Playing a key role in immunity and autoimmunity, Major Histocompatibility Complex (MHC) contains microsatellite regions and polymorphisms associated with resistance to several diseases and thermo-physiologic characteristics in farm animals. This study aims to reveal genetic diversity in four native Turkish goat populations via MHC related gene regions including MHC-linked microsatellite markers (BF1, BM1818, BM1258, SMHCC1 and DYMS1) and MHC Class II-DRB gene. A total of 120 unrelated animals belonging to Hair (HAI), Honamlı (HNM), Kabakulak (KBK) and Norduz (NRD) from different representative populations in Antalya and Van provinces were sampled and genotyped for molecular analysis. Based on MHC-linked microsatellite markers, number of alleles ranged from 8.20 (NRD) to 8.80 (HAI and KBK) across studied goat populations. Observed heterozygosity was between 0.68 (NRD) and 0.80 (KBK), whereas expected heterozygosity ranged from 0.74 (NRD) to 0.82 (KBK) in native Turkish goats. Inbreeding coefficients were 0.04, 0.13, -0.01 and 0.09 for HAI, HNM, KBK and NRD populations, respectively. A 284 bp length PCR products belonging to MHC Class II-DRB gene region were digested separately with PstI, TaqI, BsaHI and AluI restriction endonucleases to assess polymorphism status together with Hardy-Weinberg equilibrium in studied goat populations. P allele frequency ranged from 0.73 (KBK) to 0.95 (NRD), while p allele frequency was between 0.05 (NRD) and 0.27 (KBK) in PstI polymorphism. The highest and lowest frequency of T allele were detected in HNM (0.80) and KBK (0.49), respectively, whereas frequency of t allele was between 0.20 (HNM) and 0.51 (KBK) in TaqI polymorphism. G and A allele frequency were between 0.16 (HNM) - 0.39 (HAI) and 0.84 (HNM) – 0.61 (HAI), respectively in BsaHI polymorphism, while three different genotypes and two alleles, which were different from results reported in the literature, were observed in AluI polymorphism. In this study, high genetic diversity and low inbreeding were detected in native Turkish goats according to MHC-linked microsatellite markers. Similarly, native Turkish goats hold enough polymorphisms in MHC Class II-DRB gene which gives opportunity to support selection strategies against tuberculosis and heat stress in the future.

Coat color is among the most distinctive phenotypes in cattle. Worldwide, several breeds share peculiar coat color features such as the presence of a fawn pigmentation of the calf at birth, turning over time to grey, and sexual dichromatism. The aim of this study was to search for polymorphisms under differential selection by contrasting grey cattle breeds displaying the above phenotype with non-grey cattle breeds, and to identify the underlying genes. Using medium-density SNP array genotype data, a multi-cohort FST-outlier approach was adopted for a total of 60 pair-wise comparisons of the 15 grey with 4 non-grey cattle breeds (Angus, Limousin, Charolais, and Holstein), with the latter selected as representative of solid and piebald phenotypes, respectively. Overall, more than 50 candidate genes were detected, almost all were either directly or indirectly involved in pigmentation, and some of them were already known for their role in phenotypes related with hair graying in mammals. Notably, 17 relevant genes, including SDR16C5, MOS, SDCBP, and NSMAF, were located in a signal on BTA14 convergently observed in all the four considered scenarios. Overall, the key stages of pigmentation (melanocyte development, melanogenesis, and pigment trafficking/transfer) were all represented among the pleiotropic functions of the candidate genes, suggesting the complex nature of the grey phenotype in cattle.

The aim of the present study was to detect a total of 11 polymorphisms of 6 genes (OCX32, GHR, DRD, VIP, VIPR, MR) related to egg yield and quality in six brown layer lines namely Rhode Island Red-I (RIRI), Rhode Island Red-II (RIRII), Barred Rock-I (BARI), Barred Rock-II (BARII), Colombian Rock (COL) and Line-54 (L5-4) raised in Ankara Poultry Research Institute. A total of 208 samples belonging to RIRI (n= 32), RIRII (n= 32), BARI (n= 40), BARII (n= 32), COL (n= 32) and L-54 (n= 40) were genotyped by PCR-RFLP method. For this reason, a total of 11 polymorphic regions were amplified by PCR and then PCR products were digested with specific restriction endonucleases. Results of the present study showed that polymorphisms and adequate genetic variations were found in OCX32-ex4/NcoI, OCX32-ex2/HpyCH4IV, DRD2/BseGI, VIPR-1/HhaI, VIPR-2/TaqI and MR1C/MboI genes which previously reported to associated with egg quality and yield in different chicken lines. However, conducting the association analysis between these genes and egg yield and quality will be useful. On the contrary, it has been revealed that GHR-intron 2/HindIII, GHR-intron 5/NspI, VIP-501/VspI, DRD1/BseNI and VIP-I2/HinfI polymorphisms cannot be used to increase egg yield and quality due to no detection of desired genotypes or homozygous excess., Bu çalışmanın amacı Ankara Tavukçuluk Araştırma Enstitüsünde yetiştirilen Rhode Island Red-I (RIRI), Rhode Island Red-II (RIRII), Barred Rock-I (BARI), Barred Rock-II (BARII), Colombian Rock (COL) ve Line-54 (L-54) isimli altı kahverengi saf yumurtacı tavuk hattında yumurta verim ve kalitesi ile ilişkili olan 6 gende (OCX32, GHR, DRD, VIP, VIPR, MR) toplam 11 polimorfizmin belirlenmesidir. Mevcut çalışmanın sonuçları daha önce farklı tavuk ırklarında yumurta verimi ve kalitesiyle ilişkili olduğu bildirilen OCX32-ex4/NcoI, OCX32-ex2/HpyCH4IV, DRD2/BseGI, VIPR-1/HhaI, VIPR-2/TaqI ve MR1C/MboI genlerinde polimorfizmin ve yeterli genetik varyasyonun bulunduğunu göstermiştir. Bununla birlikte yumurta verim ve kalite özellikleri ile bu genler arasında ilişki analizlerinin yapılması faydalı olacaktır. Aksine, GHR-intron 2/HindIII, GHR-intron 5/NspI, VIP-501/VspI, DRD1/BseNI ve VIP-I2/HinfI gen bölgelerinde istenilen genotiplerin tespit edilememesi ya da homozigot fazlalığından dolayı bu gen bölgelerinin kullanılamayacağı tespit edilmiştir.

