A comprehensive approach to the mineral composition of black teas of different origins was studied using the Flame Atomic Absorption Spectrometry (FAAS) method, supported by chemometric tools including Principal Component Analysis PCA) and Classification and Regression Trees (CART). Significant differences between the teas from seven countries (Japan, Nepal, Kenya, Iran, Sri Lanka, India, and China) were shown. K was the main element determined in all teas, with an average concentration of 11,649 mg/kg, followed by Ca, Mg and Mn. In general, regarding all investigated black teas, the element content was ranked in the following order: K &gt, Ca &gt, Mg &gt, Mn &gt, Fe &gt, Na &gt, Zn &gt, Cu. The applied chemometric methods allowed us to recognize black tea clusters based on their mineral composition and place of cultivation, and allowed us to find correlations between particular elements in black teas. The performed analyses revealed interesting correlations between the concentration of various elements in black teas: K was negatively correlated with Na, Fe, Mn and Cu, K was positively correlated with the content of Ca and Mg. Significant positive correlations between Mn and Fe and Mn and Zn in the studied black tea samples were also revealed. It was shown that mineral composition may be a significant factor regarding the origin of the black tea, not only considering the country, but also the region or province.

This work aims to assess the recently established anti-inflammatory and antioxidant potential of melatonin of plant origin extracted from the plant matrix as a phytomelatonin complex (PHT-MLT), and compare its activity with synthetic melatonin (SNT-MLT) when used on its own or with vitamin C. For this purpose, a COX-2 enzyme inhibitory activity test, an antiradical activity in vitro and on cell lines assays, was performed on both PHT-MLT and SNT-MLT products. COX-2 inhibitory activity of PHT-MLT was found to be ca. 6.5 times stronger than that of SNT-MLT (43.3% and 6.7% enzyme inhibition, equivalent to the activity of acetylsalicylic acid in conc. 30.3 ± 0.2 and 12.0 ± 0.3 mg/mL, respectively). Higher antiradical potential and COX-2 inhibitory properties of PHT-MLT could be explained by the presence of additional naturally occurring constituents in alfalfa, chlorella, and rice, which were clearly visible on the HPLC-ESI-QTOF-MS fingerprint. The antiradical properties of PHT-MLT determined in the DPPH test (IC50 of 21.6 ± 1 mg of powder/mL) were found to originate from the presence of other metabolites in the 50% EtOH extract while SNT-MLT was found to be inactive under the applied testing conditions. However, the antioxidant studies on HaCaT keratinocytes stimulated with H2O2 revealed a noticeable activity in all samples. The presence of PHT-MLT (12.5, 25 and 50 µg/mL) and vitamin C (12.5, 25 and 50 µg/mL) in the H2O2-pretreated HaCaT keratinocytes protected the cells from generating reactive oxygen species. This observation confirms that MLT-containing samples affect the intracellular production of enzymes and neutralize the free radicals. Presented results indicated that MLT-containing products in combination with Vitamin C dosage are worth to be considered as a preventive alternative in the therapy of various diseases in the etiopathogenesis, of which radical and inflammatory mechanisms play an important role.

Numerous species of Allium genus have been used in the traditional medicine based on their vast biological effects, e.g., antimicrobial, digestion stimulant, anti-sclerotic, soothing, antiradical or wound healing properties. In this work, unpolar and polar extracts from two lesser-investigated species of Allium growing in Kazakhstan, Allium galanthum Kar. &amp, Kir. (AG) and A. turkestanicum Regel. (AT), were studied for their composition and biological effects. In the HPLC-ESI-QTOF-MS/MS analyses of water and alcoholic extracts simple organic acids, flavonoids and their glycosides were found to be the best represented group of secondary metabolites. On the other hand, in the GC-MS analysis diethyl ether, extracts were found to be rich sources of straight-chain hydrocarbons and their alcohols, fatty acids and sterols. The antimicrobial activity assessment showed a lower activity of polar extracts, however, the diethyl ether extract from AT bulbs and AG chives showed the strongest activity against Bacillus subtilis ATCC 6633, B. cereus ATCC 10876, some species of Staphylococcus (S. aureus ATCC 25923 and S. epidermidis ATCC 12228) and all tested Candida species (Candida albicans ATCC 2091, Candida albicans ATCC 10231, Candida glabrata ATCC 90030, Candida krusei ATCC 14243 and Candida parapsilosis ATCC 22019) with a minimum inhibitory concentration of 0.125–0.5 mg/mL. The highest antiradical capacity exhibited diethyl ether extracts from AG bulbs (IC50 = 19274.78 ± 92.11 mg Trolox eq/g of dried extract) in DPPH assay. In ABTS scavenging assay, the highest value of mg Trolox equivalents, 50.85 ± 2.90 was calculated for diethyl ether extract from AT bulbs. The same extract showed the highest inhibition of mushroom tyrosinase (82.65 ± 1.28% of enzyme activity), whereas AG bulb ether extract was the most efficient murine tyrosinase inhibitor (54% of the enzyme activity). The performed tests confirm possible cosmeceutical applications of these plants.

