Abstract Background High-frequency ventilation (HFV) is commonly used in neonatal intensive care units to provide respiratory support for critically ill neonates. Currently, there is no standardized procedure for weaning from HFV. Two commonly used strategies are transitioning from HFV to conventional mechanical ventilation (CMV) before extubation (HFV-CMV) and extubation after decreasing mean airway pressure during HFV (HFV-HFV). The impact of these strategies on neonatal cerebral oxygenation and hemodynamics remains incompletely understood. Methods We will conduct a prospective, single-center, randomized controlled trial to investigate the effects of two different HFV weaning strategies (HFV-CMV, HFV-HFV) on neonatal cerebral oxygenation and hemodynamics. The patients enrolled in the trial will be randomly allocated to either the HFV-CMV group or the HFV-HFV group in a 1:1 ratio. The primary outcome will be cerebral oxygen saturation (ScO2) before and after the intervention. Second outcomes are cerebral fractional tissue oxygen extraction, heart rate, blood pressure, and the incidence and severity of intraventricular hemorrhage and periventricular leukomalacia. We hypothesize that HFV-CMV results in positive impact on neonatal cerebral oxygenation compared to HFV-HFV. This study aims to identify a better weaning strategy for HFV and contribute evidence-based data to enhance its clinical application in newborns, potentially improving the care and outcomes for neonates receiving HFV. Discussion This study aims to assessing the impact of different HFV weaning strategies on neonatal cerebral oxygenation and hemodynamics, as well as the relationship between the duration of HFV under different strategies and neurological complications, to identify better weaning methods for HFV. We hope to contribute evidence-based data to enhance clinical application of HFV in newborns, potentially improving the care and outcomes for neonates receiving HFV. Abstract Background Rice body formation is an uncommon, nonspecific inflammatory process. Certain clinical features, such as chronic synovitis associated with rheumatoid arthritis, tuberculous arthritis, and osteoarthritis, can induce a non-specific response that may lead to the development of rice bodies. Currently, the etiological and prognostic significance of rice bodies remains unknown. Magnetic resonance imaging (MRI) is the preferred diagnostic imaging modality for evaluating rice body formation. Case presentation The patient, a 44-year-old female, presented with bursitis of the hip joint and the presence of numerous rice bodies. This case is exceptionally rare and unusual as it involves a combination of hip dysplasia and pelvic fracture. The patient underwent a one-stage resection of the rice body, internal fixation of the acetabular fracture, and two-stage total hip arthroplasty, resulting in an immediate improvement in her symptoms. We reviewed the relevant literature and observed that the majority of rice bodies were predominantly found within the capsules of the shoulder and knee joints, while instances of hip joint rice body bursitis were relatively infrequent. To our knowledge, there have been no previous reports documenting a case of hip joint rice body bursitis in conjunction with hip dysplasia and pelvic fracture. Conclusion This paper presents the rare case of hip joint rice body bursitis combined with hip dysplasia and pelvic fracture, and is one of the highlights.

In Chinese culture, Confucian thought about interpersonal relationships and family ethics hold significant historical and cultural value. These ideas not only affect individual moral cultivation, but also play a key role in the entire society’s harmony and stability. Beginning with Confucius’ views on concepts, such as friendship and order, the paper examines the concept of interpersonal relationships in Confucius’ thought from five aspects based on the five constant virtues—benevolence, righteousness, propriety, wisdom and sincerity. Based on the five constant virtues of interpersonal relationships, this paper further summarizes and analyzes Confucius’ family ethics concepts and reflects on them. Through reflection and outline of Confucius’ thoughts, we can easily grasp the foundation of Chinese traditional culture and give helpful reference and motivation to the construction of ethics and morality in current culture.

The significant identification of Beclin-1’s function in regulating autophagy flow signified a significant progression in our understanding of cellular operations. Beclin-1 acts as a scaffold for forming the PI3KC3 complex, controlling autophagy and cellular trafficking processes in a complicated way. This intricate protein has garnered considerable attention due to its substantial impact on the development of tumors. Strong evidence indicates Beclin-1 plays a critical role in controlling autophagy in various human cancer types and its intricate connection with apoptosis and ferroptosis. The potential of Beclin-1 as a viable target for cancer therapy is highlighted by its associations with key autophagy regulators such as AMPK, mTOR, and ATGs. Beclin-1 controls the growth and dissemination of tumors by autophagy. It also affects how tumors react to therapies such as chemotherapy and radiation therapy. The role of Beclin-1 in autophagy can influence apoptosis, depending on whether it supports cell survival or leads to cell death. Beclin-1 plays a crucial role in ferroptosis by increasing ATG5 levels, which in turn promotes autophagy-triggered ferroptosis. Finally, we analyzed the possible function of Beclin-1 in tumor immunology and drug sensitivity in cancers. In general, Beclin-1 has a significant impact on regulating autophagy, offering various potentials for medical intervention and altering our understanding of cancer biology.

