10. The Partial Substitution of Digestible Protein with Gelatinized Starch as an Energy Source Reduces Susceptibility to Lipid Oxidation in Rainbow Trout (Oncorhynchus mykiss) and Sea Bass (Dicentrarchus labrax) Muscle

Abstract

We evaluated the influence of dietary gelatinized starch and protein on the fatty acid composition of muscle in rainbow trout and European sea bass and on the susceptibility of flesh to lipid peroxidation. The possibility that flesh peroxidation could be accounted for by lipogenesis and the deposition of fat was also explored. The inclusion of gelatinized starch in the diet of rainbow trout improved growth with respect to that observed in fish fed crude starch (P < .001). This was especially noticeable at the lowest concentration of dietary protein tested (P = .037), suggesting that gelatinized starch may partially replace protein in the production of energy without inducing a negative effect on growth. However, in European sea bass, the gelatinization of starch and dietary protein concentration showed no significant effect on final body weight. The intramuscular neutral lipid concentration of the sea bass was reduced by the gelatinization of dietary starch (P = .034). The highest dietary protein concentration increased the proportion of saturated fatty acids in the neutral (P = .0742) and polar (P = .0033) lipid fractions. The dietary inclusion of high levels of protein in rainbow trout led to a lower concentration of total (n-3) (P.0457) and (n-6) (P = .0522) fatty acids and a higher concentration of total monounsaturated fatty acids (P = .0006). The inclusion of gelatinized starch led to a lower concentration of (n-3) fatty acids (P = .0034) and a higher concentration of saturated fatty acids (P = .0007). The polar fraction was hardly affected by the same treatment. A significantly lower susceptibility of the dorsal muscle to oxidation was observed in groups of European sea bass fed gelatinized starch (P < .01). A similar trend was observed in rainbow trout, although differences were not significant. The findings suggest that the digestible protein concentration of nutrient-dense diets for rainbow trout and European sea bass can be reduced with a beneficial effect on tissue lipid oxidation and no negative effects on growth and muscle composition. Our objectives were to test the potential replacement of fish meal by soy protein concentrate (SPC) in high-energy, extruded diets fed to rainbow trout (Oncorhynchus mykiss) and to evaluate the efficiency of DL-methionine supplementation of soy-based diets. Groups of trout (initial BW 103 to 106 g) were fed to visual satiety with isonitrogenous (6.6% DM) high-energy (22.8 MJ/kg DM gross energy), extruded diets, in which fish meal was progressively replaced with SPC (0, 50, 75, and 100%). Three 100% SPC diets were formulated to be either unsupplemented or supplemented with DL-methionine, so that total methionine content was .8 or 1.0% of DM. The quality of the SPC source used was assessed by measuring the antitryptic and antigenic activities and the concentrations of the isofiavones daidzein and genistein. Apparent digestibility of the diets was determined using the indirect method. A growth trial was conducted over 90 d at a water temperature of 18 degrees C. In addition to body composition analysis, plasma amino acid concentrations, anti-soy protein antibodies in the serum, and isofiavone concentrations in the bile were measured. The SPC source tested exhibited low antitryptic and antigenic activities, but it contained high concentrations of isofiavones (1,990 and 5,903 ppm for daidzein and genistein, respectively). Protein digestibility was high (92%) and was unaffected either the proportion of SPC in the diet or by DL-methionine supplementation. This was also true for the availability of amino acids, except phenylalanine. Digestibility of lipid and energy was reduced by 19% when SPC totally replaced fish meal. Growth rate was reduced when more than 50% of the dietary protein was of soy origin (daily growth coefficient of 3.2 and 2.1% for the control and the unsupplemented 100% SPC diet, respectively). The effect on growth was mainly explained by a general decline in feed intake (13.7 and 12.0 g DM.kg BW(super -l).d(super -1) for the control and the unsupplemented 100% SPC diet, respectively) and in lipid and, thus, in energy digestibility. The DL-methionine supplementation partially reversed the depressive effects of high dietary SPC incorporation (+13% growth), mainly by enhancing intake. The negative effect of SPC incorporation either may be due to the high isofiavone concentration or to an interaction between the soy protein component and the dietary lipids. Abstract

