The ability of older adults (48 to 72) with relatively intact low-frequency hearing to detect the motion of an acoustic source was investigated using dynamically varying interaural delays. Thresholds were measured using a single-interval two-alternative forced-choice task in which listeners determined if the sound source was moving or stationary. Motion thresholds were significantly larger than stationary localization thresholds. No correlation was observed between age and motion-detection ability for the age range tested. An interesting finding was that there were similar thresholds for older and younger adults. Results suggest reliance on dominant low-frequency binaural timing cues unaffected by high-frequency hearing loss in older adults.

Auditory stream segregation and informational masking were investigated in brain-lesioned individuals, age-matched controls with no neurological disease, and young college-age students. A psychophysical paradigm known as rhythmic masking release (RMR) was used to examine the ability of participants to identify a change in the rhythmic sequence of 20-ms Gaussian noise bursts presented through headphones and filtered through generalized head-related transfer functions to produce the percept of an externalized auditory image (i.e., a 3D virtual reality sound). The target rhythm was temporally interleaved with a masker sequence comprising similar noise bursts in a manner that resulted in a uniform sequence with no information remaining about the target rhythm when the target and masker were presented from the same location (an impossible task). Spatially separating the target and masker sequences allowed participants to determine if there was a change in the target rhythm midway during its presentation. RMR thresholds were defined as the minimum spatial separation between target and masker sequences that resulted in 70.7% correct-performance level in a single-interval 2-alternative forced-choice adaptive tracking procedure. The main findings were (1) significantly higher RMR thresholds for individuals with brain lesions (especially those with damage to parietal areas) and (2) a left–right spatial asymmetry in performance for lesion (but not control) participants. These findings contribute to a better understanding of spatiotemporal relations in informational masking and the neural bases of auditory scene analysis.

Tinnitus is a sound heard by 15% of the general population in the absence of any external sound. Because external sounds can sometimes mask tinnitus, tinnitus is assumed to affect the perception of external sounds, leading to hypotheses such as "tinnitus filling in the temporal gap" in animal models and "tinnitus inducing hearing difficulty" in human subjects. Here we compared performance in temporal, spectral, intensive, masking and speech-in-noise perception tasks between 45 human listeners with chronic tinnitus (18 females and 27 males with a range of ages and degrees of hearing loss) and 27 young, normal-hearing listeners without tinnitus (11 females and 16 males). After controlling for age, hearing loss, and stimulus variables, we discovered that, contradictory to the widely held assumption, tinnitus does not interfere with the perception of external sounds in 32 of the 36 measures. We interpret the present result to reflect a bottom-up pathway for the external sound and a separate top-down pathway for tinnitus. We propose that these two perceptual pathways can be independently modulated by attention, which leads to the asymmetrical interaction between external and internal sounds, and several other puzzling tinnitus phenomena such as discrepancy in loudness between tinnitus rating and matching. The present results suggest not only a need for new theories involving attention and central noise in animal tinnitus models but also a shift in focus from treating tinnitus to managing its comorbid conditions when addressing complaints about hearing difficulty in individuals with tinnitus.SIGNIFICANCE STATEMENT Tinnitus, or ringing in the ears, is a neurologic disorder that affects 15% of the general population. Here we discovered an asymmetrical relationship between tinnitus and external sounds: although external sounds have been widely used to cover up tinnitus, tinnitus does not impair, and sometimes even improves, the perception of external sounds. This counterintuitive discovery contradicts the general belief held by scientists, clinicians, and even individuals with tinnitus themselves, who often report hearing difficulty, especially in noise. We attribute the counterintuitive discovery to two independent pathways: the bottom-up perception of external sounds and the top-down perception of tinnitus. Clinically, the present work suggests a shift in focus from treating tinnitus itself to treating its comorbid conditions and secondary effects.