Hepatocellular carcinoma (HCC) is the predominant form of liver cancer and ranks as the third leading cause of cancer-related mortality globally. The liver performs a wide range of tasks and is the primary organ responsible for metabolizing harmful substances and foreign compounds. Oxidative stress has a crucial role in growth and improvement of hepatocellular carcinoma (HCC). Nuclear factor erythroid 2 (1)-related factor 2 (Nrf2) is an element that regulates transcription located in the cytoplasm. It controls the balance of redox reactions by stimulating the expression of many genes that depend on antioxidant response elements. Nrf2 has contrasting functions in the normal, healthy liver and HCC. In the normal liver, Nrf2 provides advantageous benefits, while in HCC it promotes harmful effects that support the growth and survival of HCC. Continuous activation of Nrf2 has been detected in HCC and promotes its advancement and aggressiveness. In addition, Activation of Nrf2 may lead to immune evasion, weakening the immune cells’ ability to attack tumors and thereby promoting tumor development. Furthermore, chemoresistance in HCC, which is considered a form of stress response to chemotherapy medications, significantly impedes the effectiveness of HCC treatment. Stress management is typically accomplished by activating specific signal pathways and chemical variables. One important element in the creation of chemoresistance in HCC is nuclear factor-E2-related factor 2 (Nrf2). Nrf2 is a transcription factor that regulates the activation and production of a group of genes that encode proteins responsible for protecting cells from damage. This occurs through the Nrf2/ARE pathway, which is a crucial mechanism for combating oxidative stress within cells.

Abstract The intensifying energy crisis has made it urgent to develop robust and reliable next‐generation energy systems. Except for conventional large‐scale energy sources, the imperceptible and randomly distributed energy embedded in daily life awaits comprehensive exploration and utilization. Harnessing the latent energy has the potential to facilitate the further evolution of soft energy systems. Compared with rigid energy devices, flexible energy devices are more convenient and suitable for harvesting and storing energy from dynamic and complex structures such as human skin. Stretchable conductors that are capable of withstanding strain (≫1%) while sustaining stable conductive pathways are prerequisites for realizing flexible electronic energy devices. Therefore, understanding the characteristics of these conductors and evaluating the feasibility of their fabrication strategies are particularly critical. In this review, various preparation methods for stretchable conductors are carefully classified and analyzed. Furthermore, recent progress in the application of energy harvesting and storage based on these conductors is discussed in detail. Finally, the challenges and promising opportunities in the development of stretchable conductors and integrated flexible energy devices are highlighted, seeking to inspire their future research directions.

Abstract Purpose To compare the clinical efficacy of arthroscopic TightRope loop titanium button and clavicular hook plate in the treatment of acromioclavicular joint (ACJ) dislocation of Rockwood III/IV. Methods A retrospective analysis of patients with ACJ dislocation in our hospital from January 2018 to December 2020 was conducted. The patients were assigned to be treated with arthroscopic TightRope loop titanium button (TR group) or clavicular hook plate (HP group). The preoperative, intraoperative and postoperative data and imaging findings of the two groups were compared. Results A total of 58 eligible patients were enrolled in this study. Compared with HP group, TR group had shorter incision length and less blood loss during operation. Postoperative follow-up ranged from 12 to 24 months (mean 15.4 months). At 6 months and 12months postoperatively, compared with HP group, TR group had lower VAS and higher CMS, and the difference was statistically significant. At 12 months postoperatively, compared with HP group, TR group had lower ACJ gap and coracoclavicular joint(CCJ) distance, and the difference was statistically significant.In HP group, there were 3 cases of subacromial impact, 1 case of redislocation, 2 cases of traumatic arthritis and 2 cases of wound infection. There was 1 case of redislocation in TR group. Conclusions Compared with clavicular hook plate, arthroscopic TightRope loop titanium button is minimally invasive, safe and effective in the treatment of ACJ dislocation, and has a good trend in clinical application.

Abstract Solanum lycopersicum, which is commonly known as tomato, not only serves as a dietary fruit vegetable but also a functional food that protects humans against chronic degenerative illnesses such as diabetes microvascular complications, viral diseases, and cancers. The nutritional benefit of tomato is largely attributed to its phytonutrient composition. The influence of post‐harvest processing methods, digestion process, and compounds interaction on the bio‐accessibility of phytonutrients are key aspects to consider when investigating the potential impact of tomato on human health. Besides, information on the underlying mechanism of tomato phytochemicals’ interaction and pharmacological action with other food components could pave the way for novel functional research study. To date, the impact of different processing methods and digestion effect on the extraction of phytochemicals from tomato showed contradictory findings. Although thermal processing and the application of spices and/or condiments like olive oil on tomato are common practices in domestic cooking, understanding of the changes in bioactivity in the processed tomato products is still limited. This paper aimed to summarize the current post‐harvest processing methods that maximize the health benefits of tomato and offer additional insight into the development of tomato and its related products with optimal pharmacological values. This study also reviewed several major phytochemicals in tomato with functional values and their possible interaction with other food components. The impact of in vitro digestion on the bio‐accessibility and functional activity of tomato phytochemicals were critically discussed, alongside selected post‐harvest processing methods that could maximize the health benefit and commercial values of tomato.