Nerve Growth Factor Gene (NGF)has important functions in the maintenance of viability and proliferation ofovarian cells. Recent studies on caprine NGF gene revealed two SNPs(determined by SacII/C291A and MboII/A705Gpolymorphisms) related with body length and littersize in goats. This study aimed todetermine SacII and MboII polymorphisms of NGF in nativeTurkish goat populations including Turkish Hair (HAI), Kabakulak (KBK), Honamlı(HNM) and Norduz (NRD) by using PCR-RFLP. Amplificated PCR fragments of 808were digested with SacII,while amplificated PCR fragments of 319 bp were digested with MboII restriction enzyme in order todetect different genotypes of NGF gene. The “A” allele frequency rangedfrom 0.500 (HAI, HNM and KBK) to 0.950 (NRD), while the “C” allele frequencyranged from 0.025 (NRD) to 0.500 (HAI, HNM and KBK) in NGF/SacIIpolymorphism. The “A” allele frequency ranged from 0.213 (HAI) to 1.000(NRD), while the “G” allele frequency ranged from 0.000 (NRD) to 0.787 (HAI) inNGF/MboIIpolymorphism. Asetil koenzim A dehidrogenaz (ACADVL) uzun zincirli yağ asitlerinin oksidasyonunda ve enerji salınımında önemli rol oynayan bir proteindir. Sığırlarda 19. kromozom üzerinde bulunan ACADVL geni üzerindeki bir SNP’in (g.2885C>A; Pro236Thr) bazı büyüme özellikleri (göğüs genişliği, göğüs derinliği ve sağrı genişliği) ile ilişkili olduğu belirlenmiştir. Bu gen için AA genotipli bireyler AC ve CC genotiplilere göre daha üstün büyüme özelliklerine sahiptir. Bu çalışmada Türkiye’ de yetiştirilen Siyah Alaca (SA), Yerli Kara (YK), Boz (BI) ve Doğu Anadolu Kırmızısı (DAK) sığır ırklarında ACADVL (g.2885C>A) geni üzerinde bulunan polimorfizmin ARMS-PCR yöntemiyle belirlenmesi hedeflenmiştir. Çalışmada SA (64 örnek), YK (54 örnek), BI (48 örnek) ve DAK (44 örnek) sığır ırklarına ait toplam 210 örnek incelenmiştir. ARMS-PCR analizleri sonucunda ACADVL (g.2885C>A) geni için Siyah Alaca ırkı monomorfik (CC-307-152 bç) bulunurken BI, DAK ve YK sığır ırkları polimorfik (AC: 307-211-152 bç ve CC: 307-152 bç) bulunmuştur. AC genotipinin frekansı BI, DAK ve YK sığır ırklarında sırasıyla 0.063, 0.045, 0.093 olarak hesaplanırken, CC genotipinin frekansı sırasıyla 0.937, 0.955 ve 0.907 olarak hesaplanmıştır. Yapılan bu çalışma ile SA, BI, DAK ve YK sığır ırklarında ACADVL genindeki polimorfizmler ilk defa gösterilmiştir. Çalışılan sığır ırklarında AA genotipi tespit edilememiştir.

