Abstract

The maintenance of normal body weight is disrupted in patients with anorexia nervosa (AN) for prolonged periods of time. Prior to the onset of AN, premorbid body mass index (BMI) spans the entire range from underweight to obese. After recovery, patients have reduced rates of overweight and obesity. As such, loci involved in body weight regulation may also be relevant for AN and vice versa. Our primary analysis comprised a cross-trait analysis of the 1000 single-nucleotide polymorphisms (SNPs) with the lowest P-values in a genome-wide association meta-analysis (GWAMA) of AN (GCAN) for evidence of association in the largest published GWAMA for BMI (GIANT). Subsequently we performed sex-stratified analyses for these 1000 SNPs. Functional ex vivo studies on four genes ensued. Abstract

Individuals with anorexia nervosa (AN) restrict eating and become emaciated. They tend to have an aversion to foods rich in fat. Because epoxide hydrolase 2 (EPHX2) was identified as a novel AN susceptibility gene, and because its protein product, soluble epoxide hydrolase (sEH), converts bioactive epoxides of polyunsaturated fatty acid (PUFA) to the corresponding diols, lipidomic and metabolomic targets of EPHX2 were assessed to evaluate the biological functions of EPHX2 and their role in AN. Epoxide substrates of sEH and associated oxylipins were measured in ill AN, recovered AN and gender- and race-matched controls. PUFA and oxylipin markers were tested as potential biomarkers for AN. Oxylipin ratios were calculated as proxy markers of in vivo sEH activity. Several free- and total PUFAs were associated with AN diagnosis and with AN recovery. AN displayed elevated n-3 PUFAs and may differ from controls in PUFA elongation and desaturation processes. Cytochrome P450 pathway oxylipins from arachidonic acid, linoleic acid, alpha-linolenic acid and docosahexaenoic acid PUFAs are associated with AN diagnosis. The diol:epoxide ratios suggest the sEH activity is higher in AN compared with controls. Multivariate analysis illustrates normalization of lipidomic profiles in recovered ANs. EPHX2 influences AN risk through in vivo interaction with dietary PUFAs. PUFA composition and concentrations as well as sEH activity may contribute to the pathogenesis and prognosis of AN. Our data support the involvement of EPHX2-associated lipidomic and oxylipin dysregulations in AN, and reveal their potential as biomarkers to assess responsiveness to future intervention or treatment.

3. Emotional Intelligence Skills: Significant Factors in Freshmen Achievement and Retention.

Abstract

This study investigated the role of emotional skills in the academic achievement and retention of university freshmen. The research group was a randomly selected sample of first semester freshmen students (N=165), and cumulative grade point average was used as the criterion for academic success. The study was designed to investigate: (a) the relationship of emotional skills to academic achievement and retention, (b) gender and ethnicity differences in emotional skills, and (c) the emotional skill patterns of successful and unsuccessful freshmen. The results of the study indicate that the emotional skills of Time Management, Goal Achievement, and Assertive Communication were significant factors in freshmen achievement and retention. Abstract

Anorexia nervosa (AN) and related eating disorders are complex, multifactorial neuropsychiatric conditions with likely rare and common genetic and environmental determinants. To identify genetic variants associated with AN, we pursued a series of sequencing and genotyping studies focusing on the coding regions and upstream sequence of 152 candidate genes in a total of 1205 AN cases and 1948 controls. We identified individual variant associations in the Estrogen Receptor-ß (ESR2) gene, as well as a set of rare and common variants in the Epoxide Hydrolase 2 (EPHX2) gene, in an initial sequencing study of 261 early-onset severe AN cases and 73 controls (P=0.0004). The association of EPHX2 variants was further delineated in: (1) a pooling-based replication study involving an additional 500 AN patients and 500 controls (replication set P=0.00000016); (2) single-locus studies in a cohort of 386 previously genotyped broadly defined AN cases and 295 female population controls from the Bogalusa Heart Study (BHS) and a cohort of 58 individuals with self-reported eating disturbances and 851 controls (combined smallest single locus P

