perfectly, with much better image quality than expected. In this and accompanying papers, we give a brief history, describe the observatory, outline its objectives and current observing program, and discuss the inventions and people who made it possible. Abstract: The Y-dwarf WISE 1828+2650 is one of the coldest known Brown Dwarfs with an effective temperature of $\sim$300 K. Located at a distance of just 10 pc, previous model-based estimates suggest WISE1828+2650 has a mass of $\sim$5-10 Mj, making it a valuable laboratory for understanding the formation, evolution and physical characteristics of gas giant planets. However, previous photometry and spectroscopy have presented a puzzle with the near-impossibility of simultaneously fitting both the short (0.9-2.0 microns) and long wavelength (3-5 microns) data. A potential solution to this problem has been the suggestion that WISE 1828+2650 is a binary system whose composite spectrum might provide a better match to the data. Alternatively, new models being developed to fit JWST/NIRSpec and MIRI spectroscopy might provide new insights. This article describes JWST/NIRCam observations of WISE 1828+2650 in 6 filters to address the binarity question and to provide new photometry to be used in model fitting. We also report Adaptive Optics imaging with the Keck 10 m telescope. We find no evidence for multiplicity for a companion beyond 0.5 AU with either JWST or Keck. Abstract: Retrieving images transmitted through multi-mode fibers is of growing interest, thanks to their ability to confine and transport light efficiently in a compact system. Here, we demonstrate machine-learning-based decoding of large-scale digital images (pages), maximizing page capacity for optical storage applications. Using a millimeter-sized square cross-section waveguide, we image an 8-bit spatial light modulator, presenting data as a matrix of symbols. Normally, decoders will incur a prohibitive O(n^2) computational scaling to decode n symbols in spatially scrambled data. However, by combining a digital twin of the setup with a U-Net, we can retrieve up to 66 kB using efficient convolutional operations only. We compare trainable ray-tracing-based with eigenmode-based twins and show the former to be superior thanks to its ability to overcome the simulation-to-experiment gap by adjusting to optical imperfections. We train the pipeline end-to-end using a differentiable mutual-information estimator based on the von-Mises distribution, generally applicable to phase-coding channels., Comment: 21 pages, 5 figures

Abstract: This paper provides an overview of current progress in the technological advances and the use of deep brain stimulation (DBS) to treat neurological and neuropsychiatric disorders, as presented by participants of the Fourth Annual Deep Brain Stimulation Think Tank, which was convened in March 2016 in conjunction with the Center for Movement Disorders and Neurorestoration at the University of Florida, Gainesveille FL, USA. The Think Tank discussions first focused on policy and advocacy in DBS research and clinical practice, formation of registries, and issues involving the use of DBS in the treatment of Tourette Syndrome. Next, advances in the use of neuroimaging and electrochemical markers to enhance DBS specificity were addressed. Updates on ongoing use and developments of DBS for the treatment of Parkinson’s disease, essential tremor, Alzheimer’s disease, depression, post-traumatic stress disorder, obesity, addiction were presented, and progress toward innovation(s) in closed-loop applications were discussed. Each section of these proceedings provides updates and highlights of new information as presented at this year’s international Think Tank, with a view toward current and near future advancement of the field.

Abstract: Tourette Syndrome (TS) is a neuropsychiatric disease characterized by a combination of motor and vocal tics. Deep brain stimulation (DBS), already widely utilized for Parkinson’s disease and other movement disorders, is an emerging therapy for select and severe cases of TS that are resistant to medication and behavioral therapy. Over the last two decades, DBS has been used experimentally to manage severe TS cases. The results of case reports and small case series have been variable but in general positive. The reported interventions have, however, been variable, and there remain non-standardized selection criteria, various brain targets, differences in hardware, as well as variability in the programming parameters utilized. DBS centers perform only a handful of TS DBS cases each year, making large-scale outcomes difficult to study and to interpret. These limitations, coupled with the variable effect of surgery, and the overall small numbers of TS patients with implanted DBS worldwide, have delayed regulatory agency approval (e.g. FDA and equivalent agencies around the world). The Tourette Association of America, in response to the worldwide need for a more organized and collaborative effort, launched an international TS DBS registry and database. The main goal of the project has been to share data, uncover best practices, improve outcomes, and to provide critical information to regulatory agencies. The international registry and database has improved the communication and collaboration among TS DBS centers worldwide. In this paper we will review some of the key operation details for the international TS DBS database and registry.