Macrocyclic bisbibenzyl compound, marchantin A, naturally occurring in liverworts belonging to genus Marchantia, was isolated and assessed for their cytotoxicity against two different lines of human skin cells - immortalized human keratinocytes HaCaT and human malignant melanoma cells A375. The purpose of the studies was also to examine the anti-hyperpigmentation effect of marchantin A, especially used in concentrations assessed as safe to normal human keratinocytes while simultaneously toxic for malignant melanoma cells. The data showed that marchantin A was cytotoxic for A375 melanoma cells (IC50 = 11.97, 7.45 and 9.80 μg/mL), nevertheless less cytotoxic for normal keratinocytes. Assay conducted for mushroom tyrosinase activity showed also that this compound did not affect the dopachrome formation during enzymatic reaction in concentrations previously assessed as safe for normal human cell. To our knowledge this is the first report showing anti-melanoma activity of marchantin A and assessing its activity against tyrosinase. Obtained results indicate potential application of marchantin A as a potential active ingredient for pharmaceutical and dermatological products.

Plants belonging to the Rosa genus are known for their high content of bioactive molecules and broad spectrum of healing and cosmetic activities. Rosa platyacantha Schrenk is a wild-type species abundant in the mountainous regions of Kazakhstan. The phytochemical composition as well as the bioactivity of R. platyacantha extracts have not been fully investigated to date. In this study, various parts of R.platyacantha plant, collected in Almaty region, Kazakhstan, were used to prepare five hydroalcoholic extracts (R1–R5). The extracts were compared for the content of phytochemicals and selected biological activities, which are important for the potential cosmetic application of R. platyacantha. Extract R3, prepared from flower buds, showed the most significant antioxidant and tyrosinase inhibitory potential, decreasing the monophenolase and diphenolase activities of tyrosinase. Extract R3 showed also collagenase inhibitory activity and cytotoxicity against human melanoma cells A375, being less cytotoxic for noncancerous skin keratinocytes HaCaT. Analysis of fractions E and F, obtained from R3 extracts, revealed that quercetin, kaempferol, rutin, and their derivatives are more likely responsible for the tyrosinase inhibitory properties of R. platyacantha extracts.

Growing scientific evidence indicates that Achillea biebersteinii is a valuable source of active ingredients with potential cosmetic applications. However, the data on its composition and pharmacological properties are still insufficient. This study aims to optimize the extraction procedure of the plant material, evaluate its phytochemical composition, and compare anti-tyrosinase potential of A. biebersteinii extracts obtained by various methods. In order to identify compounds responsible for the tyrosinase inhibitory activity of A. biebersteinii, the most active anti-tyrosinase extract was fractionated by column chromatography. The fractions were examined for their skin lightening potential by mushroom and murine tyrosinase inhibitory assays and melanin release assay. HPLC-ESI-Q-TOF-MS/MS analysis of the total extract revealed the presence of several phenolic acids, flavonoids, flavonoid glucosides, and carboxylic acid. Among them, fraxetin-8-O-glucoside, quercetin-O-glucopyranose, schaftoside/isoschaftoside, gmelinin B, 1,3-dicaffeoylquinic acid (1,3-DCQA), and ferulic acid were found in the fractions with the highest skin lightening potential. Based on obtained qualitative and quantitative analysis of the fractions, it was assumed that the caffeoylquinic acid derivatives and dicaffeoylquinic acid derivatives are more likely responsible for mushroom tyrosinase inhibitory activity of A. biebersteinii extracts and fractions. Ferulic acid was proposed as the most active murine tyrosinase inhibitor, responsible also for the reduced melanin release from B16F10 murine melanoma cells.

