Sodyum Dodesil sülfat poliakrilamid jel elektroforezi (SDS-PAGE), bir protein karışımının bileşenleri ayrı ayrı çözmek için en yaygın kullanılan analitik yöntemdir. Ayrıca bir proteinin saflığını elektroforetik bir yöntemle değerlendirmek neredeyse zorunlu hale gelmiştir. SDS-PAGE, proteinlerin moleküler boyuttaki farklılıkları ile jel matrisi boyunca %1' den daha az farklılık gösteren elektroforetik hareketliliklerini aynı anda kullanarak proteinleri birbirinden ayırır. Teknik aynı zamanda proteinlerin moleküler ağırlıklarını tahmin etme de güçlü bir araçtır.Protein analizinde vazgeçilmez bir araç olarak SDS-PAGE'in başarısı, elektroforetik hareketliliğin bir proteinin moleküler kütlesi ile korelasyonuna izin vermektedir. Bu çalışma, soğuk koşullarda barındırılan peripartum ineklerde ilk tohumlamada endokrinolojik ve metabolik profil ile gebelik oranları arasındaki ilişkiyi araştırmak amacıyla yapılmıştır. Ortam sıcaklığı, periparturient dönem boyunca saatlik olarak kaydedilmiştir. Anti-Müllerian hormon (AMH), β-hidroksibutirik asit (BHBA), insulin benzeri büyüme faktörü 1 (ILGF-1), insülin, kortizol, malondialdehit (MDA), progesteron (P4), tiroid uyarıcı hormon (TSH), tri-iyodotironin (T3) ve tiroksin (T4) konsantrasyonlarının analizi için 26 adet Simmental inekten doğum öncesi (-14 gün), doğum günü (0 gün) ve doğum sonrası (3, 4, 6, 8, 15, 22 ve 29 gün) farklı aralıklarla kan örnekleri alınmıştır. İnekler, ilk tohumlama sonuçlarına göre geriye dönük olarak gebe (PG) ve gebe olmayan (NPG) olmak üzere iki gruba ayrılmıştır. Çadır ahırda ortalama ortam sıcaklığı −7°C ile +4°C arasında değişmiştir. İneklerin (PG ve NPG) serum AMH (2,00 ± 0,04 vs. 1,89 ± 0,04 mU/L; P < ,006), insulin (2,10 ± 0,03 vs. 2,51 ± 0,05 ng/mL; P < ,0001), MDA (49,0 ± 1,30 vs. 44,0 ± 1,2 ng/mL; P < ,001) ve P4 (44,1 ± 2,2 vs. 41,7 ± 2,1 pg/mL; P < ,002) konsantrasyonları farklıydı. Sonuç olarak, AMH ve insulin soğuk şartlarda barındırılan ineklerde gebelik oranları üzerine belirleyici bir rol oynar., This study was performed to investigate the relationship between endocrinological and metabolic profiles and the pregnancy rate at the first insemination in peripartum dairy cows housed under cold conditions. Temperature inside the barn was recorded hourly during the periparturient period. Blood samples were collected before (last 14 day), on the day (0 day), and after parturition (3, 4, 6, 8, 15, 22, and 29 days) from 26 peripartum Simmental cows and analyzed for anti-Müllerian hormone, β-hydroxybutyric acid, insulin-like growth factor 1, insulin, cortisol, malondialdehyde, progesterone, thyroid-stimulating hormone, tri-iodothyronine, and thyroxine concentrations. The cows were divided into 2 groups: pregnant and non-pregnant based on results at the first insemination. The average ambient temperature ranged from −7°C to +11°C in the tent barn. Serum anti-Müllerian hormone (2.00 ± 0.04 vs. 1.89 ± 0.04 mU/L; P < .006), insulin (2.10 ± 0.03 vs. 2.51 ± 0.05 ng/mL; P < .0001), malondialdehyde (49.0 ± 1.30 vs. 44.0 ± 1.2 ng/mL; P < .001), and progesterone (44.1 ± 2.2 vs. 41.7 ± 2.1 pg/mL; P < .002) concentrations were different between pregnant and non-pregnant cows. In conclusion, anti-Müllerian hormone and insulin have a determinative role on pregnancy rate in peripartum cows housed under cold condition.