5. Use of Children's Literature in a Comprehensive School Guidance Program for Young Children.

Abstract

This chapter is part of a book that recounts the year's work at the Early Childhood Development Center (ECDC) at Texas A & M University-Corpus Christi. Rather than an "elitist" laboratory school for the children of university faculty, the dual-language ECDC is a collaboration between the Corpus Christi Independent School District and the university, with an enrollment representative of Corpus Christi's population. Asserting that bibliotherapy, the therapeutic use of books, can play an instrumental role in comprehensive school guidance programs, this chapter describes how ECDC school counselors promoted health care literacy using children's literature as part of a comprehensive developmental guidance program for very young children. Children's stories were used to promote a more positive sense of self, to help children learn about the world, and to help children cope with stress, as well as to provide insight into problems, affirm thoughts and feelings, stimulate discussion about problems, create an awareness that others have similar problems, provide solutions to problems, communicate new values and attitudes, and to help children find meaning in life. The chapter defines comprehensive developmental school guidance programs and emphasizes practical school counseling applications for engaging children and families in health care literacy. The chapter includes a 25-item annotated bibliography of children's books, 28 references, and appendices with lesson plans for "Oh, the Places You'll Go,""The Rainbow Fish," and "Pig Will and Pig Won't." 7. Factors Affecting the Learning Environment of Doctoral Students in Counselor Education.

Abstract

This paper describes a study designed to explore the learning environments of doctoral and master's degree counselor education programs. Counselor educators (N=8) from four states were interviewed; four Council for Accreditation of Counseling and Related Educational Program (CACREP) programs were represented. The focus of counselor education at the master's level appears to be congruent with the notion that graduate programs help students prepare for careers in order to meet the needs of the marketplace. Courses are generally survey in nature and most of the curriculum involves the transmission of knowledge from the professor to the students. Faculty report less contact with master's students than with doctoral students. The doctoral program prepares students for scholarship, although most of the research has focused on the success of graduates and not on the developmental issues of the programs. The influence of CACREP was a strong element in this study in that all interviewees had experience or knowledge of accreditation standards. Of particular interest was the realization from several interviewees that their knowledge was gained experientially. One could argue that the process of preparing counselor educators can be transmitted satisfactorily from observing an experienced counselor. Another argument could be made in light of the study's results, for establishing and publishing theoretical counselor education models based on the most important factors in constructing an effective learning environment. 10. Graphical tools for the examination of high-dimensional functions obtained as the result of Bayesian analysis

Abstract

Bayesian statistics has a tendency to produce objects that are of many more than three dimensions, typically of the same dimensionality as the parameter set of the problem. This thesis takes the idea of visual, exploratory data analysis and attempts to apply it to those objects. In order to do this it examines several areas, Monte Carlo Markov Chains (MCMC), display graphics and methods – especially Projection Pursuit methods – and Kernel Density Estimation (KDE). During the course of this work acceptable prior technology was found for MCMC and, once the decision for it had been made, Projection Pursuit. However, the current state of KDE gave rise to several objections. Not least among these came from the Bayesian background of the researcher, KDE had not been put in a suitable Bayesian framework and so clashed with the other technology. In addition it was felt that KDE needed too much user input and that is was somewhat ad hoc. This led to reformulating KDE in a Bayesian framework which had the added advantage of removing the need for a user to provide a bandwidth for each application. Chapter 6 of this thesis considers Bayesian theory and how it can be applied to KDE to produce a form more usable and satisfying in terms of Bayesian mathematics. This is shown to provide a powerful and flexible statistical tool without the need for the ad hoc choices often associated with these methods. This formulation of the KDE as a Bayesian problem is believed to be unique. As part of this work, software was produced in R to provide a usable visualisation of BKDE. A large number of examples is provided to demonstrate how this software can allow easy visualisation of a variety of types of dataset both with and without Kernel Density Estimation.