This study aimed to determine the effects of thermal manipulation during early embryogenesis (EE) and late embryogenesis (LE) on hatching weight, body weight at 5 weeks of age, hatchability and embryonic mortality rate in Japanese quails (Coturnix coturnix japonica). Incubation conditions from day 0 to day 17 were; 37.7 °C and 55 % relative humidity for control group. In the thermally treated eggs during early embryogenesis (EE6-EE8 days), incubation temperature was increased to 41 °C and relative humidity to 65 % for 3 hours (12.00-15.00) at 3 consecutive days. Also, in the late embryogenesis stage (LE12-LE14 days), incubation temperature was increased to 41 °C and relative humidity to 65 % 3 hours (12.00–15.00) at 3 consecutive days. At hatch in each trial, all chicks were wing-banded and individually weighted. Thermal manipulations had significant effect on hatching weight, and lowest hatching weights were found in late embryogenesis (LE) group in terms of male and female. In addition, thermal manipulations and gender had significant effect on body weight at 5 weeks of age and lowest body weights at 5 weeks of age were dedected in late embryogenesis (LE) group for both gender.

The aim of this study was to determine the effects of sex and slaughter ages on body weight and some carcass traits in Japanese quails (Coturnix coturnix japonica). The material used in this research was selected for 11 generations from high (HL) body weight Japanese quail line according to 5-week body weights. Age had a significant effect on body weight, carcass weight and carcass yield. Body weight was lower for the quails slaughtered at 5 weeks of age than for all the other ages examined (P< 0,01). Also, sex effect was significant on body weight and carcass weight being higher for females than for males at all ages, but males showed higher carcass yield than females (P< 0,01). Except wing percent, there was no significant difference between the sexes in terms of thigh percent, neck percent and back percent. Sex had also significant effect on liver and gizzard percent (P< 0,01). Furthermore, results showed that slaughter age affected percents of some internal organs and carcass parts of quails except thigh and wing percents, significantly., Bu çalışmanın amacı Japon bıldırcınlarında cinsiyet ve kesim yaşının (Coturnix coturnix japonica) canlı ağırlık ve bazı karkas özelliklerine etkilerini belirlemektir. Ayrıca cinsiyetin canlı ağırlık ve karkas ağırlığına etkisi önemli bulunmuş olup dişilerin erkeklerden daha yüksek canlı ağırlık ve karkas ağırlığına, buna karşılık erkeklerin ise daha yüksek karkas randımanına sahip oldukları saptanmıştır. (P< 0,01). Kanat yüzdesi dışında, but, boyun ve sırt yüzdesi bakımından cinsiyetler arasında önemli farklılıklar saptanmamıştır. Bunun yanında cinsiyetin ciğer ve taşlık yüzdelerine etkisi önemli bulunmuştur (P< 0,01). Sonuçlar, kesim yaşının but ve kanat yüzdeleri hariç, karkas parçaları ile iç organların yüzdelerini önemli derecede etkilediğini göstermiştir.

The aim of this research was to investigate the effects of selection and season on age at sexual maturity, clutch traits, and egg production in 11 generations selected Japanese quails (Coturnix coturnix japonica) of different lines. The material used in this research was selected for 11 generations from high (HL) or low (LL) body weight Japanese quail lines according to 5-week body weights and their random bred control line (C) and layer line (L) for 120 d egg production, which were bred in the Akdeniz University, Faculty of Agriculture in Turkey. First egg weight, age at sexual maturity, body weight at sexual maturity, total egg number, total egg weight, clutch length, clutch number, pause length and pause number were determined. Egg production was recorded daily throughout 12 weeks. The season had a significant effect on total egg number, age at sexual maturity, clutch size and pause size. Selection for increased or decreased 5-week body weights in Japanese quail resulted in a delay in age at sexual maturity. Also, selection for increased 5-week body weight increased body weight at sexual maturity. The season had no significant effect on first egg weight, total egg weight, body weight at sexual maturity, clutch number and pause number. Ayrıca, yüksek canlı ağırlık yönünde yapılan seleksiyon eşeysel olgunluk ağırlığını arttırmıştır. Mevsimin ilk yumurta ağırlığı, toplam yumurta ağırlığı, eşeysel olgunluk ağırlığı, klaç uzunluğu ile klaçlar arası süre sayısına etkisi önemsiz bulunmuştur. Major genes increase ovulation rate and litter size in some sheep breeds. Alleles of these major genes affecting ovulation rate in sheep are the result of a mutations. Primary mutations that increase ovulation rate in sheep were located on Bone Morphogenetic Protein Receptor IB/BMPR-IB (FecB), Bone Morphogenetic Protein 15/BMP-15 (FecXB, FecXG, FecXH, FecXL, FecXR, FecXI) and Growth Differentiation Factor 9/GDF-9 genes. In this study, the presence of FecB, FecXG and FecXH allele were investigated in Kangal and Güney Karaman sheeps reared in Turkey. In order to determine these allele in Kangal (n=42) and Güney Karaman (n=29) sheeps, a totall 71 samples were analyzed by using PCR-RFLP method. Bu araştırmada, Antalya yöresinde kış mevsiminde yetiştirilen Japon bıldırcınlarında genotipin ve canlı ağırlığın yumurta ağırlığına, yumurta sayısına, yem tüketimine ve yemden yararlanma oranına etkilerinin belirlenmesi amaçlanmıştır.