Abstract Background Health problems in neonates with gestational age (GA) ≥ 32 weeks remain a major medical concern. Respiratory distress (RD) is one of the common reasons for admission of neonates with GA ≥ 32 weeks. Abstract Background Mechanical ventilation (MV) is essential in the management of critically ill neonates, especially preterm infants. However, inappropriate or prolonged use of invasive MV may result in ventilator-associated lung injury. A systemic review comparing pressure control ventilation (PCV) with volume-targeted ventilation mode (VTV) approved that VTV reduces the incidence of death or bronchopulmonary dysplasia (BPD) in neonates; however, this study did not analyze subgroups of very low birthweight (VLBW) infants. Therefore, the aim of this study was to compare the use of VTV and PCV in VLBW infants and to provide clinical evidence for reducing mortality and complications of MV in VLBW infants. Method A single-center randomized controlled trial will be performed. All eligible infants will be randomized and assigned to either VTV or PCV group with 1:1 ratio using sealed envelopes. Death or BPD at 36 weeks’ postmenstrual age will be used as the primary outcome. Secondary outcomes include BPD, death, length of invasive MV, noninvasive mechanical ventilation, and oxygen use, length of hospital stay, failure of conventional MV, rate of using high-frequency oscillatory ventilation (HFOV) as rescue therapy, rate of reintubation within 48 h, and hospital expenses. Discussion Systemic review suggested that VTV decreases the incidence of death or BPD in neonates compared to PLV; however, this study did not specifically analyze subgroups of VLBW infants. We designed this single-center randomized controlled trials (RCT) to add a significant contribution regarding the benefits of VTV for VLBW patients.

Abstract Organic imine cage compounds have found a variety of different applications in several fields in materials science. To design tailor-made cages for corresponding applications, synthetic approaches to cages with tunable functionalities, sizes and shapes have to be found. It is well established that promoting the balance of nutrients and plant secondary metabolites (PSM) by feeding diverse forage physiologically improves ruminant production. However, the underlying mechanism remains unclear. To investigate the physiological mechanism related to the improvement of physiological stress tolerance, ruminants were fed diverse forage. Oxidative stress markers were quantified, and serum metabolomics was performed. Six crossbred Shiba wethers (32.8 ± 9.2 kg BW) were arranged in a replicated 3 × 3 Latin square design. The treatments were feeding only Sudan grass hay (100% SDN); feeding a mixture of Sudan grass and alfalfa hay (70:30, SDN-ALF); and feeding a mixture of Sudan grass, timothy grass, and alfalfa hay (35:35:30; SDN-TMT-ALF). Each diet group was fed its specific diet for 21 days with a 14-day adaptation period. Feed intake and digestibility, blood biochemistry, total antioxidant capacity (TAC), and superoxide dismutase (SOD) were analysed. Abstract Background 3D printing technology has become a research hotspot in the field of scientific research because of its personalized customization, maneuverability and the ability to achieve multiple material fabrications. The focus of this study is to use 3D printing technology to customize personalized poly L-lactic acid (PLLA) porous screws in orthopedic plants and to explore its effect on tendon-bone healing after anterior cruciate ligament (ACL) reconstruction. Methods Preparation of PLLA porous screws with good orthogonal pore structure by 3D printer. The hydroxyapatite (HA) was adsorbed on porous screws by electrostatic layer-by-layer self-assembly (ELSA) technology, and PLLA-HA porous screws were prepared. The surface and spatial morphology of the modified screws were observed by scanning electron microscopy (SEM). The porosity of porous screw was measured by liquid displacement method. Thirty New Zealand male white rabbits were divided into two groups according to simple randomization. Autologous tendon was used for right ACL reconstruction, and porous screws were inserted into the femoral tunnel to fix the transplanted tendon. PLLA group was fixed with porous screws, PLLA-HA group was fixed with HA modified porous screws. At 6 weeks and 12 weeks after surgery, 5 animals in each group were sacrificed randomly for histological examination. The remaining 5 animals in each group underwent Micro-CT and biomechanical tests. Results The pores of PLLA porous screws prepared by 3D printer were uniformly distributed and connected with each other, which meet the experimental requirements. HA was evenly distributed in the porous screw by ELSA technique. Histology showed that compared with PLLA group, mature bone trabeculae were integrated with grafted tendons in PLLA-HA group. Micro-CT showed that the bone formation index of PLLA-HA group was better than that of PLLA group. The new bone was uniformly distributed in the bone tunnel along the screw channel. Biomechanical experiments showed that the failure load and stiffness of PLLA-HA group were significantly higher than those of PLLA group. Conclusions The 3D printed PLLA porous screw modified by HA can not only fix the grafted tendons, but also increase the inductivity of bone, promote bone growth in the bone tunnel and promote bone integration at the tendon-bone interface. The PLLA-HA porous screw is likely to be used in clinic in the future.

