In this work, the accuracy of material decomposition (MD) using an energy discriminating photon counting detector was studied. An MD framework was established and validated using calcium hydroxyapatite (CaHA) inserts of known densities (50 mg/cm³, 100 mg/cm³, 250 mg/cm³, 400 mg/cm³), and diameters (1.2 mm, 3.0 mm, 5.0 mm). These inserts were placed in a cardiac rod phantom that mimics a tissue equivalent heart and measured using an experimental photon counting detector cone beam computed tomography (PCD-CBCT) setup. The quantitative coronary calcium scores (density, mass, and volume) obtained from the MD framework were compared with the nominal values. Additionally, three different calibration techniques, signal-toequivalent thickness calibration (STC), polynomial correction (PC), and projected equivalent thickness calibration (PETC) were compared to investigate the effect of the calibration method on the quantitative values. The obtained MD estimates agreed well with the nominal values for density (mass) with mean absolute percent errors (MAPEs) 8 ± 11% (9 ± 15%) and 4 ± 6% (9 ± 14%) for STC and PETC calibration methods, respectively. PC displayed large MAPEs for density (27 ± 9%), and mass (25 ± 12%). Volume estimation resulted in large deviations between true and measured values with notable MAPEs for STC (40 ± 90%), PC (40 ± 80%), and PETC (40 ± 90%). The framework demonstrated the feasibility of quantitative CaHA mass and density scoring using PCD-CBCT.

Background: Deep brain stimulation (DBS) in the subthalamic nucleus (STN) is used in advanced Parkinson’s disease (PD) for reducing motor fluctuations and the side effects of antiparkinsonian medication (APM). The development of neuroimaging has enabled the direct targeting of the STN. The aim of this study is to evaluate the outcome in patients with PD using STN DBS when changing from atlas-based indirect targeting method (iTM) to direct MRI-based targeting (dTM) assuming dTM is superior. Methods: Twenty-five consecutive PD patients underwent dTM STN DBS surgery from 2014 to 2017 with follow-up for 1 year. Most plants form symbiotic associations with various fungi in natural ecosystems. Traditionally, many of these associations are viewed as mutually beneficial, but recent studies show that symbiotic interactions can be complex and labile. I studied the variability of interactions between grasses and fungi, using root-associated fungi and foliar endophytes in grasses as study systems. First, I studied experimentally how root-associated fungi colonize their host grasses in different natural and artificial conditions. I then investigated experimentally how foliar endophytes affect their host grasses, and whether the endophyte contributes to the host’s adaptation to local and/or foreign habitats. Finally, I reviewed the current use of foliar endophytes in agriculture, with a particular focus on Europe, and critically considered their potential for practices beyond agriculture. I found root fungi to be common, but different types of root fungi had contrasting colonization patterns in natural environments. However, I found that grasses lose almost all of their root fungi in controlled and more favourable conditions, indicating that the associations are fairly loose and conditional to environmental context. My results also showed that foliar endophytes affected the host’s performance in varying ways, depending on plant origin and experimental country, indicating that the associations were context dependent and could represent conditional mutualism. I also found that endophytes contribute to plant adaptation only weakly, but that the grasses were clearly locally adapted to their sites of origin, especially in