Essential oils (EOs) and their main constituents, the terpenes, are widely studied, mostly relating to their antioxidant ability and bioactivity, such as antimicrobial, anticancer, anti-inflammatory, and range of other actions in the living systems. However, there is limited information on their bioavailability, especially upon clinical studies. Having in mind both strong biological effects and health benefits of EOs and their specific physicochemical properties (volatility, lipophilic character, low water solubility or insolubility, viscosity, expressed odor, concentration-dependent toxicity, etc.), there is a need for their encapsulation for target delivery. Encapsulation of EOs and their constituents is the prerequisite for enhancing their oxidative stability, thermostability, photostability, shelf life, and biological activity. We considered various carrier types such a (1) monophase and polyphase polysaccharide hydrogel carriers, (2) polysaccharide-protein carriers, and (3) lipid carriers in the context of physicochemical and engineering factors. Physicochemical factors are encapsulation efficiency, chemical stability under gastric conditions, mechanical stability, and thermal stability of carrier matrices. Choice of carrier material also determines the encapsulation technique. Consequently, the engineering factors are related to the advantage and disadvantage of various encapsulation techniques frequently used in the literature. In addition, it was intended to address the interactions between (1) main carrier components, such as polysaccharides, proteins, and lipids themselves (in order to form chemically and mechanically stable structure); (2) main carrier components with pepsin under gastric conditions (in order to form resistant material under gastric conditions); and (3) main carrier components with EOs (in order to enhance encapsulation efficiency), as a necessary precondition for whole process optimization. Finally, different sources for obtaining natural carrier macromolecules are surveyed, especially the agro-waste materials and agricultural and food by-products. This review article highlights the bioavailability aspects of encapsulated EOs and physicochemical and engineering factors concerning natural macromolecule carriers for their target delivery and application.

The aim of this study was to investigate polyphenolic composition of different parts (leaves, flowers and fruits) of Crataegus almaatensis Pojark, an endemic plant of Kazakhstan, and compare it to a well known European Crataegus oxyacantha L. flowers. A Qual-Quant analysis was performed based on HR-MS measurements on 22 secondary metabolites: flavonoids and phenolic acids. Another goal was to evaluate the antioxidant potency of hawthorn extracts which was expressed in the total phenolic content and DPPH scavenging potency tests. Leaf extracts from C. almaatensis were found to be the most rich in metabolites and the most active in antiradical tests (IC50 value of 48 μg/ml and TPC of 218 mg/g). The weakest potential was determined for the fruit extract of this species. According to the performed principal component analysis (PCA), the fruit extracts were not correlated with other organs of the plant, and the metabolites responsible for the extracts’ differentiation were cyanidin 3-glucoside and quetcetin 3-galactoside. Based on a high correlation factor, the flowers of the Kazakh species was found to be as rich in polyphenols as the European hawthorn. The results of this study indicate that C. almaatensis is a promising source of natural antioxidants.

Three curcuminoids: bisdemethoxycurcumin, demethoxycurcumin, and curcumin from turmeric were successfully separated by a high capacity solvent system composed of heptane: chloroform: methanol: water mixture (5: 6: 3: 2 v/v/v/v) tailored for centrifugal partition chromatographs at K-values of 0.504, 1.057, 1.644, respectively. These three ferulic acid derivatives obtained at a purity rate exceeding 95% were analysed by an HPLC-MS spectrometer. Turmeric extract inhibited the proliferation/viability of A549 human lung cancer, HT29 colon cancer, and T98G glioblastoma cell lines in (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) tetrazolium reduction assay (MTT). Single curcuminoids significantly decreased the viability/proliferation of lung cancer cells in a dose-dependent manner. However, total extract displayed the superior anticancer activity in the investigated cell lines. Crude extract in combination with cisplatin augmented the decrease in the viability of cancer cells compared with single compound treatment in A549 lung cancer cells. Total extract of Curcuma longa could be regarded as being more effective against lung cancer cells in vitro than its separated compounds.