The aim of the presented study was to detect the blood parameters and the clinical findings of disbudding process in calves which conducted with surgery and cauterization with/without analgesic drug. The material of the study was 32 calves. The cases were randomly divided into the 4 groups, and each group was consisted of 8 cases: Group 1 Operation+analgesic drug (ketoprofen 3 mg/kg im); Group 2 only Operation; Group 3- Cauterization+analgesic drug (ketoprofen 3mg/kg im); Group 4 only cauterization. Rectal temperature, pulse, respiratory rate, left and right cornea temperatures were evaluated at -1st, 0, 2nd, 6th and 24th hours. The blood samples were analyzed for plasma cortisol, fibrinogen, serum glucose, and superoxide dismutase levels. In all predetermined times, no statistical differences were observed in rectal temperature, respiratory rate and corneal temperature according to performed method. Pulse rate dropped at 0 and 2nd hours in Group 2, 3 and 4, while it approached pretreatment values at 6th hour. Fibrinogen level was detected highest in Group 4 at 0, 2nd, 6th and 24th hours. Group 4 had the highest serum glucose levels of all groups at predetermined times. While cortisol levels were higher than pretreatment values at 0 and 2nd hours in all groups, it was showed similarity between at -1st and 6th hours in all groups. SOD level was detected respectively higher in Group 4 and 2 at 0, 2nd, 6th and 24th hours than other groups. In conclusion, the surgical removing of the horns could be used as an alternative method due to occuring the less stress and the pain in calves., Sunulan çalışmanın amacı, ağrı kesici kullanılarak ve kullanılmaksızın uygulanan cerrahi ve koterizasyon yöntemiyle yapılan boynuzsuzlaştırma işleminin buzağılardaki klinik ve kan parametreleri üzerine olan etkisini belirlemekti. Çalışma materyalini toplam 32 adet buzağı oluşturdu. Olgular rastgele her grupta 8 adet buzağı olacak şekilde 4 gruba ayrıldı: Grup 1 operasyon+ağrı kesici uygulaması (ketoprofen 3mg/kg im); Grup 2 sadece operasyon; Grup 3 koterizasyon+ağrı kesici uygulaması (ketoprofen 3mg/kg im); Grup 4 sadece koterizasyon. Uygulama öncesindeki 1. saatte başlamak üzere; 0., 2., 6. ve 24. saatlerde rektal sıcaklık, kalp frekansı, solunum değerleri, sağ ve sol kornea sıcaklık değerleri kaydedildi. Laboratuar analizleri için alınan kan örneklerinden kortizol, fibrinojen, serum glikoz, süperoksid dismutaz değerlerine bakıldı. Tüm zaman dilimlerinde rektal sıcaklık, solunum sayısı ve kornea sıcaklık değerlerinde uygulamalara göre istatistiksel olarak bir farklılık gözlenmedi. 0 ve 2. saatlerde kalp frekansı Grup 2, 3 ve 4’de düşerken, 6. saatte uygulama öncesi değerlerine yaklaştı. Fibrinojen seviyesi 0, 2, 6 ve 24. saatlerde Grup 4’de en yüksek olarak belirlendi. Grup 4 belirlenen zaman dilimlerinde en yüksek serum glikoz seviyesine sahipti. Tüm gruplarda 0 ve 2. saatlerde kortizol değerleri uygulama öncesi değerlerden yüksek olarak belirlenirken, 6. saatte tüm gruplarda uygulama öncesi değerler ile benzerlik gösterdi. 0, 2, 6 ve 24. saatlerde SOD seviyesi sırasıyla Grup 4 ve Grup 2’de diğer gruplara göre yüksek olarak belirlendi. Sonuç olarak; boynuz tomurcuklarının operatif yöntemle çıkarılması buzağılarda daha az strese ve ağrıya neden olduğu için alternatif bir yöntem olarak kullanılabilir.

