24. [Effect of topical timolol on microcirculation in the rabbit optic nerve head evaluated with a laser speckle microcirculation analyser]

Abstract

The effect of topical 0.5% timolol on the microcirculation in albino rabbit optic nerve head (ONH) tissue was examined using a laser speckle microcirculation analyser. In the first experiment, normalized blue (NB) in ONH, a quantitative index of peripheral blood flow in ONH, intraocular pressure (IOP), blood pressure (BP) and pulse rate (PR) were measured under general anesthesia before, and 30, 60, 90 and 120 minutes after a 20 microliters instillation of timolol in one eye and a placebo in the other eye in a masked randomized manner. In the second experiment, one eye of another group of rabbits received timolol twice daily for 20 days and a placebo in the fellow eye in a randomized masked manner. The NB in ONH and IOP were measured before treatment and 2 hours after the last instillation on the 20th day. After a single instillation of timolol, PR showed a maximum reduction of 11% and the IOP in the timolol-treated eyes showed a maximum decrease of 18%, but the NB in ONH and BP did not show any significant change during the experiment. After the 20-day treatment, IOP showed a maximum decrease of 28% in the timolol-treated eyes and 17% in the placebo-treated eyes. The NB in the timlol-treated eyes increased significantly (p0.01), but in the placebo-treated eyes it showed no significant change. Thus, the long-term use of topical timolol may increase peripheral blood flow in ONH tissue in the living rabbit eye.

47. X-Ray Studies of the Liquid/Vapor Interface: Water and Polymer and Fatty Acid Monolayers on Water

Abstract

X-ray specular reflectivity is used to study the liquid-vapor interface of pure water and of fatty acid and polymer monolayers at that interface. For the pure water surface the reflectivity was measured for three different spectrometer resolutions and simultaneous fits with only one free parameter to all of the data are in excellent agreement with the prediction of capillary wave theory for the RMS surface roughness. Diffuse scattering away from the specular condition, at wavevectors corresponding to those of the capillary waves, yields intensities and line shapes in agreement with theory with no significant adjustable parameters. Reflectivity from separate monolayers of co-poly 1,2-butadiene/butyl alcohol (50% random substitution) and lignoceric acid \((CH_3(CH_2)_{22}COOH)\) at the water/vapor interface are interpreted to obtain profiles of the average electron density ρ(z) as a function of distance z along the surface normal. For the polymer monolayer we find the following: 1) a local maximum in the electron density approximately 10% larger than that of the bulk polymer and 2) the RMS roughness of the vapor/polymer interface agrees with capillary wave theory predictions for the lower surface pressures. For the highest surface pressure the RMS roughness exceeds the value predicted by the capillary wave model. Measurements of reflectivity from a lignoceric acid monolayer, as a function of surface pressure throughout an isotherm (near room temperature), reveal the following behavior: 1) the overall thickness of the monolayer increases with increasing pressure and 2) the head groups occupy a progressively larger region along the surface normal as the pressure increases, indicating that they rearrange normal to the interface., Engineering and Applied Sciences

48. Some Properties of Precipitating Antigens Associated with Infectious Bursal Disease Virus

Abstract

Infection of the bursa of Fabricius and chicken embryo fibroblast cell cultures with avian infectious bursal disease virus resulted in production of a number of virus-induced antigens. The antigens were specific, forming three precipitin lines by immunodiffusion with antiserum (designated PA-1, -2, and -3). To separate immunoprecipitin from the remaining viral particles, two (PA-1 and PA-3) were partially purified by subjection to two cycles of diethylaminoethyl-cellulose chromatography and filtration through a column of Sephadex G-150 gel. The precipitating antigen, PA-1, was found to migrate most slowly through the agar gel, remaining serologically active after treatment with heating (56 C for 1 h), trypsin, lipolytic solvents, deoxyribonuclease, and ribonuclease. Its density was 1.27 g/ml. Morphologically the antigen displayed a doughnut-shaped structure 8 to 12 nm in size. PA-3 migrated most rapidly through the agar gel. It was destroyed by treatment with heating and trypsin but not with lipolytic solvents, deoxyribonuclease, and ribonuclease. Density was about 1.25 g/ml. This suggests that the antigen is a part of viral structural components. PA-2 migrated through agar gel at a rate between that of PA-1 and PA-3. Because of its low concentration, PA-2 was not further characterized.