An HPLC quadrupole time-of-flight (Q-TOF) MS method was developed for the identification of secondary metabolites in Leonurus turkestanicus V.I. Krecz. et Kuprian. Ethanolic and chloroform extracts from the plant’s aerial parts were tested. A total of 16 compounds (iridoid glycosides, phenylpropanoids, flavonoids, and nitrogen-containing compounds, as well as diterpene acetate derivatives) were identified and tentatively characterized based on (or using) their retention times and UV and Q-TOF-MS data. Previously reported aucubin (1),6-deoxy-8-acetylharpagid(2), and stachydrine(13, 15)and homostachydrine isomers(14, 16)were identified, along with a lavandulifolioside isomer(3),verbascoside(4),rutin(5),3-O-kaempferol rutinoside (6), and an unknown diterpene acetate(8).Compounds3–6were detected for the first time in this plant. Additionally. antimicrobial activity was evaluated. No significant differences were found between ethanolic and chloroform extracts of L. turkestanicus; however, the alcoholic extract showed stronger antifungal activity [minimal inhibitory concentration (MIC) of 2.5–5 mg/mL], whereas the chloroform extract showed stronger activity against the tested spore-forming Bacillus species (MIC 1.25–2.5 mg/mL).

Essential oils obtained from Origanum species are characterized by high diversity in composition. In this work, thin-layer chromatography (TLC)—fingerprinting of Origanum species was performed for the first time and enabled the discrimination of three chemotypes (carvacrol, caryophyllene oxide, and terpineol/sabinyl). Gas chromatography—mass spectrometry and statistical analyses confirmed the presence of the mentioned chemotypes and deepened the characterization of the studied essential oils. TLC—bioautography showed that thymol and carvacrol are the main antioxidant compounds in Origanum sp. essential oils (EOs).

The volatile constituents from then-hexane extracts of the roots and rhizomes ofValeriana officinalis(VO) andValeriana turkestanica(VT) were investigated by GC-MS analysis. Two VO samples were obtained from cultivation, one from commercially available material, while VT was collected in a mountain of Kazakhstan. The most characteristic components present in all of the analysed samples were sesquiterpenoids. The three investigated samples of VO produced mainly valerenane and kessane sesquiterpenoids. Acetoxyvalerenic acid (33.94%), valerenic acid (15.05%), valerenal (11.93%), valeric acid 2,6-dimethylnon-1-en-3-yn-5-yl ester (5.24%), valerenol (3.31%), elemol (3.19%) and (E)-valerenyl isovalerate (2.53%), were the common components identified in then-hexane extract from the roots of VT. In comparison to VO this species does not produce kessane sesquiterpenoids and polyunsaturated fatty acids. This study shows that the roots of VT possess compounds of high biological significance, since they have the appropriate contents of valerenic acid and its derivatives, thus VT can be considered as a substitute for VO.

An optimisation of extraction towards an increased antioxidant capacity and the study on the extracts’ composition by HPLC-ESI-Q-TOF-MS were performed on different organs of a rarely studied plant: Rheum cordatum Losinsk (Polygonaceae) growing in Kazakhstan. More than 20 compounds from anthraquinones and phenolics were identified in an optimised method. The plant was proven to contain a wide variety of phenolic compounds (catechins, flavonoids, and their glucosides and phenolic acids) in contrast to the anthraquinone composition, which was mainly represented by emodin and its analogues. The results of the studies could determine the plant as a rich source of pharmacologically precious polyphenols. It was evidenced that the extracting solvents, the time of collection, and the organs tested affected both the chemical content and the antioxidant potential of the extracts. Ethanol : water (50 : 50 v/v) was selected as the most beneficial extractant for all metabolites, and based on the principal component analysis of raw data, the radical scavenging potential of the plant was strictly related to the presence of epicatechin gallate (ECG), kaempferol glucoside, and epigallocatechin gallate (EGCG) occurring in this extract at the concentration of 1.69-5%, 0.16-0.47%, and 0.001-2.93%, respectively.

Horse chestnut fruits and seeds have been traditionally used as a remedy against chronic venous insufficiency symptoms. Currently the total extracts or purified saponins are extensively used as dietary supplements or medicines’ ingredients in numerous authorized pharmaceuticals. The goal of this study was to investigate the changes in the secondary metabolites content within time. The selected optimal conditions for collection could provide the highest quantity of active compounds in the extracts, which will increase the quality of the final product. The obtained results indicate that the highest content of natural products in the horse chestnut fruits was measured between the 13th and 16th week from the beginning of blooming. The quantitative analysis of escin and two flavonoids: kaempferol and procyanidin A2 was performed on the extracts within the study. The content of the selected natural products increased of ca. 90% between the 12th and 20th week for escin and of ca. 120% between the 7th and 12th week, for flavonoids. The rise in natural products quantity was also expressed in the measured antioxidant activity and total phenolic content values in DPPH and Folin–Ciocalteu tests.

