An automated instrument to measure specific surface area (SSA) of powdered materials was designed using the Brunauer–Emmett–Teller (BET) isotherm method. A sample of known SSA was introduced to make the method simple, accurate, and fast. The cost of the instrument was reduced by operating the instrument at a fixed partial pressure. A piezoelectric sensor was introduced to detect the changes in gas flow rates with better sensitivity and low cost. The analyzer was used to analyze the SSA of an unknown sample, to compare the SSA of two samples, and to identify unknowns using the SSA. The error of the instrument was less than 0.1%. Asiatic pennyworts (AP) (Centella asiatica) are among the most popular leafy vegetable species in Sri Lanka. Five AP cultivars, namely, Lowland AP (LAP), Giant AP1 (GAP1), Giant AP2 (GAP2), Salad AP (SAP), and Medicinal AP (MAP) are grown in the country. Two other leafy vegetable species, water pennyworts (WP) (Hydrocotyle verticillata) and Korean pennyworts (KP) (Adenophora triphylla), are also designated under 'pennyworts' in Sri Lanka. The consumerpreference on the pennywort bunches available in the market and the salad properties are yet to be studied. Also, no attempts have been reported for the identification of species delimits and phylogenetic relationships among the different cultivars of C. asiatica. In the present study, consumer preference on the appearance of leaf bunches and salads of pennyworts were assessed. The intra- and interspecific variation of pennyworts grown in Sri Lanka were also studied. The genomic DNA extracted from the immature leaves was PCR amplified for the DNA barcoding markers matK-trnT, atpB-rbcL, and rbcL. The PCR products were sequenced and used to assess the intraand interspecific variation. The consumers mostly preferred to purchase the leaf bunches of GAP2 because of its large leaf size and appealing look. However, the highest preference for salads was reported for the dishes prepared using LAP, MAP, and WP. The clade structure of the phylogenetic tree drawn for AP cultivars shows an intraspecific variation. Our study also highlights the importance of producing an improved AP cultivar with larger leaf size, improved flavor properties, and sinuate leaf margin. [ABSTRACT FROM AUTHOR]

The development of phosphorous deficiency (PD) tolerant cultivars is important for rice farming. The DNA markers linked to Pup1, the major quantitative trait locus (QTL) for PD tolerance, have been developed using reference rice cultivars Kasalath and Nipponbare, however these markers cannot be directly employed in regional breeding programmes as specific cultivars that are important to a region/country may contain diverse QTL haplotypes complicating marker assisted breeding (MAB). Thus identification of the Pup1 linked DNA marker haplotypes present in local cultivars is the prerequisite for MAB of PD tolerance in rice. Therefore, the objective of this study was to identify the Pup1 haplotypes from a panel of 30 rice cultivars important for breeding programmes in Sri Lanka. A total of 17 Pup1 linked DNA markers were used to genotype the rice cultivars previously characterised for PD tolerance in comparison to Kasalath and Nipponbare. Overall 28 Pup1 haplotypes were identified in the marker allele based clustergram and the Pup1 hapoltype in Kasalath was observed in four PD tolerant cultivars Sudubalawee, Bw 364, Kaluheenati and Marss. However, the PD tolerant cultivars Bg 94-1, Bg 403, H-7, H-4 and Murungakayan contain haplotypes that are clustered separately from Kasalath indicating the possibility of conferring PD tolerance in the absence of Kasalath haplotype. The marker-trait association analysis for the PD tolerance revealed that selection of the favourable alleles such as 433 bp allele of K46-K2 marker could increase PD tolerance by 0.73. The efficacy of modern rice varieties has been challenged by global warming because of their incapability to tolerate heat stress. Breeding for heat tolerance (HT) is therefore essential. Molecular breeding studies on HT in hybrid rice is sparse although HT quantitative trait loci (QTL) have been discovered. China is the leading country for hybrid rice production, however, introgression of HT alleles to hybrids is not practiced in breeding programmes. Therefore, the objectives of this study were to introgress HT alleles from indica donors Bg 90-2 and Mengguandamagu to SH 527, a popular parent in hybrid variety development, and to identify the associated DNA markers with HT to