The aim of this study was to evaluate (1) the effects of replacement of fish oil by vegetable oils on flesh quality and (2) the effects of a washout with a return to fish oil on flesh quality of turbot. In a first period of 3 months, three isonitrogenous and isolipidic diets containing 9% of added marine fish oil (170), soybean oil (SO) or linseed oil (LO) were fed to triplicate groups of 25 marketable size turbot (initial body weight: 579 +/- 1 g) grown in sea water at the temperature of 17 degreesC. At the end of the first period, all groups of turbot were fed with the diet containing fish oil (diet FO) for a further period of 2 months. The gutted and fillet yields were not affected by the incorporation of vegetable oils. However, soybean or linseed oils significantly affected the organoleptic quality of flesh particularly odour, colour and texture. A more pronounced potatoes odour and a lower fat texture were observed in dorsal fillet of turbot fed soybean oil diet. During the washout period, the differences observed between treatments in first period on sensory attributes disappeared. This study showed that the changes in organoleptic properties occurring due to the intake of vegetable oils can be reduced with a return to a fish-oil-based diet.

The aim of the study was to investigate the replacement of fish oil by vegetable oils and the effects of a washout with a return to fish oil on growth performances and lipid metabolism. three experimental fish meal-based isonitrogenous (crude protein content: 57.5%) and isolipidic (crude lipid content: 16.5%) diets were formulated containing either 9% of added fish oil (170), soybean oil (SO), or linseed oil (LO). Each diet was distributed to triplicate groups of 25 marketable size turbot (initial body weight of 579 g) grown in seawater at water temperature of 17 T. Fish were fed once a day to visual satiety. At the end of the growth trial, which lasted 13 weeks, all groups of turbot were fed with FO diet for 8 weeks. The growth of turbot was high, but the incorporation of vegetable oils in the diets resulted in a slight decrease in growth as compared to those fed with fish oil-based diet. Feed and protein efficiency and whole body composition were not affected by dietary lipid sources. Total lipid content was low in the muscle of turbot (below 2%); ventral muscle being fatter than dorsal muscle. Liver and muscle fatty acid (FA) composition reflected dietary FA composition. Liver and muscle of fish fed with SO diet were rich in 18:2n-6, whereas those of fish fed with LO diet were rich in 18:3n-3. Liver and muscle of fish fed with SO and LO diets had lower levels of 20:5n-3 and 22:6n-3 in comparison to those of fish fed with FO diet. In turbot, hepatic lipogenic enzyme activities were low and not influenced by dietary lipid source. At the end of the second period, after transfer to FO-based diets, muscle. FA composition of fish fed previously with SO and LO diets were still different to those of fish fed with FO diet. The values of 18:2n-6 and 18:3n-3, respectively, were lower than the values found at the end of the growth period, but higher than those of fish fed with FO diet. An increase of FA levels, characteristic of fish oil, was observed in the liver and muscle of fish previously fed with vegetable oils. Data obtained show that replacement of fish oil by vegetable oils is possible with negligible impact on growth performance of turbot, that dietary lipids are an effective vector to influence the nutritional quality of finished product, and that a duration of 8 weeks is not sufficient to bring the FA profile of turbot of this size back to that of fish fed with fish oil over the whole period.

Abstract

In fish, the voluntary feed intake is influenced by dietary, environmental and/or physiological factors. It is well known that under hypoxia the concentration of oxygen in the water (DO) determines the feed intake of fish. However at non-limiting water DO levels (normoxia), several other mechanisms might play a role in feed intake regulation. Under hypoxia feed intake and oxygen consumption are interrelated. In this thesis we proposed the ‘oxystatic’ concept of feed intake regulation, which states that even at normoxia and in the absence of other constraints, the long term (weeks) voluntary feed intake of fish can be constrained by a set-point value of oxygen consumption. Dietary macronutrient composition affects the ‘dietary oxygen demand’ (i.e., amount of O2 consumed per unit of feed). This oxystatic concept implies that fish fed to satiation with diets differing in ‘dietary oxygen demand’ (mg O2/ g or kJ feed) will have a different digestible energy intake but a similar oxygen consumption. The validity of the oxystatic concept was assessed in two species, Nile tilapia and rainbow trout. These fish were fed diets which had large contrasts in nutrient composition (i.e., protein to energy ratio; type of the non-protein energy source (starch vs. fat); amino acid composition) in order to create contrasts in dietary oxygen demand. In all conducted studies with both species, the digestible energy intake was affected by the diet composition. However, in some studies oxygen consumption was similar and in others it differed between the diets, which respectively supports and contradicts the oxystatic concept. Abstract

