Rising tuition rates across colleges and universities contribute to the mounting financial strain on students. In response to rising costs, literature details that students seek to bolster their financial security by working additional hours per week, consequently leading to less available time. Previous studies have focused on the negative associations with GPA but not with in-class points. We observed this relationship within a 77-student 200-level genetics course and associated lab. Students working 20+ hours per week (59.7% of students) fared significantly worse on exams. These students scored significantly fewer total class points (p = 0.0089), exam points (p = 0.0255, p = 0.0028, p = 0.0311), and were more likely to incur a failed assignment sooner during the class than their counterparts (p = 0.0025). We observed these trends again in a 34-student 400-level immunology validation cohort (p = 0.0032). These findings represent the first quantitative analysis of employment vs. numerical biology exam and course scores. They are an important step toward identifying and treating the underlying factors associated with reduced STEM performance, retention, and well-being, and represent a forewarning of an approaching higher education crisis as institutions seek to retain and support underrepresented students.

Abstract Coordinated movement requires the nervous system to continuously compensate for changes in mechanical load across different conditions. For voluntary movements like reaching, the motor cortex is a critical hub that generates commands to move the limbs and counteract loads. How does cortex contribute to load compensation when rhythmic movements are sequenced by a spinal pattern generator? Here, we address this question by manipulating the mass of the forelimb in unrestrained mice during locomotion. While load produces changes in motor output that are robust to inactivation of motor cortex, it also induces a profound shift in cortical dynamics. This shift is minimally affected by cerebellar perturbation and significantly larger than the load response in the spinal motoneuron population. This latent representation may enable motor cortex to generate appropriate commands when a voluntary movement must be integrated with an ongoing, spinally-generated rhythm.

The COVID-19 pandemic forced higher education to develop new strategies to meet the needs of students. One of the most critical issues was delivering an online experience in undergraduate Biology laboratories aimed at meeting hands-on outcomes, specifically within the health sciences. Hybrid lab group models in which students rotated between in-person and online attendance represented one such option. A student population of 71 at Minnesota State University, Mankato was studied to observe the effects on student exam scores. Negative student perceptions were made clear in course feedback surveys, and students that attended 2+ labs via Zoom preceding an exam were associated with a lower Exam I score compared to peers that only attended 0-1 labs via Zoom. Three class strategies were introduced before Exam II, meant to improve online learning. The Exam II scores saw no overall significant difference between groups, with Multi-Zoom students closing the gap from Exam I. Importantly, Multi-Zoom students that utilized the class strategies significantly outperformed Multi-Zoom students that that did not use them. Although any classroom results represent a mix of variables, the risks of online labs intended to assess hands-on and data interpretation outcomes of Human Physiology can be detrimental when targeted strategies are absent.

During aging, microglia produce inflammatory factors, show reduced tissue surveillance, altered interactions with synapses, and prolonged responses to CNS insults, positioning these cells to have profound impact on the function of nearby neurons. We and others recently showed that microglial attributes differ significantly across brain regions in young adult mice. However, the degree to which microglial properties vary during aging is largely unexplored. Here, we analyze and manipulate microglial aging within the basal ganglia, brain circuits that exhibit prominent regional microglial heterogeneity and where neurons are vulnerable to functional decline and neurodegenerative disease. In male and female mice, we demonstrate that VTA and SNc microglia exhibit unique and premature responses to aging, compared with cortex and NAc microglia. This is associated with localized VTA/SNc neuroinflammation that may compromise synaptic function as early as middle age. Surprisingly, systemic inflammation, local neuron death, and astrocyte aging do not appear to underlie these early aging responses of VTA and SNc microglia. Instead, we found that microglial lysosome status was tightly linked to early aging of VTA microglia. Microglial ablation/repopulation normalized VTA microglial lysosome swelling and suppressed increases in VTA microglial density during aging. In contrast, CX3CR1 receptor KO exacerbated VTA microglial lysosome rearrangements and VTA microglial proliferation during aging. Our findings reveal a previously unappreciated regional variation in onset and magnitude of microglial proliferation and inflammatory factor production during aging and highlight critical links between microglial lysosome status and local microglial responses to aging.SIGNIFICANCE STATEMENT Microglia are CNS cells that are equipped to regulate neuronal health and function throughout the lifespan. We reveal that microglia in select brain regions begin to proliferate and produce inflammatory factors in late middle age, months before microglia in other brain regions. These findings demonstrate that CNS neuroinflammation during aging is not uniform. Moreover, they raise the possibility that local microglial responses to aging play a critical role in determining which populations of neurons are most vulnerable to functional decline and neurodegenerative disease.