facilitate hybrid rice molecular breeding in China. The BC2F2:4 introgression lines were produced by crossing donor and recurrent parents and their performances were evaluated at two provinces of China in 2011 and 2012 under normal and heat stress conditions. The data were subjected to ANOVA and HT was compared against the standard heat-check variety N22. Out of 600 SSR markers, 61 and 59 markers were polymorphic for the two crosses SH 527/ Bg 90 - 2 and SH 527/ Mengguandamagu, respectively and introgression lines were subsequently genotyped. The single-marker analysis was performed to detect marker trait associations. A total of 46 HT introgression lines were obtained with 88.3 % of homozygosity for HT alleles and seven high yielding HT lines were specifically selected for the provinces. Under heat stress, 42 significant marker trait associations were detected. Two of these were previously reported in HT QTLs implying their applicability in rice breeding programmes. Purpose : The aril/flesh of the fruitlets is the main edible part of jackfruit (Artocarpus heterophyllus Lam.). There are two flesh types; soft flesh (SF) (Wela) and fi rm flesh (FF) (Waraka) (i.e. highly preferred). There is no strategy available to identify FF plants at nursery or vegetative stages. Thus the present study was conducted to assess the applicability of leaf morphological variation and DNA bar-coding to differentiate SF and FF bearing jackfruit plants. Research Method : The syncarp morphology and sensory parameters of SF and FF ripen flesh were also assessed to check the variation with 10 trees from each flesh type. The trees were chosen from SF and FF types and leaves and mature syncarps were collected. The leaf and syncarp morphological variation, pH, soluble solids and ascorbic acid content were measured. The sensory parameters of the flesh types were ranked by employing a taste panel. The DNA bar-coding was carried out for two loci rbcL and ITS to identify the sequence variants. Findings : The leaf tip length was longer in SF, ascorbic acid content was higher in FF and all the other parameters were not different (P < 0.05). The SF leaves were elliptical whereas FF leaves were obovate. The association analysis revealed that the sweetness and sourness of flesh were inversely associated. The DNA bar-coding for rbcL and ITS showed that rbcL locus was not informative however, ITS locus can be used to characterize the genetic diversity of jackfruit. Research Limitations : The detected haplotypic variants must be further validated with a larger set of jackfruit trees collected from diverse sites. Originality/value : Three haplotypes were detected within ITS locus where CC haplotype is characteristic to SF and CT or TC is detected in FF enabling marker assisted seedling selection. Abstract Background Medullary nephrocalcinosis and distal renal tubular acidosis are closely associated and each can lead to the other. These clinical entities are rare in patients with nephrotic syndrome and polycythaemia is an unusual finding in such patients. We describe the presence of medullary nephrocalcinosis, distal renal tubular acidosis and polycythaemia in a patient with nephrotic syndrome due to minimal change disease. Proposed mechanisms of polycythaemia in patients with nephrotic syndrome and distal renal tubular acidosis include, increased erythropoietin production and secretion of interleukin 8 which in turn stimulate erythropoiesis. Case presentation A 22 year old Sri Lankan Sinhala male with nephrotic syndrome due to minimal change disease was investigated for incidentally detected polycythaemia. Investigations revealed the presence of renal tubular acidosis type I and medullary nephrocalcinosis. Despite extensive investigation, a definite cause for polycythaemia was not found in this patient. Treatment with potassium and bicarbonate supplementation with potassium citrate led to correction of acidosis thereby avoiding the progression of nephrocalcinosis and harmful effects of chronic acidosis. Conclusion The constellation of clinical and biochemical findings in this patient is unique but the pathogenesis of erythrocytosis is not clearly explained. The proposed mechanisms for erythrocytosis in other patients with proteinuria include increased erythropoietin secretion due to renal hypoxia and increased secretion of interleukin 8 from the kidney. This case illustrates that there may exist hitherto unknown connections between tubular and glomerular dysfunction in patients with nephrotic syndrome.