Abstract: JWST holds great promise in characterizing atmospheres of transiting exoplanets, potentially providing insights into Earth-sized planets within the habitable zones of M dwarf host stars if photon-limited performance can be achieved. Here, we discuss the systematic error sources that are expected to be present in grism time series observations with the NIRCam instrument. We find that pointing jitter and high gain antenna moves on top of the detectors' subpixel crosshatch patterns will produce relatively small variations (less than 6 parts per million, ppm). The time-dependent aperture losses due to thermal instabilities in the optics can also be kept to below 2 ppm. To achieve these low noise sources, it is important to employ a sufficiently large (more than 1.1 arcseconds) extraction aperture. Persistence due to charge trapping will have a minor (less than 3 ppm) effect on time series 20 minutes into an exposure and is expected to play a much smaller role than it does for the HST WFC3 detectors. We expect temperature fluctuations to be less than 3 ppm. In total, our estimated noise floor from known systematic error sources is only 9 ppm per visit. We do however urge caution as unknown systematic error sources could be present in flight and will only be measurable on astrophysical sources like quiescent stars. We find that reciprocity failure may introduce a perennial instrument offset at the 40 ppm level, so corrections may be needed when stitching together a multi-instrument multi-observatory spectrum over wide wavelength ranges., Comment: Published in AJ, 25 pages

13. A novel diagnostic sign of hip fracture mechanism in ground level falls: two case reports and review of the literature

Abstract: Abstract Introduction Most elderly hip fractures are the result of a ground level fall. Defining high risk falls and fracture mechanisms are important to develop successful hip fracture prevention programs. This case series presents a previously unreported diagnostic sign and for the first time documents a hip fracture mechanism for a knee impact injury from a ground level fall in two elderly patients. Case presentation Case 1 was a 65-year-old Caucasian woman who fell forward with initial contact to her left knee, sustaining an impacted femoral neck fracture of her ipsilateral left hip. Case 2 was a 92-year-old Caucasian woman who fell bending forward, impacting her left knee and sustaining a comminuted intertrochanteric fracture of her ipsilateral left hip. The fractures occurred as a result of unprotected ground level falls in a forward direction with initial impact to the knee. The knee contusions were located near Gerdy’s tubercle and appear characteristic of a direct impact injury. Conclusion The physical finding of a small localized site of impact and/or contusion in the anterior aspect of the knee in both of these patients with radiographic evidence of an ipsilateral hip fracture would strongly suggest that a knee impact injury can transmit enough energy to the proximal femur by axial loading to result in the hip fracture. The physical finding described is a reliable indicator of this hip fracture mechanism.