Amaç: Bu çalışma Theileria equi seropozitif ve seronegatif atlar üzerinde, serum protein ve lipid profillerini ortaya koymak amacıyla yürütülmüştür. Gereç ve Yöntem: Theileria equi seropozitif ve seronegatif atlar IFAT ile test edilerek belirlendi. Bu atlara ait kan se- rumlarında SDS-PAGE ve HPTLC analizleri yapılarak sıra- sıyla serum protein ve lipit profilleri ortaya çıkarıldı ve bu analiz sonuçlarına göre seropozitif ve seronegatif atlar ara- sındaki farklılıklar ortaya konulmaya çalışıldı. Bulgular: Seropozitif hayvanların 23, 52, 103, 176, 200 ve 240 kDa ağırlığındaki proteinlerinde önemli oranda artış olduğu tespit edildi. Ayrıca hem sağlıklı hem de T. equi se- ropozitif hayvanların albumin nktarları benzer düzeyler- de olmasına karşın T. equi seropozitif hayvanların globulin miktarlarının arttığı görüldü. Serum lipid profilinde ise se- ropozitif hayvanların genel bir lipoliz durumunda oldukları, toplam lipid miktarı içinde özellikle hidrokarbon oranının azaldığı ve diacilgliserol düzeyinin arttığı belirlendi. Yine seropozitif hayvanlarda fosfolipid miktarında hafif artış ve kolesterol oranında ise hafif bir düşme tespit edildi. Öneri: Protein ve lipid profillerindeki bu değişiklikler, hay- vanların akut enfeksiyonu atlatmalarına rağmen hala tam olarak iyileşemediklerini göstermektedir., Aim: This study aims at determining serum protein and li- pid profiles in Theileria equi seropositive and seronegative horses. Materials and Methods: Theileria equi seropositivity and seronegativity at horses were tested by IFAT Having ana- lyzed the blood sera of horses by SDS-PAGE and HPTLC, se- rum protein and lipid profiles were detected respectively. Differences between seropositive and seronegative horses were tried to be determined based on the results obtained by said analysis. Results: It was detected that there was a significant in- crease in 23, 52, 103, 176, 200 and 240 kDa protein levels of seropositive horses. Additionally, globulin levels of T. equi seropositive animals were observed to have increased, al- though albumin levels of both healthy and T. equi seroposi- tive horses were similar. At serum lipid profile, seropositive horses were observed to be at overall lipolysis; among the total amount of lipid, particularly reduced hydrocarbon rates as well as increased diacylglycerol levels were detect- ed. A slight increase in phospholipids and a slight decrease in cholesterol levels were found in seropositive horses. Conclusion: Mentioned changes at both protein and lipid profiles suggest that these horses have not fully recovered despite the fact that they survived acute infection.

Koyun ve sığır karaciğer ve akciğerlerinden alınan kistlere ait kist sıvısı, germinal membran, kız kesesi ve protoskoleksler homojenize edildikten sonra, protein ve lipid profillerini belirlemek için SDS-PAGE ve HPTLC analizleri gerçekleştirildi. HPTLC analizi sonucunda, membran yapılarda, genel olarak tüm membranların oluşumuna katılan başlıca üç lipid sınıfı (fosfolipid, serbest yağ asidi, kolesterol) görülürken, sıvılarda ise sadece fosfolipidlerin varlığı tespit edildi. Numunelerin SDS-PAGE analizi sonucunda ise, moleküler büyüklükleri 8-205 kDa arasında, sığırlara ait karaciğer germinal membranında 16, akciğer germinal membranında 19, akciğer kız kese germinal membranında 15, akciğer kız kesesi sıvısında 17, akciğer steril kist sıvısında 4 adet, koyunlara ait akciğer germinal membranında 19, akciğer steril kist sıvısında 5 ve koyun akciğer fertil kist sıvısında 12 adet protein tespit edildi.

