Histopathological staining of human tissue is essential in the diagnosis of various diseases. The recent advances in virtual tissue staining technologies using AI alleviate some of the costly and tedious steps involved in the traditional histochemical staining process, permitting multiplexed rapid staining of label-free tissue without using staining reagents, while also preserving tissue. However, potential hallucinations and artifacts in these virtually stained tissue images pose concerns, especially for the clinical utility of these approaches. Quality assessment of histology images is generally performed by human experts, which can be subjective and depends on the training level of the expert. Here, we present an autonomous quality and hallucination assessment method (termed AQuA), mainly designed for virtual tissue staining, while also being applicable to histochemical staining. AQuA achieves 99.8% accuracy when detecting acceptable and unacceptable virtually stained tissue images without access to ground truth, also presenting an agreement of 98.5% with the manual assessments made by board-certified pathologists. Besides, AQuA achieves super-human performance in identifying realistic-looking, virtually stained hallucinatory images that would normally mislead human diagnosticians by deceiving them into diagnosing patients that never existed. We further demonstrate the wide adaptability of AQuA across various virtually and histochemically stained tissue images and showcase its strong external generalization to detect unseen hallucination patterns of virtual staining network models as well as artifacts observed in the traditional histochemical staining workflow. Human epidermal growth factor receptor 2 (HER2) is a critical protein in cancer cell growth that signifies the aggressiveness of breast cancer (BC) and helps predict its prognosis. Accurate assessment of immunohistochemically (IHC) stained tissue slides for HER2 expression levels is essential for both treatment guidance and understanding of cancer mechanisms. Nevertheless, the traditional workflow of manual examination by board-certified pathologists encounters challenges, including inter- and intra-observer inconsistency and extended turnaround times. Here, we introduce a deep learning-based approach utilizing pyramid sampling for the automated classification of HER2 status in IHC-stained BC tissue images. Our approach analyzes morphological features at various spatial scales, efficiently managing the computational load and facilitating a detailed examination of cellular and larger-scale tissue-level details. This method addresses the tissue heterogeneity of HER2 expression by providing a comprehensive view, leading to a blind testing classification accuracy of 84.70%, on a dataset of 523 core images from tissue microarrays. We present and characterize a method to accelerate the relaxation of a Brownian object between two distinct equilibrium states. Instead of relying on a deterministic time-dependent control parameter, we use stochastic resetting to guide and accelerate the transient evolution. The protocol is investigated numerically, and its thermodynamic cost is evaluated with the tools of stochastic thermodynamics. Remarkably, we show that stochastic resetting significantly accelerates the relaxation to the final state. This stochastic protocol exhibits energetic and temporal characteristics that align with the scales observed in previously investigated deterministic protocols. Histological examination is a crucial step in an autopsy; however, the traditional histochemical staining of post-mortem samples faces multiple challenges, including the inferior staining quality due to autolysis caused by delayed fixation of cadaver tissue, as well as the resource-intensive nature of chemical staining procedures covering large tissue areas, which demand substantial labor, cost, and time. These challenges can become more pronounced during global health crises when the availability of histopathology services is limited, resulting in further delays in tissue fixation and more severe staining artifacts. Here, we report the first demonstration of virtual staining of autopsy tissue and show that a trained neural network can rapidly transform autofluorescence images of label-free autopsy tissue sections into brightfield equivalent images that match hematoxylin and eosin (H&E) stained versions of the same samples, eliminating autolysis-induced severe staining artifacts inherent in traditional histochemical staining of autopsied tissue. Our virtual H&E model was trained using >0.7 TB of image data and a data-efficient collaboration scheme that integrates the virtual staining network with an image registration network. The trained model effectively accentuated nuclear, cytoplasmic and extracellular features in new autopsy tissue samples that experienced severe autolysis, such as COVID-19 samples never seen before, where the traditional histochemical staining failed to provide consistent staining quality. This virtual autopsy staining technique can also be extended to necrotic tissue, and can rapidly and cost-effectively generate artifact-free H&E stains despite severe autolysis and cell death, also reducing labor, cost and infrastructure requirements associated with the standard histochemical staining., Comment: 24 Pages, 7 Figures

Sketches are a family of streaming algorithms widely used in the world of big data to perform fast, real-time analytics. A popular sketch type is Quantiles, which estimates the data distribution of a large input stream. We present Quancurrent, a highly scalable concurrent Quantiles sketch. Quancurrent's throughput increases linearly with the number of available threads, and with $32$ threads, it reaches an update speedup of $12$x and a query speedup of $30$x over a sequential sketch. Quancurrent allows queries to occur concurrently with updates and achieves an order of magnitude better query freshness than existing scalable solutions.

Cordial Miners are a family of efficient Byzantine Atomic Broadcast protocols, with instances for asynchrony and eventual synchrony. They improve the latency of state-of-the-art DAG-based protocols by almost 2X and achieve optimal good-case complexity of O(n) by forgoing Reliable Broadcast as a building block. Rather, Cordial Miners use the blocklace -- a partially-ordered counterpart of the totally-ordered blockchain data structure -- to implement the three algorithmic components of consensus: Dissemination, equivocation-exclusion, and ordering.