regard to reproduction. However, the grasses of subalpine origin performed well vegetatively also in Northern Finland, suggesting that they may have high adaptive potential under changing climates. The literary review showed that foliar endophytes are successfully used in agriculture, e.g. in the USA and New Zealand, and that they possess the potential for several practical applications. However, the intentional use of endophyte-enhanced grasses is non-existent in Europe, although many European grass cultivars have great potential for improvement via endophytes. Taken together, these results show that plant–fungal interactions are highly variable along sites, environmental contexts and origins of the symbiotum, making predictions for these interactions difficult. Tiivistelmä Luonnollisissa ekosysteemeissä melkein kaikilla kasveilla on symbionttisia sienikumppaneita. Perinteisesti monien näiden vuorovaikutusten on oletettu olevan molemmille hyödyllisiä, mutta viimeaikaiset tutkimukset osoittavat symbionttisten yhteyksien olevan vaihtelevia. Väitöskirjassani tutkin tätä heinien ja sienien välisten yhteyksien vaihtelevuutta, käyttäen heinissä esiintyviä juurisieniä ja lehtiendofyyttejä tutkimuskohteinani. Ensiksi tutkin kokeellisesti, kuinka juurisienet kolonisoivat heiniä erilaisissa luonnon- ja kasvihuoneolosuhteissa. Seuraavaksi tutkin kokeellisesti, kuinka lehtiendofyytit vaikuttivat heinien menestymiseen ja edesauttoivatko endofyytit heinien sopeutumista paikallisiin ja/tai vieraisiin elinympäristöihin. Viimeiseksi selvitin kirjallisuuskatsauksen avulla, kuinka lehtiendofyyttejä hyödynnetään maataloudessa ja arvioin endofyyttien potentiaalista käyttöarvoa maatalouden ulkopuolella, erityisesti keskittyen niiden hyödyntämiseen Euroopassa. Tutkimukseni osoitti, että heinien juurisienet ovat yleisiä, mutta niiden keskinäiset runsaussuhteet vaihtelivat luonnollisissa ympäristöissä. Kasvihuoneolosuhteissa heinät kuitenkin menettivät juurisienensä, viitaten siihen, että kyseinen yhteys on melko löyhä ja riippuvainen ympäristöstä. Tutkimukseni osoitti myös, että lehtiendofyytit vaikuttivat heinien menestykseen vaihtelevasti riippuen heinien alkuperästä sekä koemaasta, viitaten siihen, että nämä yhteydet ovat niin ikään olosuhteista riippuvaisia. Endofyytti vaikutti vain heikosti heinien sopeutumiseen, mutta heinät olivat selkeästi paikallisesti sopeutuneita niiden alkuperäisiin elinympäristöihin, erityisesti heinien lisääntymisen suhteen. Alppien heinät kuitenkin menestyivät vegetatiivisesti myös Pohjois-Lapissa, mikä viittaa siihen, että näillä heinillä saattaa olla korkea sopeutumispotentiaali muuttuvissa olosuhteissa. Kirjallisuuskatsaukseni osoitti, että lehtiendofyyttejä käytetään menestyksekkäästi mm. USA:n ja Uudessa-Seelannin maataloudessa. Euroopassa niiden käyttö on kuitenkin lähes olematonta, vaikka endofyyteillä voisi olla monia käyttömahdollisuuksia, sekä maataloudessa että ympäristön hoidossa. Kaiken kaikkiaan tutkimukseni osoittavat, että heinien ja sienten väliset vuorovaikutukset ovat hyvin vaihtelevia riippuen ympäristöstä ja heinien alkuperästä, minkä vuoksi näiden sienien vaikutuksia heiniin on vaikea ennustaa.

The ERL Emergency Challenge requires teams of land, underwater and flying robots to work together to survey the scene, collect environmental data, and identify critical hazards. To prepare teams for this multidisciplinary task a series of summer schools and workshops have been arranged. In this paper the challenges and hands-on results of bringing students and researchers collaborating successfully in unknown environments and in new research areas are explained. As a case study results from the euRathlon/SHERPA workshop 2015 in Oulu are given.