Background: In the UK, African communities are a focus of public health efforts to increase uptake of HIV testing. Mobile phone interventions may be an innovative way of reaching migrant groups who are known to face multiple obstacles in accessing mainstream health services. This paper presents findings from a feasibility study that used participatory approaches to investigate the use of a text messaging intervention to encourage HIV testing among migrant African communities. Methods: Participants were recruited in the city of Nottingham by a team of community researchers. They were sent two text messages per week (one on HIV and one on general health) for 12 weeks. Baseline and follow-up questionnaires were completed to measure HIV testing behaviour, HIV-related knowledge and attitudes and general health. Participants’ views on the intervention were solicited. Results: One hundred and sixty-nine participants were enrolled in the study. Follow-up data on HIV testing were obtained for 76 participants (45%) and complete follow-up measures were available from 60 participants (36%). Eight reported seeking an HIV test during the study period. There were statistically significant positive changes in attitudes about HIV and a trend towards increased knowledge about HIV. One-third of participants reported improvements in physical activity levels, diet and stress management following the intervention. The intervention messages and structure were positively evaluated. Conclusion: Well-designed mobile phone messaging proved to be a feasible and acceptable intervention to promote both HIV testing and lifestyle behaviours among African migrant communities in the UK. When co-constructed with communities, they hold considerable promise for overcoming some of the health-related barriers faced by migrant populations in new countries. Future research and service development should focus on exploiting and evaluating this potential in relation to other key health priorities.

Comparison of day-neutral temperate maize and photoperiod-dependent teosinte revealed divergent leaf gene expression networks that implicate different genes of the same gene families in response to flowering., Temperate maize was domesticated from its tropical ancestor, teosinte. Whereas temperate maize is an autonomous day-neutral plant, teosinte is an obligate short-day plant that requires uninterrupted long nights to induce flowering. Leaf-derived florigenic signals trigger reproductive growth in both teosinte and temperate maize. To study the genetic mechanisms underlying floral inductive pathways in maize and teosinte, mRNA and small RNA genome-wide expression analyses were conducted on leaf tissue from plants that were induced or not induced to flower. Transcriptome profiles reveal common differentially expressed genes during floral induction, but a comparison of candidate flowering time genes indicates that photoperiod and autonomous pathways act independently. Expression differences in teosinte are consistent with the current paradigm for photoperiod-induced flowering, where changes in circadian clock output trigger florigen production. Conversely, differentially expressed genes in temperate maize link carbon partitioning and flowering, but also show altered expression of circadian clock genes that are distinct from those altered upon photoperiodic induction in teosinte. Altered miRNA399 levels in both teosinte and maize suggest a novel common connection between flowering and phosphorus perception. These findings provide insights into the molecular mechanisms underlying a strengthened autonomous pathway that enabled maize growth throughout temperate regions.

The aim of this study was to explore the impact of Sleep Tight, a behavioural sleep programme, on child sleep problems and family wellbeing. A service evaluation of 40 families with a child aged 0–12 years who completed Sleep Tight through Sure Start children's centres was performed. A before and after within-subjects design was used and outcomes assessed using the Sleep Disturbance Index ( Quine, 1991 ). Evidence of a statistical effect was found for settling, night waking, sleeping in parents' bed, and parents up at night. On average, children had 2.5 hours more sleep per night after the intervention. Parents reported changes in family life and child's daytime behaviour. The findings support the use of Sleep Tight as an effective, time-limited behavioural sleep programme.

Introduction: The use of seclusion as a means of managing the extreme behaviours forensic patients in secure settings is a controversial yet often common practice, despite there being little evidence that seclusion as a practice has any significant therapeutic value for the patient. The aim of this study was to explore the use of seclusion and whether this differs as a function of gender and diagnosis across secure services. Method: This study collated data from 11 medium and low secure hospitals that admit male and female patients, with some services providing services for patients with intellectual disability (with or without co-morbid disorders), and others for patients with mental illness and/or personality disorder only. Results: Both gender and diagnosis were associated with differential seclusion rates. Seclusions were three times longer for patients in the non-ID compared to the ID service. Background: There is a public health need to tackle high levels of undiagnosed (or late diagnosed) HIV amongst the UK's African communities. This research aimed to assess the feasibility and acceptability of using a text messaging intervention to encourage uptake of HIV testing amongst this population in a UK city. Methods: Participatory research adopting a mixed-methods design. Four distinct stages included: [1] Formative Research: Six focus group discussions (FGDs) were conducted with diverse sections of the African community to assess perceptions about HIV and to inform message development (n=48). The Health Belief Model (HBM) was used as an organising framework for data analysis and interpretation. [2] Message Development: SMS messages were developed based on HBM constructs, existing HIV campaigns and FGD findings, and tailored according to language, gender and religion. 12 HIV-related and 12 generic health-related text messages were developed and piloted using elicitation interview processes. [3] Intervention and outcomes: 172 participants were recruited. They received 2 messages per week for 12 weeks. Data was collected in pre and post questionnaire surveys assessing uptake of HIV testing, HIV-related attitudes and knowledge and perceived general health. [4] Evaluation: Acceptability and meaningfulness of the intervention were explored via semi-structured telephone interviews (n=21). Data were analysed using thematic content analysis.