Abstract Background Cytomegalovirus associated transverse myelitis among immunocompetent adults has been rarely reported. We report a patient presenting with clinical myelitis followed by previously unreported finding of cytomegalovirus deoxyribonucleic acid in cerebrospinal fluid. Case report A forty year old immunocompetent male presented with acute onset progressive bilateral lower limb weakness. His spinal magnetic resonance imaging findings, cerebrospinal fluid analysis and clinical picture were compatible with transverse myelitis. Polymerase chain reaction of the cerebrospinal fluid for cytomegalovirus was positive while other infectious agents were not detected by serology or polymerase chain reaction. He was treated with intravenous ganciclovir with partial clinical response. Conclusion Viral genome detection in the cerebrospinal fluid was performed but negative in five out of ten reported cases of cytomegalovirus associated transverse myelitis in the immunocompetent host. In previous cases the inability to isolate the virus in cerebrospinal fluid was considered favouring an immunological mechanism leading to pathogenesis rather than direct viral toxicity but this case is against that theory. This case highlights the fact that Cytomegalovirus should be considered as an aetiological agent in patients with transverse myelitis and that the virus may cause serious infections in immunocompetent host. Therefore this report is of importance to neurologists and physicians in general.

Abstract A sixty year old male presented with dark urine, symptomatic anaemia and peripheral gangrene following cold exposure. Investigations revealed that he had haemolysis and serological evidence of recent Epstein Barr virus infection. Although acrocyanosis is commonly associated with cold agglutinin disease, gangrene is a rare complication. Management of secondary cold agglutinin disease is mainly supportive.

Abstract Introduction Post-infectious cerebellitis is an acute form of inflammatory encephalitis mainly limited to the cerebellum. It is commonly found in children, especially after viral infections such as Epstein-Barr virus. Post-infectious cerebellitis presents with acute onset dysarthria and ataxia. To the best of our knowledge, this is the first case report of post-infectious cerebellitis in a patient with both dengue and Epstein-Barr viral infection confirmed on serology. Case presentation A 43-year-old Sri Lankan Sinhala man presented with an acute febrile illness associated with thrombocytopenia. While being managed as uncomplicated dengue fever, our patient developed dysarthria, ataxia and cerebellar signs. Our patient's infectious disease screen was positive for both dengue and Epstein-Barr specific immunoglobulin M. A cerebrospinal fluid analysis was suggestive of viral meningoencephalitis while cerebrospinal fluid serology was positive for dengue immunoglobulin G. T2-weighted magnetic resonance images were consistent with post-viral cerebellitis. The patient was given full supportive care and he made an uneventful complete recovery. Conclusion There have been no previously reported cases of post-infectious cerebellitis associated with both Epstein-Barr and dengue viral infections confirmed by serology. Our patient's clinical features and findings on the imaging studies were consistent with post-viral cerebellitis. This report highlights the need to screen for other possible more common etiologies of a particular presentation before coming to a specific diagnosis based on initial findings. Uncomplicated cases of cerebellitis can be successfully managed with appropriate supportive measures with good prognosis, as in this case.