Tiivistelmä. Terveysteknologian teollisuudessa tuotteiden ja tuotannon suunnittelu, toteutus ja laadunhallinta ovat kansainvälisesti säännösteltyä, ohjeistettua ja valvottua toimintaa. Pienelle ja aloittelevalle yritykselle lakisääteiset vaatimukset ja asiakasvaatimukset täyttävän laatujärjestelmän rakentaminen on raskas ja vaativa kehitysvaihe, missä organisaation tulee olla kyvykäs tunnistamaan asiakkaiden tarpeet sekä lainsäädännön ja viranomaisohjeistuksen asettamat rajoitukset ja suositukset järjestelmällisellä ja suunnitelmallisella tavalla. Viranomaisvaatimukset huomioivan ja toteuttavan laatujärjestelmän avulla yritys varmistaa, että tuotteet täyttävät asiakasvaatimukset, projektit täyttävät asiakkaiden tarpeet ja koko organisaatio on sitoutunut toimittamaan regulatiiviset vaatimukset täyttäviä, turvallisia ja tehokkaita terveysteknologian tuotteita ja palveluja. Tutkimuksen tavoitteena oli selvittää mitä laatuvaatimuksia terveysteknologian tuotantoon automaatiojärjestelmiä myyvän toimittajan tulee ottaa huomioon uuden laatujärjestelmän suunnittelussa ja rakentamisessa. Laatujärjestelmän viitekehys suunniteltiin teorian pohjalta laaditun konstruktion avulla. Teorettisessa ja empiirisessä tutkimuksessa selvitettiin tuotteisiin kohdistuvia laatuvaatimuksia ja laatujärjestelmävaatimuksia viranomaisvaatimuksien, organisaation omien tarpeiden ja asiakasvaatimuksien näkökulmista. Tutkimus kokonaisuutena muodosti viitekehyksen, jonka avulla organisaatio kykenee sopeuttamaan, laajentamaan, uudistamaan ja parantamaan laatujärjestelmää myös tulevaisuudessa muuttuvien asiakastarpeiden ja liiketoiminnan kehitystarpeiden mukaan. GxP-periaatteiden mukaiset viranomaisvaatimukset eivät suoraan sido automaatiojärjestelmän toimittajaa, mutta ne sitovat asiakkaiden tuotantoprosesseja ja järjestelmiä. Asiakkaat kuitenkin odottavat, että automaatiojärjestelmän toimittaja noudattaa tai vähintään soveltaa viranomaisvaatimuksia laatujärjestelmässä, tuotteissa ja palveluissa. Tutkimuksen merkittävimpänä tuloksena laadittiin ehdotus laatujärjestelmästä Global Innovation Network Oy:n valmistamille automaatiojärjestelmille. Laatujärjestelmä suunniteltiin täyttämään ISO 9001-standardin vaatimukset sekä soveltamaan ISO 13485 -standardin vaatimuksia, FDA:n lääkinnällisille laitteille asettamia laatujärjestelmävaatimuksia ja teollisuuden luomaa ohjeistoa. Moderneja regulatiivisia laatusuunnittelun periaatteita noudattaen tuotteiden ja palvelujen toteutus sidottiin riskitasoon perustuvaan elinkaarimalliin, joten laatujärjestelmä sisältää tuotekohtaisten riskien arvioinnin ja hallinnan menetelmät.Design of quality management system framework within the health tech industry. Abstract. In health tech and life science industries, product and production design, implementation and quality control are covered by international laws, regulations, guidelines and monitored inspections. For a start-up business to fulfill statutory and client requirements is a strenuous and demanding phase, during which the organization should be capable to recognize the customer needs as well as the legislative limitations and recommendations systematically. By building-up a quality control and quality management system implementing laws, directives and regulatory requirements, an organization assures commitment in delivering projects which meet customer needs as well as products and services which are safe and effective to use, and fulfill customer requirements. The aim of the research was to clarify what quality requirements automated systems supplier for health tech and life science industries has to take into account while designing and implementing a new quality management system. The framework of the quality system was designed using constructive research process and a novel construct developed according to theoretical findings. Theoretic and empiric studies clarified product specific quality requirements and quality system requirements from the perspectives of regulatory and customer requirements as well as organization and business needs. The study itself formed a framework which supports the organization’s capability to utilize, adapt, expand and enhance the quality management system according to changing customer and business needs in the future. Regulatory requirements following global GxP principles do not confine the supplier, but they have that effect on the system owner’s production processes and automated systems. Customers however expect that the automated systems supplier implements or applies regulatory requirements in the supplier’s quality system as well as in the products and services. The most significant result of this study was a proposal of a new quality management system for automated systems manufactured by Global Innovation Network Oy. The quality system was designed to fulfill the requirements of ISO 9001 standard and to apply to requirements of ISO 13485 standard. Accordingly, proposed quality system applies to quality requirements of 21 CFR Part 820 for medical devices set by the FDA as well as regulatory guidelines and practical guidance created by authorities and industry representatives. The design, development and manufacturing phases of the products and services were bound to modern life cycle and validation models based on the Quality by Design principles and identification of risk levels. Hence, the quality system contains procedures and methods for a product specific risk assessment and control.