OBJECTIVE To examine the measurement properties of the Chronic Pain Acceptance Questionnaire Revised (CPAQ-R) and its subscales with people with knee osteoarthritis using Rasch analysis. DESIGN Cross-sectional questionnaire study. PATIENTS A total of 176 participants with radiographic evidence of osteoarthritis of the knee, as identified by a Kellgren-Lawrence grade ≥ 2, and pain on most days for at least the past month. METHODS Participants completed the CPAQ-R at home within a set of measures covering different aspects of osteoarthritis pain. The questionnaires were returned by pre-paid envelope. Rasch analysis was conducted on the Activity Engagement and Pain Willingness subscales and the Total scale using Rasch Unidimensional Measurement Models (RUMM2020). RESULTS The Activity Engagement and Pain Willingness subscales fit the Rasch model following minimal changes, including re-scoring and removal of item 14 due to misfit. Both subscales passed tests of unidimensionality. Although the Total scale could be adjusted to yield adequate fit statistics, it demonstrated multidimensionality. CONCLUSION The Activity Engagement and Pain Willingness subscales have good measurement properties for 2 distinct factors relevant to pain acceptance. CPAQ-R is a valid measurement tool that may help target and evaluate response to treatments that address low activity engagement and pain willingness in people with osteoarthritis.

Mennonite Economic Development Associates (MEDA) was launched as an investment club in 1953 when a group of North American Mennonite business people joined together to support the development of communities in Paraguay, Uruguay, and Argentina. With their business background, this group of early ‘impact investors’ determined that they would provide loans to small to medium enterprises (SME) in order to catalyse sustainable economic growth. They offered the loans as high-risk venture capital and mitigated the risks with the provision of business coaching and technical assistance. Since those early days, MEDA and the SME investment fund managers which it has co-founded (Microvest and Sarona Asset Management) have continued to make impactful investments and to work towards a common development goal, ‘to help people help themselves’ (Fretz, 1978 : 19). This paper presents a case study of the 60-year ‘MEDA experiment’, (Fretz, 1978), describes specific activities and innovations, and identifies MEDA's learnings that have emerged from this SME investment experience.

PurposeAre peaches, Caesar salad and chocolate masculine or feminine food? Literature suggests that there is a clear association between certain types of food, portion sizes and gendered identities. This research paper and short film aims to explore the theory in practice of food consumption for young consumers, particularly impression management required to create/maintain an attractive identity to the opposite sex.Design/methodology/approachThe authors adopt an interpretive approach to an in‐depth analysis of the food practices of an all male and an all female household. They use a theory in practice methodology to explore their food consumption.FindingsIt is found that despite enlightenment in many areas, gendered identities are still strongly associated with food consumption. The experiment in which each household consumed a meal associated with the opposite gender offers insight into the association between food consumption and gendered identity. The social implications of the research demonstrate that masculine identity is supported and negotiated through what he is eating, whereas feminine identity is being constructed by what she is not eating. It is concerning that an attractive feminine identity is premised on omission rather than consumption and traps many females into a negative and potentially harmful relationship with food consumption.Originality/valueThe use of videography allows insight into the negotiation of an underpinning cultural attitude where women eat less to be what consumer culture has defined as an attractive feminine identity which means being slimmer and smaller than males.