13. Comparison of Music-Based Curriculum versus an Eclectic Curriculum for Speech Acquisition in Students with Autism Spectrum Disorder

Abstract

The purpose of this study was to explore whether students with Autism Spectrum Disorders (ASD) could develop enhanced expressive speech acquisition through intervention via "Precision Songs" (Kantz & Kantz, 2004) and an eclectic curricula versus using only an eclectic curricula. Three hypotheses guided the study: (1) there is no statistically significant difference in expressive language acquisition between week 1 and week 6 for students with ASD receiving both eclectic and Precision Songs curriculum; (2) there is no statistically significant difference in expressive language acquisition between students with ASD first receiving eclectic curricula during weeks 1-6, then an addition of the Precision Songs (Kantz & Kantz, 2004) curriculum to the eclectic curricula during weeks 7-12; (3) there is no statistically significant difference in expressive language acquisition between students with ASD receiving eclectic and Precision Songs curriculum versus other students receiving eclectic curricula only during weeks 1-6. This comparative study included "ex post facto" analysis of existing data. The data available for this study included pre-test scores with "Autism Diagnostic Observation Schedule" (ADOS; Lord, Rutter, DiLavore, & Risi, 1999) and post-test ADOS scores of the identified students with ASD during the targeted time period. Two groups of identified students with ASD participated in an intervention of music-based speech acquisition curriculum, "Precision Songs" by Kantz and Kantz (2004), in addition to the provided eclectic curricula under the supervision of their classroom teachers. Pre-intervention and post-intervention data collected at predetermined intervals by the school district's autism team were analyzed. The Wilcoxon signed rank test was used for the non-normally distributed data to test the first null hypothesis. The second null hypothesis required using the repeated measures Analysis of Variance (ANOVA). An ANOVA was conducted to test the third null hypothesis. A statistically significant difference in expressive language acquisition was found for one group of students with ASD receiving an additional intervention of music-based curriculum. While the first null hypothesis was rejected, the second and third hypotheses were retained. The significant finding for the first hypothesis offers the hope that teachers can make a difference in the successful acquisition of expressive language by students with ASD because a specific group of students with ASD showed improved expressive language skills when pieces of practical information were taught to them through simple music. 16. Becoming a Supervisor: Doctoral Student Perceptions of the Training Experience

Thirteen doctoral students, 7 Caucasians and 6 Hispanics, participated over 3 semesters of practicum/internship in a qualitative research project exploring the experience of becoming supervisors. A follow-up study with 5 different doctoral students confirmed and refined findings 1 1/2 years later. The research team of 2 female faculty members and 1 male research assistant (the authors) identified 6 themes emerging from individual and group interviews with supervisors-in-training regarding the process of becoming a supervisor: (a) learning, (b) supervisee growth, (c) individual uniqueness, (d) reflection, (e) connections, and (f) putting it all together.

Abstract

We demonstrate the first ever recorded positron-emission tomography (PET) imaging and dosimetry of a FLASH proton beam at the Proton Center of the MD Anderson Cancer Center. Two scintillating LYSO crystal arrays, read out by silicon photomultipliers, were configured with a partial field of view of a cylindrical poly-methyl methacrylate (PMMA) phantom irradiated by a FLASH proton beam. The proton beam had a kinetic energy of 75.8 MeV and an intensity of about 3.5 × 1010 protons that were extracted over 101.5 ms-long spills. The radiation environment was characterized by cadmium–zinc–telluride and plastic scintillator counters. Preliminary results indicate that the PET technology used in our tests can efficiently record FLASH beam events. The instrument yielded informative and quantitative imaging and dosimetry of beam-activated isotopes in a PMMA phantom, as supported by Monte Carlo simulations. These studies open a new PET modality that can lead to improved imaging and monitoring of FLASH proton therapy. [ABSTRACT FROM AUTHOR]

23. Dynamics of Interracial Relationships Involving White Faculty in Black Colleges: Review, Systematization, and Directives.