For the first time, the separation of coumarin derivatives from the petroleum ether extract of the fruits of Mutellina purpurea through high-performance countercurrent chromatography (HPCCC) is described. Four compounds, pteryxin (1; 2.72 mg), hyuganin C (2; 7.94 mg), osthol (3; 4.30 mg), and hyuganin A (4; 3.09 mg), were obtained in a single run following the injection of crude extract (300 mg). Additionally, auraptenol (5) and hyuganin D (6) were identified using LC-ESI-(Q)TOF-MS. The structures of the isolated compounds were elucidated through spectroscopic (NMR and MS) methods. This is apparently the first report of the identification of dihydropyranocoumarins in this plant, and the first time that HPCCC was used to separate them.

Over the past five years, phytochemical and pharmacological studies have been conducted on material extracted from members of the Rutaceae family. In such work, new furochinoline-structured alkaloids were isolated from Ruta sp. and Dictamnus sp. Beyond the aforementioned, other substances with promising activity were isolated from the less-known species of Zanthoxylum, Evodia, Lonchocarpus, Myrthopsis and Teclea. Currently used forms of extraction, as well as methods of isolation and detection, allow the obtaining of pure, biologically active compounds. Many of these have antifungal, anti-bacterial and anti-plasmodial properties. Others are still being researched as potential drugs, which, in future, may be used in treating those afflicted with HIV and cancer. This article is designed to give the readers a thorough review of the active natural products from the Rutaceae family.

Many phytochemical investigations have been focused recently on the antioxidant activity of herbal extracts which can be used in phytotherapy. The previous study revealed antioxidative properties of Mutellina purpurea extract, but the constituents responsible for this action were not described yet. The aim of this study was activityguided separation and identification of antioxidant compounds from M. purpurea herb. Thin-layer chromatography-2,2-diphenyl-1-picrylhydrazyl (TLC-DPPH•) assay was used to detect compounds of interest; liquid chromatography-diode array detection-mass spectrometry (LC-DAD-MS) analysis allowed to identify antioxidants. The active fractions analyzed with LC-DAD-MS contained as a main compound chlorogenic acid accompanied with p-coumaric acid, ferulic acid, three dicaffeoylquinic acids, and caffeoylferuloylquinic acid. The fast TLC-DPPH• assay with LC-DAD-MS identification enabled the accurate identification of antioxidants in M. purpurea herb, which was done for the first time.

Background: In the search for novel antidepressive drug candidates, bioguided fractionation of nonpolar constituents present in the oleoresin from ginger rhizomes (Zingiber officinale Roscoe, Zingiberaceae) was performed. This particular direction of the research was chosen due to the existing reports on the antidepressive properties of ginger total extract. The search for individual metabolites acting as MAO-A inhibitors, which correspond to the apparent effect of the total extract, is the subject of this work. Methods: Hexane extracts from ginger rhizomes were fractionated by using column chromatography (including silica gel impregnated with silver nitrate) and semi-preparative high-performance chromatography. For the activity assessment, an in vitro monoamine oxidase A (MAO-A) inhibition luminescence assay was performed on 10 purified terpenes: 1,8-cineole, α-citronellal, geraniol, β-sesquiphellandrene, γ-terpinen, geranyl acetate, isobornyl acetate, terpinen-4-ol, (E,E)-α-farnesene, and α-zingiberene. Results: Geraniol and (−)-terpinen-4-ol were found to be the strongest enzyme inhibitors with inhibition of 44.1% and 42.5%, respectively, at a concentration of 125 µg/mL. No differences in the inhibition potential were observed for the different groups of terpenes: sesquiterpenes, monoterpenes, or sesquiterpene alcohols; however, the two most active compounds contained a hydroxyl moiety. Conclusions: Terpene constituents from ginger’s extract were found to exhibit moderate inhibitory properties against the MAO-A enzyme in in vitro tests.