Aflatoxins (AFs) are a group of mycotoxins produced by certain fungi of Aspergillus spp. AFs of major concern are B1 (AFB1), B2 (AFB2), G1 (AFG1), G2 (AFG2), and M1 (AFM1). AFM1 is a hydroxylated metabolite of AFB1 formed inside the animal's body which is excreted into milk of cows that consumed AFB1 contaminated feed. Consumption of AFM1-contaminated milk and subsequent dairy products causes negative health effects in consumers. This study was conducted to determine the occurrence and levels of AFM1 in cheese products available in the Sri Lankan market where AFM1 is not regularly monitored in milk while having an outdated regulatory limit of 1 ppb established for dairy products. Processed cheese (n = 28), hard cheese (n = 14), semi-hard cheese (n = 5), and soft cheese (n = 3) representing seven popular brands were collected. The samples were analyzed by Ultra High-Performance Liquid Chromatography Fluorescence Detection. AFM1 was detected in 40 samples (80 %), while 17 (34 %) and 37 (74 %) of the samples had AFM1 levels exceeding the maximum permitted limit set by Codex Alimentarius Commission (0.5 ppb) and the Netherlands (0.2 ppb). Further, 10 samples violated the Sri Lankan maximum limit of 1 ppb. Thirteen out of the 14 hard cheese (92.9 %, 0.11-14.43 ppb) and all semi-hard cheese samples (100 %, 0.29-0.65 ppb) contained AFM1. Most of the soft (66.7 %, 0.35-0.45 ppb) and processed (71.4 %, 0.11-1.35 ppb) cheese samples had AFM1. Most of the locally manufactured cheese products in Sri Lankan market may pose health risks to consumers. Li-air battery (LAB) technology is making continuous progress toward its theoretical capacity, which is comparable to gasoline. However, the sluggish reaction at the cathode is still a challenge. We propose a simple strategy to optimize the surface e g occupancy by adjusting the stoichiometric ratios of transition metal-based spinel structures through a controlled hydrothermal synthesis. Three distinct stoichiometries of Ni-Co oxides were used to demonstrate the direct correlation between stoichiometry and catalytic performance. The groundsel flower-like structure having a 1 : 1.4 Ni : Co atomic ratio with high surface area, high defect density, and an abundance of Ni 3+ at the surface with semi-filled e g orbitals was found to benefit the structure promoting high catalytic activities in aqueous and aprotic media. The assembled LAB cells employing this cathode demonstrate an exceptional lifespan, operating for 3460 hours and completing 173 cycles while achieving the highest discharge capacity of 13 759 mA h g -1 and low charging overpotentials. The key to this prolonged performance lies in the full reversibility of the cell, attributed to its excellent OER performance. A well-surface adsorbed, amorphous LiO 2 /Li 2 O 2 discharge product is found to possess high diffusivity and ease of decomposition, contributing significantly to the enhanced longevity of the cell.

The rational design of cost-effective and efficient electrocatalysts for electrochemical water splitting is essential for green hydrogen production. Utilizing nanocatalysts with abundant active sites, high surface area, and deliberate stacking faults is a promising approach for enhancing catalytic efficiency. In this study, we report a simple strategy to synthesize a highly efficient electrocatalyst for the hydrogen evolution reaction (HER) using carbonized luffa cylindrica as a conductive N-doped carbon skeleton decorated with Ag nanorings that are activated by introducing stacking faults. The introduction of stacking faults and the resulting tensile strain into the Ag nanorings results in a significant decrease in the HER overpotential, enabling the use of Ag as an efficient HER electrocatalyst. Our findings demonstrate that manipulating the crystal properties of electrocatalysts, even for materials with intrinsically poor catalytic activity such as Ag, can result in highly efficient catalysts. Further, applying a conductive carbon backbone can lower the quantities of metal needed without compromising the HER activity. This approach opens up new avenues for designing high-performance electrocatalysts with very low metallic content, which could significantly impact the development of sustainable and cost-effective electrochemical water-splitting systems.

Abiotic stresses, predominately drought, heat, salinity, cold, and waterlogging, adversely affect cereal crops. They limit barley production worldwide and cause huge economic losses. In barley, functional genes under various stresses have been identified over the years and genetic improvement to stress tolerance has taken a new turn with the introduction of modern gene-editing platforms. In particular, clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) is a robust and versatile tool for precise mutation creation and trait improvement. In this review, we highlight the stress-affected regions and the corresponding economic losses among the main barley producers. We collate about 150 key genes associated with stress tolerance and combine them into a single physical map for potential breeding practices. We also overview the applications of precise base editing, prime editing, and multiplexing technologies for targeted trait modification, and discuss current challenges including high-throughput mutant genotyping and genotype dependency in genetic transformation to promote commercial breeding. The listed genes counteract key stresses such as drought, salinity, and nutrient deficiency, and the potential application of the respective gene-editing technologies will provide insight into barley improvement for climate resilience.