1. ÖZET Birçok protozoon enfeksiyonlarında hücre yüzeylerine bağlanma büyük öneme sahiptir. Glikoprotein ve glikolipidlerin ökaryotik hücre yüzeylerinde önemli ölçüde bulunmaları dolayısıyla protozoon lektinleri hedef hücreleri tanıma ve bağlanmada etkin vasıtalardır. Protozoon lektini ile hedef hücrelerin yüzey karbohidratları arasındaki interaksiyona dair önemli bir örnek olarak Entamoeba histolytica Gal/GalNAc (galaktoz/N- asetilgalaktozamin) lektini ile konakçı hücre yüzeylerinin Gal veya GalNAc' leri arasındaki interaksiyon verilebilir. Bu tür etkileşimler aynı zamanda Plasmodium falciparum, Acanthamoeba ve Trypanosoma cruzi için ve diğer bazı protozoonlar için de bildirilmiştir. Theileria annulata invazyonu ile ilgili olarak ligant-reseptör etkileşiminin moleküler detayları bilinmemekle birlikte, bu prosesin bazı yönleri incelenmiştir. Liganta bağlı proteinler bir kısa santrifüj işlemi sonrası elüe edildi ve sodyum dodesil sülfat-poliakrilamit jel elektroforezi ile tespit edildi. Sonuçta piroplazma membranında 32 kDa molekül ağırlığındaki piroplazma kaba membran ekstraktı proteininin galaktoza bağlandığı ve bu protein ile galaktoz etkileşiminde hidrofobik güçlerin de rol alabileceği kanaatine varıldı. 32 kDa Theileria annulata piroplazma proteininin tespiti gelecekte Theileriozis' ten korunma amacıyla yeni ilaç ve/veya aşı üretiminin temelini oluşturabilir. 2. ABSTRACT Adhesion to cell surface is of prime importance in the pathogenesis of many protozoan infection. Because glycoproteins and glycolipids are prevalent on the surface of eukaryotic cells, protozoan lectins can provide an efficient means to recognize and adhere to target cells. The prominent example of protozoa lectin-surface carbohydrates of target cell interactions includes the interactions of Entamoeba histolytica Gal/GalNAc (galactose/N-acetyl galactosamine) lectin with Gal/GalNAc of host cell surfaces. Such interactions have also been reported for Plasmodium falciparum, Acanthamoeba and Trypanasoma cruzi. The moleculer details of the ligand-receptor interactions involved in Theileria annulata are unknown, but some aspects of the process have been elucidated. Elastin receptor ligand in Theileria annulata sporozoite surface antigen, SPAG-1, has been postulated as prospective ligands for target host cell. But this hypothesis was weakened. The main objectives of the present project proposal are to search for carbohydrate binding proteins (lectins) among Theileria annulata piroplasm proteins and to evaluate the role of lectin binding in the recognition and adherence of Theileria annulata to target cells. To this end, numerous immobilized lectin ligands (L-fucose-sepharose, N-acetyl- neuraminic acid-sepharose, N-acetyl-D-galactosamine-agarose, N-acetyl- D-glucosamine-agarose, D-mannose-agarose, p-D-glucose,-agarose, a- methyl-D-mannoside-agarose) were incubated with Theileria annulatapiroplasm membrane proteins isolated. The ligand-bound proteins were eluted and separated by a brief centrifugation and determined by sodium dodecyl sulphate-polyacrylamide gel electrophoresis. It was found that 32 kDa protein of piroplasm crude membran extract binds galactose and that the hydrophobic forces may play a part in the interaction of the protein and galactose. Characterization of 32 kDa protein of Theileria annulata piroplasms could form the basis of novel drug and/or vaccine designs targeted at the prevention of theileriosis in the future. 127