Abstract Background Studies focusing on the relationship between gastric volume and facemask oxygenation without ventilation during apnea in anesthesia induction are scarce. This study compared the change in gastric volume during apnea in anesthesia induction using facemask ventilation and facemask oxygenation without ventilation in adults undergoing laparoscopic surgery. Methods In this prospective, randomized, double-blinded trial, 70 adults undergoing laparoscopic surgery under general anesthesia were divided into two groups to receive facemask oxygenation with and without ventilation for 60 seconds after loss of consciousness. Before anesthesia induction and after endotracheal intubation, the gastric antral cross-sectional area was measured with ultrasound imaging. Arterial blood gases were tested at baseline (T1), after preoxygenation (T2), after loss of consciousness (T3), and before and after endotracheal intubation (T4 and T5, respectively). Results Sixty patients were included (ventilation n = 30; non ventilation n = 30, 10 patients were excluded). The median [IQR] change of gastric antral cross-sectional area in ventilation group was significantly higher than in non ventilation group (0.83 [0.20 to 1.54] vs. 0.10 [− 0.11 to 0.56] cm2, P = 0.001). Local anesthetics are used clinically for the control of postoperative pain management. This study aimed to develop chitosan (CS) with genipin (GP) hydrogels as the hydrophilic lipid shell loaded poly(ε-caprolactone) (PC) nanocapsules as the hydrophobic polymeric core composites (CS-GP/PC) to deliver bupivacaine (BPV) for the prolongation of anesthesia and pain relief. The swelling ratio, in vitro degradation, and rheological properties enhancement of CS-GP/PC polymeric hydrogel. The incorporation of PC nanocapsules into CS-GP hydrogels was confirmed by SEM, FTIR, and XRD analysis. Scanning electron microscopy results demonstrated that the CS-GP hydrogels and CS-GP/PC polymeric hydrogels have a porous structure, the pore dimensions being non-uniform with diameters between 25 and 300 μm. The in vitro drug release profile of CS-GP/PC polymeric hydrogel has been achieved 99.2 ± 1.12% of BPV drug release in 36 h. Cellular viability was evaluated using the CCK-8 test on 3T3 fibroblast cells revealed that the obtained CS-GP/PC polymeric hydrogel with BPV exhibited no obvious cytotoxicity. The CS-GP/PC polymeric hydrogel loaded with BPV showed significant improvement in pain response compared to the control group animals for at least 7 days. When compared with BPV solution, CS-GP hydrogel and CS-GP/PC polymeric hydrogel improved the skin permeation of BPV 3-fold and 5-fold in 24 h, respectively. In vitro and in vivo results pointed out PC nanocapsules loaded CS-GP hydrogel can act as effective drug carriers, thus prolonging and enhancing the anesthetic effect of BPV. Histopathological results demonstrated the excellent biodegradability and biocompatibility of the BPV-loaded CS-GP/PC polymeric hydrogel system on 7, 14, and 21 days without neurotoxicity.HIGHLIGHTS Preparation and characterization of CS-GP/PC polymeric hydrogel system. Background: Musculoskeletal pain is the most common form of chronic pain. Anxiety increases pain intensity and appears to have a major impact on the prevalence and also disability of musculoskeletal pain in women. We examined the effect of dexmedetomidine (DEX) on anxiety-like behaviors associated with musculoskeletal pain and the underlying molecular mechanism in female mice. Methods: Musculoskeletal pain was induced by injection of acidified saline into the gastrocnemius muscle in adult female mice, and the von Frey filament test is used to measure mechanical sensitivity. DEX and EX527 (SIRT1 inhibitor) were administered after modelling. Behavioral tests were used for anxiety and motor activity tests. SIRT1, p53 and acetyl-p53 were quantified by Western blot. Results: Adult female mice with musculoskeletal pain exhibit increased fear-like behavior by reducing SIRT1 expression in the medial prefrontal cortex (mPFC). While administration of DEX was able to alleviate mechanical hypersensitivity and anxiety-like behaviors by blocking SIRT1 decline and acetyl-p53 upregulation in mPFC, EX527 inhibited acetyl-p53 rise and reversed the antinociceptive and anxiolytic effects of DEX. Conclusion: DEX may alleviate anxiety-like behaviors in mice with musculoskeletal pain via the SIRT1/p53 axis. These results suggest that DEX may have a potential therapeutic role in musculoskeletal pain-induced anxiety.

According to the structural characteristics and the surface quality requirements of aspheric optical elements, a hemispherical head polishing head with radial magnetized permanent magnet as rotating magnetic field source is designed and manufactured on the basis of magnetic compound fluid (MCF) polishing. Firstly, through Ansoft Maxwell magnetic field simulation, the distributions of magnetic field around each magnet with different shapes, sizes and eccentricities are analyzed and compared, and the cylindrical magnet with diameter of 10.0 mm, height of 5.0 mm, eccentricity of 2.5 mm and radial magnetization is selected. Secondly, by observing and comparing the behaviors of MCF with different compositions, formula and supplies on the polishing head, the compositions of MCF polishing slurry is determined. Finally, the aspheric PMMA workpiece is polished with the prepared MCF polishing slurry and the self-made polishing head. After polishing for 15 min, the surface quality of the PMMA workpiece is significantly improved, its surface accuracy Rq is decreased from 0.703 μm to 2.433 nm, and the surface roughness Ra is reduced from 0.545 μm to 1.786 nm, indicating that the developed polishing head can realize the nano-polishing of aspheric workpiece.