To determine aerosol administration capability and therapeutic efficacy of the new formulation of hyaluronan cisplatin conjugates, HylaPlat™ (HA-Pt), for lung cancer treatment.In vitro formulation stability test, 2D and 3D spheroid cell culture and in vivo efficacy studies using mouse orthotopic allograft models were conducted.The HA-Pt effectively attenuated cell growth in 2D and 3D cultures with IC50 of 2.62 and 5.36 μM, respectively, which were comparable to those with unconjugated control cisplatin-dependent growth inhibition (IC50 1.64 and 4.63 μM, respectively). A single dose of either 7.5 or 15 mg/kg HA-Pt (cisplatin equivalent) by intratracheal aerosol spray 7 days after Lewis lung carcinoma (LLC) cell inoculation markedly inhibited growth of LLC allografts in mouse lungs and resulted in a 90 or 94% reduction of tumor nodule numbers, respectively, as compared to those from the PBS control. Cancer stem cells and cisplatin resistant cells marker, CD44 expression decreased in the tumor nodules of the HA-Pt but not in those of cisplatin treated groups.The current study suggests that an intratracheal aerosol administration of the HA-Pt nanoparticles offers an effective strategy for lung cancer treatment and this treatment may induce only limited cisplatin resistance.

Purpose To examine the measurement properties of measures of psychological constructs in people with knee osteoarthritis. Method Participants with osteoarthritis of the knee completed the beck depression inventory (BDI-II), state-trait anxiety inventory (STAI), arthritis helplessness index (AHI), fatigue severity scale (FSS), coping strategies questionnaire (CSQ), beliefs about pain control questionnaire (BPCQ), illness perceptions questionnaire-revised (IPQ-R), pain self-efficacy questionnaire (PSEQ) at home as part of a set of measures covering different aspects of osteoarthritis pain. The questionnaires were returned by pre-paid envelope. Rasch analysis was used to check the psychometric properties of the scales in people with osteoarthritis. Results The STAI-SF was an acceptable measure of anxiety and the revised FSS an acceptable measure of fatigue, with removal of items 1 and 2. The BDI subscales were acceptable for measuring negative thoughts and behaviours related to depressive symptomatology with some modifications to the scale. The helplessness scale of the AHI was acceptable as a measure of helplessness. The PSEQ was an acceptable measure of self-efficacy and the CSQ as a measure of cognitive coping strategies. The BPCQ and IPQ-R did not fit the Rasch model. Conclusions These findings indicate that questionnaires need to be checked for their ability to measure psychological constructs in the clinical groups to which they will be applied. Implications for Rehabilitation For people with osteoarthritis, the STAI-SF is an acceptable measure of anxiety and the revised FSS an acceptable measure of fatigue with removal of items 1 and 2. The BDI subscales, but not the total score, are acceptable for measuring depressive symptomatology with some modifications to the scoring of the scale. And helplessness can be measured using the Helplessness subscale of the AHI. The PSEQ was an acceptable measure of self-efficacy and cognitive coping strategies can be measured with the CSQ. Rasch analysis highlighted lack of unidimensionality, disordered response thresholds and poor targeting in some measures commonly used for people with osteoarthritis.