Amaç: Bu çalışmanın amacı; doğum öncesi 14. günden itibaren doğum sonrası 21. güne kadarki dönemde bulunan süt ineklerindeki AMH, Ca, P, KOL, total protein, magnezyum, triaçilgliserol, BHB, serbest gliserol düzeyleri, serum lipid ve protein profilleri ile ineklerin bir sonraki tohumlama dönemindeki tohumlama sayıları arasındaki ilişkiyi incelemektir.Materyal ve Metod: Hayvan materyali olarak 2. laktasyon dönemindeki 36 adet Simental ırkı inek kullanıldı. Doğuma 14 gün kala, doğum esnasında, doğum sonrası 7., 14. ve 21. günlerde vena jugularis' ten jelli biyokimya tüpüne kan alınıp santrifüj edilerek serum elde edildi, örnekler -80°C' de analiz edilene kadar saklandı ve analizden 1 gün önce çözdürülerek ölçümler gerçekleştirildi. Bu ineklerin tekrar tohumlama sayıları takip edilerek çalışma tamamlandı. Bulgular: Gruplara göre değerlendirildiğinde anti-müllerian hormonun ve enerji durumunu yansıtan parametrelerin (TP, TAG) tohumlama sayısı üzerinde önemli bir etkisi olduğu gözlendi. Serum toplam yağı içerisindeki KOLE oranının doğumla birlikte aşırı yükseldiği KOL oranında ve magnezyum konsantrasyonundaki değişimin kısıtlı olduğu görüldü. Doğum esnasında yüksek miktarda β-Hidroksibütirat tespit edilen hayvanların tohumlama sayılarında önemli bir fark tespit edildi.Sonuç: Bu çalışmadan elde edilen sonuçlar değerlendirildiğinde ineğin enerji durumunun AMH düzeyini doğrudan etkilediği bunun ise bir sonraki tohumlama döneminde gebe kalma oranını değiştirdiği görülmektedir. Aim: The aim of this study was to investigate the relationship between AMH, Ca, P, cholesterol, total protein, magnesium, triacylglycerol, BHB, free glycerol levels and serum lipid and protein profiles of the dairy cattle which are within the period from 14th day to 21st day and insemination numbers in the next insemination period. Material and method: 36 Simental cattle, which were at the second lactation period, were used as animal material. The blood were drawn into the jelly biochemistry tube from vena jugularis at the 14th days before the parturition, during parturition, 7th, 14th and 21st days after the postpartum, and then it was centrifuged to obtain serum and samples were kept at -80°C until the analysis. However, parameters were measured after thawing 1 day before the analysis. The study was completed by following the re-insemination numbers of these cows.Results: When it was analyzed according to the groups, it was observed that anti- mullerian hormone and the parameters reflecting the energy (TP, TAG) had a significant effect upon insemination. Cholesterol ester level was highly increased in the birth period while it was seen that cholesterol and magnesium concentration changes were limited. It was determined that there was a significant difference among the insemination levels of the cattle high-level β-hydroxybutyrate during the parturition period. Conclusion: According to the results of the study, the energy level of the cattle affected directly to AHM levels, moreover it was seen that it would affect the probability being pregnant in the next insemination period. 122

Biyokimya laboratuvarlarda dislipidemilerin teşhisinde rutin olarak trigliserid ile total kolesterol analizleri yapılmaktadır. Hasta türü ne olursa olsun lipid metabolizması ile temel diagnoz bu iki belirteç üzerinden gerçekleştirilmektedir. Fakat her iki test de prensibinde büyük handikap vardır. Trigliserid ölçümünde, önce lipoprotein lipaz enzimi ile trigliseridler gliserol ve serbest yağ asidine çevrilmektedir. Daha sonra gliserol kinaz enzimi `gliserol` e ATP kullanarak gliserol 1 fosfata dönüştürmektedir. Oluşan gliserol 1-P ise enzim yardımıyla renkli bir bileşik oluşturmakta, oluşan renk ise 500 nm dalga boyunda ölçülerek trigliserid analizi yapılmaktadır. Ancak aşırı stres ve diyabet gibi bir durumda aşırı miktardaki trigliserid parçalanmış aynı zamanda aşırı miktarda gliserol açığa çıkmıştır. Aynı