Highlights The processing methods of functional polymer composites (FPCs) are systematically summarized in “Toolbox”. The relationship of processing method-structure-property is discussed and the selection and combination of tools in processing among different FPCs are analyzed. A promising prospect is provided regarding the design principle for high performance FPCs for further investigation. Abstract Functional polymer composites (FPCs) have attracted increasing attention in recent decades due to their great potential in delivering a wide range of functionalities. These functionalities are largely determined by functional fillers and their network morphology in polymer matrix. In recent years, a large number of studies on morphology control and interfacial modification have been reported, where numerous preparation methods and exciting performance of FPCs have been reported. Despite the fact that these FPCs have many similarities because they are all consisting of functional inorganic fillers and polymer matrices, review on the overall progress of FPCs is still missing, and especially the overall processing strategy for these composites is urgently needed. Herein, a “Toolbox” for the processing of FPCs is proposed to summarize and analyze the overall processing strategies and corresponding morphology evolution for FPCs. From this perspective, the morphological control methods already utilized for various FPCs are systematically reviewed, so that guidelines or even predictions on the processing strategies of various FPCs as well as multi-functional polymer composites could be given. This review should be able to provide interesting insights for the field of FPCs and boost future intelligent design of various FPCs.

Research on water confinement in small hydrophobic pores remains scarce because of a preconception that small hydrophobic pores repulse water molecules. Here, the authors demonstrate water confinement across hydrophobic microporous channels in crystalline covalent organic frameworks.

With the shortage of fossil energy, more attention is paid to two-phase flow due to the high heat transfer efficiency and uniform surface temperature. However, flow distribution in parallel channels is more complex than that of single-phase flow, which usually leads to heat transfer deterioration. Thus, a 3D numerical model of flow distribution and heat transfer of two-phase flow in parallel channels is developed and validated in the present study. The effect of channel cross section was investigated, and three typical cross section shapes are discussed. The results show that the flow distribution in parallel channels of the triangular cross section is the worst, and of the circular section is the best. Similarly, the heat transfer performance of the circular channel is the best because the bubbles are difficult to separate at the junction of adjacent channel surfaces in the rectangular and triangular channel. Besides, flow distribution in the U-type heat exchanger is better than that in the Z-type heat exchanger.

Abstract Background To investigate osteointegration at the graft-bone interface and the prevention of osteoarthritis after anterior cruciate ligament (ACL) reconstruction using a silk-collagen scaffold with both ends modified by hydroxyapatite (HA) in a rabbit model. Methods The HA/silk-collagen scaffold was fabricated using a degummed, knitted silk scaffold, collagen I matrix, and simulated body fluid (SBF). The HA/silk-collagen scaffold was rolled up to make a graft for replacing the native ACL in the experimental group (HA group), and the silk-collagen scaffold was used in the control (S group). All specimens were harvested at 16 weeks postoperatively to evaluate graft-bone healing and osteoarthritis prevention. Results Histological staining revealed the massive formation of more mature bone at the tendon-bone interface, and immunohistochemistry staining revealed more collagen I and osteocalcin deposition in the HA group than in the S group. Higher signals indicating more bone mineral formation were detected in the HA group than in the S group, which was consistent with the results of biomechanical testing. Better osteoarthritis prevention was also observed in the HA group, indicating a more stable knee joint in the HA group than in the S group. Conclusion The HA/silk-collagen scaffold promotes osteointegration at the tendon-bone interface after ACL reconstruction and has great potential for clinical applications.

Abstract Background This study aimed to investigate the correlation of chaperonin containing t-complex polypeptide 1 subunit 6A (CCT6A) expression with clinicopathological features and survival profiles in non-small cell lung carcinoma (NSCLC) patients. Methods A total of 381 NSCLC patients with Tumor, Node, Metastasis (TNM) stage I–IIIA who underwent tumor resection were retrospectively screened. Immunohistochemistry staining and semi-quantitative scoring were used to evaluate CCT6A expression in tumor and adjacent tissues. Clinicopathological features were retrieved. Disease-free survival (DFS) and overall survival (OS) were calculated. Results CCT6A expression was elevated in tumor tissue (CCT6A high 47.5% vs. low 52.5%) compared with adjacent tissue (CCT6A high 30.4% vs. low 69.6%) (P < 0.001), and ROC curve displayed that CCT6A could distinguish tumor tissue from adjacent tissue. Moreover, tumor CCT6A high expression was associated with lymph node metastasis (P = 0.001), elevated TNM stage (P = 0.002), and abnormal carcinoembryonic antigen (P = 0.022). Kaplan–Meier curves displayed that tumor CCT6A high expression was negatively correlated with DFS and OS (all P < 0.001). Cox’s regression analysis disclosed that tumor CCT6A high expression independently predicted worse DFS (P < 0.001) (hazard ratio (HR) 1.659 (95% confidence interval (CI) 1.318–2.089)), and OS (P < 0.001) (HR 1.779 (95%CI 1.378–2.298)). Conclusions CCT6A may present some clinical value in the management of NSCLC.