Abstract: The James Webb Space Telescope near-infrared camera (JWST NIRCam) has two 2.'2 $\times$ 2.'2 fields of view that are capable of either imaging or spectroscopic observations. Either of two $R \sim 1500$ grisms with orthogonal dispersion directions can be used for slitless spectroscopy over $\lambda = 2.4 - 5.0$ $\mu$m in each module, and shorter wavelength observations of the same fields can be obtained simultaneously. We present the latest predicted grism sensitivities, saturation limits, resolving power, and wavelength coverage values based on component measurements, instrument tests, and end-to-end modeling. Short wavelength (0.6 -- 2.3 $\mu$m) imaging observations of the 2.4 -- 5.0 $\mu$m spectroscopic field can be performed in one of several different filter bands, either in-focus or defocused via weak lenses internal to NIRCam. Alternatively, the possibility of 1.0 -- 2.0 $\mu$m spectroscopy (simultaneously with 2.4 -- 5.0 $\mu$m) using dispersed Hartmann sensors (DHSs) is being explored. The grisms, weak lenses, and DHS elements were included in NIRCam primarily for wavefront sensing purposes, but all have significant science applications. Operational considerations including subarray sizes, and data volume limits are also discussed. Abstract: This thesis reports on work performed in the development and application of an automated plate reading microscope implementing wide field time gated fluorescence lifetime imaging technology. High content analysis (HCA) imaging assays enabled by automated microscopy platforms allow hundreds of conditions to be tested in a single experiment. Though fluorescence lifetime imaging (FLIM) is established in life sciences applications as a method whereby quantitative information may be extracted from time-resolved fluorescence signals, FLIM has not been widely adopted in an HCA context. The FLIM plate reader developed throughout this PhD has been designed to allow HCA-FLIM experiments to be performed and has been demonstrated to be capable of recording multispectral, FLIM and bright field data from 600 fields of view in less than four hours. FLIM is commonly used as a means of reading out Förster resonance energy transfer (FRET) between fluorescent fusion proteins in cells. Using the FLIM plate reader to investigate large populations of cells per experimental condition without significant user input has allowed statistically significant results to be obtained in FRET experiments that present relatively small changes in mean fluorescent lifetime. This capability has been applied to investigations of FOXM1 SUMOylation in response to anthracycline treatment, and to studies of the spatiotemporal activation profiles of small GTPases. Furthermore, the FLIM plate reader allows FLIM-FRET to be applied to protein-protein interaction screening. The application of the instrument to screening RASSF proteins for interaction with MST1 is discussed. The FLIM plate reader was also configured to utilise ultraviolet excitation radiation and optimised for the measurement of autofluorescence lifetime for label-free assays of biological samples. Experiments investigating the autofluorescence lifetime of live cells under the influence of metabolic modulators are presented alongside the design considerations necessary when using UV excitation for HCA-FLIM.

Abstract: BackgroundThe purpose of this study was to implement and evaluate the utility of a multi-echo sequence at 7 Tesla (T) for simultaneous time-of-flight (TOF) MR-angiography (MRA) and susceptibility-weighted imaging (SWI) of radiation-induced cerebral microbleeds (CMBs), intracranial arteries, and veins.MethodsA four-echo gradient-echo sequence was implemented on a 7T scanner. The first echo was used to create TOF-MRA images, while the remaining echoes were combined to visualize CMBs and veins on SWI images. The sequence was evaluated on eight brain tumor patients with known radiation-induced CMBs. Single-echo images were also acquired to visually and quantitatively compare the contrast-to-noise ratio (CNR) of small- and intermediate-vessels between acquisitions. The number of CMBs detected with each acquisition was also quantified. Statistical significance was determined using a Wilcoxon signed-rank test.ResultsCompared with the single-echo sequences, the CNR of small and intermediate arteries increased 7.6% (P < 0.03) and 9.5% (P = 0.06), respectively, while the CNR of small and intermediate veins were not statistically different between sequences (P = 0.95 and P = 0.46, respectively). However, these differences were not discernible by visual inspection. Also the multi-echo sequence detected 18.3% more CMBs (P < 0.008) due to higher slice resolution.ConclusionThe proposed 7T multi-echo sequence depicts arteries, veins, and CMBs on a single image to facilitate quantitative evaluation of radiation-induced vascular injury.

24. IR observations of MS 1054-03: Star Formation and its Evolution in Rich Galaxy Clusters

Abstract: We study the infrared (IR) properties of galaxies in the cluster MS 1054-03 at z=0.83 by combining MIPS 24 micron data with spectra of more than 400 galaxies and a very deep K-band selected catalog. 19 IR cluster members are selected spectroscopically, and an additional 15 are selected by their photometric redshifts. We derive the IR luminosity function of the cluster and find strong evolution compared to the similar-mass Coma cluster. The best fitting Schechter function gives L*_{IR}=11.49 +0.30/-0.29 L_sun with a fixed faint end slope, about one order of magnitude larger than that in Coma. The rate of evolution of the IR luminosity from Coma to MS 1054-03 is consistent with that found in field galaxies, and it suggests that some internal mechanism, e.g., the consumption of the gas fuel, is responsible for the general decline of the cosmic star formation rate (SFR) in different environments. The mass-normalized integrated SFR within 0.5R_200 in MS 1054-03 also shows evolution compared with other rich clusters at lower redshifts, but the trend is less conclusive if the mass selection effect is considered. A nonnegligible fraction (13%) of cluster members, are forming stars actively and the overdensity of IR galaxies is about 20 compared to the field. It is unlikely that clusters only passively accrete star forming galaxies from the surrounding fields and have their star formation quenched quickly afterward; instead, many cluster galaxies still have large amounts of gas, and their star formation may be enhanced by the interaction with the cluster., Comment: 49 pages, 9 figures, accepted by ApJ