Abstract

Factors that affect interracial relationships of white faculty at predominantly black colleges are considered. Based on theoretical writings and research, five dynamics influencing black-white interaction are identified: prejudice and stereotyping, dominance by whites, racial role-playing, social acceptance/social distance, and value differences. Factors at the individual, small group, college, and community levels that influence white faculty members' adjustment within the black college are also addressed. Stereotyped perceptions of white faculty at black colleges as academic rejects who are there for a paycheck or as paternalistic "know it alls" can impede effective interpersonal functioning. The social environment of black colleges, where blacks rather than whites are dominant, can lead to ambiguities and subtle conflicts between the two races. The phenomenon of racial role-playing refers to pretense and a lack of sincerity in relating to those of another race. Some research has concluded that black students distance themselves more from white instructors than black instructors. Value-related concerns include the influence of social class, differing perspectives on race consciousness, and value conflicts that may be experienced by white faculty when grading black students. Abstract

Genome-wide association studies have discovered hundreds of loci associated with complex brain disorders, but it remains unclear in which cell types these loci are active. Here we integrate genome-wide association study results with single-cell transcriptomic data from the entire mouse nervous system to systematically identify cell types underlying brain complex traits. We show that psychiatric disorders are predominantly associated with projecting excitatory and inhibitory neurons. Neurological diseases were associated with different cell types, which is consistent with other lines of evidence. Notably, Parkinson’s disease was genetically associated not only with cholinergic and monoaminergic neurons (which include dopaminergic neurons) but also with enteric neurons and oligodendrocytes. Using post-mortem brain transcriptomic data, we confirmed alterations in these cells, even at the earliest stages of disease progression. Our study provides an important framework for understanding the cellular basis of complex brain maladies, and reveals an unexpected role of oligodendrocytes in Parkinson’s disease. Abstract

International audience; BACKGROUND:Although attention-deficit/hyperactivity disorder (ADHD) and eating disorders (EDs) frequently co-occur, little is known about the shared etiology. In this study, we comprehensively investigated the genetic association between ADHD and various EDs, including anorexia nervosa (AN) and other EDs such as bulimia nervosa.METHODS:We applied different genetically informative designs to register-based information of a Swedish nationwide population (N = 3,550,118). We first examined the familial coaggregation of clinically diagnosed ADHD and EDs across multiple types of relatives. We then applied quantitative genetic modeling in full-sisters and maternal half-sisters to estimate the genetic correlations between ADHD and EDs. We further tested the associations between ADHD polygenic risk scores and ED symptoms, and between AN polygenic risk scores and ADHD symptoms, in a genotyped population-based sample (N = 13,472).RESULTS:Increased risk of all types of EDs was found in individuals with ADHD (any ED: odds ratio [OR] = 3.97, 95% confidence interval [CI] = 3.81, 4.14; AN: OR = 2.68, 95% CI = 2.15, 2.86; other EDs: OR = 4.66, 95% CI = 4.47, 4.87; bulimia nervosa: OR = 5.01, 95% CI = 4.63, 5.41) and their relatives compared with individuals without ADHD and their relatives. The magnitude of the associations decreased as the degree of relatedness decreased, suggesting shared familial liability between ADHD and EDs. Quantitative genetic models revealed stronger genetic correlation of ADHD with other EDs (.37, 95% CI = .31, .42) than with AN (.14, 95% CI = .05, .22). ADHD polygenic risk scores correlated positively with ED symptom measures overall and with the subscales Drive for Thinness and Body Dissatisfaction despite small effect sizes.CONCLUSIONS:We observed stronger genetic association with ADHD for non-AN EDs than for AN, highlighting specific genetic correlation beyond a general genetic factor across psychiatric disorders.

Abstract

Genome-wide association studies have discovered hundreds of loci associated with complex brain disorders, but it remains unclear in which cell types these loci are active. Here we integrate genome-wide association study results with single-cell transcriptomic data from the entire mouse nervous system to systematically identify cell types underlying brain complex traits. We show that psychiatric disorders are predominantly associated with projecting excitatory and inhibitory neurons. Neurological diseases were associated with different cell types, which is consistent with other lines of evidence. Notably, Parkinson’s disease was genetically associated not only with cholinergic and monoaminergic neurons (which include dopaminergic neurons) but also with enteric neurons and oligodendrocytes. Using post-mortem brain transcriptomic data, we confirmed alterations in these cells, even at the earliest stages of disease progression. Our study provides an important framework for understanding the cellular basis of complex brain maladies, and reveals an unexpected role of oligodendrocytes in Parkinson’s disease. © 2020, The Author(s), under exclusive licence to Springer Nature America, Inc.