Abstract Background Vestibular migraine has recently been recognized as a novel subtype of migraine. However, the mechanism that relate vestibular symptoms to migraine had not been well elucidated. Thus, the present study investigated vestibular dysfunction in a rat model of chronic migraine (CM), and to dissect potential mechanisms between migraine and vertigo. Methods Rats subjected to recurrent intermittent administration of nitroglycerin (NTG) were used as the CM model. Migraine- and vestibular-related behaviors were analyzed. Immunofluorescent analyses and quantitative real-time polymerase chain reaction were employed to detect expressions of c-fos and calcitonin gene-related peptide (CGRP) in the trigeminal nucleus caudalis (TNC) and vestibular nucleus (VN). Morphological changes of vestibular afferent terminals was determined under transmission electron microscopy. FluoroGold (FG) and CTB-555 were selected as retrograde tracers and injected into the VN and TNC, respectively. Lentiviral vectors comprising CGRP short hairpin RNA (LV-CGRP) was injected into the trigeminal ganglion. Results CM led to persistent thermal hyperalgesia, spontaneous facial pain, and prominent vestibular dysfunction, accompanied by the upregulation of c-fos labeling neurons and CGRP immunoreactivity in the TNC (c-fos: vehicle vs. CM = 2.9 ± 0.6 vs. 45.5 ± 3.4; CGRP OD: vehicle vs. CM = 0.1 ± 0.0 vs. 0.2 ± 0.0) and VN (c-fos: vehicle vs. CM = 2.3 ± 0.8 vs. 54.0 ± 2.1; CGRP mRNA: vehicle vs. CM = 1.0 ± 0.1 vs. 2.4 ± 0.1). Furthermore, FG-positive neurons was accumulated in the superficial layer of the TNC, and the number of c-fos+/FG+ neurons were significantly increased in rats with CM compared to the vehicle group (vehicle vs. CM = 25.3 ± 2.2 vs. 83.9 ± 3.0). Meanwhile, CTB-555+ neurons dispersed throughout the VN. The structure of vestibular afferent terminals was less pronounced after CM compared with the peripheral vestibular dysfunction model. In vivo knockdown of CGRP in the trigeminal ganglion significantly reduced the number of c-fos labeling neurons (LV-CGRP vs. LV-NC = 9.9 ± 3.0 vs. 60.0 ± 4.5) and CGRP mRNA (LV-CGRP vs. LV-NC = 1.0 ± 0.1 vs. 2.1 ± 0.2) in the VN, further attenuating vestibular dysfunction after CM. Conclusions These data demonstrates the possibility of sensitization of vestibular nucleus neurons to impair vestibular function after CM, and anti-CGRP treatment to restore vestibular dysfunction in patients with CM.

Abstract Background MicroRNA-125b (miR-125b) is downregulated in human cutaneous squamous cell carcinoma (CSCC). However, its function in CSCC has yet to be extensively explored. Here, we analyze the relationship between signal transducer and activator of transcription 3 (STAT3) and miR-125b in CSCC. Methods Western blotting and quantitative RT-PCR were used to determine the expression of the miR-125b–STAT3 axis in human CSCC tissues and cell lines. The direct regulatory effect of miR-125b on STAT3 expression was assessed using a luciferase reporter gene assay and RNA immunoprecipitation assay. The MTT assay and flow cytometry were used to determine the role of the miR-125b–STAT3 axis in CSCC cell proliferation and apoptosis. Results MiR-125b expression levels were significantly lower in CSCC cell lines and tissues than in normal cell lines and tissues. STAT3 was identified as the direct target of miR-125b. Upregulation of miR-125b and downregulation of STAT3 suppressed cell proliferation and promoted cell apoptosis. Cyclin D1 and Bcl2 were identified as the downstream targets of the miR-125–STAT3 axis. Conclusions Our findings indicate that miR-125b acts as a tumor suppressor in CSCC by targeting the STAT3 pathway. This observation increases our understanding of the molecular mechanisms of CSCC. Therapies aimed at activating miR-125b or inhibiting STAT3 signaling should be explored as potential treatments for CSCC.

This paper proposes an unsupervised way for Phonocardiogram (PCG) analysis, which uses a revised auto encoder based on distribution density estimation in the latent space. Auto encoders especially Variational Auto-Encoders (VAEs) and its variant β−VAE are considered as one of the state-of-the-art methodologies for PCG analysis. VAE based models for PCG analysis assume that normal PCG signals can be represented by latent vectors that obey a normal Gaussian Model, which may not be necessary true in PCG analysis. This paper proposes two methods DBVAE and DBAE that are based on estimating the density of latent vectors in latent space to improve the performance of VAE based PCG analysis systems. Examining the system performance with PCG data from the a single domain and multiple domains, the proposed systems outperform the VAE based methods. The representation of normal PCG signals in the latent space is also investigated by calculating the kurtosis and skewness where DBAE introduces normal PCG representation following Gaussian-like models but DBVAE does not introduce normal PCG representation following Gaussian-like models.