Leaf-level gas exchange data support the mechanistic understanding of plant fluxes of carbon and water. These fluxes inform our understanding of ecosystem function, are an important constraint on parameterization of terrestrial biosphere models, are necessary to understand the response of plants to global environmental change, and are integral to efforts to improve crop production. Collection of these data using gas analyzers can be both technically challenging and time consuming, and individual studies generally focus on a small range of species, restricted time periods, or limited geographic regions. The high value of these data is exemplified by the many publications that reuse and synthesize gas exchange data, however the lack of metadata and data reporting conventions make full and efficient use of these data difficult. Here we propose a reporting format for leaf-level gas exchange data and metadata to provide guidance to data contributors on how to store data in repositories to maximize their discoverability, facilitate their efficient reuse, and add value to individual datasets. For data users, the reporting format will better allow data repositories to optimize data search and extraction, and more readily integrate similar data into harmonized synthesis products. The reporting format specifies data table variable naming and unit conventions, as well as metadata characterizing experimental conditions and protocols. For common data types that were the focus of this initial version of the reporting format, i.e., survey measurements, dark respiration, carbon dioxide and light response curves, and parameters derived from those measurements, we took a further step of defining required additional data and metadata that would maximize the potential reuse of those data types. To aid data contributors and the development of data ingest tools by data repositories we provided a translation table comparing the outputs of common gas exchange instruments. Extensive consultation with data collectors, data users, instrument manufacturers, and data scientists was undertaken in order to ensure that the reporting format met community needs. The reporting format presented here is intended to form a foundation for future development that will incorporate additional data types and variables as gas exchange systems and measurement approaches advance in the future. The reporting format is published in the U.S. Department of Energy's ESS-DIVE data repository, with documentation and future development efforts being maintained in a version control system.

Data quality control is one of the most time consuming activities within Research Infrastructures (RIs), especially when involving observational data and multiple data providers. In this work we report on our ongoing development of data rogues, a scalable approach to manage data quality issues for observational data within RIs. The motivation for this work started with the creation of the FLUXNET2015 dataset, which includes carbon, water, and energy fluxes plus micrometeorological and ancillary data measured in over 200 sites around the world. To create an uniform dataset, including derived data products, extensive work on data quality control was needed. The unpredictable nature of observational data quality issues makes the automation of data quality control inherently difficult. Developed based on this experience, the data rogues methodology allows for increased automation of quality control activities by systematically identifying, cataloging, and documenting implementations of solutions to data issues. We believe this methodology can be extended and applied to others domains and types of data, making the automation of data quality control a more tractable problem.

As a result of climate change warmer temperatures are projected through the 21st century and are already increasing above modelled predictions. Apart from increases in the mean, warm/hot temperature extremes are expected to become more prevalent in the future, along with an increase in the frequency of droughts. It is crucial to better understand the response of terrestrial ecosystems to such temperature extremes for predicting land-surface feedbacks in a changing climate. While land-surface feedbacks in drought conditions and during heat waves have been reported from Europe and the US, direct observations of the impact of such extremes on the carbon and water cycles in Australia have been lacking. During the 2012/2013 summer, Australia experienced a record-breaking heat wave with an exceptional spatial extent that lasted for several weeks. In this study we synthesised eddy-covariance measurements from seven woodlands and one forest site across three biogeographic regions in southern Australia. These observations were combined with model results from BIOS2 (Haverd et al., 2013a, b) to investigate the effect of the summer heat wave on the carbon and water exchange of terrestrial ecosystems which are known for their resilience toward hot and dry conditions. We found that water-limited woodland and energy-limited forest ecosystems responded differently to the heat wave. During the most intense part of the heat wave, the woodlands experienced decreased latent heat flux (23ĝ€% of background value), increased Bowen ratio (154ĝ€%) and reduced carbon uptake (60ĝ€%). At the same time the forest ecosystem showed increased latent heat flux (151ĝ€%), reduced Bowen ratio (19ĝ€%) and increased carbon uptake (112ĝ€%). Higher temperatures caused increased ecosystem respiration at all sites (up to 139ĝ€%). During daytime all ecosystems remained carbon sinks, but carbon uptake was reduced in magnitude. The number of hours during which the ecosystem acted as a carbon sink was also reduced, which switched the woodlands into a carbon source on a daily average. Precipitation occurred after the first, most intense part of the heat wave, and the subsequent cooler temperatures in the temperate woodlands led to recovery of the carbon sink, decreased the Bowen ratio (65ĝ€%) and hence increased evaporative cooling. Gross primary productivity in the woodlands recovered quickly with precipitation and cooler temperatures but respiration remained high. While the forest proved relatively resilient to this short-Term heat extreme the response of the woodlands is the first direct evidence that the carbon sinks of large areas of Australia may not be sustainable in a future climate with an increased number, intensity and duration of heat waves.