Summary. The presence of phenolic content in overground extracts of Euonymus verrucosus Scop. – commonly growing in Europe – has been reported recently. The chromatographical and spectral data revealed the presence of several simple phenolic acids (gallic, protocatechuic, p-hydroxybenzoic, vanillic, syringic, caffeic, p-coumaric, ferulic, and m-coumaric acids), both as free and conjugated with other secondary metabolites. The comparison of two-dimensional TLC systems on cellulose stationary phases with HPLC– DAD reversed-phase chromatography was performed to assess a cheap and rapid technique in the identification process of major phenolic constituents. 2,2-Diphenyl-1picrylhydrazyl (DPPH) radical-scavenging tests, expressed as IC50, revealed the most beneficial results for the fraction after alkaline hydrolysis and yielded 205 ± 8 μg mL−1.

The effects of xanthotoxin (8-methoxypsoralen) on the anticonvulsant activity of four classical antiepileptic drugs (carbamazepine, phenobarbital, phenytoin and valproate) were studied in the mouse maximal electroshock seizure model. Tonic hind limb extension (seizure activity) was evoked in adult male albino Swiss mice by a current (25 mA, 500 V, 50 Hz, 0.2 s stimulus duration) delivered via auricular electrodes. Total brain concentrations of antiepileptic drugs were measured by fluorescence polarization immunoassay to ascertain any pharmacokinetic contribution to the observed anticonvulsant effects. Results indicate that xanthotoxin (50 and 100 mg/kg, i.p.) significantly potentiated the anticonvulsant activity of carbamazepine against maximal electroshock-induced seizures (P0.05 and P0.001, respectively). Similarly, xanthotoxin (100 mg/kg, i.p.) markedly enhanced the anticonvulsant action of valproate in the maximal electroshock seizure test (P0.001). In contrast, xanthotoxin (100 mg/kg, i.p.) did not affect the protective action of phenobarbital and phenytoin against maximal electroshock-induced seizures in mice. Moreover, xanthotoxin (100 mg/kg, i.p.) significantly increased total brain concentrations of carbamazepine (P0.001) and valproate (P0.05), but not those of phenytoin and phenobarbital, indicating pharmacokinetic nature of interactions between drugs. In conclusion, the combinations of xanthotoxin with carbamazepine and valproate, despite their beneficial effects in terms of seizure suppression in mice, were probably due to a pharmacokinetic increase in total brain concentrations of these antiepileptic drugs in experimental animals.

Polygonum tinctorium Ait. (dyer’s knotweed) is a commonly known Asian perennial containing indole alkaloids – indigo and indirubin. Its leaves constitute a pharmacopoeial material (Chinese Pharmacopoeia) known for its antineoplastic, antipsoriatic, antiviral and antibacterial actions. This article presents the results of a detailed study into the optimization of the extraction and separation (CPC-based purification) process, and also the quantitative analysis of the extracts. The applied extraction parameters considered the impact of the extraction temperature, its duration and the solvent selection on the quantitative analysis of both indigo and indirubin. The study concludes that a 30 min long extraction of the leaves, in a 2% solution of chloral hydrate in dichloromethane, was found to be most beneficial and reproductive. The compounds’ poor solubility, low stability and high toxicity of the so far used solvents, were some of the challenges encountered. CPC purification by heptane:ethyl acetate:methanol:water (6:1:6:1) in ascending mode led to the most satisfactory results in terms of purity, solvent consumption and duration. The elaborated method can be used as a suitable tool for the detailed quantitative indigo and indirubin standardization in the leaves of the P. tinctorium. The indirubin recovery model may serve for further scale-up for industrial applications.

Scientists around the world are constantly working on new drugs to fight many incurable diseases. Great hopes come with triterpene compounds and sterols. Refinement and selection of the appropriate techniques of isolation and identification, allows for a more satisfactory recovery sought from plant matter. This paper presents the most commonly used techniques for the isolation and identification of triterpenes and sterols.

The present paper discusses the analysis of flavonoids in the methanol extract from the flowering herbs of Carduus acanthoides (L.), where flavonois were indentified by TLC and SPE HPLC. The quantitative analysis was performed using spectrophotometric Christ-Müller’s method. The samples containing flavonoids and those released after acid hydrolysis were investigated by 1D TLC on silica gel and polyamide. After purification by SPE, the samples were also analyzed by RP-HPLC. Apigenin-7-glucoside, luteolin-7-glucoside, kaempferol-3-rhamnoglucoside, kaempferol-3-glucoside, apigenin, luteolin and chlorogenic acid and p-coumaric acid were detected in the fractions of methanol extracts obtained from the plant. This method is very simple for the analysis and suitable for rapid screening of flavonoids in plants.