In this work, XLPE/MWCNT and XLPE/CB nanocomposites have been prepared in order to investigate AC electric field and water effects on electrical aging of XLPE. The mechanical, AC breakdown strength and AC conductivity were tested and the morphologies after 30 days electrical aging were observed using an optical microscope. The results showed that all samples exhibit excellent insulation properties and mechanical properties. Compared with CB addition, the MWCNT composites exhibit better resistance to electrical aging, with the length of electrical aging-induced microcracks in the MWCNT blends decreasing from 104 to 22 µm, and the width decreasing from 87 to 17 µm, which means a reduction of ∼80% compared of values for neat XLPE in both length and width. However, the XLPE/CB composites have a tendency to promote electrical aging. The mechanism is revealed by comparing the results of the fibrous MWCNTs with the spherical CBs.

Anterior cruciate ligament (ACL) reconstruction was realized using a combination of bone mesenchymal stem cells (BMSCs) and silk–collagen scaffold, and an in vivo evaluation of this combination was performed. By combining type I collagen and degummed silk fibroin mesh, silk–collagen scaffolds were prepared to simulate ligament components. BMSCs isolated from bone marrow of rabbits were cultured for a homogenous population and seeded on the silk–collagen scaffold. In the scaffold and BMSC (S/C) group, scaffolds were seeded with BMSCs for 72 h and then rolled and used to replace the ACL in 20 rabbits. In the scaffold (S) group, scaffolds immersed only in culture medium for 72 h were used for ACL reconstruction. Specimens were collected at 4 and 16 weeks postoperatively to assess ligament regeneration and bone integration. HE and immunohistochemical staining (IHC) were performed to assess ligament regeneration in the knee cavity. To assess bone integration at the graft–bone interface, HE, Russell–Movat staining, micro-CT, and biomechanical tests were performed. After 4 weeks, vigorous cell proliferation was observed in the core part of the scaffold in the S/C group, and a quantity of fibroblast-like cells and extracellular matrix (ECM) was observed in the center part of the graft at 16 weeks after surgery. Abstract Investigating the pharmacology is key to the modernization of Chinese Medicine (CM) formulas. However, identifying which are the active compound(s) of CM formulas, which biological entities they target, and through which signaling pathway(s) they act to modify disease symptoms, are still difficult tasks for researchers, even when equipped with an arsenal of advanced modern technologies. Multiple approaches, including network pharmacology, pharmaco-genomics, -proteomics, and -metabolomics, have been developed to study the pharmacology of CM formulas. They fall into two general categories in terms of how they tackle a problem: bottom-up and top-down. In this article, we compared these two different approaches in several dimensions by using the case of MaZiRenWan (MZRW, also known as Hemp Seed Pill), a CM herbal formula for functional constipation. Multiple hypotheses are easy to be proposed in the bottom-up approach (e.g. network pharmacology); but these hypotheses are usually false positives and hard to be tested. In contrast, it is hard to suggest hypotheses in the top-down approach (e.g. pharmacometabolomics); however, once a hypothesis is proposed, it is much easier to be tested. Merging of these two approaches could results in a powerful approach, which could be the new paradigm for the pharmacological study of CM formulas.

In this letter, an Nd3+-doped compound fluorosilicate glass was successfully fabricated with the method of melt-quenching. Under the excitation of a 808 nm laser, enhanced near-infrared photoluminescence emission at the range from 1,065 to 1,140 nm was observed in the glass sample. To characterize its stability and resistance to environmental effects, transmission spectra at the range of NIR-MIR were measured under different environments, including humidity and temperature. In addition, the obtained fluorosilicate glass was also developed as a microsphere resonator by using CO2 laser heating method. When the diameter of the microsphere was controlled at 61.5 μm, coupling with a tapered fiber, single-multimode lasing in the wavelength range λ1,056–1,071 nm was achieved with a low lasing threshold of 1.5 mW. Compared to silica and phosphate glasses, this fluorosilicate glasses have lower phonon energy, which can reduce the probability of non-radiative transitions and improve the photoluminescence efficiency. Therefore, using it as the raw material, the developed microsphere resonator offers a high transition temperature and with a low lasing threshold, which are promising it for high performance sensing and detection applications.

The present study was aimed at investigating the effects of sodium butyrate and sodium β-hydroxybutyrate on lactation and health of dairy cows fed a high-concentrate (HC) diet. Eighty mid-lactation dairy cows with an average milk yield of 33.75 ± 5.22 kg/d were randomly allocated to four groups (n = 20 per group) and were fed either a low-concentrate (LC) diet, a HC diet, the HC diet with 1% sodium butyrate (HCSB), or the HC diet with 1% sodium β-hydroxybutyrate (HCHB). The feeding trial lasted for 7 weeks, with a 2-week adaptation period and a 5-week measurement period, and the trial started from 96 ± 13 d in milk. Sodium butyrate supplementation delayed the decline in milk production and improved milk synthesis efficiency and milk fat content. Additionally, it decreased the proinflammatory cytokines and acute phase proteins (APPs) in plasma, the leucocytes in blood, the somatic cell count (SCC) in milk, and the gene expression of pattern recognition receptors (PRRs) and proinflammatory cytokines in the mammary gland, due to decreasing the contents of bacterial cell wall components (lipopolysaccharide, LPS; peptidoglycan, PGN; and lipoteichoic acid, LTA) in the rumen and plasma, compared with the HC diet. Sodium β-hydroxybutyrate supplementation also improved milk yield, milk synthesis efficiency and milk fat content and partially reduced the adverse effects caused by the HC diet, but it had no effect on decreasing bacterial cell wall components in the rumen and plasma, compared with the HC diet. Collectively, both sodium butyrate and sodium β-hydroxybutyrate mitigated the negative effects of HC diet on lactation and health of dairy cows, with sodium butyrate being more effective than sodium β-hydroxybutyrate.