Utilising the solid-state synthesis method is an easy and effective way to recycle spent lithium-ion batteries. However, verifying its direct repair effects on completely exhausting cathode materials is necessary. In this work, the optimal conditions for direct repair of completely failed cathode materials by solid-state synthesis are explored. The discharge capacity of spent LiCoO 2 cathode material is recovered from 21.7 mAh g -1 to 138.9 mAh g -1 under the optimal regeneration conditions of 850 °C and n(Li)/n(Co) ratio of 1:1. The regenerated materials show excellent electrochemical performance, even greater than the commercial LiCoO 2 . In this work we have designed and developed a low cost and simple instrument to purify air in an enclosure. The device sucks up the air in the enclosed area, kills the microorganisms and let clean air flow out. A combination of an ultra violet light and an electric field are used to kill the microorganisms in air. Three electric field chambers (radial, parallel, perpendicular) are used to clean air more effectively. Stainless steel meshes were used to increase the density of the electric fields. The outer covers were made with plastic and wood. The instrument was tested against an evaporated bacterial solution (Staphylococcus aureus) by letting it flow through the instrument and measuring the bacterial concentration of the output air. The results showed the instrument is extremely effective even when tested against high bacterial concentrations. Aim: This study evaluates the quality of care for patients admitted with acute myocardial infarction (AMI) in a tertiary hospital in Colombo using the European Society of Cardiology Quality of Care Working Group's guidelines (2017)., Methods: A recently implemented electronic AMI registry m-Health tool was used for prospective data collection. Each patient was assessed for eligibility for each of the six domains of quality. Global Registry of Acute Coronary Events Risk Model for predicted probability of mortality, and scores for risk of bleeding complications (CRUSADE) and severity of heart failure (Killip classification) were calculated as per published guidelines. A composite measure of quality was derived from compliance with the six domains. Patients were followed up via telephone at 30 days following discharge to evaluate outcome and satisfaction. Organisational information was assessed by administrative review and interview., Results: Between March 2017 and April 2018, 934 patients with AMI presented to the cardiology department. The majority of patients (90.4%) presented with features of ST-elevation myocardial infarction (STEMI). Mean (SD) overall compliance with the composite quality indicator (CQI) was 44% (0.07). Compliance of ≥50% to the CQI was achieved in 9.8% of STEMI patients. The highest compliance was observed for antithrombotics during hospitalisation (79.1%) and continuous measure of patient satisfaction (76.1%). The lowest compliance was for organisational structure and care processes (22.4%)., Conclusion: This study reports a registry-based continuous evaluation of the quality of AMI care from a low and middle-income country. The wood adulteration is a common problem and under-studied aspect in the timber industry of Sri Lanka. Hence we conducted a survey to assess the status of timber adulteration and check the applicability of morphometric parameters and DNA barcoding to detect the adulterated timber sources. We interviewed the stakeholders of the timber industry to collect information regarding timber adulterations. We measured the morphometric parameters; wood density and sizes of the xylem elements of the standard and adulterant species. For DNA barcoding, DNA was extracted from the wood of the selected standard and adulterant species and subjected to PCR using the markers, matK-trnT and atpB-rbcL. The PCR products were subjected to DNA sequencing. According to the survey, 92.5% of patrons, 73.7% of manufacturers and 96.7% of carpenters said timber adulteration is taking place in the country. The respondents said that the standard timber species; Tectona grandis, Artocarpus heterophyllus, and Swietenia macrophylla, profoundly undergo adulteration in Sri Lanka. The morphometric parameters did not discriminate the adulterant species from the standard species. The DNA barcodes matK-trnT and atpB-rbcL provided unique polymorphic DNA sequences with specific lengths for each species permitting the precise establishment of species identity and enabling the accurate detection of timber adulterations.