Natural products obtained from various resources are often found to be new sources of effective drugs for disease including cancer. The novel extracellular polysaccharides (exopolysaccharides) purified from Parachlorella kessleri (PCEPS), a generous gift by Panac ADVANCE Co. Ltd. (Tokyo, Japan), were evaluated on their antitumor and immunomodulatory effects in cell culture and mouse colon carcinoma peritoneal dissemination model. In two-dimensional cell culture, the PCEPS treatment inhibited cell growth of both murine and human colon carcinoma cells in a dose- and time-dependent manner. In contrast, the cell proliferation of mouse splenocytes (SPLs) and bone marrow cells (BMCs) were stimulated by the treatment with PCEPS. The treatment with PCEPS also increased specific subpopulations of the cells in BMCs: antigen presenting cells (CD19+ B cells, 33D1+ dendritic cells and CD68+ macrophage) and CD8+ cytotoxic T cells. In three-dimensional spheroid culture, spheroid growth of CT26 cells co-cultured with HL-60 human neutrophilic promyeloblasts and Jurkat cells (human T lymphoblasts), but not THP-1 human monocytes/macrophages, was significantly attenuated by PCEPS treatment. In transwell co-culture of CT26 and HL-60, THP-1 or Jurkat cells, mRNA level of NOX2 in HL-60 and THP-1 cells, and TNFα, IFNγ, IL-2 and granzyme B (GZMB) in Jurkat cells was significantly increased by PCEPS treatment. In a mouse CT26 colon carcinoma peritoneal dissemination model, intraperitoneal injection of PCEPS (10 mg/kg, started from 1 week after CT26 inoculation for 11 days, twice per week) significantly attenuated the growth of CT26 colon carcinoma in syngeneic mice by PCEPS treatment-associated apoptosis without any noticeable adverse effect. Analysis of subpopulation of intraperitoneally infiltrating immune cells collected from CT26 cell tumor-bearing mice found that PCEPS treatment increased CD4+ and CD8+ T lymphocytes, CD19+ B cell, and CD68+ macrophage populations. In contrast, 33D1+ dendritic cell and LY6G+ neutrophil populations were decreased as compared to PBS-treated group. The present study suggests that PCEPS inhibits colon carcinoma growth via direct cell growth inhibition and a stimulation of the host antitumor immune responses. Taken together, the current study suggests that exopolysaccharides derived from Parachlorella kessleri contain significant bioactive materials that inhibit colon carcinoma growth. Aggressive behaviour is a problem for services providing care for people with intellectual disabilities, affecting the quality of life of the individual and the quality of care provided. Current research trends, which focus on risk factors and mental health problems, are discussed. Other factors that could contribute to aggression in people with intellectual disability (PWID), such as lifestyle and environmental issues are examined. A methodology that would allow for the integration of all these factors, Behavioural Sequence Analysis, is a suitable investigative approach to this problem.

Within the social constructionist (narrative, solution-focused and collaborative language systems) therapies, therapists have increasingly focused on identifying and mobilizing clients’ resources and competencies. How such client resources and competencies are identified and elaborated can be considered empirical matters for analysts of therapeutic discourse. The present paper examines resourceful sequences of dialogue in 12 single session lifestyle consultations for how counsellors conversationally engaged consultees, and for what transpired within these sequences of dialogue. These sequences were micro-analyzed using conversation analysis and retrospective comments from consultees and therapists involved in these sequences were solicited using a videotaped replay procedure known as comprehensive process analysis [Elliott (1989). The combined conversation analyses and related comments for three of the 23 videotaped passages analyzed are reported with the aim of enhancing therapists’ conversational practices in hosting resourceful dialogues with clients.

The concept of ecological 'traps' is based in theory from ecology and conservation biology that has now found application to infectious diseases with a study from Paul Turner's group. This study is important because it offers a mathematical model of ecological traps, applies this model to viruses, and tests the model in a bacteria-phage system. Although there will be technical hurdles to overcome, this concept might lead to benefits for both health and industry.

The activity of the maize (Zea mays) florigen gene ZEA CENTRORADIALIS8 (ZCN8) is associated with the floral transition in both day-neutral temperate maize and short-day (SD)-requiring tropical maize. We analyzed transcription and chromatin modifications at the ZCN8 locus and its nearly identical paralog ZCN7 during the floral transition. This analysis was performed with day-neutral maize (Zea mays ssp. mays), where flowering is promoted almost exclusively via the autonomous pathway through the activity of the regulatory gene indeterminate1 (id1), and tropical teosinte (Zea mays ssp. parviglumis) under floral inductive and noninductive photoperiods. Comparison of ZCN7/ZCN8 histone modification profiles in immature leaves of nonflowering id1 mutants and teosinte grown under floral inhibitory photoperiods reveals that both id1 floral inductive activity and SD-mediated induction result in histone modification patterns that are compatible with the formation of transcriptionally competent chromatin environments. Specific histone modifications are maintained during leaf development and may represent a chromatin signature that favors the production of processed ZCN7/ZCN8 messenger RNA in florigen-producing mature leaf. However, whereas id1 function promotes histone H3 hyperacetylation, SD induction is associated with increased histone H3 dimethylation and trimethylation at lysine-4. In addition, id1 and SD differently affect the production of ZCN7/ZCN8 antisense transcript. These observations suggest that distinct mechanisms distinguish florigen regulation in response to autonomous and photoperiod pathways. Finally, the identical expression and histone modification profiles of ZCN7 and ZCN8 in response to floral induction suggest that ZCN7 may represent a second maize florigen.