As a vital power propulsion device, gas turbines have been widely applied in aircraft. However, fly ash is easily ingested by turbine engines, causing blade abrasion or even film hole blockage. In this study, a three-dimensional turbine cascade model is conducted to analyze particle trajectories at the blade leading edge, under a film-cooled protection. A deposition mechanism, based on the particle sticking model and the particle detachment model, was numerically investigated in this research. Additionally, the invasion efficiency of the AGTB-B1 turbine blade cascade was investigated for the first time. The results indicate that the majority of the impact region is located at the leading edge and on the pressure side. In addition, small particles (1 μm and 5 μm) hardly impact the blade’s surface, and most of the impacted particles are captured by the blade. With particle size increasing, the impact efficiency increases rapidly, and this value exceeds 400% when the particle size is 50 μm. Invasion efficiencies of small particles (1 μm and 5 μm) are almost zero, and the invasion efficiency approaches 12% when the particle size is 50 μm.

Negative curvature hollow core fiber (NCHCF) is a promising candidate for sensing applications; however, research on NCHCF based fiber sensors starts only in the recent two years. In this work, an all-fiber interferometer based on an NCHCF structure is proposed for the first time. The interferometer was fabricated by simple fusion splicing of a short section of an NCHCF between two singlemode fibers (SMFs). Both simulation and experimental results show that multiple modes and modal interferences are excited within the NCHCF structure. Periodic transmission dips with high spectral extinction ratio (up to 30 dB) and wide free spectral range (FSR) are produced, which is mainly introduced by the modes coupling between HE11 and HE12. A small portion of light guiding by means of Anti-resonant reflecting optical waveguide (ARROW) mechanism is also observed. The transmission dips, resulting from multimode interferences (MMI) and ARROW effect have a big difference in sensitivities to strain and temperature, thus making it possible to monitor these two parameters with a single sensor head by using a characteristic matrix approach. In addition, the proposed sensor structure is experimentally proven to have a good reproducibility.

A popular astronomical concept covered by projection learning programs in the planetarium is seasonal constellation. However, a planetarium’s learning environment is limited to virtual scenes, where learners can observe seasonal constellations, but there is a significant difference between reality and the learners’ imagination regarding constellations. It is important to create a real-world oriented observation learning environment for observing seasonal constellations. Augmented reality has proved to be a powerful tool for astronomical observation learning. In this paper, augmented reality (AR) contents and 2D contents are used to develop a smartphone-based learning system called the Real-World Oriented Smartphone AR Learning System (R-WOSARLS) for seasonal constellation observation, which is based on the planetarium contents of the planetarium of the Nagoya City Science Museum, for seasonal constellation learning. Two experiments were conducted to evaluate the usefulness, usability, and learner satisfaction of our system in university and junior high school, respectively. The results show that R-WOSARLS is an effective learning tool for constellation observation and learning, and it enhances learners’ motivation to pursue seasonal constellation learning. Moreover, R-WOSARLS could be a teaching tool not only to help students learn more than with traditional instruction, but also to stimulate their interest in astronomical phenomena outside of school.

Adequate maintenance and taking active preventive measures can effectively prevent the early disease of asphalt pavements before significant damage occurs. By developing a light screening preventive maintenance agent (LS pre-maintenance agent) for strong ultraviolet (UV) radiation areas, based on the asphalt aging and regeneration mechanism, we analyzed the function and basic components and determined the optimum components ratio based on the best proportion of penetrant and solvent oil for solubility. The optimum ratio for quick-drying and long-term storage ability is the mass ratio of rock asphalt, reducing agent, penetrant, and solvent oil, which is 30:20:20:30. The light-shield agent is 5% of the total mass of the rock asphalt, reducing agent, penetrant, and solvent oil, and the dispersant is 0.4%. Digital image technology was used to provide an accurate measurement of the LS pre-maintenance agent penetration depth, evaluate its permeability and reasonable amount. We then used the rolling thin film oven test (RTFOT) to analyze its effect on aged asphalt and evaluated the restoration performance. Using the strong UV radiation aging test, we analyzed its anti-light aging performance. The results showed that pavement must be closed at least 2 h after the brushing LS pre-maintenance agent has been applied and this can be extended to upwards of 8 h time permitting. A dosage of 0.5 kg/m2 can ensure sufficient penetration depth and curing effect. Furthermore, the agent shows excellent restorative and anti-light aging abilities, which can effectively improve the low-temperature performance of aged asphalt and meet the pre-maintenance requirements for asphalt pavement, especially in strong UV radiation areas.
