Im Gegensatz für die biotechnologische Produktion von L-Lactat, gibt es noch kein industrielles Verfahren zur Herstellung von D-Lactat mit agrarischen Reststoffen. Der Fokus dieser Arbeit lag daher auf der Entwicklung eines biotechnologischen Verfahrens zur Herstellung von D-Lactat auf der Basis von erneuerbaren Ressourcen. Für die Umsetzung dieses Ziels sollte zuerst nach entsprechenden Biokatalysatoren gescreent werden, die die Fertigkeiten haben, D-Lactat in hoher Konzentration, Ausbeute und hoher optischer Reinheit zu produzieren. Anhand dieser Kriterien wurden zwei Sporolactobacillus Spezies für eine biotechnologische Anwendung charakterisiert. Dazu wurden die allgemeinen Kultivierungsbedingungen untersucht sowie die optimale Temperatur für die D-Lactatproduktion bestimmt. Im Folgenden sind die im Hauptkulturmedium enthaltenden Stickstoffquellen sowie Spurenelemente und Salze qualitativ und quantitativ auf ihre Relevanz für die Kultivierung getestet worden. Darüber hinaus wurde der Substrattoleranzbereich mit Hilfe verschiedener Glucosestartkonzentrationen für die Bakterien ermittelt und verschiedene Zucker wurden auf die Nutzbarkeit zur D-Lactatproduktion untersucht. Anhand dieser Erkenntnisse wurden darauffolgend kostengünstige, in Deutschland verfügbare Ersatzstoffe auf ihre Eignung zur D-Lactatherstellung getestet. Weiterhin wurden zur Prozessoptimierung die optimalen pH-Werte der Bakterien zur Steigerung der Produktionsgeschwindigkeit durch Experimente im Bioreaktor definiert und Fermentationsstrategien im Batch- bzw. Fed-Batch-Verfahren, mit Calciumcarbonat sowie Ammoniakwasser als Neutralisationsmittel, entwickelt. Die Verwendung von Ammoniakwasser führte zudem zu einer Produktinhibition der Bakterien infolge der steigenden Konzentration an nicht-dissoziierter Säure während der Kultivierung bei konstanten pH-Wert. Dieses Problem konnte durch einen produktkonzentrationsabhängigen pH-Shift gelöst werden, wodurch die Gesamtproduktivität gesteigert werden konnte. Zusätzlich konnte die Prozessstabilität und die Gesamtproduktivität durch Zellimmobilisierung, unter Zuhilfenahme von Lentikats(R), deutlich erhöht werden. Letztendlich konnte aus der Kombination der gewonnenen Erkenntnisse ein kostengünstiger Fermentationsprozess auf der Grundlage von agrarischen Restoffen zur Produktion von D-Lactat realisiert werden., In contrast to the biotechnological production of L-lactic acid, there is no existing industrial procedure for the manufacture of D-lactic acid based on renewable resources. For this reason, the aim of this study was to develop a procedure for biotechnological production of D-lactic acid on the basis of agricultural residual materials. In the very first step appropriate biocatalysts were screened, which are capable to produce optically pure D-lactic acid in a high amount. Furthermore, two Sporolactobacillus species were selected and characterized for a potential biotechnological application. For this purpose, general cultivation conditions were evaluated and the optimal growth temperature of the bacteria was determined. Moreover, the ingredients of the culture medium were analyzed for their importance of D-lactic acid biosynthesis. Therefore, regarding to the compounds of the culture medium, the nitrogen sources, trace elements and salt ingredients were tested in a qualitative and quantitative manner. Additionally, the optimal initial Glucose concentration was specified and different sugar species were investigated for their usability by the bacteria. Based on these results, cost-effective sugar substitutes were selected and examined for their suitability for D-lactic acid production. Furthermore, the productivity of the bacteria could be increased by determining their optimal pH-value and by the development of batch and fed batch fermentation strategies with calcium carbonate and ammonia as neutralizing agents. Unfortunately, using ammonia as neutralizer, led to an inhibition due to the increase of undissociated acid during the fermentation at constant pH. Finally, this issue could be circumvented by a D-lactic acid concentration dependent pH-shift. In Addition, the bacterial lag-phase could be eliminated and the productivity was increased by cell immobilization, when using a polyvinyl alcohol matrix (Lentikats (R)) for bacteria entrapment. Eventually, combining all results, a high yielding procedure for the production of D-lactic acid on the basis of renewable resources could be realized using ammonia as neutralizing agent.