Abstract: We are performing a uniform and unbiased, ~7x7 degrees imaging survey of the Large Magellanic Cloud (LMC), using the IRAC and MIPS instruments on board the Spitzer Space Telescope in order to survey the agents of a galaxy's evolution (SAGE), the interstellar medium (ISM) and stars in the LMC. The detection of diffuse ISM with column densities >1.2x10^21 H cm^-2 permits detailed studies of dust processes in the ISM. SAGE's point source sensitivity enables a complete census of newly formed stars with masses >3 solar masses that will determine the current star formation rate in the LMC. SAGE's detection of evolved stars with mass loss rates >1x10^-8 solar masses per year will quantify the rate at which evolved stars inject mass into the ISM of the LMC. The observing strategy includes two epochs in 2005, separated by three months, that both mitigate instrumental artifacts and constrain source variability. The SAGE data are non-proprietary. The data processing includes IRAC and MIPS pipelines and a database for mining the point source catalogs, which will be released to the community in support of Spitzer proposal cycles 4 and 5. We present initial results on the epoch 1 data with a special focus on the N79 and N83 region. The SAGE epoch 1 point source catalog has ~4 million sources. The point source counts are highest for the IRAC 3.6 microns band and decrease dramatically towards longer wavelengths consistent with the fact that stars dominate the point source catalogs and that the dusty objects, e.g. young stellar objects and dusty evolved stars that detected at the longer wavelengths, are rare in comparison. We outline a strategy for identifying foreground MW stars, that may comprise as much as 18% of the source list, and background galaxies, that may comprise ~12% of the source list., Comment: Accepted by the Astronomical Journal

26. Infrared Luminosity Function of the Coma Cluster

Abstract: Using mid-IR and optical data, we deduce the total infrared (IR) luminosities of galaxies in the Coma cluster and present their infrared luminosity function (LF). The shape of the overall Coma IR LF does not show significant differences from the IR LFs of the general field, which indicates the general independence of global galaxy star formation on environment up to densities $\sim$ 40 times greater than in the field (we cannot test such independence above $L_{ir} \approx 10^{44} {\rm ergs s}^{-1}$). However, a shallower faint end slope and a smaller $L_{ir}^{*}$ are found in the core region (where the densities are still higher) compared to the outskirt region of the cluster, and most of the brightest IR galaxies are found outside of the core region. The IR LF in the NGC 4839 group region does not show any unique characteristics. By integrating the IR LF, we find a total star formation rate in the cluster of about 97.0 $M_{\sun}{\rm yr}^{-1}$. We also studied the contributions of early- and late-type galaxies to the IR LF. The late-type galaxies dominate the bright end of the LF, and the early-type galaxies, although only making up a small portion ($\approx$ 15%) of the total IR emission of the cluster, contribute greatly to the number counts of the LF at $L_{ir} < 10^{43} {\rm ergs s}^{-1}$., Comment: 33 pages, 8 figures, 1 table. 27. The [FeII]1.644micron emission in M82 and NGC253: Is it a measure of the supernova rate?