Compromisation of food intake when confronted with diets deficient in essential amino acids is a common response of fish and other animals, but the underlying physiological factors are poorly understood. We hypothesize that oxygen consumption of fish is a possible physiological factor constraining food intake. To verify, we assessed the food intake and oxygen consumption of rainbow trout fed to satiation with diets which differed in essential amino acid (methionine and lysine) compositions: a balanced vs. an imbalanced amino acid diet. Both diets were tested at two water oxygen levels: hypoxia vs. normoxia. Trout consumed 29% less food under hypoxia compared to normoxia (p0.05). This difference in food intake between diets under normoxia together with the identical oxygen consumption supports the hypothesis that food intake in fish can be constrained by a set-point value of oxygen consumption, as seen here on a six-week time scale.

44. The Positive Impact of the Early-Feeding of a Plant-Based Diet on Its Future Acceptance and Utilisation in Rainbow Trout

Abstract

Sustainable aquaculture, which entails proportional replacement of fish-based feed sources by plant-based ingredients, is impeded by the poor growth response frequently seen in fish fed high levels of plant ingredients. This study explores the potential to improve, by means of early nutritional exposure, the growth of fish fed plant-based feed. Rainbow trout swimup fry were fed for 3 weeks either a plant-based diet (diet V, V-fish) or a diet containing fishmeal and fish oil as protein and fat source (diet M, M-fish). After this 3-wk nutritional history period, all V- or M-fish received diet M for a 7-month intermediate growth phase. Both groups were then challenged by feeding diet V for 25 days during which voluntary feed intake, growth, and nutrient utilisation were monitored (V-challenge). Three isogenic rainbow trout lines were used for evaluating possible family effects. The results of the V-challenge showed a 42% higher growth rate (P = 0.002) and 30% higher feed intake (P = 0.005) in fish of nutritional history V compared to M (averaged over the three families). Besides the effects on feed intake, V-fish utilized diet V more efficiently than M-fish, as reflected by the on average 18% higher feed efficiency (P = 0.003). We noted a significant family effect for the above parameters (P,0.001), but the nutritional history effect was consistent for all three families (no interaction effect, P.0.05). In summary, our study shows that an early shortterm exposure of rainbow trout fry to a plant-based diet improves acceptance and utilization of the same diet when given at later life stages. This positive response is encouraging as a potential strategy to improve the use of plant-based feed in fish, of interest in the field of fish farming and animal nutrition in general. Future work needs to determine the persistency of this positive early feeding effect and the underlying mechanisms.

Abstract

International audience; Genistein enriched diets at 500 ppm and 1000 ppm were prepared from a fish meal based diet used as control. Male and female rainbow trout were fed with these diets for one year while they undergo their first sexual development. Testosterone (T), 11-keto-testosterone (11KT), estradiol (E2)and vitellogenin (VTG) were monitored every 1,5 months. Fish were sacrificed at the same periodicity for histological control of the gonad. At the end of the reproductive maturation process, 17α,20β-(OH)2-progesterone (17α,20βP), GTH1 and GTH2 were measured. Reproductive efficiency was assessed on the basis of sperm characteristics (sperm volume, spermatocrit measurements, sperm motility), on oocyte characteristics (oocyte size, layer mass, mass of remaining gonad after spawning). Fertilisation was realised mixing sperm obtained on male fed one defined diet with eggs from females fed on the same diet. Endocrine disruptions were noticed on 17α,20βP GTH1 and GTH2. VTG was increased in a dose response manner in female after natural yolk production. In fish naive to estradiol the effect was also dose-dependant but of much less importance. In male, spermatogenesis was accelerated, while in females, oogenesis was slowed down. Reproductive efficiency was impaired in the 500 ppm fed fish.
