Abstract

Numerous cultivars of Japanese flowering cherry (Prunus subgenus Cerasus) are recognized, but in many cases they are difficult to distinguish morphologically. Therefore, we evaluated the clonal status of 215 designated cultivars using 17 SSR markers. More than half the cultivars were morphologically distinct and had unique genotypes. However, 22 cultivars were found to consist of multiple clones, which probably originate from the chance seedlings, suggesting that their unique characteristics have not been maintained through propagation by grafting alone. We also identified 23 groups consisting of two or more cultivars with identical genotypes. Most members of these groups were putatively synonymously related and morphologically identical. However, some of them were probably derived from bud sport mutants and had distinct morphologies. SSR marker analysis provided useful insights into the clonal status of the examined Japanese flowering cherry cultivars and proved to be a useful tool for cultivar characterization.

26. Nomenclature of Tokyo cherry (Cerasus× yedoensis'Somei-yoshino', Rosaceae) and allied interspecific hybrids based on recent advances in population genetics

Abstract

The flowering cherries, Cerasus (=Prunus subg. Cerasus; Rosaceae), including C. × yedoensis 'Somei-yoshino', are popular ornamental trees in temperate regions of the world. Recent advances in molecular and population genetics have revealed that this group originated as an interspecific hybrid. We used these findings and the verification of type specimens to examine the nomenclature of nothotaxa including C. × yedoensis. Four nothotaxa, C. × yedoensis, C. × nudiflora , C. × sacra and C. × kashioensis, were confirmed to be hybrids resulting from crosses between C. itosakura and taxa of the C. serrulata complex (C. speciosa, C. sargentii, C. jamasakura and C. leveilleana, respectively). In addition, the hybrids found on Jeju Island are C. × nudiflora, a new combination for an interspecific hybrid shown to be genetically distinct from C. × yedoensis. A new nothospecies, C. × kashioensis is proposed for a hybrid between C. itosakura and C. leveilleana.

Abstract

Prunus jamasakura is a species of Prunus native to eastern Asia. We determined the first complete chloroplast genome of Prunus jamasakura using genome skimming approach. The cp genome was 157,905 bp long, with a large single-copy region (LSC) of 85,910 bp and a small single-copy region (SSC) of 19,123 bp separated by a pair of inverted repeats (IRs) of 26,436 bp. It encodes 129 genes, including 84 protein-coding genes, 37 tRNA genes, and 8 ribosomal RNA genes. We also reconstructed the phylogeny of Prunus sensu lato using maximum likelihood (ML) method, including our data and previously reported cp genomes of related taxa. The phylogenetic analysis indicated that P. jamasakura is closely related with Prunus speciosa. [ABSTRACT FROM AUTHOR]

Abstract The optimal conditions for fusion of leaf protoplasts of Populus alba, Betula platyphylla, and Alnus firma by electric treatment were alternate current (AC) 200 V cm−1 in 2.5 mM CaCl2 for a pearl chain formation and direct current (DC) pulse of 100 μs at 2 kV  cm−1 After interfamilial cell fusion treatment, colonies were obtained using liquid media containing 2,4-D or NAA as an auxin and BA or CPPU as a cytokinin at 0.1, 1, or 10 µM in MS (Murashige and Skoog (1962) Physiol. Plant. 15: 473--497), 1/2salt MS, or NH4NO3-free MS containing 0.6 M mannitol and 3% sucrose (totaling 147 combinations). Two shoots after electric cell fusion treatment between P. alba and B. platyphylla, and 12 regenerated plants after electric cell fusion between P. alba and A. firma, were obtained from colonies induced on agar medium containing NAA, IBA, CPPU, and BA. Seven lines of the latter 12 plants which were regenerated later cultured in vitro had serrated leaves different from those of P. alba.