Background: Utilisation of endobronchial ultrasound-guided transbronchial needle aspiration (EBUS-TBNA) and guide sheath (EBUS-GS) for diagnosis and staging of lung cancer is gaining popularity, however, its impact on clinical practice is unclear. This study aimed to determine the impact of the introduction of endobronchial ultrasound-guided procedures (EBUS) on time to management decision for lung cancer patients, and on the utilisation of other invasive diagnostic modalities, including CT-guided trans-thoracic needle aspiration (CT-TTNA), bronchoscopy, and mediastinoscopy., Methods: Hospital records of new primary lung cancer patients presenting in 2007 and 2008 (Pre-EBUS cohort) and in 2010 and 2011 (Post-EBUS cohort) were reviewed retrospectively., Results: The Pre-EBUS cohort included 234 patients. Of the 326 patients in the Post-EBUS cohort, 90 had an EBUS procedure (EBUS-TBNA for 19.0% and EBUS-GS for 10.4% of cases). The number of CT-TTNAs and bronchoscopies decreased following the introduction of EBUS (p = 0.015 and p < 0.001 respectively). Of 162 CT-TTNAs, 59 (36%) resulted in complications compared to 1 complication each for bronchoscopy and EBUS-GS, and no complications from EBUS-TBNA. Fewer complications occurred overall in the Post-EBUS cohort compared to the Pre-EBUS cohort (p = 0.0264). The median time to management decision was 17 days (IQR 24) for the Pre-EBUS and 13 days (IQR 21) for the Post-EBUS cohort (p = 0.07). Within the Post-EBUS cohort, median time to management decision was longer for the EBUS group (n = 90) than the Non-EBUS group (17 days (IQR 29) vs. 10 days (IQR 10), p < 0.001). For half of EBUS-TBNA patients (n = 28, 50.0%) and EBUS-GS patients (n = 14, 50.0 %), EBUS alone provided sufficient diagnostic and/or staging information; these patients had median time to management decision of 10 days. Regression analysis revealed that the number of imaging events, inpatient, and outpatient visits were significant predictors of time to management decision of >28 days; EBUS was not a predictor of time to management decision., Conclusions: The introduction of EBUS led to fewer CT-TTNAs and bronchoscopies and did not impact on the time to management decision. EBUS-TBNA or EBUS-GS alone provided sufficient information for diagnosis and/or regional staging in half of the lung cancer patients referred for this investigation.

Background and Objective: Flexi-rigid pleuroscopy is a useful tool in the work-up of pleural effusions, but pleural biopsy using flexible forceps can be difficult in some patients. This study evaluated the feasibility, safety and diagnostic value of using a flexible cryoprobe to obtain parietal pleural biopsies during pleuroscopy., Methods: This was a single-centre retrospective study. In patients undergoing diagnostic pleuroscopy, pleural biopsy using flexible forceps, followed by a flexible cryoprobe introduced through the pleuroscope, were performed. A pathologist independently reviewed all biopsies. Any complications, particularly bleeding, were recorded. All patients were followed up for ≥ 6 months (median 12 months (range 7-26))., Results: Twenty-two patients (21 males; median age 72 years; 14 right-sided effusions) were included. All underwent flexible forceps biopsies (FFB) and cryoprobe biopsies (CB) of pleura. FFB and CB established a definitive diagnosis in 20/22 (90%). CB successfully obtained pleural tissue suitable for histopathological analysis in all patients. CB was larger than FFB (median, 25-75 IQR of 10, 7-15.8mm vs 4, 3-8mm), and had better preserved cellular architecture and tissue integrity. Crush artefacts were less common with CB (2/22) compared with FFB (21/22). No significant bleeding was reported., Conclusions: CB during flexi-rigid pleuroscopy is feasible, safe and effective.