The aim of the present study was to evaluate the possible mechanisms of testicular toxicity of GR40370X, a follow-up 5-hydroxytryptamine (5-HT) receptor agonist. Administration to adult male rats of a single (toxic) dose of 750 mg/kg GR40370X induced marked distension of seminiferous tubules and an associated increase in testis weight at 12-24 h with a gradual recovery to normal by 96 h. Seminiferous tubule distension was due to expansion of the lumen, which occurred at all stages of the spermatogenic cycle and was accompanied by vacuolation of the cytoplasm of elongating spermatids. Seminiferous tubule distension was preceded/accompanied by distension of the efferent ducts and rete testis with maximal changes evident at 24-48 h. These changes could not be explained by increases in seminiferous tubule fluid or interstitial fluid production, as both were reduced (15-20%), rather than increased, by treatment. Examination of the vasculature after treatment with 750 mg/kg GR40370X revealed significant changes that were maximal at 4 h and thus preceded rete/testicular changes. Veins of the mediastinal venous plexus, which overlies the rete, were constricted and arteriovenous anastomoses in the spermatic cord were shut/constricted, as determined (indirectly) by measurement of the dilution of outflowing testicular venous blood by incoming arterial blood. The latter effect of GR40370X could be blocked by co-administration of minoxidil, a vasodilator. Vascular effects of GR40370X had normalised by 24-48 h. It was also noted that administration of a toxic dose of GR40370X significantly lowered blood levels of LH and testosterone, though these changes were considered to be incidental and not involved in the other changes described above. None of the above changes were induced by a pharmacologically active dose (1 mg/kg) of GR40370X. It is concluded that the mechanism of testicular toxicity induced by 750 mg/kg GR40370X results from primary effects on the vasculature of the testis/neighbouring region, which in turn lead to impaired fluid resorption from the efferent ducts and rete and thence to accumulation of seminiferous tubule fluid in the rete and testis.

Nanoparticle-based chemotherapy, while promising, remains clinically unsuccessful, mainly due to a lack of targeted delivery methods of the therapeutics into cancer tissues and the side effect of chemotherapeutics. We have demonstrated that the nanoparticle formulation with hyaluronan-cisplatin conjugates (HylaPlat) is suggested to be an effective chemotherapeutic delivery method in breast cancer mouse models (Cai, et al., J Surg Res., 2008; Cai, et al., Ther Deliv, 2011). The objectives of the present study were to examine the growth inhibition efficacy of HylaPlat formulation on lung adenocarcinoma cells in cell culture and to examine the therapeutic efficiency of the local administration of HylaPlat on lung adenocarcinoma in mice. Effect of the HylaPlat on the growth of lung carcinoma was evaluated using Lewis Lung carcinoma (LLC) cells in 2D culture and 3D spheroid cell culture, as well as, orthotopic autografts in C57BL/6 mice lungs. Cell culture studies clarified that the HylaPlat effectively attenuated cell growth in 2D and 3D spheroid culture with IC50 of 0.35μM and 1.35 μM, respectively; whereas control cisplatin-dependent growth inhibition was achieved with several fold higher concentrations (IC50 1.64 and 3.6 μM, respectively). The 3D spheroid cell culture study also revealed that the treatment with HylaPlat induced apoptosis in the cells located on the peripheral area of the spheroid first, this effect lasted for several days. A single intratracheal administration of 7.5mg/kg HylaPlat (1mg cisplatin equivalent/kg) seven days after LLC cell inoculation almost completely inhibited growth of LLC autografts in the mouse lungs. Histological analysis of dissected lungs revealed that a small number of microscopic tumor nodules were detected in the treated mouse lungs, whereas several large tumors were detected in the untreated control mouse lungs. Normal lung architectures of the trachea, bronchioles and alveoli were maintained in the treated mice although small inflammation spots can be detected at peripheral areas. Apoptotic index was significantly higher in the treated tumors than PBS treated control tumors, suggesting that cisplatin was successfully delivered to the tumor tissues by nanoparticle formulated HylaPlat and caused apoptosis of tumor cells. The pharmacokinetics of cisplatin 14 days after intratracheal administration of HylaPlat is under investigation. Taken together, the current study suggests that an intratracheal administration of HylaPlat nanoparticles offer an effective strategy for lung cancer treatment.