Abstract: We present HST/NICMOS [FeII]1.644micron, Pa-alpha (1.87micron) and continuum images of the starburst galaxies M82 and NGC253 at an unprecedented spatial resolution. In both galaxies we detect [FeII] compact sources superimposed on a diffuse background in the disk of the galaxies together with a component above and below the plane of the galaxy. The radio and [FeII] emissions perpendicular to the disk of M82 show a remarkable similarity to each other, suggesting that both emissions originate in shocks from supernova explosions. We find a spatial correspondence between bright compact [FeII] emitting regions and the location of radio supernova remnants (SNR) for approximately 30-50% of radio SNRs in M82 and NGC253. This lack of a one-to-one correspondence, more than being indicative of a different origin for the radio and [FeII] emission in starbursts, suggests two populations of SNRs: an older population(<10^4yr) traced by the [FeII] emission and a younger population (a few hundred years old) traced by the radio SNRs. We therefore conclude that the [FeII] emission in starburst galaxies provides a good estimate of the supernova activity. Using our newly determined [FeII] luminosities (corrected for extinction) of M82 and NGC253, we reevaluate the calibration of the supernova rate in terms of the [FeII] luminosity for starburst galaxies., Comment: Accepted for publication in AJ, 2003. 18 pages, 10 figures. Abstract: We report results from a Hubble Space Telescope (HST) and Near-Infrared Camera and Multiobject Spectrometer (NICMOS) program to study the distribution of hot neutral (molecular hydrogen) and ionized circumstellar material in the young planetary nebulae NGC 7027. HST/NICMOS provided very high spatial resolution imaging in line and continuum emission, and the stability and large dynamic range needed for investigating detailed structures in the circumstellar material. We present dramatic new images of NGC 7027 that have led to a new understanding of the structure in this important planetary nebula. The central star is clearly revealed, providing near-infrared fluxes that are used to directly determine the stellar temperature very accurately (T = 198,000 K). It is found that the photodissociation layer as revealed by near-infrared molecular hydrogen emission is very thin, and is biconical in shape. The interface region is structured and filamentary, suggesting the existence of hydrodynamic instabilities. We discuss evidence for the presence of one or more highly collimated, off-axis jets that might be present in NGC 7027. Abstract: Images of Hubble 12 were obtained with the HST/NICMOS instrument in the F110W, F164N, and F166N filters of NIC1, and the F160W, F187N, F190N, F212N, F215N filters of NIC2. The images show the structure of the inner ``torus'' and lobes much clearer than previous ground-based images. In particular, the [Fe II] lobes are clearly resolved and shown to be distinct from the molecular hydrogen emission structures. Apparent changes in the inner ionized and neutral bipolar shell implies periodic mass loss or changes in stellar wind shape or direction. The molecular hydrogen in the ``eye'' is radiatively excited and shows a complex morphology that suggests that several mass ejection events are responsible for producing this structure. Abstract: We present the results of a near-infrared narrowband imaging survey of planetary nebulae. Objects were selected in a way that complements similar surveys done at visible and near-infrared wavelengths. No new detections of molecular hydrogen emission were made. The H2 is frequently found to be extended, except in young, visibly compact objects. Our results are consistent with the already determined correlation of H2 emission with planetary nebula morphological type. Abstract: Image retrieval after propagation through multi-mode fibers is gaining attention due to their capacity to confine light and efficiently transport it over distances in a compact system. Here, we propose a generally applicable information-theoretic framework to transmit maximal-entropy (data) images and maximize the information transmission over sub-meter distances, a crucial capability that allows optical storage applications to scale and address different parts of storage media. To this end, we use millimeter-sized square optical waveguides to image a megapixel 8-bit spatial-light modulator. Data is thus represented as a 2D array of 8-bit values (symbols). Transmitting 100000s of symbols requires innovation beyond transmission matrix approaches. Deep neural networks have been recently utilized to retrieve images, but have been limited to small (thousands of symbols) and natural looking (low entropy) images. We maximize information transmission by combining a bandwidth-optimized homodyne detector with a differentiable hybrid neural-network consisting of a digital twin of the experiment setup and a U-Net. For the digital twin, we implement and compare a differentiable mode-based twin with a differentiable ray-based twin. Importantly, the latter can adapt to manufacturing-related setup imperfections during training which we show to be crucial. Our pipeline is trained end-to-end to recover digital input images while maximizing the achievable information page size based on a differentiable mutual-information estimator. We demonstrate retrieval of 66 kB at maximum with 1.7 bit per symbol on average with a range of 0.3 - 3.4 bit., Comment: 17 pages, 5 figures