Aerial parts and roots of Crithmum maritimum L. fertilized with Tytanit were investigated on the presence of phenolic acids (PhAs). Cinnamic and benzoic acid derivatives were quantified by use of validated RP-HPLC/DAD method. The amount of PhAs in fertilized plants (T) was higher than in control (C) plants (in the aerial parts: 2.16 mg/g and 1.28 mg/g dry weight, respectively, and in roots: 4.05 mg/g and 2.78 mg/g dry weight, respectively). The predominant PhA was the caffeic acid (83.2–94.2% of the total PhAs). After Tytanit treatment, amount of the caffeic acid rose from 667.41 µg/g in C to 1463.83 µg/g dry weight in the aerial parts of T, and in roots from 2251.74 µg/g in C to 3451.86 µg/g dry weight in T. Tytanit had also influence on the qualitative composition of PhAs; in extracts from aerial parts, some of PhAs (ferulic, chlorogenic, and syringic acids), absent in control, appeared after fertilization.

An HPLC quadrupole time-of-flight (Q-TOF) MS method was developed for the identification of secondary metabolites in Leonurus turkestanicus V.I. Krecz. et Kuprian. Ethanolic and chloroform extracts from the plant's aerial parts were tested. A total of 16 compounds (iridoid glycosides, phenylpropanoids, flavonoids, and nitrogen-containing compounds, as well as diterpene acetate derivatives) were identified and tentatively characterized based on (or using) their retention times and UV and Q-TOF-MS data. Previously reported aucubin (1), 6-deoxy-8-acetylharpagid (2), and stachydrine (13, 15) and homostachydrine isomers (14, 16) were identified, along with a lavandulifolioside isomer (3), verbascoside (4), rutin (5), 3-O-kaempferol rutinoside (6), and an unknown diterpene acetate (8). Compounds 3-6 were detected for the first time in this plant. Additionally. antimicrobial activity was evaluated. No significant differences were found between ethanolic and chloroform extracts of L. turkestanicus; however, the alcoholic extract showed stronger antifungal activity [minimal inhibitory concentration (MIC) of 2.5-5 mg/mL], whereas the chloroform extract showed stronger activity against the tested spore-forming Bacillus species (MIC 1.25-2.5 mg/mL).

In this study, the optimisation of horse chestnut seed extraction was performed to determine the best extraction solvent in the recovery of pharmacologically active escin from raw material. All extracts were obtained by accelerated solvent extraction and were analysed for their antioxidant potential (DPPH test), phenolic content (Folin-Ciocalteu (FC) assay) and escin content (ESI-TOF-MS extracts profiling). Methanol was found to recover the highest amount of escin from the plant matrix as well as to produce the most active antiradical solutions rich in polyphenols. Radical scavenging properties of methanol extracts were calculated as 2 GAE in DPPH test and 23 GAE (mg/g) in FC assay. Quantitative analysis of extracts performed by ESI-TOF-MS measurements in ion-positive mode confirmed the presence of four major escin isomers in the extracts and showed that the high antioxidant potential of methanolic extracts went hand in hand with the highest content of escin (8.92%).

BACKGROUND: In this study we examined the influence of various forms of selenium (organic and inorganic) on the vivacity of Hericium erinaceum mycelium and structural changes and ultrastructure occurring during its development in submerged culture. RESULTS: The mycelium was grown on sodium selenite (Na2SeO3), Selol (with 20 and 50 g kg −1 Se, respectively) and a mixture of Na2SeO3 and Selol. Samples of the mycelium were collected on day 3 and day 24 of the incubation and viewed under an electron microscope. Selol at concentration 20 g kg −1 did not cause any damage to the cell ultrastructure, but it contributed to the thickening of the cell wall, which implied an influence on polysaccharide production. In the other cases, degradation changes appeared in the protoplasm and the thickness of the cell wall did not increase. CONCLUSION: The nature of the effect exerted by various sources of selenium in the culture medium on the formation of polysaccharides probably results from the differences in their chemical composition and differences in the toxicity of these compounds towards the cells, but is also connected with the decomposition of the wall surrounding degraded fungal cells. © 2014 Society of Chemical Industry