zamanda, açlığa bağlı lipoliz sonucu trigliseridlerin aşırı parçalanıp yine aşırı miktarda gliserol açığa çıktığında kan trigliserid ölçümü sonucunda aç olması gereken bireye lipemi tanısı konmaktadır.Total kolesterol ölçümünde de buna benzer bir durum söz konusudur. Bu testte ise kolesterol-esterleri kolesterol esteraz enzimi ile kolesterole çevrilmektedir. Oluşan kolesterol ile zaten var olan kolesterol bir havuz oluşturur. Kolesterol oksidaz enzimi bu havuzdaki kolesterolleri kimyasal reaksiyona sokarak renkli bir bileşik oluşturmaktadır. Bu renkli bileşik ise 540 nm dalga boyunda ölçülerek toplam kolesterol analizi yapılmaktadır. Miktarı artınca sentezi durur. Ve genellikle düzeyi sabittir, önemli ölçüde-fazla miktarda değişmez. Ancak, ör. negatif enerji dengesi sonucu karaciğere aşırı miktarda lipid gelip karaciğerden çok fazla miktarda lipid çıkmaktadır. Karaciğerin üretip dolaşıma gönderdiği lipidler (lipoproteinler) ekstrahepatik dokulara gitmekte (LDL) bu dokulardan ise fazla kolesterolü üzerlerine almaktadır (HDL). Daha sonra HDL yapısındaki bu kolesterol LCAT enzimi ile kolesterol esterine dönüştürülüp diğer lipoproteinlere transfer edilmektedir. Karaciğerden gönderilen lipid miktarına paralel olarak lipoprotein metabolizması artmakta ve bunun sonucunda kolesterol esteri oranı da artmaktadır. Ancak böyle bireylerde toplam kolesterol ölçümü yapıldığında kolesterol düzeyi normal olması gerekirken aşırı kolesterol tanısı konmaktadır.Bu sebeplerden dolayı, planlanan bu tezde dislipidemi oluşturulacak sıçanlarda trigliserid ve toplam kolesterol ölçümleri yerine serbest gliserol ile kolesterol ester ölçümünün kullanımı değerlendirilecektir. The glycerol kinase enzyme then converts glycerol to glycerol 1 phosphate using ATP. The resulting glycerol 1-P forms a colored compound with the help of enzyme and and triglyceride analysis is performed by measuring the resulting wavelength at 500 nm.However, an excessive amount of triglycerides has been cleaved and excessive amounts of glycerol have been exposed in a situation such as excessive stress and diabetes. At the same time, when lipolysis caused by fasting causes excessive triglycerides to break up and when excess glycerol is released, lipemia is diagnosed to the individual who should be hungry as a result of blood triglyceride measurement.There is a similar situation in the measurement of total cholesterol. In this test, cholesterol-esters are converted to cholesterol by cholesterol esterase. The cholesterol that is already formed creates a pool with other cholesterol forms. The cholesterol oxidase enzyme forms a colored compound by chemically reacting cholesterol in this pool. This colored compound is measured at 540 nm and total cholesterol analysis is performed. However, the level of cholesterol is controlled by itself; When its quantity decreases, its synthesis increases, When the amount increases, the synthesis stops. And usually the level of colesterol is fixed, it does not change significantly-much amount.However, eg. as a result of negative energy balance, excessive amounts of lipid come into the liver and a lot of lipoproteins come out of the liver. Lipids (lipoproteins), which are produced and circulated by the liver, export to extrahepatic tissues (LDL) and take over cholesterol from these tissues (HDL). This cholesterol in HDL structure is then converted to cholesterol ester by LCAT enzyme and transferred to other lipoproteins. The more lipid is removed from the liver, the more cholesterol ester ratio increases. Lipoprotein metabolism is increased in parallel with the amount of lipid delivered from the liver, resulting in an increased cholesterol ester ratio.