Languages are continuously undergoing changes, and the mechanisms that underlie these changes are still a matter of debate. In this work, we approach language evolution through the lens of causality in order to model not only how various distributional factors associate with language change, but how they causally affect it. In particular, we study slang, which is an informal language that is typically restricted to a specific group or social setting. We analyze the semantic change and frequency shift of slang words and compare them to those of standard, nonslang words. With causal discovery and causal inference techniques, we measure the effect that word type (slang/nonslang) has on both semantic change and frequency shift, as well as its relationship to frequency, polysemy and part of speech. Byzantine Agreement is a key component in many distributed systems. While Dolev and Reischuk have proven a long time ago that quadratic communication complexity is necessary for worst-case runs, the question of what can be done in practically common runs with fewer failures remained open. In this paper we present the first Byzantine Broadcast algorithm with $O(n(f+1))$ communication complexity, where $0\leq f\leq t$ is the actual number of process failures in a run. And for BA with strong unanimity, we present the first optimal-resilience algorithm that has linear communication complexity in the failure-free case and a quadratic cost otherwise.

Money transfer is an abstraction that realizes the core of cryptocurrencies. It has been shown that, contrary to common belief, money transfer in the presence of Byzantine faults can be implemented in asynchronous networks and does not require consensus. Nonetheless, existing implementations of money transfer still require a quadratic message complexity per payment, making attempts to scale hard. In common blockchains, such as Bitcoin and Ethereum, this cost is mitigated by payment channels implemented as a second layer on top of the blockchain allowing to make many off-chain payments between two users who share a channel. Such channels only require on-chain transactions for channel opening and closing, while the intermediate payments are done off-chain with constant message complexity. But payment channels in-use today require synchrony, therefore they are inadequate for asynchronous money transfer systems. In this paper, we provide a series of possibility and impossibility results for payment channels in asynchronous money transfer systems. We first prove a quadratic lower bound on the message complexity of on-chain transfers. Then, we explore two types of payment channels, unidirectional and bidirectional. We define them as shared memory abstractions and prove that in certain cases they can be implemented as a second layer on top of an asynchronous money transfer system whereas in other cases it is impossible.

Cold atmospheric plasma (CAP) is a near room temperature ionized gas, generated under non-equilibrium discharge conditions. Here we show that a short exposure of rat peritoneal exudate macrophages and T-cells to CAP in vitro, triggered an inflammatory phenotype leading to better antigen-presenting and effector cell function respectively. Different from previous studies mainly using immortalized cell lines, both macrophage and T-cells in this study were primary cells isolated from mice. Furthermore, ex-vivo exposure of T-cells to CAP, followed by their adoptive transfer into tumor-bearing mice resulted in a strong antitumor effect in vivo. Mechanistically, CAP seems to disrupt tolerogenic pathways leading to enhanced production of pro-inflammatory cytokines while limiting the production of anti-inflammatory cytokines and the expression of inhibitory molecules such as programmed death-ligand 1 (PD-L1). CAP represents therefore a novel, non-toxic and easy to deliver technology to augment the function of immune cells and enhance antitumor responses when used as a component of T-cell adoptive immunotherapies strategies or, potentially in combination with other cancer immunotherapeutic approaches.

In recent years, Byzantine Agreement is being considered in increasing scales due to the proliferation of blockchains and other decentralized financial technologies. Consequently, a number of works have improved its communication complexity in various network models. In this short paper we survey recent advances and outline some open research questions on the subject.

With the recent surge in social applications relying on knowledge graphs, the need for techniques to ensure fairness in KG based methods is becoming increasingly evident. Previous works have demonstrated that KGs are prone to various social biases, and have proposed multiple methods for debiasing them. However, in such studies, the focus has been on debiasing techniques, while the relations to be debiased are specified manually by the user. As manual specification is itself susceptible to human cognitive bias, there is a need for a system capable of quantifying and exposing biases, that can support more informed decisions on what to debias. To address this gap in the literature, we describe a framework for identifying biases present in knowledge graph embeddings, based on numerical bias metrics. We illustrate the framework with three different bias measures on the task of profession prediction, and it can be flexibly extended to further bias definitions and applications. A new type of material with embedded intelligence, namely 'intelligent plasma', is introduced. Such new material exhibits programmable chemical pathway networks resembling artificial neural networks. As a Markov process of chemistry, the chemical pathway network can be customized and thus the intelligent plasmas can be programmed to make their own decisions to react to the dynamic external and internal conditions. It finally can accomplish complex missions without any external controls from the humans while relying on its preprogrammed chemical network topology before the mission. To that end, only basic data input and readings are required without any external controls during the mission. The approach to 'if' conditions and 'while' loops of the programmable intelligent plasmas are also discussed with examples of applications including automatic workflows, and signal processing.

Plasma, the fourth and most pervasive state of matter in the visible universe, is a fascinating medium that is connected to the beginning of our universe itself. Man-made plasmas are at the core of many technological advances that include the fabrication of semiconductor devices, which enabled the modern computer and communication revolutions. The introduction of low temperature, atmospheric pressure plasmas to the biomedical field has ushered a new revolution in the healthcare arena that promises to introduce plasma-based therapies to combat some thorny and long-standing medical challenges. This paper presents an overview of where research is at today and discusses innovative concepts and approaches to overcome present challenges and take the field to the next level. It is written by a team of experts who took an in-depth look at the various biomedical applications, made critical analysis, and proposed ideas and concepts that should help the research community focus their efforts on clear and practical steps necessary to keep the field advancing for decades to come.
